    We reconsider the Affleck-Dine mechanism for baryogenesis and show that the baryonic isocurvature fluctuations are generated in many inflation models in supergravity.     Haploinsufficiency of A20 impairs protein–protein interactome and leads into caspase-8-dependent enhancement of NLRP3 inflammasome activation

    Objectives TNFAIP3 encodes A20 that negatively regulates nuclear factor kappa light chain enhancer of activated B cells (NF-ÎșB), the major transcription factor coordinating inflammatory gene expression. TNFAIP3 polymorphisms have been linked with a spectrum of inflammatory and autoimmune diseases and, recently, loss-of-function mutations in A20 were found to cause a novel inflammatory disease ‘haploinsufficiency of A20’ (HA20). Here we describe a family with HA20 caused by a novel TNFAIP3 loss-of-function mutation and elucidate the upstream molecular mechanisms linking HA20 to dysregulation of NF-ÎșB and the related inflammasome pathway.Methods NF-ÎșB activation was studied in a mutation-expressing cell line using luciferase reporter assay. Physical and close-proximity protein–protein interactions of wild-type and TNFAIP3 p.(Lys91*) mutant A20 were analysed using mass spectrometry. NF-ÎșB -dependent transcription, cytokine secretion and inflammasome activation were compared in immune cells of the HA20 patients and control subjects.Results The protein–protein interactome of p.(Lys91*) mutant A20 was severely impaired, including interactions with proteins regulating NF-ÎșB activation, DNA repair responses and the NLR family pyrin domain containing 3 (NLRP3) inflammasome. The p.(Lys91*) mutant A20 failed to suppress NF-ÎșB signalling, which led to increased NF-ÎșB -dependent proinflammatory cytokine transcription. Functional experiments in the HA20 patients’ immune cells uncovered a novel caspase-8-dependent mechanism of NLRP3 inflammasome hyperresponsiveness that mediated the excessive secretion of interleukin-1ÎČ and interleukin-18.Conclusions The current findings significantly deepen our understanding of the molecular mechanisms underlying HA20 and other diseases associated with reduced A20 expression or function, paving the way for future therapeutic targeting of the pathway.Peer reviewe

    Non-Gaussianity from isocurvature perturbations

    We develop a formalism to study non-Gaussianity in both curvature and isocurvature perturbations. It is shown that non-Gaussianity in the isocurvature perturbation between dark matter and photons leaves distinct signatures in the CMB temperature fluctuations, which may be confirmed in future experiments, or possibly, even in the currently available observational data. As an explicit example, we consider the QCD axion and show that it can actually induce sizable non-Gaussianity for the inflationary scale, H_{inf} = O(10^9 - 10^{11})GeV.Comment: 24 pages, 6 figures; references added; version to appear in JCA

    Seasonal particulate organic carbon dynamics of the Kolyma River tributaries, Siberia

    Arctic warming is causing permafrost thaw and release of organic carbon (OC) to fluvial systems. Permafrost-derived OC can be transported downstream and degraded into greenhouse gases that may enhance climate warming. Susceptibility of OC to decomposition depends largely upon its source and composition, which vary throughout the seasonally distinct hydrograph. Most studies on carbon dynamics to date have focused on larger Arctic rivers, yet little is known about carbon cycling in lower-order rivers and streams. Here, we characterize the composition and sources of OC, focusing on less studied particulate OC (POC), in smaller waterways within the Kolyma River watershed. Additionally, we examine how watershed characteristics control carbon concentrations. In lower-order systems, we find rapid initiation of primary production in response to warm water temperatures during spring freshet, shown by decreasing ÎŽ13C-POC, in contrast to larger rivers. This results in CO2 uptake by primary producers and microbial degradation of mainly autochthonous OC. However, if terrestrially derived inorganic carbon is assimilated by primary producers, part of it is returned via CO2 emissions if the autochthonous OC pool is simultaneously degraded. As Arctic warming and hydrologic changes may increase OC transfer from smaller waterways to larger river networks, understanding carbon dynamics in smaller waterways is crucial.</p

    Constraints on generating the primordial curvature perturbation and non-Gaussianity from instant preheating

    We analyse models of inflation in which isocurvature perturbations present during inflation are converted into the primordial curvature perturbation during instant preheating. This can be due to an asymmetry between the fields present either during inflation or during preheating. We consider all the constraints that the model must satisfy in order to be theoretically valid and to satisfy observations. We show that the constraints are very tight in all of the models proposed and special initial conditions are required for the models to work.     The effects of smoking on obesity: Evidence from Indonesian panel data

    Background: It has been known that smoking is negatively related to weight-related outcomes. However, it has been difficult to determine whether the relationship is causal, and if so, how strong it is. We attempted to estimate the approximately causal effects of smoking on weight, body mass index (BMI), and obesity. Methods: The Indonesian Family Life Survey provided a sample of over 9000 men aged 15-55 years - each of them was observed in 1993, 1997, 2000 and 2007. The preferred method was a fixed effects model; that is, we related changes in smoking status or smoking intensity to changes in weight-related outcomes, while controlling for time-varying covariates. We also compared these results to those estimated by ordinary least squares and assessed the importance of controlling for time invariant individual heterogeneity. Results: Although the effects of smoking were precisely estimated in a statistical sense, their size was minuscule: a quitter would gain weight by at most 1 kg, or a smoker would lose weight by the same amount. The results were similar for BMI and obesity. When we did not control for time invariant individual heterogeneity, the size of the relationship was overestimated at least three times. Conclusions: Smoking exerted little influence on weight, and it was important to control for bias stemming from time invariant individual heterogeneity

    Heterozygous premature termination in zinc-finger domain of Kruppel-like factor 2 gene associates with dysregulated immunity

    Kruppel-like factor 2 (KLF2) is a transcription factor with significant roles in development, maturation, differentiation, and proliferation of several cell types. In immune cells, KLF2 regulates maturation and trafficking of lymphocytes and monocytes. KLF2 participates in regulation of the nuclear factor kappa-light-chain-enhancer of activated B cells (NF-kappa B) pathway. Although pulmonary arterial hypertension (PAH) related to KLF2 genetic variant has been suggested, genetic role of KLF2 associated with immune dysregulation has not been described. We identified a family whose members suffered from lymphopenia, autoimmunity, and malignancy. Whole exome sequencing revealed a KLF2 p.(Glu318Argfs*87) mutation disrupting the highly conserved zinc finger domain. We show a reduced amount of KLF2 protein, defective nuclear localization and altered protein-protein interactome. The phenotypically variable positive cases presented with B and T cell lymphopenia and abnormalities in B and T cell maturation including low naive T cell counts and low CD27(+)IgD(-)IgM(-) switched memory B cells. KLF2 target gene (CD62L) expression was affected. Although the percentage of (CD25(+)FOXP3(+), CD25(+)CD127(-)) regulatory T cells (Treg) was high, the naive Treg cells (CD45RA(+)) were absent. Serum IgG1 levels were low and findings in one case were consistent with common variable immunodeficiency (CVID). Transcription of NF-kappa beta pathway genes and p65/RelA phosphorylation were not significantly affected. Inflammasome activity, transcription of genes related with JAK/STAT pathway and interferon signature were also comparable to controls. Evidence of PAH was not found. In conclusion, KLF2 variant may be associated with familial immune dysregulation. Although the KLF2 deficient family members in our study suffered from lymphopenia, autoimmunity or malignancy, additional study cohorts are required to confirm our observations.Peer reviewe

    Development and characterization of the readout system for POLARBEAR-2

    POLARBEAR-2 is a next-generation receiver for precision measurements of the polarization of the cosmic microwave background (Cosmic Microwave Background (CMB)). Scheduled to deploy in early 2015, it will observe alongside the existing POLARBEAR-1 receiver, on a new telescope in the Simons Array on Cerro Toco in the Atacama desert of Chile. For increased sensitivity, it will feature a larger area focal plane, with a total of 7,588 polarization sensitive antenna-coupled Transition Edge Sensor (TES) bolometers, with a design sensitivity of 4.1 uKrt(s). The focal plane will be cooled to 250 milliKelvin, and the bolometers will be read-out with 40x frequency domain multiplexing, with 36 optical bolometers on a single SQUID amplifier, along with 2 dark bolometers and 2 calibration resistors. To increase the multiplexing factor from 8x for POLARBEAR-1 to 40x for POLARBEAR-2 requires additional bandwidth for SQUID readout and well-defined frequency channel spacing. Extending to these higher frequencies requires new components and design for the LC filters which define channel spacing. The LC filters are cold resonant circuits with an inductor and capacitor in series with each bolometer, and stray inductance in the wiring and equivalent series resistance from the capacitors can affect bolometer operation. We present results from characterizing these new readout components. Integration of the readout system is being done first on a small scale, to ensure that the readout system does not affect bolometer sensitivity or stability, and to validate the overall system before expansion into the full receiver.
