BACKGROUND: intrinsic capacity (IC) is a construct encompassing people\u27s physical and mental abilities. There is an implicit link amongst IC domains: cognition, locomotion, nutrition, sensory and psychological. However, little is known about the integration of the domains. OBJECTIVES: to investigate patterns in the presentation and evolution of IC domain impairments in low-and-middle-income countries and if such patterns were associated with adverse outcomes. METHODS: secondary analyses of the first two waves of the 10/66 study (population-based surveys conducted in eight urban and four rural catchment areas in Cuba, Dominican Republic, Puerto Rico, Venezuela, Peru, Mexico and China). We applied latent transition analysis on IC to find latent statuses (latent clusters) of IC domain impairments. We evaluated the longitudinal association of the latent statuses with the risk of frailty, disability and mortality, and tested concurrent and predictive validity. RESULTS: amongst 14,923 participants included, the four latent statuses were: high IC (43%), low deterioration with impaired locomotion (17%), high deterioration without cognitive impairment (22%), and high deterioration with cognitive impairment (18%). A total of 61% of the participants worsened over time, 35% were stable, and 3% improved to a healthier status.Participants with deteriorated IC had a significantly higher risk of frailty, disability and dementia than people with high IC. There was strong concurrent and predictive validity. (Mortality Hazard Ratio = 4.60, 95%CI 4.16; 5.09; Harrel\u27s C = 0.73 (95%CI 0.72;0.74)). CONCLUSIONS: half of the study population had high IC at baseline, and most participants followed a worsening trend. Four qualitatively different IC statuses or statuses were characterised by low and high levels of deterioration associated with their risk of disability and frailty. Locomotion and cognition impairments showed other trends than psychological and nutrition domains across the latent statuses

The effect of Voisin restricted grazing and portion grazing upon indexes determining load capacity of Brachiaria humidicola cv. CIAT 679 without watering and fertilizing was evaluated for 4 years on a Vertisol soil in Cauto valley. 40 cows 5/8 Holstein- 3/8 Zebu were sampled by a completely randomized design. Grazing was done “in line” using cattle yards with the same size for Voisin restricted grazing, and grazing strips for portion grazing in which each herd fed daily out of a strip according to its avail- ability in order to increase maximum grazing consumption. Strip management in portion grazing significantly increased grazing efficiency during both seasons compared to Voisin restricted grazing, and had a much better impact upon grazing consumption as well. Neither yield indexes nor rest time were affected; they changed according to season with a marked depletion during dry season. Annual yield average was 11,7 t dry matter ha-1, out of which only a 23% was obtained during dry season. A higher graz- ing consumption index in portion grazing showed a 24 UGM ha-1 increase in load capacity due to rotation during rainy season compared to Voisin restricted grazing (242 vs. 218). Results clearly showed a marked effect of season and grazing consumption upon portion grazing.

Enabling high-quality transparent conductive oxide on 3D printed ZrO2 architectures through atomic layer deposition

The conformal atomic layer deposition of a transparent conductive oxide composed of Al-doped ZnO (AZO) over three-dimensional (3D) shaped ZrO2 microarchitectures produced using two-photon lithography (TPL) is reported here for the first time. The effect of ZrO2 morphology, surface roughness, and crystallographic phase (tetragonal and monoclinic) on the quality and properties of the deposited ZnO and AZO thin films is investigated. No discontinuities, domains, or areas differing from the desired chemical composition have been found in films grown over the 3D structures. Three different Al dopant concentrations (4.0 %, 4-5 %, and 5.0 % Al doping cycles) are examined and compared to undoped ZnO. AZO and ZnO optical and electrical properties are studied using cathodoluminescence (CL) and Hall effect measurements. The CL study confirms that the observed emissions from the ZnO and AZO films are associated with the near band emission of ZnO and defects, i.e., zinc and oxygen vacancies and interstitial oxygen. The AZO films exhibit n-type semiconductor behavior, and a minimum resistivity of 1.2 x 10-3 Ω cm is achieved. From a broad perspective, AZO deposition on 3D microarchitectures opens a new route towards dimensionally refined optoelectronic devices in which the ZrO2/AZO can serve a key enabling role for the production of electrodes

Experimental practices handle data that cause an error in comparison with the theoretical data, however, the use of numerical methods in the resolution of experimental guides limits the error that can be caused from using few points to determine a slope, in the case of analyzing two variables with direct proportionality, for example. This article seeks to guide the calculations towards a better understanding and approximation of the results to conclude and deduce the physical laws within the experimental guides, which are part of didactic-pedagogical processes in the teaching-learning of Physics Experimental. Using a high-resolution antiparallel double-crystal x-ray spectrometer, we obtained the line shapes, that is, asymmetry index and natural linewidths. We found that the corrected full width at half maximum of the Kα1 and Kα2 lines as a function of Z is in good agreement with the data in the literature. Furthermore, satellite lines arising from shake-off appear in the low-energy side of the Kα1 and Kα2 lines in Se but, in Y and Zr, it was very difficult to identify the contribution of the shake process to the overall lines. The Kβ1,3 natural linewidth of these elements was also corrected using the appropriate instrumental function for this type of x-ray spectrometer, and the spin doublet energies were obtained from the peak positions. The corrected full width at half maximum (FWHM) of the Kβ1 x-ray lines increases linearly with Z, but this tendency was found to be, in general, not linear for Kβ3 x-ray lines. This behavior may be due to the existence of satellite lines originated from shake processes. Simulated line profiles, obtained using the multiconfiguration Dirac-Fock formalism, accounting for radiative and radiationless transitions and shake-off processes, show a very good agreement with the high-resolution experimental spectra.publishersversionpublishe

Tumor regression and resistance mechanisms upon CDK4 and RAF1 inactivation in KRAS/P53 mutant lung adenocarcinomas.

KRAS mutant lung adenocarcinomas remain intractable for targeted therapies. Genetic interrogation of KRAS downstream effectors, including the MAPK pathway and the interphase CDKs, identified CDK4 and RAF1 as the only targets whose genetic inactivation induces therapeutic responses without causing unacceptable toxicities. Concomitant CDK4 inactivation and RAF1 ablation prevented tumor progression and induced complete regression in 25% of KRAS/p53-driven advanced lung tumors, yet a significant percentage of those tumors that underwent partial regression retained a population of CDK4/RAF1-resistant cells. Characterization of these cells revealed two independent resistance mechanisms implicating hypermethylation of several tumor suppressors and increased PI3K activity. Importantly, these CDK4/RAF1-resistant cells can be pharmacologically controlled. These studies open the door to new therapeutic strategies to treat KRAS mutant lung cancer, including resistant tumors. We thank S. Ortega for the generation of the Cdk4FxKD mouse model; and M. Evaluation of dose-dependent treatment effects after mid-trial dose escalation in biomarker, clinical, and cognitive outcomes for gantenerumab or solanezumab in dominantly inherited Alzheimer\u27s disease

Get PDF
INTRODUCTION: While the Dominantly Inherited Alzheimer Network Trials Unit (DIAN-TU) was ongoing, external data suggested higher doses were needed to achieve targeted effects; therefore, doses of gantenerumab were increased 5-fold, and solanezumab was increased 4-fold. We evaluated to what extent mid-trial dose increases produced a dose-dependent treatment effect. METHODS: Using generalized linear mixed effects (LME) models, we estimated the annual low- and high-dose treatment effects in clinical, cognitive, and biomarker outcomes. RESULTS: Both gantenerumab and solanezumab demonstrated dose-dependent treatment effects (significant for gantenerumab, non-significant for solanezumab) in their respective target amyloid biomarkers (Pittsburgh compound B positron emission tomography standardized uptake value ratio and cerebrospinal fluid amyloid beta 42), with gantenerumab demonstrating additional treatment effects in some downstream biomarkers. No dose-dependent treatment effects were observed in clinical or cognitive outcomes. CONCLUSIONS: Mid-trial dose escalation can be implemented as a remedy for an insufficient initial dose and can be more cost effective and less burdensome to participants than starting a new trial with higher doses, especially in rare diseases. HIGHLIGHTS: We evaluated the dose-dependent treatment effect of two different amyloid-specific immunotherapies.Dose-dependent treatment effects were observed in some biomarkers. No dose-dependent treatment effects were observed in clinical/cognitive outcomes, potentially due to the fact that the modified study may not have been powered to detect such treatment effects in symptomatic subjects at a mild stage of disease exposed to high (or maximal) doses of medication for prolonged durations
