    Spectroscopy of a Cooper-Pair box in the Autler-Townes configuration

    A theoretical spectroscopic analysis of a microwave driven superconducting charge qubit (Cooper-pair box coupled) to an RLC oscillator model is performed. By treating the oscillator as a probe through the backreaction effect of the qubit on the oscillator circuit, we extract frequency splitting features analogous to the Autler-Townes effect from quantum optics, thereby extending the analogies between superconducting and quantum optical phenomenology. These features are found in a frequency band that avoids the need for high frequency measurement systems and therefore may be of use in qubit characterization and coupling schemes. In addition we find this frequency band can be adjusted to suit an experimental frequency regime by changing the oscillator frequency.Comment: 13 pages, 7 figures. v2: Revised version after referee comments.     Experimental Verification of 3D Plasmonic Cloaking in Free-Space

    We report the experimental verification of metamaterial cloaking for a 3D object in free space. We apply the plasmonic cloaking technique, based on scattering cancellation, to suppress microwave scattering from a finite-length dielectric cylinder. We verify that scattering suppression is obtained all around the object in the near- and far-field and for different incidence angles, validating our measurements with analytical results and full-wave simulations. Our near-field and far-field measurements confirm that realistic and robust plasmonic metamaterial cloaks may be realized for elongated 3D objects with moderate transverse cross-section at microwave frequencies.Comment: 12 pages, 8 figures, published in NJ

    HIV-Associated Mycobacterium tuberculosis Bloodstream Infection Is Underdiagnosed by Single Blood Culture

    ABSTRACT We assessed the additional diagnostic yield for Mycobacterium tuberculosis bloodstream infection (BSI) by doing more than one tuberculosis (TB) blood culture from HIV-infected inpatients. In a retrospective analysis of two cohorts based in Cape Town, South Africa, 72/99 (73%) patients with M. tuberculosis BSI were identified by the first of two blood cultures during the same admission, with 27/99 (27%; 95% confidence interval [CI], 18 to 36%) testing negative on the first culture but positive on the second. In a prospective evaluation of up to 6 blood cultures over 24 h, 9 of 14 (65%) patients with M. tuberculosis BSI had M. tuberculosis grow on their first blood culture; 3 more patients (21%) were identified by a second independent blood culture at the same time point, and the remaining 2 were diagnosed only on the 4th and 6th blood cultures. Additional blood cultures increase the yield for M. tuberculosis BSI, similar to what is reported for nonmycobacterial BSI. </jats:p

    Investigation and Assessment of Resource Consumption of Process Chains

    AbstractMany different technologies and processes have been established in production within the last decades. These technologies have to be integrated into sophisticated process chains to achieve today's requirements of high performance products. For most of these products the costs can be determined or at least estimated accurately. However, resource intensive and thus cost intensive processes and their potential within the process chains are often neither identified nor quantified. For identifying, measuring and subsequently assessing the need of resources, like energy or material and their monetary as well as environmental impact, four different process chains of high industrial relevance have been chosen and investigated with regards to their resource consumption. These process chains are used for manufacturing turbine blades made of Inconel and titanium aluminide as well as for comparisons of a conventional and an innovative process chain to manufacture an insert for an injection mold. By measuring and assessing their resource consumption the most resource intensive and thus influential processes have been identified and their potential for resource reduction has been evaluated. Due to the change of single processes to reduce resource consumption and thus the conditions for subsequent processes, the requirements might change and lead to adaptions within the entire process chain. For the assessment of the process chains and the changes within the processes themselves, a scenario based assessment has been modelled. This results in an economic and ecologic evaluation of these process chains and enables a comparison of these to choose the most meaningful process chain

    Wir zeigen, dass negativ geladene Polysulfate durch elektrostatische Wechselwirkungen an das Spike-Protein von SARS-CoV-2 binden. Durch einen Plaquereduktionstest verglichen wir die hemmende Wirkung von Heparin, Pentosanpolysulfat, linearem Polyglycerolsulfat (LPGS) und hyperverzweigtem Polyglycerolsulfat (HPGS) gegengber SARSCoV-2. Dabei ist das synthetische LPGS der vielversprechendste Inhibitor mit IC50=67 ÎŒgmL-1 (ca. 1,6 ÎŒm) und zeigt eine 60-fach hçhere virushemmende AktivitĂ€t als Heparin (IC50=4084 ÎŒgmL-1) bei zugleich deutlich geringerer gerinnungshemmender AktivitĂ€t. Außerdem konnten wir durch MolekĂŒldynamiksimulationen bestĂ€tigen, dass LPGS stĂ€rker an das Spike-Protein bindet als Heparin selbst und dass LPGS sogar noch stĂ€rker an die Spike-Proteine der neuen N501Yund E484K-Varianten bindet. Unsere Studien belegen, dass die Aufnahme von SARS-CoV-2 in Wirtzellen ĂŒber elektrostatische Wechselwirkungen blockiert werden kann. Deshalb kann LPGS als vielversprechender Prototyp fĂŒr das Design weiterer neuartiger viraler Inhibitoren von SARS-CoV-2 herangezogen werden

    Optimising integrated stroke care in regional networks:A nationwide self-assessment study in 2012, 2015 and 2019

    Background: To help enhance the quality of integrated stroke care delivery, regional stroke services networks in the Netherlands participated in a self-assessment study in 2012, 2015 and 2019. Methods: Coordinators of the regional stroke services networks filled out an online self-assessment questionnaire in 2012, 2015 and 2019. The questionnaire, which was based on the Development Model for Integrated Care, consisted of 97 questions in nine clusters (themes). Cluster scores were calculated as proportions of the activities implemented. Associations between clusters and features of stroke services were assessed by regression analysis. Results: The response rate varied from 93.1% (2012) to 85.5% (2019). Over the years, the regional stroke services networks increased in ‘size’: the median number of organisations involved and the volume of patients per network increased (7 and 499 in 2019, compared to 5 and 364 in 2012). At the same time, fewer coordinators were appointed for more than 1 day a week in 2019 (35.1%) compared to 2012 (45.9%). Between 2012 and 2019, there were statistically significantly more elements implemented in four out of nine clusters: ‘Transparent entrepreneurship’ (MD = 18.0% F(1) = 10.693, p = 0.001), ‘Roles and tasks’ (MD = 14.0% F(1) = 9.255, p = 0.003), ‘Patient-centeredness’ (MD = 12.9% F(1) = 9.255, p = 0.003), and ‘Commitment’ (MD = 11.2%, F(1) = 4.982, p = 0.028). A statistically significant positive correlation was found for all clusters between implementation of activities and age of the network. In addition, the number of involved organisations is associated with better execution of implemented activities for ‘Transparent entrepreneurship’, ‘Result-focused learning’ and ‘Quality of care’. Conversely, there are small but negative associations between the volume of patients and implementation rates for ‘Interprofessional teamwork’ and ‘Patient-centredness’. Conclusion: This long-term analyses of stroke service development in the Netherlands, showed that between 2012 and 2019, integrated care activities within the regional stroke networks increased. Experience in collaboration between organisations within a network benefits the uptake of integrated care activities

    Polysulfates block SARS-CoV-2 uptake through electrostatic interactions

    Here we report that negatively charged polysulfates can bind to the spike protein of SARS-CoV-2 via electrostatic interactions. Using a plaque reduction assay, we compare inhibition of SARS-CoV-2 by heparin, pentosan sulfate, linear polyglycerol sulfate (LPGS) and hyperbranched polyglycerol sulfate (HPGS). Highly sulfated LPGS is the optimal inhibitor, with a half-maximal inhibitory concentration (IC50) of 67 ÎŒg/mL (approx.1.6 ÎŒM). This synthetic polysulfates exhibit more than 60-fold higher virus inhibitory activity than heparin (IC50: 4084ÎŒg/mL), along with much lower anticoagulant activity. Furthermore, in molecular dynamics simulations, we verified that LPGS can bind stronger to the spike protein than heparin, and that LPGS can interact even morewith the spike protein of the new N501Y and E484K variants. Our study demonstrates that the entry of SARS-CoV-2 into host cells can be blocked via electrostatic interaction, therefore LPGS can serve as a blueprint for the design of novel viral inhibitors of SARS-CoV-2

    Surveying the Dynamic Radio Sky with the Long Wavelength Demonstrator Array

    This paper presents a search for radio transients at a frequency of 73.8 MHz (4 m wavelength) using the all-sky imaging capabilities of the Long Wavelength Demonstrator Array (LWDA). The LWDA was a 16-dipole phased array telescope, located on the site of the Very Large Array in New Mexico. The field of view of the individual dipoles was essentially the entire sky, and the number of dipoles was sufficiently small that a simple software correlator could be used to make all-sky images. From 2006 October to 2007 February, we conducted an all-sky transient search program, acquiring a total of 106 hr of data; the time sampling varied, being 5 minutes at the start of the program and improving to 2 minutes by the end of the program. We were able to detect solar flares, and in a special-purpose mode, radio reflections from ionized meteor trails during the 2006 Leonid meteor shower. We detected no transients originating outside of the solar system above a flux density limit of 500 Jy, equivalent to a limit of no more than about 10^{-2} events/yr/deg^2, having a pulse energy density >~ 1.5 x 10^{-20} J/m^2/Hz at 73.8 MHz for pulse widths of about 300 s. This event rate is comparable to that determined from previous all-sky transient searches, but at a lower frequency than most previous all-sky searches. We believe that the LWDA illustrates how an all-sky imaging mode could be a useful operational model for low-frequency instruments such as the Low Frequency Array, the Long Wavelength Array station, the low-frequency component of the Square Kilometre Array, and potentially the Lunar Radio Array.Comment: 20 pages; accepted for publication in A

    Use of mitogenic cascade blockers for treatment of C-Raf induced lung adenoma in vivo: CI-1040 strongly reduces growth and improves lung structure

    BACKGROUND: Signaling networks promoting cell growth and proliferation are frequently deregulated in cancer. Tumors often are highly dependent on such signaling pathways and may become hypersensitive to downregulation of key components within these signaling cascades. The classical mitogenic cascade transmits stimuli from growth factor receptors via Ras, Raf, MEK and ERK to the cell nucleus and provides attractive molecular targets for cancer treatment. For example, Ras and Raf kinase inhibitors are already in a number of ongoing phase II and phase III clinical trials. In this study the effect of the Raf kinase inhibitor BAY 43-9006 and of the MEK inhibitor CI-1040 (PD184352) on a Raf dependent lung tumor mouse model was analyzed in detail. METHODS: We have generated a lung cancer mouse model by targeting constitutively active C-Raf kinase to the lung. These mice develop adenomas within 4 months of life. At this time-point they received daily intraperitoneal injections of either 100 mg/kg BAY 43-9006 or CI-1040 for additional 21 days. Thereafter, lungs were isolated and the following parameters were analyzed using histology and immunohistochemistry: overall lung structure, frequency of adenoma foci, proliferation rate, ERK activity, caspase-3 activation, and lung differentiation. RESULTS: Both inhibitors were equally effective in vitro using a sensitive Raf/MEK/ERK ELISA. In vivo, the systemic administration of the MEK inhibitor CI-1040 reduced adenoma formation to a third and significantly restored lung structure. The proliferation rate of lung cells of mice treated with CL-1040 was decreased without any obvious effects on differentiation of pneumocytes. In contrast, the Raf inhibitor BAY 43-9006 did not influence adenoma formation in vivo. CONCLUSION: The MEK inhibitor CI-1040 may be used for the treatment of Ras and/or Raf-dependent human malignancies

    Recovery of visual fields in brain-lesioned patients by reaction perimetry treatment

    <p>Abstract</p> <p>Background</p> <p>The efficacy of treatment in hemianopic patients to restore missing vision is controversial. So far, successful techniques require laborious stimulus presentation or restrict improvements to selected visual field areas. Due to the large number of brain-damaged patients suffering from visual field defects, there is a need for an efficient automated treatment of the total visual field.</p> <p>Methods</p> <p>A customized treatment was developed for the reaction perimeter, permitting a time-saving adaptive-stimulus presentation under conditions of maximum attention. Twenty hemianopic patients, without visual neglect, were treated twice weekly for an average of 8.2 months starting 24.2 months after the insult. Each treatment session averaged 45 min in duration.</p> <p>Results</p> <p>In 17 out of 20 patients a significant and stable increase of the visual field size (average 11.3° ± 8.1) was observed as well as improvement of the detection rate in the defective visual field (average 18.6% ± 13.5). A two-factor cluster analysis demonstrated that binocular treatment was in general more effective in augmenting the visual detection rate than monocular. Four out of five patients with a visual field increase larger than 10° suffered from hemorrhage, whereas all seven patients with an increase of 5° or less suffered from infarction. Most patients reported that visual field restoration correlated with improvement of visual-related activities of daily living.</p> <p>Conclusion</p> <p>Rehabilitation treatment with the Lubeck Reaction Perimeter is a new and efficient method to restore part of the visual field in hemianopia. Since successful transfer of treatment effects to the occluded eye is achieved under monocular treatment conditions, it is hypothesized that the damaged visual cortex itself is the structure in which recovery takes place.</p
