    mtDNA variation among Greenland Eskimos: the edge of the Beringian expansion.

    The Eskimo-Aleut language phylum is distributed from coastal Siberia across Alaska and Canada to Greenland and is well distinguished from the neighboring Na Dene languages. Genetically, however, the distinction between Na Dene and Eskimo-Aleut speakers is less clear. In order to improve the genetic characterization of Eskimos in general and Greenlanders in particular, we have sequenced hypervariable segment I (HVS-I) of the mitochondrial DNA (mtDNA) control region and typed relevant RFLP sites in the mtDNA of 82 Eskimos from Greenland. A comparison of our data with published sequences demonstrates major mtDNA types shared between Na Dene and Eskimo, indicating a common Beringian history within the Holocene. We further confirm the presence of an Eskimospecific mtDNA subgroup characterized by nucleotide position 16265G within mtDNA group A2. This subgroup is found in all Eskimo groups analyzed so far and is estimated to have originated !3,000 years ago. A founder analysis of all Eskimo and Chukchi A2 types indicates that the Siberian and Greenland ancestral mtDNA pools separated around the time when the Neo-Eskimo culture emerged. The Greenland mtDNA types are a subset of the Alaskan mtDNA variation: they lack the groups D2 and D3 found in Siberia and Alaska and are exclusively A2 but at the same time lack the A2 root type. The data are in agreement with the view that the present Greenland Eskimos essentially descend from Alaskan Neo-Eskimos. European mtDNA types are absent in our Eskimo sample

    Impact of Warning Pop-Up Messages on the Gambling Behavior, Craving, and Cognitions of Online Gamblers: A Randomized Controlled Trial

    International audienceBackground: Many features of Internet gambling may impact problem severity, particularly for vulnerable populations (availability, anonymity, a convenience and ease of play, digital forms of payment, and a higher level of immersion). To prevent the risks associated with excessive gambling and to inform gamblers, we need responsible gambling strategies. Gambling-related warning messages are one possible strategy that can help minimizing gambling-related harm. Methods: Our experimental study aimed to evaluate the effectiveness of self-appraisal and informative pop-up messages compared to a control condition (blank pop-up messages), for both at-risk (ARG) and low risk/non-problem Internet gamblers (LR/NPG) according to their favorite type of game, in a semi naturalistic setting and with a 15-day follow-up. During the experimental session, participants were invited to gamble on their favorite website with their own money in the laboratory. Effectiveness was investigated through the impact of pop-ups on gambling behavior (money wagered and time spent), craving, cognitive distortions, and gambling experience, taking into account message recall. We analyzed data from 58 participants, playing preferentially either to skill and chance bank games (sports betting, horse race betting) and skill and chance social games (poker). Results: We observed a significant decrease in the illusion of control for ARG in the informative pop-up condition at the 15-day follow-up. A significant effect of self-appraisal pop-ups compared to blank pop-up messages was also demonstrated only for sport and horse bettors, with a decrease on time spent gambling and an increase of gambling-related expectancies at the follow-up. Finally, we also observed that a majority of the participants were disturbed and irritated by pop-ups during their gambling session. Conclusions: The results of our study demonstrated the limited impact of pop-up warning messages on gambling behavior and cognition in Internet gamblers according to the type of game and the status of gamblers. The limited impact of warning messages on gambling behavior and the inconvenience of the pop-ups for Internet gamblers lead us to only consider warning messages as one piece of a larger responsible gambling strategy.     Impact of Wagering Inducements on the Gambling Behaviors, Cognitions, and Emotions of Online Gamblers: A Randomized Controlled Study

    International audienceWagering inducements are part of loyalty/reward programs implemented by online gambling operators to retain or attract consumers. They constitute incentives to bet that are offered to gamblers provided that they perform certain betting-related activities. They are often considered risk factors for gambling problems, but studies exploring the actual impacts of such incentives are scarce. The objective of the present study was to assess the actual impact of wagering inducements on gambling behaviors, cognitions, and emotions of online gamblers. One hundred seventy-one adults (18-65 years old) who gamble on a regular basis on the Internet, including at-risk and recreational gamblers, were recruited through media announcements and in panels from survey institutes. Participants were randomly assigned to one of four experimental conditions, in which a defined amount of money was given to the gambler with a bank e-card system during an experimental gambling session to simulate a wagering inducement (€10, €50, €100, or €200), or the control condition, in which no incentive was given. The experimental gambling session was designed to be as natural as possible (participants gambled with their own gambling account and their own money). Participants completed a pretest interview, took part in the experimental gambling session, and then completed a post-test interview. The impact of wagering inducements was estimated on objective (money wagered and time spent gambling during the gambling session) and subjective (cognitive distortions, enjoyment of gambling, loss of control, and respect of usual gambling habits) gambling endpoints that were compared between conditions. Two-thirds of participants reported having already received wagering inducements at some point of their gambling course. Although no effect was demonstrated on time spent gambling, inducements increased money wagered, gambling-related expectancies and perceived loss of control. In particular, it seems that wagering inducements could lead to extreme expenses, especially for at-risk gamblers. This research suggests that regulating wagering inducements could be helpful for prevention and early intervention. Future research on the impacts of wagering inducements is still needed, especially more ecological studies based on behavioral tracking data and studies assessing the differential impacts of various incentive types. We provide here a summary of 70 years of data collection including registered lines and subjects by the end of 2012.METHODS:Affected individuals were identified from a national register of hereditary eye diseases at the National Eye Clinic (NEC), a tertiary low vision rehabilitation center for the entire Danish population. The assembling of LHON pedigrees was based on the reconstruction of published families and newly diagnosed cases from 1980 to 2012 identified in the files of NEC. Genealogic follow-up on the maternal ancestry of all affected individuals was performed to identify a possible relation to an already known maternal line. A full genotypic characterization of the nation-based LHON cohort is provided.RESULTS:Forty different lines were identified. The number of live affected individuals with a verified mitochondrial DNA mutation was 104 on January 1, 2013, which translates to a prevalence rate of 1:54,000 in the Danish population.CONCLUSIONS:Haplogroup distribution as well as mutational spectrum of the Danish LHON cohort do not deviate from those of other European populations. The genealogic follow-up reveals a relatively high turnover among families with approximately 15 newly affected families per century and the dying out of earlier maternal lines

    Practices in research, surveillance and control of neglected tropical diseases by One Health approaches: A survey targeting scientists from French-speaking countries

    International audienceOne health (OH) approaches have increasingly been used in the last decade in the fight against zoonotic neglected tropical diseases (NTDs). However, descriptions of such collaborations between the human, animal and environmental health sectors are still limited for French-speaking tropical countries. The objective of the current survey was to explore the diversity of OH experiences applied to research, surveillance and control of NTDs by scientists from French-speaking countries, and discuss their constraints and benefits. Six zoonotic NTDs were targeted: echinococcoses, trypanosomiases, leishmaniases, rabies, Taenia solium cysticercosis and leptospiroses. Invitations to fill in an online questionnaire were sent to members of francophone networks on NTDs and other tropical diseases. Results from the questionnaire were discussed during an international workshop in October 2019. The vast majority (98%) of the 171 respondents considered OH approaches relevant although only 64% had implemented them. Among respondents with OH experience, 58% had encountered difficulties mainly related to a lack of knowledge, interest and support for OH approaches by funding agencies, policy-makers, communities and researchers. Silos between disciplines and health sectors were still strong at both scientific and operational levels. Benefits were reported by 94% of respondents with OH experience, including increased intellectual stimulation, stronger collaborations, higher impact and cost-efficiency of interventions. Recommendations for OH uptake included advocacy, capacity-building, dedicated funding, and higher communities’ involvement. Improved research coordination by NTD networks, production of combined human-animal health NTD impact indicators, and transversal research projects on diagnostic and reservoirs were also considered essential
