Method of identifying inhibitors of glutathione S-transferase (GST) gene expression
Complementary DNA and genomic clones for three variants of GST-.pi. are disclosed. It is demonstrated that certain of these variants are overexpressed in gliomas, thereby indicating an involvement with that form of cancer. At the 2016 All Campus Forum President Hinton called for us to create an ecosystem of inclusion at CSB/SJU. Faculty members\u27 work with students is central to creating such an ecosystem, but what, after all, do we mean by inclusion? In this presentation, FYS and Humanities Mellon cohort members provide an overview of our evolving understanding of inclusive pedagogy, the research that supports it, as well as examples of such pedagogies that we are implementing in our fall 2016 courses
Uniform approximation for diffractive contributions to the trace formula in billiard systems
We derive contributions to the trace formula for the spectral density
accounting for the role of diffractive orbits in two-dimensional billiard
systems with corners. This is achieved by using the exact Sommerfeld solution
for the Green function of a wedge. We obtain a uniformly valid formula which
interpolates between formerly separate approaches (the geometrical theory of
diffraction and Gutzwiller's trace formula). It yields excellent numerical

Aeroelastic Response and Protection of Space Shuttle External Tank Cable Trays
Sections of the Space Shuttle External Tank Liquid Oxygen (LO2) and Liquid Hydrogen (LH2) cable trays are shielded from potentially damaging airloads with foam Protuberance Aerodynamic Load (PAL) Ramps. Flight standard design LO2 and LH2 cable tray sections were tested with and without PAL Ramp models in the United States Air Force Arnold Engineering Development Center s (AEDC) 16T transonic wind tunnel to obtain experimental data on the aeroelastic stability and response characteristics of the trays and as part of the larger effort to determine whether the PAL ramps can be safely modified or removed. Computational Fluid Dynamic simulations of the full-stack shuttle launch configuration were used to investigate the flow characeristics around and under the cable trays without the protective PAL ramps and to define maximum crossflow Mach numbers and dynamic pressures experienced during launch. These crossflow conditions were used to establish wind tunnel test conditions which also included conservative margins. For all of the conditions and configurations tested, no aeroelastic instabilities or unacceptable dynamic response levels were encountered and no visible structural damage was experienced by any of the tested cable tray sections. Based upon this aeroelastic characterization test, three potentially acceptable alternatives are available for the LO2 cable tray PAL Ramps: Mini-Ramps, Tray Fences, or No Ramps. All configurations were tested to maximum conditions, except the LH2 trays at -15 deg. crossflow angle. This exception is the only caveat preventing the proposal of acceptable alternative configurations for the LH2 trays as well. Structural assessment of all tray loads and tray response measurements from launches following the Shuttle Return To Flight with the existing PAL Ramps will determine the acceptability of these PAL Ramp alternatives
Twirling and Whirling: Viscous Dynamics of Rotating Elastica
Motivated by diverse phenomena in cellular biophysics, including bacterial
flagellar motion and DNA transcription and replication, we study the overdamped
nonlinear dynamics of a rotationally forced filament with twist and bend
elasticity. Competition between twist injection, twist diffusion, and writhing
instabilities is described by a novel pair of coupled PDEs for twist and bend
evolution. Analytical and numerical methods elucidate the twist/bend coupling
and reveal two dynamical regimes separated by a Hopf bifurcation: (i)
diffusion-dominated axial rotation, or twirling, and (ii) steady-state
crankshafting motion, or whirling. The consequences of these phenomena for
self-propulsion are investigated, and experimental tests proposed.Comment: To be published in Physical Review Letter
The CO-to-H2 Conversion Factor From Infrared Dust Emission Across the Local Group
We estimate the conversion factor relating CO emission to H2 mass, alpha_CO,
in five Local Group galaxies that span approximately an order of magnitude in
metallicity - M31, M 33, the Large Magellanic Cloud (LMC), NGC 6822, and the
Small Magellanic Cloud (SMC). We model the dust mass along the line of sight
from infrared (IR) emission and then solve for the alpha_CO that best allows a
single gas-to-dust ratio (delta_GDR) to describe each system. This approach
remains sensitive to CO-dark envelopes of H2 surrounding molecular clouds. In M
31, M 33, and the LMC we find alpha_CO \approx 3-9 M_sun pc^-2 (K km s^-1)^-1,
consistent with the Milky Way value within the uncertainties. The two lowest
metallicity galaxies in our sample, NGC 6822 and the SMC (12 + log(O/H) \approx
8.2 and 8.0), exhibit a much higher alpha_CO. Our best estimates are
\alpha_NGC6822 \approx 30 M_sun/pc^-2 (K km s^-1)^-1 and \alpha_SMC \approx 70
M_sun/pc^-2 (K km s-1)-1. These results are consistent with the conversion
factor becoming CO a strong function of metallicity around 12 + log(O/H) \sim
8.4 - 8.2. We favor an interpretation where decreased dust-shielding leads to
the dominance of CO-free envelopes around molecular clouds below this

Negative correlation of single-cell PAX3:FOXO1 expression with tumorigenicity in rhabdomyosarcoma
Rhabdomyosarcomas (RMS) are phenotypically and functionally heterogeneous. Both primary human RMS cultures and low-passage Myf6Cre,Pax3:Foxo1,p53 mouse RMS cell lines, which express the fusion oncoprotein Pax3:Foxo1 and lack the tumor suppressor Tp53 (Myf6Cre,Pax3:Foxo1,p53), exhibit marked heterogeneity in PAX3:FOXO1 (P3F) expression at the single cell level. In mouse RMS cells, P3F expression is directed by the Pax3 promoter and coupled to eYFP. YFPlow/P3Flow mouse RMS cells included 87% G0/G1 cells and reorganized their actin cytoskeleton to produce a cellular phenotype characterized by more efficient adhesion and migration. This translated into higher tumor-propagating cell frequencies of YFPlow/P3Flow compared with YFPhigh/P3Fhigh cells. Both YFPlow/P3Flow and YFPhigh/P3Fhigh cells gave rise to mixed clones in vitro, consistent with fluctuations in P3F expression over time. Exposure to the anti-tropomyosin compound TR100 disrupted the cytoskeleton and reversed enhanced migration and adhesion of YFPlow/P3Flow RMS cells. Heterogeneous expression of PAX3:FOXO1 at the single cell level may provide a critical advantage during tumor progression
