    Gas-phase hydrodechlorination of mixtures of chloromethanes with activated carbon-supported platinum catalysts

    Platinum catalysts supported on activated carbon (Pt/C) at different metal loadings (0.5-2% Pt) have been tested in the gas-phase hydrodechlorination (HDC) of mixtures of dichloromethane (DCM) and chloroform (TCM), with a total feed concentration of 1000ppmv. Almost complete dechlorination was achieved at 250°C, 1kghmol-1 space time and a H2/CM molar ratio of 25 with the 2% Pt catalyst. At a reaction temperature of 250°C, scarce inhibition in the conversion of both compounds was observed compared to the HDC of the individual chloromethanes. The TOF values for DCM increase with the Pt content, which can be attributed to the higher proportion of the zero-valent species. The selectivity to reaction products suggests a parallel reactions scheme. The catalysts showed a high stability, demonstrated by the unchanged conversion of both chloromethanes upon time on stream in long-term experiments carried out at up to 26h.The authors gratefully acknowledge financial support from the Spanish Ministerio de Economía y Competitividad (MINECO) through the project CTM2011-2835

    Effect of the Pt–Pd molar ratio in bimetallic catalysts supported on sulfated zirconia on the gas-phase hydrodechlorination of chloromethanes

    Bimetallic Pt:Pd catalysts with different molar ratios and 0.5 wt.% overall metal load supported on sulfated zirconia catalysts were synthesized and tested in the gas-phase hydrodechlorination (HDC) of chloromethanes and their mixtures. The catalysts were characterized by adsorption–desorption of N2 at −196 °C, X-ray diffraction, X-ray photoelectronic spectroscopy, temperature-programmed reduction, and aberration-corrected scanning transmission electron microscopy (STEM). The effect of the Pt:Pd molar ratio on the activity, stability, and selectivity was analyzed. The high acidity of the sulfated zirconia results in metal particles of small size (mainly <5 nm), as confirmed by STEM. The bimetallic catalysts showed higher stability than the monometallic ones, as demonstrated in long-term experiments (80 h on stream), confirming the positive effect of combining the two metallic phases. Turnover frequency (TOF) values in the range 0.0007–0.0168 s−1 and apparent activation energies between ≈41 and 44 kJ·mol−1 were obtained. TOF values for dichloromethane HDC increased with increasing mean metal particle size within the range of this work (≈1.2–2.3 nm).     Delayed Corneal Epithelial Healing after Intravitreal Bevacizumab: A Clinical and Experimental Study

    Purpose: To report corneal epithelial defects (CEDs) and delayed epithelial healing after intravitreal bevacizumab (IVB) injection and to describe delayed corneal epithelial healing with topical administration of bevacizumab in an experimental rabbit model. Methods: A retrospective chart review was performed on 850 eyes of 850 patients with neovascular eye disease and diabetic macular edema who had received 1.25 to 2.5 mg IVB. In the experimental arm of the study, photorefractive keratectomy was used to create a 3 mm CED in the right eyes of 18 New Zealand rabbits which were then randomized to three equal groups. All rabbits received topical antibiotics, additionally those in group A received topical bevacizumab and animals in group B were treated with topical corticosteroids. The rate of epithelial healing was assessed at different time points using slitlamp photography. Results: In the clinical study, seven eyes of seven subjects developed CEDs the day after IVB injection. All of these eyes had preexisting corneal edema. The healing period ranged from 3 to 38 days (average 11 days) despite appropriate medical management. In the experimental study, topical bevacizumab and corticosteroids both significantly hindered corneal epithelial healing at 12 and 24 hours. Conclusion: Bevacizumab was demonstrated to cause CEDs in clinical settings. Moreover, corneal epithelial healing was delayed by topical application of bevacizumab, in the experimental model. These short-term results suggest that corneal edema may be considered as a risk factor for epithelial defects after IVB

    Ivermectin reduces in vivo coronavirus infection in a mouse experimental model

    SARS-CoV2 is a single strand RNA virus member of the type 2 coronavirus family, responsible for causing COVID-19 disease in humans. The objective of this study was to test the ivermectin drug in a murine model of coronavirus infection using a type 2 family RNA coronavirus similar to SARS-CoV2, the mouse hepatitis virus (MHV). BALB/cJ female mice were infected with 6,000 PFU of MHV-A59 (Group Infected; n=20) and immediately treated with one single dose of 500 μg/kg of ivermectin (Group Infected + IVM; n=20), or were not infected and treated with PBS (Control group; n=16). Five days after infection/treatment, mice were euthanized to obtain different tissues to check general health status and infection levels. Overall results demonstrated that viral infection induces the typical MHV disease in infected animals, with livers showing severe hepatocellular necrosis surrounded by a severe lymphoplasmacytic inflammatory infiltration associated with a high hepatic viral load (52,158 AU), while ivermectin administration showed a better health status with lower viral load (23,192 AU; p<0.05) and few livers with histopathological damage (p<0.05), not showing statistical differences with control mice (P=NS). Furthermore, serum transaminase levels (aspartate aminotransferase and alanine aminotransferase) were significantly lower in treated mice compared to infected animals. In conclusion, ivermectin seems to be effective to diminish MHV viral load and disease in mice, being a useful model for further understanding new therapies against coronavirus diseases.Fil: Arevalo, A.     Relación del perfil salival con el grado de inmunosupresión en pacientes infectados con VIH con y sin tratamiento antirretroviral

    The aim of this study was to evaluate the salivary profile of HIV-infected patients with and without Highly Active Antiretroviral Treatment (HAART), evaluating the Symptoms of salivary hypofunction (Xerostomia, Dysgeusia, Dysphagia, and Susceptibility to develop ulcers oral), the Unstimulated Salivary Flow Rate, Salivary pH and Salivary Viscosity, establishing their relationship with the degree of immunosuppression as assessed by the Counting of lymphocyte CD4/μl. We evaluated 126 patients with HIV infection with HAART and without HAART who were attended at the Health Strategy for Prevention and Control of STI, HIV and AIDS at the Hospital Nacional Dos de Mayo, Lima- Peru. Patients without HAART had significantly higher rates of some salivary hypofunction Symptoms: Xerostomia , Dysgeusia Dysphagia) and Susceptibility to develop oral ulcers and significantly higher values of Salivary viscosity but values were also significantly lower of Unstimulated Salivary Flow Rate and Salivary pH compared to patients with HAART. Furthermore, patients with a greater degree of immunosuppression had significantly higher rates of Symptoms of salivary hypofunction and Salivary Viscosit and significantly lower values of Unstimulated Salivary Flow Rate and Salivary pH ,compared to patients with a lower degree of immunosuppression. It was concluded that there was an alteration of the salivary profile of patients with HIV infection, which is significantly higher in patients without HAART and those with a greater degree of immunosuppression.El objetivo del estudio fue evaluar el perfil salival de pacientes con infección por VIH con y sin Tratamiento Antirretroviral de Gran Actividad (TARGA), por medio de la evaluación de los síntomas de hipofunción salival (Xerostomía, Disgeusia, Disfagia y Susceptibilidad de desarrollar úlceras orales), la Tasa de Flujo salival no estimulado, el pH salival y la viscosidad salival; estableciendo su relación con el Grado de Inmunosupresión, evaluado por medio del Recuento de Linfocitos CD4/μl. Se evaluaron 126 pacientes con infección por VIH con TARGA y sin TARGA que asistieron a la Estrategia Sanitaria de Prevención y Control de ITS, VIH y SIDA del Hospital Nacional Dos de Mayo, Lima-Perú. Los pacientes sin TARGA presentaron porcentajes significativamente mayores de algunos síntomas de hipofunción salival: xerostomía , disgeusia , disfagia y susceptibilidad a desarrollar úlceras orales así como valores significativamente más altos de viscosidad salival, sin embargo, presentaban también valores significativamente más bajos de Tasa de flujo salival no estimulada y de pH salival respecto a los pacientes con TARGA. Los pacientes con un mayor grado de inmunosupresión presentaron mayores síntomas de hipofunción salival y viscosidad salival así como valores significativamente más bajos de Tasa de Flujo salival no estimulado y de pH salival respecto a los pacientes con un menor Grado de Inmunosupresión. Se concluyó que existe una alteración del perfil salival de los pacientes con Infección por VIH, la cual es significativamente mayor en los pacientes sin TARGA y en aquellos que tienen un mayor grado de inmunosupresión

    Activation of Type 1 Cannabinoid Receptor (CB1R) promotes neurogenesis in murine subventricular zone cell cultures

    The endocannabinoid system has been implicated in the modulation of adult neurogenesis. Here, we describe the effect of type 1 cannabinoid receptor (CB1R) activation on self-renewal, proliferation and neuronal differentiation in mouse neonatal subventricular zone (SVZ) stem/progenitor cell cultures. Expression of CB1R was detected in SVZ-derived immature cells (Nestin-positive), neurons and astrocytes. Stimulation of the CB1R by (R)-(+)-Methanandamide (R-m-AEA) increased self-renewal of SVZ cells, as assessed by counting the number of secondary neurospheres and the number of Sox2+/+ cell pairs, an effect blocked by Notch pathway inhibition. Moreover, R-m-AEA treatment for 48 h, increased proliferation as assessed by BrdU incorporation assay, an effect mediated by activation of MAPK-ERK and AKT pathways. Surprisingly, stimulation of CB1R by R-m-AEA also promoted neuronal differentiation (without affecting glial differentiation), at 7 days, as shown by counting the number of NeuN-positive neurons in the cultures. Moreover, by monitoring intracellular calcium concentrations ([Ca2+](i)) in single cells following KCl and histamine stimuli, a method that allows the functional evaluation of neuronal differentiation, we observed an increase in neuronal-like cells. This proneurogenic effect was blocked when SVZ cells were co-incubated with R-m-AEA and the CB1R antagonist AM 251, for 7 days, thus indicating that this effect involves CB1R activation. In accordance with an effect on neuronal differentiation and maturation, R-m-AEA also increased neurite growth, as evaluated by quantifying and measuring the number of MAP2-positive processes.     Comparative Gene Expression Analysis throughout the Life Cycle of Leishmania braziliensis: Diversity of Expression Profiles among Clinical Isolates

    Leishmania is a group of parasites (Protozoa, Trypanosomatidae) responsible for a wide spectrum of clinical forms. Among the factors explaining this phenotypic polymorphism, parasite features are important contributors. One approach to identify them consists in characterizing the gene expression profiles throughout the life cycle. In a recent study, the transcriptome of 3 Leishmania species was compared and this revealed species-specific differences, albeit in a low number. A key issue, however, is to ensure that the observed differences are indeed species-specific and not specific of the strains selected for representing the species. In order to illustrate the relevance of this concern, we analyzed here the gene expression profiles of 5 clinical isolates of L. braziliensis at seven time points of the life cycle. Our results clearly illustrate the unique character of each isolate in terms of gene expression dynamics: one Leishmania strain is not necessarily representative of a given species

    The advent of intravitreal anti-vascular endothelial growth factor (anti-VEGF) medications has revolutionized the treatment of diabetic eye diseases. Herein, we report the outcomes of clinical studies carried out by the Pan-American Collaborative Retina Study Group (PACORES), with a specific focus on the efficacy of intravitreal bevacizumab in the management of diabetic macular edema and proliferative diabetic retinopathy. We will also discuss the use of intravitreal bevaci-zumab as a preoperative, adjuvant therapy before vitrectomy for prolif-erative diabetic retinopathy.
