Correcting a Nonparametric Two-sample Graph Hypothesis Test for Graphs with Different Numbers of Vertices
Random graphs are statistical models that have many applications, ranging
from neuroscience to social network analysis. Of particular interest in some
applications is the problem of testing two random graphs for equality of
generating distributions. Tang et al. (2017) propose a test for this setting.
This test consists of embedding the graph into a low-dimensional space via the
adjacency spectral embedding (ASE) and subsequently using a kernel two-sample
test based on the maximum mean discrepancy. However, if the two graphs being
compared have an unequal number of vertices, the test of Tang et al. (2017) may
not be valid. We demonstrate the intuition behind this invalidity and propose a
correction that makes any subsequent kernel- or distance-based test valid. Our
method relies on sampling based on the asymptotic distribution for the ASE. We
call these altered embeddings the corrected adjacency spectral embeddings
(CASE). We show that CASE remedies the exchangeability problem of the original
test and demonstrate the validity and consistency of the test that uses CASE
via a simulation study. We also apply our proposed test to the problem of
determining equivalence of generating distributions of subgraphs in Drosophila
connectome
Upfront dilution of ferritin samples to reduce hook effect, improve turnaround time and reduce costs
Introduction: Sandwich immunoassays offer advantages in the clinical laboratory but can yield erroneously low results due to hook (prozone)
effect, especially with analytes whose concentrations span several orders of magnitude such as ferritin. This study investigated a new approach to
reduce the likelihood of hook effect in ferritin immunoassays by performing upfront, five-fold dilutions of all samples for ferritin analysis. The impact
of this change on turnaround time and costs were also investigated.
Materials and methods: Ferritin concentrations were analysed in routine clinical practice with and without upfront dilutions on Siemens Centaur®
XP (Siemens Healthineers, Erlang, Germany) immunoanalysers. In addition, one month of baseline data (1026 results) were collected prior to implementing
upfront dilutions and one month of data (1033 results) were collected after implementation.
Results: Without upfront dilutions, hook effect was observed in samples with ferritin concentrations as low as 86,028 μg/L. With upfront dilutions,
samples with ferritin concentrations as high as 126,050 μg/L yielded values greater than the measurement interval and would have been diluted
until an accurate value was obtained. The implementation of upfront dilution of ferritin samples led to a decrease in turnaround time from a median
of 2 hours and 3 minutes to 1 hour and 18 minutes (P = 0.002).
Conclusions: Implementation of upfront dilutions of all ferritin samples reduced the possibility of hook effect, improved turnaround time and saved
the cost of performing additional dilutions
Normal Aging does Not Impair Orbitofrontal-Dependent Reinforcer Devaluation Effects
Normal aging is associated with deficits in cognitive flexibility thought to depend on prefrontal regions such as the orbitofrontal cortex (OFC). Here, we used Pavlovian reinforcer devaluation to test whether normal aging might also affect the ability to use outcome expectancies to guide appropriate behavioral responding, which is also known to depend on the OFC. Both young and aged rats were trained to associate a 10-s conditioned stimulus (CS+) with delivery of a sucrose pellet. After training, half of the rats in each age group received the sucrose pellets paired with illness induced by LiCl injections; the remaining rats received sucrose and illness explicitly unpaired. Subsequently, responding to the CS+ was assessed in an extinction probe test. Although aged rats displayed lower responding levels overall, both young and aged rats conditioned to the CS+ and developed a conditioned taste aversion following reinforcer devaluation. Furthermore, during the extinction probe test, both young and aged rats spontaneously attenuated conditioned responding to the cue as a result of reinforcer devaluation. These data show that normal aging does not affect the ability to use expected outcome value to appropriately guide Pavlovian responding. This result indicates that deficits in cognitive flexibility are dissociable from other known functions of prefrontal – and particularly orbitofrontal – cortex
Single or Double Degenerate Progenitors? Searching for Shock Emission in the SDSS-II Type Ia Supernovae
From the set of nearly 500 spectroscopically confirmed type~Ia supernovae and
around 10,000 unconfirmed candidates from SDSS-II, we select a subset of 108
confirmed SNe Ia with well-observed early-time light curves to search for
signatures from shock interaction of the supernova with a companion star. No
evidence for shock emission is seen; however, the cadence and photometric noise
could hide a weak shock signal. We simulate shocked light curves using SN Ia
templates and a simple, Gaussian shock model to emulate the noise properties of
the SDSS-II sample and estimate the detectability of the shock interaction
signal as a function of shock amplitude, shock width, and shock fraction. We
find no direct evidence for shock interaction in the rest-frame -band, but
place an upper limit on the shock amplitude at 9% of supernova peak flux ( mag). If the single degenerate channel dominates type~Ia progenitors,
this result constrains the companion stars to be less than about 6
on the main sequence, and strongly disfavors red giant companions.Comment: 28 pages, 3 figure
Perceptual size discrimination requires awareness and late visual areas: A continuous flash suppression and interocular transfer study
We applied continuous flash suppression (CFS) during an interocular transfer paradigm to evaluate the importance of awareness and the contribution of early versus late visual structures in size recognition. Specifically, we tested if size judgements of a visible target could be influenced by a congruent or incongruent prime presented to the same or different eye. Without CFS, participants categorised a target as “small” or “large” more quickly when it was preceded by a congruent prime – regardless of whether the prime and target were presented to the same or different eye. Interocular transfer enabled us to infer that the observed priming was mediated by late visual areas. In contrast, there was no priming under CFS, which underscores the importance of awareness. We conclude that awareness and late visual structures are important for size perception and that any subconscious processing of the stimulus has minimal effect on size recognition
Impaired cholecystokinin-induced gallbladder emptying incriminated in spontaneous “black” pigment gallstone formation in germfree Swiss Webster mice
“Black” pigment gallstones form in sterile gallbladder bile in the presence of excess bilirubin conjugates (“hyperbilirubinbilia”) from ineffective erythropoiesis, hemolysis, or induced enterohepatic cycling (EHC) of unconjugated bilirubin. Impaired gallbladder motility is a less well-studied risk factor. We evaluated the spontaneous occurrence of gallstones in adult germfree (GF) and conventionally housed specific pathogen-free (SPF) Swiss Webster (SW) mice. GF SW mice were more likely to have gallstones than SPF SW mice, with 75% and 23% prevalence, respectively. In GF SW mice, gallstones were observed predominately in heavier, older females. Gallbladders of GF SW mice were markedly enlarged, contained sterile black gallstones composed of calcium bilirubinate and <1% cholesterol, and had low-grade inflammation, edema, and epithelial hyperplasia. Hemograms were normal, but serum cholesterol was elevated in GF compared with SPF SW mice, and serum glucose levels were positively related to increasing age. Aged GF and SPF SW mice had deficits in gallbladder smooth muscle activity. In response to cholecystokinin (CCK), gallbladders of fasted GF SW mice showed impaired emptying (females: 29%; males: 1% emptying), whereas SPF SW females and males emptied 89% and 53% of volume, respectively. Bilirubin secretion rates of GF SW mice were not greater than SPF SW mice, repudiating an induced EHC. Gallstones likely developed in GF SW mice because of gallbladder hypomotility, enabled by features of GF physiology, including decreased intestinal CCK concentration and delayed intestinal transit, as well as an apparent genetic predisposition of the SW stock. The Subluminous Supernova 2007qd: A Missing Link in a Family of Low-Luminosity Type Ia Supernovae
We present multi-band photometry and multi-epoch spectroscopy of the peculiar
Type Ia supernova (SN Ia) 2007qd, discovered by the SDSS-II Supernova Survey.
It possesses physical properties intermediate to those of the peculiar SN
2002cx and the extremely low-luminosity SN 2008ha. Optical photometry indicates
that it had an extraordinarily fast rise time of <= 10 days and a peak absolute
B magnitude of -15.4 +/- 0.2 at most, making it one of the most subluminous SN
Ia ever observed. Follow-up spectroscopy of SN 2007qd near maximum brightness
unambiguously shows the presence of intermediate-mass elements which are likely
caused by carbon/oxygen nuclear burning. Near maximum brightness, SN 2007qd had
a photospheric velocity of only 2800 km/s, similar to that of SN 2008ha but
about 4000 and 7000 km/s less than that of SN 2002cx and normal SN Ia,
respectively. We show that the peak luminosities of SN 2002cx-like objects are
highly correlated with both their light-curve stretch and photospheric
velocities. Its strong apparent connection to other SN 2002cx-like events
suggests that SN 2007qd is also a pure deflagration of a white dwarf, although
other mechanisms cannot be ruled out. It may be a critical link between SN
2008ha and the other members of the SN 2002cx-like class of objects.Comment: To be published in the Astrophysical Journal; 37 pages, 13 figures, 4
