Evaluation of dental therapists undertaking dental examinations in a school setting in Scotland
Objective: To measure agreement between dental therapists and the
Scottish gold-standard dentist undertaking National Dental Inspection
Programme (NDIP) examinations. Methods: A study of interexaminer
agreement between 19 dental therapists and the national gold-standard dentist
was carried out. Pre-calibration training used the caries diagnostic criteria and
examination techniques agreed by the British Association for the Study of
Community Dentistry (BASCD). Twenty-three 5-year-old children (Primary 1)
and 17 11-year-old children (Primary 7) children were examined. Agreement
was assessed using kappa statistics on d 3 mft and D 3 MFT for P1 and P7 children, sensitivity and specificity values, and kappa statistics on d 3 t/D 3 T and
ft/FT. Calibration data on P1 and P7 children from 2009–2012 involving
dentists as examiners were used for comparison. Economic evaluation was
undertaken using a cost minimization analysis approach. Results: The mean
kappa score was 0.84 (SD 0.07) ranging from 0.69 to 0.94. All dental therapists
scored good or very good agreement with the gold-standard dentist. This
compares with historic NDIP calibration data with dentists, against the same
gold-standard dentist, where the mean kappa value was 0.68 (SD 0.22) with a
range of 0.35-1.00. The mean sensitivity score was 0.98 (SD 0.04) (range 0.88-1.0)
and mean specificity score was 0.90 (SD 0.06) (range 0.78-0.96). Health
economic analysis estimated that salary costs would be 33.6% lower if dental
therapists were substituted for dentists in the year 2013, with an estimated
saving of approximately £103 646 per annum on the national budget.
Conclusion: We conclude that dental therapists show a high level of
interexaminer agreement, and with the appropriate annual training and
calibration, they could undertake dental examinations as part of the NDIP
programme
Finding qualitative research: an evaluation of search strategies
BACKGROUND: Qualitative research makes an important contribution to our understanding of health and healthcare. However, qualitative evidence can be difficult to search for and identify, and the effectiveness of different types of search strategies is unknown. METHODS: Three search strategies for qualitative research in the example area of support for breast-feeding were evaluated using six electronic bibliographic databases. The strategies were based on using thesaurus terms, free-text terms and broad-based terms. These strategies were combined with recognised search terms for support for breast-feeding previously used in a Cochrane review. For each strategy, we evaluated the recall (potentially relevant records found) and precision (actually relevant records found). RESULTS: A total yield of 7420 potentially relevant records was retrieved by the three strategies combined. Of these, 262 were judged relevant. Using one strategy alone would miss relevant records. The broad-based strategy had the highest recall and the thesaurus strategy the highest precision. Precision was generally poor: 96% of records initially identified as potentially relevant were deemed irrelevant. Searching for qualitative research involves trade-offs between recall and precision. CONCLUSIONS: These findings confirm that strategies that attempt to maximise the number of potentially relevant records found are likely to result in a large number of false positives. The findings also suggest that a range of search terms is required to optimise searching for qualitative evidence. This underlines the problems of current methods for indexing qualitative research in bibliographic databases and indicates where improvements need to be made

Inference of single-cell phylogenies from lineage tracing data using Cassiopeia.
The pairing of CRISPR/Cas9-based gene editing with massively parallel single-cell readouts now enables large-scale lineage tracing. However, the rapid growth in complexity of data from these assays has outpaced our ability to accurately infer phylogenetic relationships. First, we introduce Cassiopeia-a suite of scalable maximum parsimony approaches for tree reconstruction. Second, we provide a simulation framework for evaluating algorithms and exploring lineage tracer design principles. Finally, we generate the most complex experimental lineage tracing dataset to date, 34,557 human cells continuously traced over 15 generations, and use it for benchmarking phylogenetic inference approaches. We show that Cassiopeia outperforms traditional methods by several metrics and under a wide variety of parameter regimes, and provide insight into the principles for the design of improved Cas9-enabled recorders. Together, these should broadly enable large-scale mammalian lineage tracing efforts. Cassiopeia and its benchmarking resources are publicly available at www.github.com/YosefLab/Cassiopeia
Influence Of Environmental Conditions On Performance And Heart Rate Responses To The 30-15 Incremental Fitness Test In Rugby Union Athletes
The purpose of this study was to examine the differences in performance and heart rate responses between a high heat outdoor condition (34.0°C, 64.1% humidity) and a temperate indoor condition (22.0°C, 50.0% humidity) during the 30-15 intermittent fitness test (30-15IFT). Eight highly trained Rugby Union players (28.1 +/- 1.5 years, 181.4 +/- 8.8 cm, 88.4 +/- 13.3kg) completed the 30-15IFT in two different temperature conditions. Dependant variables recorded and analysed included; final running speed of the 30-15IFT, heart rate (HR) at rest (HR rest), maximum HR (Max HR), HR recovery (HRR), average HR (HR ave) and sub-maximal HR corresponding to 25%, 50% and 75% of final test speed (HR 25%, HR 50% and HR 75%) and HR at 13 km[middle dot]h-1 (HR 13 km[middle dot]h-1). Greater running speeds were achieved when the test was conducted indoors (19.4 +/- 0.7 km[middle dot]h-1 vs. 18.6 +/- 0.6 km·h-1, p = 0.002, d = 1.67). HR ave and HR 13 km·h-1 were greater when the test was conducted outdoors (p 0.85). Large effect sizes were observed for the greater HR at submaximal intensities (d > 0.90). The results of this study highlight the influence of temperature on 30-15IFT performance and cardiac responses. It is recommended that prescription of training based on 30-15IFT results reflects the temperature that the training will be performed in and that practitioners acknowledge that a meaningful change in assessment results can be the result of seasonal temperature change rather than training induced change
What Do You Think Would Make You Happier? What Do You Think You Would Choose?
Would people choose what they think would maximize their subjective well-being (SWB)? We present survey respondents with hypothetical scenarios and elicit both choice and predicted SWB rankings of two alternatives. While choice and predicted SWB rankings usually coincide in our data, we find systematic reversals. We identify factors—such as predicted sense of purpose, control over one\u27s life, family happiness, and social status—that help explain hypothetical choice controlling for predicted SWB. We explore how our findings vary by SWB measure and by scenario. Our results have implications regarding the use of SWB survey questions as a proxy for utility
miRBase: microRNA sequences, targets and gene nomenclature.
The miRBase database aims to provide integrated interfaces to comprehensive microRNA sequence data, annotation and predicted gene targets. miRBase takes over functionality from the microRNA Registry and fulfils three main roles: the miRBase Registry acts as an independent arbiter of microRNA gene nomenclature, assigning names prior to publication of novel miRNA sequences. miRBase Sequences is the primary online repository for miRNA sequence data and annotation. miRBase Targets is a comprehensive new database of predicted miRNA target genes. miRBase is available at http://microrna.sanger.ac.uk/
Linking Heterogeneous Biodiversity Information Systems on the GRID: the GRAB Prototype
In the field of biodiversity informatics a wide range of diverse databases and tools already exists. The challenge is to integrate such resources in order to support scientists wishing to explore complex problems of relevance to biodiversity, and to create new resources where necessary. In this paper we outline the relevance of biodiversity informatics requirements to the future development of the GRID, identifying the main issues that need to be addressed in this area. We present GRAB (GRid And Biodiversity), which is a prototype demonstrator illustrating how one particular biodiversity-related task, namely bioclimatic modelling, can be supported in a Globus-based environment. We also describe a much larger-scale GRID application project that is just commencing (BiodiversityWorld) in which a flexible problem-solving environment is to be built for full-scale investigations by scientists working in a number of biodiversity research areas
Face Familiarity and Image-Specific Memory
Face familiarity produces advantages for both memory and matching. By developing an internal representation through repeated experience, viewers extract identity-specific information that aids subsequent recognition. However, researchers have recently argued that this process may also result in a familiarity disadvantage, whereby specific instances of the face are more difficult to remember, perhaps due to this process of prioritising identity- over image-specific information. Although previous experiments found no evidence of this disadvantage in working memory, initial research has demonstrated an effect in longer term storage. Here, we attempted to replicate this finding by focussing on the ability to learn images of a single (un)familiar identity. Our results failed to demonstrate a familiarity disadvantage while replicating the finding that familiarity influences response bias. As researchers continue to investigate how familiarity alters both internal representations and associated processes, it is important to establish which processes may or may not be affected

Are subjective well-being (SWB) measures a good empirical proxy for utility? We evaluate one necessary assumption: that people’s preferences coincide with what they predict will maximize their SWB. Our method is to present survey respondents with hypothetical scenarios and elicit both choice and predicted SWB rankings of two alternatives. While choice and predicted SWB rankings usually coincide, we find systematic reversals. Furthermore, we identify factors—such as predicted sense of purpose, control over one‘s life, family happiness, and social status—that help explain choice controlling for predicted SWB. We explore how our findings vary with the SWB measure and the choice situation.
