    Effect of in situ soil moisture conservation practices on maize and its crop residue incorporation on yield and economics of succeeding transplanted rice Var. ADT 46

    Crop residue incorporation is a key component of sustainable cropping systems. It reduces the adverse effects of residue burning and enhances soil fertility. Effective usage of crop residue in the field and proper management are required. With this background, a field experiment was conducted during 2020 – 21 in the maize-rice cropping sequence at Annamalai University Experimental Farm, Department of Agronomy, Faculty of Agriculture, Annamalai University, Annamalai Nagar, Tamil Nadu to find out the residual effect of different mulching practices adopted in preceding maize crop and maize stubble incorporation on the growth, yield and economics of transplanted rice Var. ADT 46. The field experiment was conducted in Factorial Randomized Block Design with three replications. In factor I, soil moisture conservation in preceding maize crop viz., mulching of sugarcane trash, water hyacinth, hydrogel and control were allotted. In factor II, different levels of maize crop stubble incorporation on rice viz., 0, 33, 66 and 100% were provided. Water hyacinth mulched plot (M3) to the preceding crop registered significantly (þ <0.05) higher yield parameters, yield and economic returns of succeeding rice. The lower values were observed in unmulched (M1) plot. With respect to maize crop stubble incorporation on rice crop, the incorporation of 66% (SI3) of maize stubble registered higher yield parameters, yield and economic returns. In the interaction effects, mulching with water hyacinth to preceding maize + maize crop stubble incorporation at 66% in rice crop (M3SI3) recorded significantly (þ <0.05) higher yield parameters, yield and economic returns than other treatments. The lowest values were recorded with an unmulched + 100% crop residue incorporated (M1SI4) plot. Mulching the preceding maize crop with water hyacinth at a rate of 12 t ha-1 and incorporating 66% maize stubble into the transplanted rice (M3SI3) had a remarkable yield advantage and financial rewards

    High performance thin-layer chromatography and in vitro cytotoxic studies on ethanol extract of Matricaria chamomilla L (Asteraceae) flowers

    Purpose: To develop a high performance thin-layer chromatography (HPTLC procedure for quantitation of apigenin in ethanol extract of Matricaria chamomilla (Babunaj) flowers, and to evaluate the extract for in vitro cytotoxic effect on MCF-7 cell lines. Methods: Quantification of apigenin was carried out using a CAMAG TLC system. A combination of toluene, ethyl acetate and formic acid (4.5:3.5:0.2 v/v/v) was used as mobile phase, with densitometry detection at 336 nm. The HPTLC procedure was subjected to validation as per ICH guidelines. The cytotoxicity of the extract was assessed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay. Results: A sharp apigenin band at Rf of 0.51 was obtained, and the content of apigenin in the extract was 0.062 % w/w. The detection limit (LOD) and quantification limit (LOQ) were 0.19 and 0.57 ng/band, respectively. MTT assay results indicate that M. chamomilla was cytotoxic to Michigan Cancer Foundation-7 (MCF-7) cells, with half-maximal concentration (IC50) of 74 µg/mL. Conclusion: The developed HPTLC method is linear, precise, accurate and specific for the determination of apigenin. M. chamomilla exerts cytotoxic effect on MCF-7 cell line via induction of apoptosis

    Database Search Strategies for Proteomic Data Sets Generated by Electron Capture Dissociation Mass Spectrometry

    Large data sets of electron capture dissociation (ECD) mass spectra from proteomic experiments are rich in information; however, extracting that information in an optimal manner is not straightforward. Protein database search engines currently available are designed for low resolution CID data, from which Fourier transform ion cyclotron resonance (FT-ICR) ECD data differs significantly. ECD mass spectra contain both z-prime and z-dot fragment ions (and c-prime and c-dot); ECD mass spectra contain abundant peaks derived from neutral losses from charge-reduced precursor ions; FT-ICR ECD spectra are acquired with a larger precursor m/z isolation window than their low-resolution CID counterparts. Here, we consider three distinct stages of postacquisition analysis: (1) processing of ECD mass spectra prior to the database search; (2) the database search step itself and (3) postsearch processing of results. We demonstrate that each of these steps has an effect on the number of peptides identified, with the postsearch processing of results having the largest effect. We compare two commonly used search engines: Mascot and OMSSA. Using an ECD data set of modest size (3341 mass spectra) from a complex sample (mouse whole cell lysate), we demonstrate that search results can be improved from 630 identifications (19% identification success rate) to 1643 identifications (49% identification success rate). We focus in particular on improving identification rates for doubly charged precursors, which are typically low for ECD fragmentation.     A randomized controlled trial of the effects of a prudent diet on cardiovascular risk factors, gene expression, and DNA methylation - the Diet and Genetic Intervention (DIGEST) Pilot study

    Background Risk of cardiovascular disease (CVD) can be increased by single-nucleotide polymorphisms (SNPs) in the 9p21 region of the genome. However, observational studies have shown that the deleterious effect of 9p21 SNPs on CVD might be offset by consuming a diet rich in fresh fruits and vegetables. This association may be driven by diet-influenced modifications in epigenetic and gene expression profiles. In this pilot study, we aimed to: i. test the feasibility of provision of a ‘Prudent’ and ‘Western’ diet outside of a specialized clinic, ii. assess the impact of each diet on cardiovascular risk factors. Methods A single centre, parallel two-arm, pilot randomized controlled trial (RCT) with food provision was conducted in a university teaching hospital outpatient clinic (McMaster university, Hamilton, ON, Canada). The aim was to recruit 80 participants, which allowed for a 10 % dropout. The actual study consisted of 84 apparently healthy participants (69 % women, 18 to 77 years) at low cardiovascular risk. Participants were randomly assigned to follow one of two weight-maintaining diets: ‘Prudent’ or ‘Western’ for 2-weeks. The Prudent diet provided 92 % of provided food consumed). The Prudent diet was 48 % more palatable than the Western diet (P < 0.05). Participants receiving the Prudent diet showed a trend toward reduced systolic (-4 mmHg; P = 0.10) and diastolic (-3 mmHg; P = 0.07) blood pressure, and total cholesterol (-0.24 mmol/L; P = 0.08), compared to individuals receiving the Western diet. Data collection from all randomized participants was completed within 18 months. Conclusions Recruitment, and retention of apparently healthy, normotensive adults into a feeding study for a 2-week duration is feasible outside of specialized dietary clinic, and modest diet-related changes in biomarkers begin to appear after two weeks

    Tree defence and bark beetles in a drying world: carbon partitioning, functioning and modelling

    Drought has promoted large‐scale, insect‐induced tree mortality in recent years, with severe consequences for ecosystem function, atmospheric processes, sustainable resources and global biogeochemical cycles. However, the physiological linkages among drought, tree defences, and insect outbreaks are still uncertain, hindering our ability to accurately predict tree mortality under on‐going climate change. Here we propose an interdisciplinary research agenda for addressing these crucial knowledge gaps. Our framework includes field manipulations, laboratory experiments, and modelling of insect and vegetation dynamics, and focuses on how drought affects interactions between conifer trees and bark beetles. We build upon existing theory and examine several key assumptions: 1) there is a trade‐off in tree carbon investment between primary and secondary metabolites (e.g. growth vs. defence); 2) secondary metabolites are one of the main component of tree defence against bark beetles and associated microbes; and 3) implementing conifer‐bark beetle interactions in current models improves predictions of forest disturbance in a changing climate. Our framework provides guidance for addressing a major shortcoming in current implementations of large‐scale vegetation models, the under‐representation of insect‐induced tree mortality

    Investigating Bayesian optimization for rail network optimization

    Optimizing the operation of rail networks using simulations is an on-going task where heuristic methods such as Genetic Algorithms have been applied. However, these simulations are often expensive to compute and consequently, because the optimization methods require many (typically >104) repeat simulations, the computational cost of optimization is dominated by them. This paper examines Bayesian Optimization and benchmarks it against the Genetic Algorithm method. By applying both methods to test-tasks seeking to maximize passenger satisfaction by optimum resource allocation, it is experimentally determined that a Bayesian Optimization implementation finds ‘good’ solutions in an order of magnitude fewer simulations than a Genetic Algorithm. Similar improvement for real-world problems will allow the predictive power of detailed simulation models to be used for a wider range of network optimization tasks. To the best of the authors’ knowledge, this paper documents the first application of Bayesian Optimization within the field of rail network optimization
