    Developmental, morphological, and behavioural plasticity in the reproductive strategies of stink bugs and their egg parasitoids

    L’environnement façonne la physiologie, la morphologie et le comportement des organismes par l’entremise de processus écologiques et évolutifs complexes et multidimensionnels. Le succès reproducteur des animaux est déterminé par la valeur adaptative d’un phénotype dans un environnement en modification constante selon une échelle temporelle d’une à plusieurs générations. De plus, les phénotypes sont façonnés par l’environnement, ce qui entraine des modifications adaptatives des stratégies de reproduction tout en imposant des contraintes. Dans cette thèse, considérant des punaises et leurs parasitoïdes comme organismes modèles, j’ai investigué comment plusieurs types de plasticité peuvent interagir pour influencer la valeur adaptative, et comment la plasticité des stratégies de reproduction répond à plusieurs composantes des changements environnementaux (qualité de l’hôte, radiation ultraviolette, température, invasion biologique). Premièrement, j’ai comparé la réponse comportementale et de traits d’histoire de vie à la variation de taille corporelle chez le parasitoïde Telenomus podisi Ashmead (Hymenoptera : Platygastridae), démontrant que les normes de réaction des comportements étaient plus souvent positives que celles des traits d’histoires de vie. Ensuite, j’ai démontré que la punaise prédatrice Podisus maculiventris Say (Hemiptera : Pentatomidae) peut contrôler la couleur de ses œufs, et que la pigmentation des œufs protège les embryons du rayonnement ultraviolet; une composante d’une stratégie complexe de ponte qui a évoluée en réponse à une multitude de facteurs environnementaux. Puis, j’ai testé comment le stress thermique affectait la dynamique de la mémoire du parasitoïde Trissolcus basalis (Wollaston) (Hymenoptera : Platygastridae) lors de l’apprentissage de la fiabilité des traces chimiques laissées par son hôte. Ces expériences ont révélé que des températures hautes et basses prévenaient l’oubli, affectant ainsi l’allocation du temps passé par les parasitoïdes dans des agrégats d’hôtes contenant des traces chimiques. J’ai aussi développé un cadre théorique général pour classifier les effets de la température sur l’ensemble des aspects comportementaux des ectothermes, distinguant les contraintes des adaptations. Finalement, j’ai testé l’habileté d’un parasitoïde indigène (T. podisi) à exploiter les œufs d’un nouveau ravageur invasif en agriculture, Halyomorpha halys Stål (Hemiptera : Pentatomidae). Les résultats ont montré que T. podisi attaque les œufs de H. halys, mais qu’il ne peut s’y développer, indiquant que le ravageur invasif s’avère un « piège évolutif » pour ce parasitoïde. Cela pourrait indirectement bénéficier aux espèces indigènes de punaises en agissant comme un puits écologique de ressources (œufs) et de temps pour le parasitoïde. Ces résultats ont des implications importantes sur la réponse des insectes, incluant ceux impliqués dans les programmes de lutte biologique, face aux changements environnementaux.The environment shapes the physiology, morphology, and behaviour of organisms through complex, multidimensional ecological and evolutionary processes. The reproductive success of individual animals is determined by how well their phenotype is suited to an environment that is constantly changing over single and multi-generational time scales. At the same time, phenotypes are shaped by the environment, which triggers adaptive modifications of animal reproductive strategies while also imposing important constraints. In this thesis, using stink bugs and their parasitoids as model organisms, I considered how several types of plasticity can interact to influence biological fitness, and how plasticity in reproductive strategies responds to several important components of environmental change (host quality, ultraviolet radiation, temperature, biological invasions). Firstly, I compared the response of behavioural and life history traits to body size variation in the parasitoid Telenomus podisi Ashmead (Hymenoptera: Platygastridae), finding that reaction norms of behavioural traits more often had positive slopes than life history traits. Next, I found that the predatory stink bug Podisus maculiventris Say (Hemiptera: Pentatomidae) can selectively control the colouration of its eggs. Egg pigmentation in this species protects embryos against ultraviolet radiation as part of a complex oviposition strategy that evolved in response to a suite of environmental factors. Then, I tested how thermal stress affects the memory dynamics of the parasitoid Trissolcus basalis (Wollaston) (Hymenoptera: Platygastridae) learning the reliability of chemical traces left by its host. These experiments revealed that both high and low stressful temperatures prevented forgetting, affecting the time allocation of parasitoids on patches of host chemical traces. I also developed a general framework to classify temperature’s effects on all aspects of ectotherm behaviour, distinguishing constraints from adaptive behavioural adjustments. Finally, I tested the ability of an indigenous parasitoid (T. podisi) to attack the eggs of a new invasive pest of agriculture, Halyomorpha halys Stål (Hemiptera: Pentatomidae). The results showed that T. podisi attacks the eggs of H. halys but cannot develop, demonstrating that the invasive pest is an “evolutionary trap” for indigenous parasitoids, which could indirectly benefit native stink bug species by acting as an egg and time sink for the parasitoid. These findings have important implications for how insects, including those involved in biological control programs, respond to environmental change

    Hurricane storm surges are influenced by several factors, including wind intensity, surface pressure, forward speed, size, angle of approach, ocean bottom depth and slope, shape and geographical features of the coastline. The relative influence of each factor may be amplified or abated by other factors that are acting at the time of the hurricane’s approach to the land. To understand the individual and combined influence of wind intensity, surface pressure and forward speed, a numerical experiment is conducted using Advanced CIRCulation + Simulating Waves Nearshore (ADCIRC + SWAN) by performing hindcasts of Hurricane Rita storm surges. The wind field generated by Ocean Weather Inc. (OWI) is used as the base meteorological forcing in ADCIRC + SWAN. All parameters are varied by certain percentages from those in the OWI wind field. Simulation results are analyzed for maximum wind intensity, wind vector pattern, minimum surface pressure, forward speed, maximum water elevation, station water elevation time series, and high water marks. The results for different cases are compared against each other, as well as with observed data. Changes in the wind intensity have the greatest impact, followed by the forward speed and surface pressure. The combined effects of the wind intensity and forward speed are noticeably different than their individual effects

    Identification of an optimal method for extracting RNA from human skin biopsy, using domestic pig as a model system

    To evaluate skin tissue gene expression patterns correctly, extracting sufficient quantities of good quality RNA is essential. However, RNA extraction from skin tissue is challenging, as the hyaluronic acid-collagen matrix is extremely difficult to homogenize. Although there are multiple ways to extract RNA from skin, there are no comparative studies that identify the most critical steps, e.g. sample collection, storage and homogenization. We analysed the various steps involved in RNA extraction (i.e. biopsy collection as dry biopsy or into nucleotide stabilizing reagents, different storage conditions, enzymatic digestion, stator-rotor and bead motion-based homogenizing combined with column-based RNA purification). We hypothesised that domestic pig skin is applicable as a model for human skin studies. Altogether twenty different workflows were tested on pig skin and the four most promising workflows were tested on human skin samples. The optimal strategy for extracting human skin RNA was to collect, store and homogenize the sample in RLT lysis buffer from the RNeasy Fibrous Tissue Kit combined with beta-mercaptoethanol. Both stator-rotor and bead motion-based homogenizing were found to result in high quality and quantity of extracted RNA. Our results confirmed that domestic pig skin can be successfully used as a model for human skin RNA studies

    Transcriptional landscape of psoriasis identifies the involvement of IL36 and IL36RN

    Background In present study we performed whole transcriptome analysis in plaque psoriasis patients and compared lesional skin with non-lesional skin and with the skin from healthy controls. We sequenced total RNA from 12 lesional (LP), 12 non-lesional (NLP) and from 12 normal (C) skin biopsies. Results Compared with previous gene expression profiling studies we had three groups under analysis - LP, NLP and C. Using NLP samples allows to see the transcriptome of visually normal skin from psoriasis patient. In LP skin S100A12, S100A7A, LCE3E, DEFB4A, IL19 were found up regulated. In addition to already these well-described genes, we also found several other genes related to psoriasis. Namely, KLK9, OAS2, OAS3, PLA2G, IL36G, IL36RN were found to be significantly and consistently related to the psoriatic lesions and this finding is supported also by previous studies. The genes up-regulated in the LP samples were related to the innate immunity, IL17 and IL10 networks. In NLP samples innate immunity and IL17 network were activated, but activation of IL10 network was not evident. The transcriptional changes characteristic in the NLP samples can be considered as a molecular signature of “dormant psoriasis”. Conclusions Taken together, our study described the transcriptome profile characteristic for LP and NLP psoriatic skin. RNA profile of the NLP skin is in between the lesional and healthy skin, with its own specific pattern. We found that both LP and NLP have up-regulated IL17 network, whereas LP skin has up regulated IL10 related cytokines (IL19, IL20, IL24). Moreover, IL36G and IL36RN were identified as strong regulators of skin pathology in both LP and NLP skin samples, with stronger influence in LP samples

    Educators’ Perceptions of Integrated STEM: A Phenomenological Study

    The study utilized a semistructured interview approach to identify phenomena that are related to integrated STEM education by addressing the question: What are the critical components of an integrated STEM definition and what critical factors are necessary for an integrated STEM definition’s implementation? Thirteen expert practitioners were identified and interviewed. The interviews were transcribed and analyzed for content in three different ways: by person, by interview question, and across all interviews using exploratory data analysis methods. Ten identified phenomena were grouped into two classes: structural implementation phenomena and interpersonal implementation phenomena. The structural implementation phenomena were: subject integration, project-based learning, and design-based education; nontraditional assessment; STEM content; time; professional development; and outside support (from businesses and industry). The interpersonal implementation phenomena include: leadership; collaboration; willingness; authentic, relevant, and meaningful experiences for participants; and outside support (from people in business and industry). The analysis concluded that these phenomena could be considered both critical components and key implementation factors due to their interconnected nature. The data showed that the identified phenomena are necessary as part of an integrated STEM curriculum, which makes them critical components, and that the identified phenomena are critical to create and implement an integrated STEM setting, making them implementation factors as well. Implications for further research include: the possibility of looking at the interconnectedness of the phenomena, examining how each phenomenon contributes to integrated STEM, and measuring current STEM implementations to see if they incorporate the identified phenomena. Additionally, inclusion of an absent phenomenon could be researched to see if integrated STEM education is improved

    Understanding the Effects of Wind Intensity, Forward Speed, Pressure and Track on Generation and Propagation of Hurricane Irma Surges around Florida

    In this study, it is demonstrated that hurricane wind intensity, forward speed, pressure, and track play an important role on the generation and propagation of coastal storm surges. Hurricane Irma, which heavily impacted the entire Florida peninsula in 2017, is used to study the storm surge sensitivity to varying storm characteristics. Results show that the west coast experiences a negative surge due to offshore wind of the approaching storm, but the positive surge returns after the hurricane eye passes over a location and wind became onshore. In the west coast peak, surges are intensified by an increase in onshore wind intensity and forward speed. In the Florida Keys, peak surges are intensified by an increase in wind intensity, a decrease in forward speed and a decrease in pressure. In southeast and east Florida, peak surges are intensified by decrease in pressure, although overall surges are less significant as the water can slide along the coastline. In the recessed coastline of Georgia-Carolinas, maximum surge is elevated by an increase in onshore wind intensity. Shifting the track westward increases peak surges on the west coast, while shifting the track eastward increases peak surge on the east coast. The results demonstrate a new understanding about the sensitivity of surge to varying parametric conditions and the importance of considering changes in the coastline orientation in storm surge predictions

    Thermal stress affects patch time allocation by preventing forgetting in a parasitoid wasp

    Learning and memory allow animals to adjust their foraging strategies through experience. Despite the known impact of temperature on many aspects of the behavioral ecology of animals, memory retention in the face of realistic thermal stress has seldom been assessed. In the laboratory, we studied the behavioral expression of an egg parasitoid's (Trissolcus basalis) memory when exposed to thermal stress that could be encountered in nature. We hypothesized that thermal stress would disrupt memory consolidation and/or modify the optimality of memory retention, thus affecting patch time allocation strategies. Memory consolidation was resilient to 1h of thermal stress following an unrewarded experience (learning) on a patch of host-associated infochemicals. Neither high (40 °C) or low (10 °C) thermal stress changed the intensity of the experienced wasps' behavioral response relative to those held at a moderate temperature (25 °C). Next, we investigated how temperature stress could affect the parasitoids' memory retention ("forgetting"). When kept at a constant moderate temperature after learning, residence times of wasps retested on host cues increased relative to controls (naive wasps) over a period of 4 days as they presumably "forgot." However, both hot and cool daily temperature cycles prevented forgetting; the residence times of retested experienced wasps in these treatments did not change relative to controls over time. We discuss to what extent this may be an adaptive response by the parasitoids versus a physiological constraint imposed by temperature. Our findings contribute to an understanding of the impact of thermal stress on foraging strategies that involve learning and memory

    Tropical sea surface temperatures for the past four centuries reconstructed from coral archives

    Most annually resolved climate reconstructions of the Common Era are based on terrestrial data, making it a challenge to independently assess how recent climate changes have affected the oceans. Here as part of the Past Global Changes Ocean2K project, we present four regionally calibrated and validated reconstructions of sea surface temperatures in the tropics, based on 57 published and publicly archived marine paleoclimate data sets derived exclusively from tropical coral archives. Validation exercises suggest that our reconstructions are interpretable for much of the past 400 years, depending on the availability of paleoclimate data within, and the reconstruction validation statistics for, each target region. Analysis of the trends in the data suggests that the Indian, western Pacific, and western Atlantic Ocean regions were cooling until modern warming began around the 1830s. The early 1800s were an exceptionally cool period in the Indo-Pacific region, likely due to multiple large tropical volcanic eruptions occurring in the early nineteenth century. Decadal-scale variability is a quasi-persistent feature of all basins. Twentieth century warming associated with greenhouse gas emissions is apparent in the Indian, West Pacific, and western Atlantic Oceans, but we find no evidence that either natural or anthropogenic forcings have altered El Ni ˜no–Southern Oscillation-related variance in tropical sea surface temperatures. Our marine-based regional paleoclimate reconstructions serve as benchmarks against which terrestrial reconstructions as well as climate model simulations can be compared and as a basis for studying the processes by which the tropical oceans mediate climate variability and change
