    Sustainability and Maturation of School Turnaround: A Multiyear Evaluation of Tennessee’s Achievement School District and Local Innovation Zones

    Recent evaluations of reforms to improve low-performing schools have almost exclusively focused on shorter term effects. In this study, we extend the literature by examining the sustainability and maturation of two turnaround models in Tennessee: the state-led Achievement School District (ASD) and district-led local Innovation Zones (iZones). Using difference-in-differences models, we find overall positive effects on student achievement in iZone schools and null effects in ASD schools. Additional findings suggest a linkage between staff turnover and the effectiveness of reforms. ASD schools experienced high staff turnover in every cohort, and iZone schools faced high turnover in its latest cohort, the only one with negative effects. We discuss how differences in the ASD and iZone interventions may help explain variation in the schools’ ability to recruit and retain effective teachers and principals

    Unmanned aerial vehicle (UAV) derived structure-from-motion photogrammetry point clouds for oil palm (Elaeis guineensis) canopy segmentation and height estimation

The final version is available from Taylor & Francis via the DOI in this record.The vast size of oil palm (Elaeis guineensis) plantations has led to lightweight unmanned aerial vehicles (UAVs) being identified as cost effective tools to generate inventories for improved plantation management, with proximal aerial data capable of resolving single palm canopies at potentially, centimetric resolution. If acquired with sufficient overlap, aerial data from UAVs can be processed within structure-from-motion (SfM) photogrammetry workflows to yield volumetric point cloud representations of the scene. Point cloud-derived structural information on individual palms can benefit not only plantation management but is also of great environmental research interest, given the potential to deliver spatially contiguous quantifications of aboveground biomass, from which carbon can be accounted. Using lightweight UAVs we captured data over plantation plots of varying ages (2, 7 and 10 years) at peat soil sites in Sarawak, Malaysia, and we explored the impact of changing spatial resolution and image overlap on spatially variable uncertainties in SfM derived point clouds for the ten year old plot. Point cloud precisions were found to be in the decimetre range (mean of 26.7 31 cm) for a 10 year old plantation plot surveyed at 100 m flight altitude and >75% image overlap. Derived canopy height models were used and evaluated for automated palm identification using local height maxima. Metrics such as maximum canopy height and stem height, derived from segmented single palm point clouds were tested relative to ground validation data. Local maximum identification performed best for palms which were taller than surrounding undergrowth but whose fronds did not overlap significantly (98.2% mapping accuracy for 7 year old plot of 776 palms). Stem heights could be predicted from point cloud derived metrics with root-mean-square errors (RMSEs) of 0.27 m (R2= 0.63) for 7 year old and 0.45 m (R2=0.69) for 10 year old palms. It was also found that an acquisition designed to yield the minimal required overlap between images (60%) performed almost as well as higher overlap acquisitions (>75%) for palm identification and basic height metrics which is promising for operational implementations seeking to maximise spatial coverage and minimise processing costs. We conclude that UAV-based SfM can provide reliable data not only for oil palm inventory generation but allows the retrieval of basic structural parameters which may enable per-palm above-ground biomass estimations.European CommissionMarie Skłodowska-Curi

    Epigenetic inactivation of mir-34b/c in addition to mir-34a and DAPK1 in chronic lymphocytic leukemia

    BACKGROUND: TP53 mutation/deletion is uncommon in chronic lymphocytic leukemia (CLL). We postulated that components of TP53-centered tumor suppressor network, miR-34b/c, in addition to DAPK1 and miR-34a might be inactivated by DNA hypermethylation. Moreover, we tested if miR-34b/c methylation might correlate with miR-203 or miR-124-1 methylation in CLL. METHODS: miR-34b/c, miR-34a and DAPK1 methylation was studied in 11 normal controls, 7 CLL cell lines, and 78 diagnostic CLL samples by methylation-specific polymerase chain reaction. MEC-1 cells were treated with 5-Aza-2'-deoxycytidine for reversal of methylation-associated miRNA silencing. Tumor suppressor properties of miR-34b were demonstrated by over-expression of precursor miR-34b in MEC-1 cells. RESULTS: miR-34b/c promoter was unmethylated in normal controls, but completely methylated in 4 CLL cell lines. miR-34b/c expression was inversely correlated with miR-34b/c methylation. Different MSP statuses of miR-34b/c, including complete methylation and complete unmethylation, were verified by quantitative bisulfite pyrosequencing. 5-Aza-2'-deoxycytidine treatment resulted in promoter demethylation and miR-34b re-expression in MEC1 cells. Moreover, over-expression of miR-34b resulted in inhibition of cellular proliferation and increased cell death. In primary CLL samples, miR-34a, miR-34b/c and DAPK1 methylation was detected in 2.6%, 17.9% and 34.6% of patients at diagnosis respectively. Furthermore, 39.7%, 3.8% and 2.6% patients had methylation of one, two or all three genes respectively. Overall, 46.2% patients had methylation of at least one of these three genes. Besides, miR-34b/c methylation was associated with methylation of miR-34a (P = 0.03) and miR-203 (P = 0.012) in CLL. CONCLUSIONS: Taken together, miR-34b/c is a tumor suppressor miRNA frequently methylated, and hence silenced in CLL. Together with DAPK1 methylation, miR-34b/c methylation is implicated in the disruption of the TP53-centered tumor suppressor network.     Quantifying tropical peatland dissolved organic carbon (DOC) using UV-visible spectroscopy

    UV–visible spectroscopy has been shown to be a useful technique for determining dissolved organic carbon (DOC) concentrations. However, at present we are unaware of any studies in the literature that have investigated the suitability of this approach for tropical DOC water samples from any tropical peatlands, although some work has been performed in other tropical environments. We used water samples from two oil palm estates in Sarawak, Malaysia to: i) investigate the suitability of both single and two-wavelength proxies for tropical DOC determination; ii) develop a calibration dataset and set of parameters to calculate DOC concentrations indirectly; iii) provide tropical researchers with guidance on the best spectrophotometric approaches to use in future analyses of DOC. Both single and two-wavelength model approaches performed well with no one model significantly outperforming the other. The predictive ability of the models suggests that UV–visible spectroscopy is both a viable and low cost method for rapidly analyzing DOC in water samples immediately post-collection, which can be important when working at remote field sites with access to only basic laboratory facilities

    Mapping peat soil moisture under oil palm plantation and tropical forest in Sarawak

    Water table conditions in drained peatlands affect peat decomposition, fluvial carbon and greenhouse gas emissions, and plant growth in oil palm plantations. This study illustrates the spatial heterogeneity of soil moisture profiles in cultivated tropical peat under oil palm plantation and uncultivated secondary forest, using maps. At a study plot under each land use the geographical coordinates of sampling points, tree locations and other features were recorded. Peat soil samples were taken at depths of 0–50 cm, 50–100 cm, 100–150 cm and 150–200 cm, and their moisture contents were determined. Overall, soil moisture content was higher in secondary forest than in oil palm plantation due to land management activities such as drainage and peat compaction in the latter. Significant differences were observed between the topsoil (0–50 cm) and deeper soil layers under both land uses. Soil moisture maps of the study plots interpolated using geographical information system (GIS) software were used to visualise the spatial distributions of moisture content in soil layers at different depths (0–50 cm, 50–100 cm, 100–150 cm, 150–200 cm). Moisture content in the 0–50 cm soil layer appeared to be inversely related to elevation, but the correlation was not statistically significant. On the other hand, there was a significant positive correlation between soil moisture content and the diameters of oil palm trunks. Palm trees with negative growth of trunk diameter were mostly located in subplots which were relatively dry and/or located near drains. The results of this study indicate that soil moisture mapping using GIS could be a useful tool in improving the management of peatland to promote oil palm growth.Peer reviewe
