    Rapid and reliable extraction of genomic DNA from various wild-type and transgenic plants

    BACKGROUND: DNA extraction methods for PCR-quality DNA from calluses and plants are not time efficient, since they require that the tissues be ground in liquid nitrogen, followed by precipitation of the DNA pellet in ethanol, washing and drying the pellet, etc. The need for a rapid and simple procedure is urgent, especially when hundreds of samples need to be analyzed. Here, we describe a simple and efficient method of isolating high-quality genomic DNA for PCR amplification and enzyme digestion from calluses, various wild-type and transgenic plants. RESULTS: We developed new rapid and reliable genomic DNA extraction method. With our developed method, plant genomic DNA extraction could be performed within 30 min. The method was as follows. Plant tissue was homogenized with salt DNA extraction buffer using hand-operated homogenizer and extracted by phenol:chloroform:isoamyl alcohol (25:24:1). After centrifugation, the supernatant was directly used for DNA template for PCR, resulting in successful amplification for RAPD from various sources of plants and specific foreign genes from transgenic plants. After precipitating the supernatant, the DNA was completely digested by restriction enzymes. CONCLUSION: This DNA extraction procedure promises simplicity, speed, and efficiency, both in terms of time and the amount of plant sample required. In addition, this method does not require expensive facilities for plant genomic DNA extraction

    Comparison of Two Mice Strains, A/J and C57BL/6, in Caspase-1 Activity and IL-1β Secretion of Macrophage to Mycobacterium leprae Infection

    A/J mice were found to have amino acid differences in Naip5, one of the NOD-like receptors (NLRs) involved in the cytosolic recognition of pathogen-associated molecular patterns and one of the adaptor proteins for caspase-1 activation. This defect was associated with a susceptibility to Legionella infection, suggesting an important role for Naip5 in the immune response also to other intracellular pathogens, such as Mycobacterium leprae. In this study, the immune responses of macrophages from A/J mice against M. leprae were compared to those of macrophages from C57BL/6 mice. Infection with M. leprae induced high levels of TNF-α production and NF-κB activation in A/J and C57BL/6 macrophages. Caspase-1 activation and IL-1β secretion were also induced in both macrophages. However, macrophages from A/J mice exhibited reduced caspase-1 activation and IL-1β secretion compared to C57BL/6 macrophages. These results suggest that NLR family proteins may have a role in the innate immune response to M. leprae

    In this paper, we have conducted classical molecular dynamics simulations for DWCNTs of various wall lengths to investigate their use as ultrahigh frequency nano-mechanical resonators. We sought to determine the variations in the frequency of these resonators according to changes in the DWCNT wall lengths. For a double-walled carbon nanotube resonator with a shorter inner nanotube, the shorter inner nanotube can be considered to be a flexible core, and thus, the length influences the fundamental frequency.     Tissue-specific down-regulation of RIPK 2 in Mycobacterium leprae-infected nu/nu mice.

    RIPK 2 is adapter molecule in the signal pathway involved in Toll-like receptors. However, there has been no reported association between receptor-interacting serine/threonine kinase 2 (RIPK 2) expression and the infectious diseases involving mycobacterial infection. This study found that its expression was down-regulated in the footpads and skin but was up-regulated in the liver of Mycobacterium leprae-infected nu/nu mice compared with those of the M. leprae non-infected nu/nu mice. It was observed that the interlukin-12p40 and interferon-gamma genes involved in the susceptibility of M. leprae were down-regulated in the skin but were up-regulated in the liver. Overall, this suggests that regulation of RIPK 2 expression is tissue-specifically associated with M. leprae infection

    AbstractPrecipitates, present in most commercial alloys, can have a strong influence on strength and hardening behavior of a single crystal. The effect of thin precipitates on the anisotropy of initial slip resistance and hardening behavior of crystals is modeled in this article. For the convenience of the computational derivation and implementation, the material formulation is given in the unrotated intermediate configuration mapped by the plastic part of the deformation gradient. Material descriptions for the considered two phased aggregates consisting in lattice hardening as well as isotropic hardening and kinematic hardening are suggested. The corresponding elastic–plastic rate-independent algorithmic treatment is derived and numerical simulations of various loading cases are presented to discuss and assess the performance of the suggested model and its rate-independent algorithmic treatment

    FEASIBILITY OF BREWING MAKGEOLLI (TURBID RICE WINE) USING PARTIALLY GELATINIZED WHEAT FLOUR AND TAPIOCA FLOUR

    Makgeolli is made from cooked rice or wheat, then brewed with nuruk (Korean fermentation starter) for several days. But, nowadays, attempts have been made to use various raw materials and process innovations to make makgeolli for particular purposes.  This study aimed to evaluate the quality of makgeolly made from partially gelatinized wheat flour and tapioca flour. Five different combination of wheat flour and tapioca flour were used to manufacture makgeolli. The results showed that different combination of partially gelatinized wheat flour and tapioca flour significantly affected the chemical and sensorial characteristics of makgeolli. Increasing proportion of wheat flour produced higher level of total acid, amino acidity, reducing sugar and total solid of makgeolli. Inversely, alcohol content was higher when higher level of tapioca flour was used. In general, sensorial characteristics of makgeolli made from partially gelatinized wheat flour and tapioca flour didn’t acceptable by panelists. Thus, brewing makgeolli by using partially gelatinized wheat flour and tapioca flour isn’t acceptable in term of sensorial characteristics
