The coordination of ruthenium(II) complexes to anionic oxygen-based donors are very rare. This study describes a simple, one-pot method for obtaining [ruthenium(II)(trithiacyclononane)(curcumin)(S-DMSO)]Cl (1) in 37% yield. The structural characterization of complex 1 by elemental analysis, FT-IR, 1-D and 2-D NMR, ESI+-MS as well as UV–vis and fluorescence spectroscopies are presented. The DNA-melting temperature (Tm) assay shows that salmon sperm DNA (smDNA) in the presence of complex 1 has a higher melting temperature, with ΔTm = 7.4 °C, while in the presence of curcumin the melting temperature remains unaltered. The in vitro cytotoxic activities of curcumin and complex 1 were investigated using the tumor human prostate cell line, PC-3, and the healthy cell line, PNT-2. Complex 1 is innocuous toward normal prostate epithelial cells and, whereas curcumin is toxic, with inhibition rates of ca. 35 and 65% at 50 and 80 μM, respectively. On the tumor cell line PC-3, complex 1 did not cause viability changes, whereas curcumin exhibited dose-dependent inhibition, with ca. 73% inhibition at the highest concentration tested, i.e. 80 μM. This study suggests that coordination with the trithiacyclononane ruthenium(II) scaffold stabilizes the photochemical properties of curcumin and strongly changes its biologic activity.publishe

A species list of amphibians from Santa Isabel do Rio Negro in Brazilian Amazonia is provided. Collections were made from March–April 2012 along each of two 3-km trails with the following sampling methods: (1) pitfall traps with drift fences; (2) visual and auditory surveys; and (3) chance encounters. The trail at Daraá is north of the Rio Negro, whereas the other in Ayuanã is south of the river. Forty species of anurans and one salamander species representing 20 genera and nine families were recorded. The species composition was compared with those of 16 other studies conducted in the Guiana, Imeri, and Jaú areas of endemism, where species richness varies from 21–63, and similarity indices range from 23–100%. The anuran fauna at our sites resembles that of Flota Faro in eastern Amazonia more than it does that of the nearest site in the Departamento del Guainía of Colombia. The index of similarity is extremely variable between sites of the same and distinct areas of endemism. This pattern also was observed in the cluster analysis. As expected, geographically close areas have similar faunal compositions. However, the anuran fauna of Parque Nacional do Jaú (Jaú area of endemism) resembles that of Manaus (Guiana area of endemism) more closely than it does that of the Ayuanã River, which belongs to the same area of endemism as Parque Nacional do Jaú. The limits of the areas of endemism are better defined by the presence / absence of other terrestrial vertebrates, such as birds and mammals, than by the assemblage of amphibians and squamate reptiles

Secreted extracellular vesicle molecular cargo as a novel liquid biopsy diagnostics of central nervous system diseases

Secreted extracellular vesicles (EVs) are heterogeneous cell-derived membranous granules which carry a large diversity of molecules and participate in intercellular communication by transferring these molecules to target cells by endocytosis. In the last decade, EVs role in several pathological conditions, from etiology to disease progression or therapy evasion, has been consolidated, including in central nervous system (CNS)-related disorders. For this review, we performed a systematic search of original works published, reporting the presence of molecular components expressed in the CNS via EVs, which have been purified from plasma, serum or cerebrospinal fluid. Our aim is to provide a list of molecular EV components that have been identified from both nonpathological conditions and the most common CNS-related disorders. We discuss the methods used to isolate and enrich EVs from specific CNS-cells and the relevance of its components in each disease context. This research was funded by the MindGaP-H2020-FETOPEN-2018-2020, Grant agreement ID: 829040. The respiratory protection equipment (RPE) used by health professionals consists of an essential device to prevent infectious diseases, especially those caused by biological agents such as the coronavirus (SARS-CoV-2). The current epidemiological panorama is worrying, and the context of creation and production of the mask has emerged as an alternative to RPE to face the public health crisis worldwide. The aim of this work is to present a low-cost alternative as an FFP2-like filter for a reusable respirator face mask. This study presents the comparison of different cellulose-based filtering materials performed by retention testing, time saturation testing, aerosol penetration testing, nanoparticle (~140 nm) filtration testing, bacterial filtration efficiency (BFE), analysis of material morphology and usability. The reusable respirator face mask used in this study is an open-source innovation, using 3D printing. Cotton disc proved to be the best filter material for the reusable mask, with satisfactory results and a performance similar to that shown by the N95-type mask. The cotton disc ensured effectiveness over 6 h of use, and after that, the reusable respirator face mask (here, Delfi-TRON®) needed to be sanitized and replenished with a new cotton disc. Upon preliminary analyses of filtration efficiency, the selected filter was shown to be a low-cost biodegradable and biocompatible alternative.info:eu-repo/semantics/publishedVersio
