    Intermolecular interactions in N-(ferrocenylmethyl)anthracene-9-carboxamide

    The title compound, [Fe(C₅H₅)(C₂₁H₁₆NO)], was synthesized from the coupling reaction of anthracene-9-carboxylic acid and ferrocenylmethylamine. The ferrocenyl (Fc) group and the anthracene ring system both lie approximately orthogonal to the amide moiety. An amide-amide interaction (along the a axis) is the principal interaction [N...O = 2.910 (2) Å]. A C-H...π(arene) interaction [C...centroid = 3.573 (2) Å] and a C-H...O interaction [C...O = 3.275 (3) Å] complete the hydrogen bonding; two short (Fc)C...C(anthracene) contacts are also present

    ACOLHIMENTO: UM OLHAR INCLUSIVO DA POLÍTICA NACIONAL DE HUMANIZAÇÃO COMO ESTRATÉGIA DE INCLUSÃO SOCIAL

    A inclusão social é um assunto de discussão predominantemente das Ciências Humanas e Socais Aplicadas. Entretanto, tem ganhado espaço na Área da Saúde. Ela é uma prática indissociável da atenção à saúde e da prática terapêutica, sendo aplicada a partir da percepção dos fenômenos que compõem o fazer em saúde. Nesse contexto, o presente artigo se propôs a fazer uma reflexão acadêmica junto a estudantes de graduação da área da saúde, sobre a temática da inclusão social, a partir das estratégias inclusivas do Sistema Único de Saúde– SUS, utilizando-se das diretrizes da Política Nacional de Humanização– PNH como referencial teórico. O objetivo do presente trabalho foi discutir a prática da inclusão social no contexto da PNH, a partir do acolhimento, como estratégia de inclusão social. A metodologia utilizada para a realização da pesquisa foi revisão bibliográfica, a partir dos documentos oficiais do Ministério da Saúde que abordam a temática da humanização em saúde. Foram apresentados e discutidos os conceitos de humanização e sua aplicabilidade à saúde. Discutiram-se ainda os conceitos de acolhimento, como diretriz norteadora da PNH. Identificou-se que o acolhimento pode ser considerado uma estratégia de inclusão social, como também de exclusão e que, para se praticar a humanização na saúde, é necessário envolver não só usuários da saúde púbica como também gestores e trabalhadores. Entretanto, para que haja mudança nos modelos de gestão e cuidado, inscritos na humanização, é necessário mudar conceitos e comportamentos dos futuros profissionais e gestores da saúde. Nesse contexto, urge uma reflexão sobre a formação profissional de estudantes de graduação da área da saúde no âmbito da inclusão social.Palavras-chave: acolhimento; humanização; inclusão social

    Identification of a Genomic Region Between SLC29A1 and HSP90AB1 Associated With Risk of Bevacizumab-Induced Hypertension: CALGB 80405 (Alliance)

    Purpose: Bevacizumab is a VEGF-specific angiogenesis inhibitor indicated as an adjunct to chemotherapy for the treatment of multiple cancers. Hypertension is commonly observed during bevacizumab treatment, and high-grade toxicity can limit therapy or lead to cardiovascular complications. The factors that contribute to interindividual variability in blood pressure rise during bevacizumab treatment are not well understood.Experimental Design: To identify genomic regions associated with bevacizumab-induced hypertension risk, sequencing of candidate genes and flanking regulatory regions was performed on 61 patients treated with bevacizumab (19 cases developed early-onset grade 3 hypertension and 42 controls had no reported hypertension in the first six cycles of treatment). SNP-based tests for common variant associations and gene-based tests for rare variant associations were performed in 174 candidate genes.Results: Four common variants in independent linkage disequilibrium blocks between SLC29A1 and HSP90AB1 were among the top associations. Validation in larger bevacizumab-treated cohorts supported association between rs9381299 with early grade 3+ hypertension (P = 0.01; OR, 2.4) and systolic blood pressure >180 mm Hg (P = 0.02; OR, 2.1). rs834576 was associated with early grade 3+ hypertension in CALGB 40502 (P = 0.03; OR, 2.9). These SNP regions are enriched for regulatory elements that may potentially increase gene expression. In vitro overexpression of SLC29A1 in human endothelial cells disrupted adenosine signaling and reduced nitric oxide levels that were further lowered upon bevacizumab exposure.Conclusions: The genomic region between SLC29A1 and HSP90AB1 and its role in regulating adenosine signaling are key targets for further investigation into the pathogenesis of bevacizumab-induced hypertension

    New mutations at the imprinted Gnas cluster show gene dosage effects of Gsα in postnatal growth and implicate XLαs in bone and fat metabolism, but not in suckling

    The imprinted Gnas cluster is involved in obesity, energy metabolism, feeding behavior, and viability. Relative contribution of paternally expressed proteins XLαs, XLN1, and ALEX or a double dose of maternally expressed Gsα to phenotype has not been established. In this study, we have generated two new mutants (Ex1A-T-CON and Ex1A-T) at the Gnas cluster. Paternal inheritance of Ex1A-T-CON leads to loss of imprinting of Gsα, resulting in preweaning growth retardation followed by catch-up growth. Paternal inheritance of Ex1A-T leads to loss of imprinting of Gsα and loss of expression of XLαs and XLN1. These mice have severe preweaning growth retardation and incomplete catch-up growth. They are fully viable probably because suckling is unimpaired, unlike mutants in which the expression of all the known paternally expressed Gnasxl proteins (XLαs, XLN1 and ALEX) is compromised. We suggest that loss of ALEX is most likely responsible for the suckling defects previously observed. In adults, paternal inheritance of Ex1A-T results in an increased metabolic rate and reductions in fat mass, leptin, and bone mineral density attributable to loss of XLαs. This is, to our knowledge, the first report describing a role for XLαs in bone metabolism. We propose that XLαs is involved in the regulation of bone and adipocyte metabolism

    Antimicrobial evaluation of quinones and heterocyclic compounds against mycobacterium marinum, M. kansasii and M. abscessus

    The resistance to antimicrobials and biocides observed in mycobacteria which do not cause tuberculosis (MNT) determines the necessity to develop drugs. The present study evaluated the activity of naphthoquinones and heterocyclic derivatives obtained from lapachol against Mycobacterium kansasii, M. marinum, and M. abscessus, through the REMA method. It was observed that lapachol was inactive against the three mycobateria species, while β-lapachone and nor-β-lapachone showed activity only against M. marinum. The most active substances for M. kansasii were the derivates 2, 3, 7, and 11, in which compound 2 (CMI = 0.96 μM) was the most active. For M. marinum, 2, 11, and 14 were the most active, while against M. abcessus the compound 3 was the only active. The results showed a wide and diversified resistance spectrum among the species studied, which could be related to the molecular structure and position of the substituting groups, indicating the potentiality of these molecules as antimicrobial prototypes.Colegio de Farmacéuticos de la Provincia de Buenos Aire

    Intermediate-affinity LFA-1 binds α-actinin-1 to control migration at the leading edge of the T cell

    T lymphocytes use LFA-1 to migrate into lymph nodes and inflammatory sites. To investigate the mechanisms regulating this migration, we utilize mAbs selective for conformational epitopes as probes for active LFA-1. Expression of the KIM127 epitope, but not the 24 epitope, defines the extended conformation of LFA-1, which has intermediate affinity for ligand ICAM-1. A key finding is that KIM127-positive LFA-1 forms new adhesions at the T lymphocyte leading edge. This LFA-1 links to the cytoskeleton through α-actinin-1 and disruption at the level of integrin or actin results in loss of cell spreading and migratory speed due to a failure of attachment at the leading edge. The KIM127 pattern contrasts with high-affinity LFA-1 that expresses both 24 and KIM127 epitopes, is restricted to the mid-cell focal zone and controls ICAM-1 attachment.     Background Findings from the RESTART trial suggest that starting antiplatelet therapy might reduce the risk of recurrent symptomatic intracerebral haemorrhage compared with avoiding antiplatelet therapy. Brain imaging features of intracerebral haemorrhage and cerebral small vessel diseases (such as cerebral microbleeds) are associated with greater risks of recurrent intracerebral haemorrhage.     Pharmacogenetic Discovery in CALGB (Alliance) 90401 and Mechanistic Validation of a VAC14 Polymorphism That Increases Risk of Docetaxel-Induced Neuropathy

    Purpose Discovery of single nucleotide polymorphisms (SNPs) that predict a patient\u27s risk of docetaxel-induced neuropathy would enable treatment individualization to maximize efficacy and avoid unnecessary toxicity. The objectives of this analysis were to discover SNPs associated with docetaxel-induced neuropathy and mechanistically validate these associations in preclinical models of drug-induced neuropathy. Experimental Design A genome-wide association study was conducted in metastatic castrate-resistant prostate cancer patients treated with docetaxel, prednisone and randomized to bevacizumab or placebo on CALGB 90401. SNPs were genotyped on the Illumina HumanHap610-Quad platform followed by rigorous quality control. The inference was conducted on the cumulative dose at occurrence of grade 3+ sensory neuropathy using a cause-specific hazard model that accounted for early treatment discontinuation. Genes with SNPs significantly associated with neuropathy were knocked down in cellular and mouse models of drug-induced neuropathy. Results 498,081 SNPs were analyzed in 623 Caucasian patients, 50 (8%) of whom experienced grade 3+ neuropathy. The 1000 SNPs most associated with neuropathy clustered in relevant pathways including neuropathic pain and axonal guidance. A SNP in VAC14 (rs875858) surpassed genome-wide significance (p=2.12×10-8 adjusted p=5.88×10-7). siRNA knockdown of VAC14 in stem cell derived peripheral neuronal cells increased docetaxel sensitivity as measured by decreased neurite processes (p=0.0015) and branches (p\u3c0.0001). Prior to docetaxel treatment VAC14 heterozygous mice had greater nociceptive sensitivity than wild-type litter mate controls (p=0.001). Conclusions VAC14 should be prioritized for further validation of its potential role as a predictor of docetaxel-induced neuropathy and biomarker for treatment individualization
