    The virtual landing pad: facilitating rotary-wing landing operations in degraded visual environments

    The safety of rotary-wing operations are significantly affected by the local weather conditions, especially during key phases of flight including hover and landing. Despite the operational flexibility of rotary-wing craft, such craft account for a significantly greater proportion of accidents than their fixed-wing counterparts. A key period of risk when operating rotary-wing aircraft is during operations that occur in degraded visual environments, for example as a result of thick fog. During such conditions, pilots’ workload significantly increases and their situation awareness can be greatly impeded. The current study examines the extent to which providing information to pilots via the use of a Head-Up display (HUD) influenced perceived workload and situation awareness, when operating in both clear and degraded visual environments. Results suggest that whilst the HUD did not benefit pilots during clear conditions, workload was reduced when operating in degraded visual conditions. Overall results demonstrate that access to the HUD reduces the difficulties associated with flying in degraded visual environments

    Applying the AcciMap methodology to investigate the tragic Mirsharai road accident in Bangladesh

    A serious road accident happened on Monday, 11 July, 2011 afternoon in Mirsharai sub-district of Bangladesh's Chittagong district, that killed at least 44 schoolboys injured many more when the truck in which they were traveling skidded off a highway and plunged into a canal. In this study, the AcciMap methodology is applied, an extension to Rasmussen’s Risk Management Framework. This sociotechnical framework provides an insight into the interactions between different levels of a system, and supports the development of recommendations that could mitigate or prevent the outcomes of such an incident in the future. In the case of Mirsharai accident, the police and social media put the blame on the truck driver alone; however, this study, based on popular media reports, accident investigation reports and local interviews revealed mismanagement and an apparent disregard for safety across different levels of the socio-technical system. By integrating the different levels of the system, from international committees to end users, the road accident scenario of Bangladesh can be improved through developing a systematic approach to accident analysis

    Regional fat depot masses are influenced by protein-coding gene variants

    Waist-to-hip ratio (WHR) is a prominent cardiometabolic risk factor that increases cardio-metabolic disease risk independently of BMI and for which multiple genetic loci have been identified. However, WHR is a relatively crude proxy for fat distribution and it does not capture all variation in fat distribution. We here present a study of the role of coding genetic variants on fat mass in 6 distinct regions of the body, based on dual-energy X-ray absorptiometry imaging on more than 17k participants. We find that the missense variant CCDC92(S70C), previously associated with WHR, is associated specifically increased leg fat mass and reduced visceral but not subcutaneous central fat. The minor allele-carrying transcript of CCDC92 is constitutively more highly expressed in adipose tissue samples. In addition, we identify two coding variants in SPATA20 and UQCC1 that are associated with arm fat mass. SPATA20(K422R) is a low-frequency variant with a large effect on arm fat only, and UQCC1(R51Q) is a common variant reaching significance for arm but showing similar trends in other subcutaneous fat depots. Our findings support the notion that different fat compartments are regulated by distinct genetic factors.Peer reviewe

    Tropical Pacific observing system

    This paper reviews the design of the Tropical Pacific Observing System (TPOS) and its governance and takes a forward look at prospective change. The initial findings of the TPOS 2020 Project embrace new strategic approaches and technologies in a user-driven design and the variable focus of the Framework for Ocean Observing. User requirements arise from climate prediction and research, climate change and the climate record, and coupled modeling and data assimilation more generally. Requirements include focus on the upper ocean and air-sea interactions, sampling of diurnal variations, finer spatial scales and emerging demands related to biogeochemistry and ecosystems. One aim is to sample a diversity of climatic regimes in addition to the equatorial zone. The status and outlook for meeting the requirements of the design are discussed. This is accomplished through integrated and complementary capabilities of networks, including satellites, moorings, profiling floats and autonomous vehicles. Emerging technologies and methods are also discussed. The outlook highlights a few new foci of the design: biogeochemistry and ecosystems, low-latitude western boundary currents and the eastern Pacific. Low latitude western boundary currents are conduits of tropical-subtropical interactions, supplying waters of mid to high latitude origin to the western equatorial Pacific and into the Indonesian Throughflow. They are an essential part of the recharge/discharge of equatorial warm water volume at interannual timescales and play crucial roles in climate variability on regional and global scales. The tropical eastern Pacific, where extreme El Niño events develop, requires tailored approaches owing to the complex of processes at work there involving coastal upwelling, and equatorial cold tongue dynamics, the oxygen minimum zone and the seasonal double Intertropical Convergence Zone. A pilot program building on existing networks is envisaged, complemented by a process study of the East Pacific ITCZ/warm pool/cold tongue/stratus coupled system. The sustainability of TPOS depends on effective and strong collaborative partnerships and governance arrangements. Revisiting regional mechanisms and engaging new partners in the context of a planned and systematic design will ensure a multi-purpose, multi-faceted integrated approach that is sustainable and responsive to changing needs

    The structural basis of bacterial manganese import

In prokaryotes, ATP-binding cassette (ABC) permeases serve as the primary import pathway for many micronutrients including the first-row transition metal manganese. However, the structural features of ionic metal transporting ABC permeases have remained undefined. Here, we present the crystal structure of the manganese transporter PsaBC from Streptococcus pneumoniae in an open-inward conformation. The type II transporter has a tightly closed transmembrane channel due to “extracellular gating” residues that prevent water permeation or ion reflux. Below these residues, the channel contains a hitherto unreported metal coordination site, which is essential for manganese translocation. Mutagenesis of the extracellular gate perturbs manganese uptake, while coordination site mutagenesis abolishes import. These structural features are highly conserved in metal-specific ABC transporters and are represented throughout the kingdoms of life. Collectively, our results define the structure of PsaBC and reveal the features required for divalent cation transport
