Profiles and technological requirements of urogenital probiotics
Probiotics, defined as live microorganisms that, when administered in adequate amounts, confer a health benefit on the host, are considered a valid and novel alternative for the prevention and treatment of female urogenital tract infections. Lactobacilli, the predominant microorganisms of the healthy human vaginal microbiome, can be included as active pharmaceutical ingredients in probiotics products. Several requirements must be considered or criteria fulfilled during the development of a probiotic product or formula for the female urogenital tract. This review deals with the main selection criteria for urogenital probiotic microorganisms: host specificity, potential beneficial properties, functional specifications, technological characteristics and clinical trials used to test their effect on certain physiological and pathological conditions. Further studies are required to complement the current knowledge and support the clinical applications of probiotics in the urogenital tract. This therapy will allow the restoration of the ecological equilibrium of the urogenital tract microbiome as well as the recovery of the sexual and reproductive health of women.Fil: Nader, Maria Elena Fatima. Microbial production of beneficial indoleamines (serotonin and melatonin) with potential application to biotechnological products for human health
Micro-organisms have showed the ability to produce biologically active compounds associated with neurotransmission in higher organisms. In particular, serotonin- and melatonin-producing microbes are valuable sources for the development of eco-friendly bioproducts. Serotonin and melatonin are indoleamines that have received special attention due to their positive effects on human health. These biomolecules exert a critical role in several physiological or pathological processes, including some mental and neurological disorders. This article includes a review of the microbial production of serotonin and melatonin, their functions in micro-organisms and their potential uses as therapeutic and/or preventive agents to improve human health. A description of the quantification methods employed to detect indoleamines and the evidence found concerning their microbial production at laboratory and industrial scale—for application in biotechnological products—is also provided. The microbial ability to synthesize beneficial indoleamines should be further studied and harnessed, to allow the development of sustainable bioprocesses to produce foods and pharmaceuticals for human health.Fil: Danilovich, Mariana Elizabeth.
Ibuprofen sensitive, Sodium Monocarboxylate Transporter (SMCT1) is expressed in the apical membrane of diverse epithelia. Its physiological role in the thyroid remains however unknown, but it has been proposed that SMCT1 could act as an apical iodide transporter required for the main function of the gland: the thyroid hormone synthesis.
We previously reported that SMCT1 exhibit a [Na+]e sensible anionic leak current that could account for the electrogenic transport of iodide. However, an iodine efflux sensitive to ibuprofen but independent of [Na+]e, was also observed in primary cultures of porcine thyrocytes, suggesting another mechanism of iodine transport mediated by SMCT1. This work aims to understand the characteristics of this type of transport using Xenopus laevis oocytes as an SMCT1 expression system. By realising 125I uptakes, we found that SMCT1 transports iodide in an ibuprofen sensitive manner (30nmol/oocyte/h). While nonsaturable uptake iodide kinetics were observed, SMCT1 iodide transport was Na+ dependent as shown by the transport reduction when the [Na+]e is replaced by NMDG. The possibility of artifacts, such as non specific binding and the overexpression of endogenous proteins, was analysed. By observing that the vast majority of the radioactive iodide is found in the soluble portion of the oocyte, we excluded non-specific binding of iodide to the cell membrane. However, it is believed that most of the iodide entering the cell is not free and must be bound to some intracellular proteins. Indeed, there is a significant reduction of SMCT1-mediated iodide transport when 2% BSA is present at the extracellular solution. Furthermore, the lack of iodide transport when overexpressing other proteins than SMCT1, precludes the possibility of an overexpression of endogenous transport proteins like chloride channels for example. In fact, the transport of iodide appears to be specific to the expression of SMCT1 and interestingly of another monocarboxylate transporter MCT1. The analysis of all trials, including the fact that the amplitude of the observed transport is 20 times larger than the leak current lead us to propose that SMCT1 can carry iodide in an electroneutral manner. However, the mechanism by which this is accomplished is not easy to identify and future experiments will be necessary to determine whether this transport is observed in other SMCT1 expression systems
Encapsulation and subsequent freeze-drying of Lactobacillus reuteri CRL 1324 for its potential inclusion in vaginal probiotic formulations
Probiotic formulations must include a high number of viable and active microorganisms. In this work, the survival of human vaginal Lactobacillus reuteri CRL 1324 during encapsulation, lyophilization and storage, and the activity of encapsulated and/or freeze-dried bacterial cells were evaluated. Extrusion-ionic gelation technique was applied to encapsulate L. reuteri CRL 1324, using xanthan and gellan. Encapsulated and free bacterial cells were freeze-dried with or without lactose and skim milk as lyoprotectors. The different systems obtained were stored at room temperature and at 4°C for 150 days. The following determinations were performed: L. reuteri CRL 1324 viability, microorganism released from capsules, survival in a medium simulating the vaginal fluid and maintenance of beneficial properties (growth inhibition of opportunistic pathogenic Streptococcus agalactiae NH 17 and biofilm formation). L. reuteri CRL 1324 encapsulation was efficient, allowing the recovery of a high number of entrapped lactobacilli. The survival of encapsulated L. reuteri during lyophilization and storage was significantly higher in the presence of lyoprotectors. At the end of storage, highest numbers of viable cells were obtained in free or encapsulated cells freeze-dried with lyoprotectors, stored at 4°C. Encapsulated and/or liophilized L. reuteri cells maintained their viability in simulated vaginal fluid as well as the ability to inhibit S. agalactiae NH 17 growth and to form biofilm. Encapsulated and freeze-dried L. reuteri CRL 1324 can be included in a suitable pharmaceutical form for vaginal application to prevent or treat urogenital infections in women.Fil: Juárez Tomás, MarĂa Silvina.
