    RNA–protein binding kinetics in an automated microfluidic reactor

    Microfluidic chips can automate biochemical assays on the nanoliter scale, which is of considerable utility for RNA–protein binding reactions that would otherwise require large quantities of proteins. Unfortunately, complex reactions involving multiple reactants cannot be prepared in current microfluidic mixer designs, nor is investigation of long-time scale reactions possible. Here, a microfluidic ‘Riboreactor’ has been designed and constructed to facilitate the study of kinetics of RNA–protein complex formation over long time scales. With computer automation, the reactor can prepare binding reactions from any combination of eight reagents, and is optimized to monitor long reaction times. By integrating a two-photon microscope into the microfluidic platform, 5-nl reactions can be observed for longer than 1000 s with single-molecule sensitivity and negligible photobleaching. Using the Riboreactor, RNA–protein binding reactions with a fragment of the bacterial 30S ribosome were prepared in a fully automated fashion and binding rates were consistent with rates obtained from conventional assays. The microfluidic chip successfully combines automation, low sample consumption, ultra-sensitive fluorescence detection and a high degree of reproducibility. The chip should be able to probe complex reaction networks describing the assembly of large multicomponent RNPs such as the ribosome

    Examining the role of menthol cigarettes in progression to established T smoking among youth

    Background: Menthol, a flavoring compound added to cigarettes, makes cigarettes more appealing to youth and inexperienced smokers and increases cigarettes\u27 abuse liability. However, limited studies are available on menthol\u27s role in smoking progression. Methods: To assess the association between menthol in cigarettes and progression to established smoking, we used five waves of data from the Evaluation of Public Education Campaign on Teen Tobacco Cohort Study, a nationally representative longitudinal survey of U.S. youth conducted as part of “The Real Cost” evaluation. We used discrete time survival analysis to model the occurrence of two event outcomes—progression to established, current smoking and progression to established, frequent smoking—using a logit model with a menthol use indicator as the key explanatory variable. Based on this framework, we estimated the effect of prior menthol use on the odds of smoking progression. Results: In the progression to established, current smoking model, prior menthol use was significantly associated with progression [adjusted odds ratio (aOR) = 1.80, p \u3c .05, confidence interval (CI) = (1.03–3.16)]. While results were in a similar direction for the model of progression to established, frequent smoking, the association between prior menthol use and this progression model did not reach significance [aOR=1.56, CI = (0.80–3.03)]. Conclusion: The results suggest a relationship between using menthol cigarettes and progression from experi- mental to established, current smoking among youth. This study adds to a growing literature base that supports that menthol cigarettes, compared to nonmenthol cigarettes, put youth at increased risk for regular cigarette use

    Snow information is required in subcontinental scale predictions of mountain plant distributions

    Aim To examine how snow cover and permafrost affect plant species distributions at a subcontinental extent. Location Mountain realm of Fennoscandia, northern Europe. Time period Species data from 1 January 1990-25 February 2019. Major taxa studied Arctic-alpine and boreal vascular plants. Methods We examined the effect of snow persistence and permafrost occurrence on the distributions of arctic-alpine and boreal plant species while controlling for climate, topography and geological factors. Data comprised 475,811 observations from 671 species in the Fennoscandian mountains. We investigated the relationships between species distributions and environmental variables using four modelling methods and ensemble modelling building on both non-spatial and spatial models. Results Snow persistence was the most important driver of plant species distributions, with the greatest variable importance for both arctic-alpine (38.2%) and boreal (49.9%) species. Permafrost had a consistent minor effect on the predicted distributions. Arctic-alpine plants occur in areas with long snow persistence and permafrost, whereas boreal species showed the opposite habitat preferences. Main conclusions Our results highlight the importance of snow persistence in driving the distribution of vascular plant species in cold environments at a subcontinental scale. The notable contribution of the cryosphere to plant species distribution models indicates that the inclusion of snow information in particular may improve our understanding and model predictions of biogeographical patterns in cold regions.Peer reviewe

    A 180 Kpc Tidal Tail in the Luminous Infrared Merger Arp 299

    We present VLA HI observations and UH88 deep optical B- and R-band observations of the IR luminous merger Arp 299 (= NGC 3690 + IC 694). These data reveal a gas-rich, optically faint tidal tail with a length of over 180 kpc. The size of this tidal feature necessitates an old interaction age for the merger (~750 Myr since first periapse), which is currently experiencing a very young star burst (~20 Myr). The observations reveal a most remarkable structure within the tidal tail: it appears to be composed of two parallel filaments separated by ~20 kpc. One of the filaments is gas rich with little if any starlight, while the other is gas poor. We believe that this bifurcation results from a warped disk in one of the progenitors. The quantities and kinematics of the tidal HI suggest that Arp 299 results from the collision of a retrograde Sab-Sb galaxy (IC 694) and a prograde Sbc-Sc galaxy (NGC 3690) that occurred 750 Myr ago and which will merge into a single object in ~60 Myr. We suggest that the present IR luminous phase in this system is due in part to the retrograde spin of IC 694.     In the 1990s and early 2000s, there was much discussion about the democratic and anti-democratic implications of the Internet. The latter particularly focused on the ways in which the far-right were using the Internet to spread hate and recruit members. Despite this common assumption, the American far-right did not harness the Internet quickly, effectively or widely. More recently, however, they have experienced a resurgence and mainstreaming, benefitting greatly from social media. This chapter examines the history of their use of the Internet with respect to: (1) how this developed in response to political changes and emerging technologies; (2) how it reflected and changed the status of such movements and their brand of hate; and (3) the relationship between online activity and traditional methods of communication

    Surgical management of mesh-related complications after prior pelvic floor reconstructive surgery with mesh

    Contains fulltext : 96379.pdf (publisher's version ) (Closed access)INTRODUCTION AND HYPOTHESIS: The objective of this study is to evaluate the complications and anatomical and functional outcomes of the surgical treatment of mesh-related complications. METHODS: A retrospective cohort study of patients who underwent complete or partial mesh excision to treat complications after prior mesh-augmented pelvic floor reconstructive surgery was conducted. RESULTS: Seventy-three patients underwent 30 complete and 51 partial mesh excisions. Intraoperative complications occurred in 4 cases, postoperative complications in 13. Symptom relief was achieved in 92% of patients. Recurrence of pelvic organ prolapse (POP) occurred in 29% of complete and 5% of partial excisions of mesh used in POP surgery. De novo stress urinary incontinence (SUI) occurred in 36% of patients who underwent excision of a suburethral sling. CONCLUSIONS: Mesh excision relieves mesh-related complications effectively, although with a substantial risk of serious complications and recurrence of POP or SUI. More complex excisions should be performed in skilled centers
