    The biomechanics of fast-starts during ontogeny in the common carp Cyprinus carpio

    Common carp Cyprinus carpio L. were reared a constant temperature of 20 degrees C from the larval (7 mm total length) to the juvenile (80 mm) stage. Body morphology and white muscle mass distribution were measured. Fast-start escape responses were recorded using high-speed cinematography from which the velocities, accelerations and hydrodynamic power requirements were estimated. All three measures of fast-start performance increased during development. White muscle contraction regimes were calculated from changes in body shape during the fast-starts and used to predict the muscle force and power production for all longitudinal positions along the body. Scaling arguments predicted that increases in body length would constrain the fish to bend less rapidly because the cross-sectional muscle area, and hence force production, does not increase at the same rate as the inertial mass that resists bending. As predicted, the increases in body length resulted in decreases in muscle shortening velocity, and this coincided with increases in both the force and power produced by the muscles. The hydrodynamic efficiency, which relates the mechanical power produced by the muscles to the inertial power requirements in the direction of travel, showed no significant change during ontogeny. The increasing hydrodynamic power requirements were thus met by increases in the power available from the muscles. The majority of the increases in fast-start swimming performance during ontogeny can be explained by size-dependent increases in muscle power output. For all sizes, there was a decrease in muscle-mass-specific power output and an increase in muscle stress in a posterior direction along the body due to systematic variations in fibre strain. These changing strain regimes result in the central muscle bulk producing the majority of the power requirements during the fast-start, and this power is transmitted to the tail region of the fish and ultimately to the water via muscle in the caudal myotomes

    Antibodies to the Mr 64,000 (64K) protein in islet cell antibody positive non-diabetic individuals indicate high risk for impaired Beta-cell function

    A prospective study of a normal childhood population identified 44 islet cell antibody positive individuals. These subjects were typed for HLA DR and DQ alleles and investigated for the presence of antibodies to the Mr 64,000 (64K) islet cell antigen, complement-fixing islet cell antibodies and radiobinding insulin autoantibodies to determine their potency in detecting subjects with impaired Beta-cell function. At initial testing 64K antibodies were found in six of 44 islet cell antibody positive subjects (13.6%). The same sera were also positive for complement-fixing islet cell antibodies and five of them had insulin autoantibodies. During the follow-up at 18 months, islet cell antibodies remained detectable in 50% of the subjects studied. In all six cases who were originally positive, 64K antibodies were persistently detectable, whereas complement-fixing islet cell antibodies became negative in two of six and insulin autoantibodies in one of five individuals. HLA DR4 (p < 0.005) and absence of asparic acid (Asp) at position 57 of the HLA DQ chain (p < 0.05) were significantly increased in subjects with 64K antibodies compared with control subjects. Of 40 individuals tested in the intravenous glucose tolerance test, three had a first phase insulin response below the first percentile of normal control subjects. Two children developed Type 1 (insulin-dependent) diabetes mellitus after 18 and 26 months, respectively. Each of these subjects was non-Asp homozygous and had persistent islet cell and 64K antibodies. We conclude that 64K antibodies, complement-fixing islet cell antibodies and insulin autoantibodies represent sensitive serological markers in assessing high risk for a progression to Type 1 diabetes in islet cell antibody positive non-diabetic individuals

    Predicting participation in group parenting education in an Australian sample: The role of attitudes, norms, and control factors

    We examined the theory of planned behavior (TPB) in predicting intentions to participate in group parenting education. One hundred and seventy-six parents (138 mothers and 38 fathers) with a child under 12 years completed TPB items assessing attitude, subjective norms, perceived behavioral control (PBC), and two additional social influence variables (self-identity and group norm). Regression analyses supported the TPB predictors of participation intentions with self-identity and group norm also significantly predicting intentions. These findings offer preliminary support for the TPB, along with additional sources of social influence, as a useful predictive model of participation in parenting education

    A momentum-dependent perspective on quasiparticle interference in Bi_{2}Sr_{2}CaCu_{2}O_{8+\delta}

    Angle Resolved Photoemission Spectroscopy (ARPES) probes the momentum-space electronic structure of materials, and provides invaluable information about the high-temperature superconducting cuprates. Likewise, the cuprate real-space, inhomogeneous electronic structure is elucidated by Scanning Tunneling Spectroscopy (STS). Recently, STS has exploited quasiparticle interference (QPI) - wave-like electrons scattering off impurities to produce periodic interference patterns - to infer properties of the QP in momentum-space. Surprisingly, some interference peaks in Bi_{2}Sr_{2}CaCu_{2}O_{8+\delta} (Bi-2212) are absent beyond the antiferromagnetic (AF) zone boundary, implying the dominance of particular scattering process. Here, we show that ARPES sees no evidence of quasiparticle (QP) extinction: QP-like peaks are measured everywhere on the Fermi surface, evolving smoothly across the AF zone boundary. This apparent contradiction stems from different natures of single-particle (ARPES) and two-particle (STS) processes underlying these probes. Using a simple model, we demonstrate extinction of QPI without implying the loss of QP beyond the AF zone boundary

    Lewis x (Lex, CD15), also known as SSEA-1 (stage specific embryonic antigen-1), is a trisaccharide with the structure Galβ(1–4)Fucα(1–3)GlcNAc, which is expressed on glycoconjugates in human polymorphonuclear granulocytes and various tumors such as colon and breast carcinoma. We have investigated the role of Lex in the adhesion of MCF-7 human breast cancer cells and PMN to human umbilical endothelial cells (HUVEC) and the effects of two different anti-Lex mAbs (FC-2.15 and MCS-1) on this adhesion. We also analyzed the cytolysis of Lex+-cells induced by anti-Lex mAbs and complement when cells were adhered to the endothelium, and the effect of these antibodies on HUVEC. The results indicate that MCF-7 cells can bind to HUVEC, and that MCS-1 but not FC-2.15 mAb inhibit this interaction. Both mAbs can efficiently lyse MCF-7 cells bound to HUVEC in the presence of complement without damaging endothelial cells. We also found a Lex-dependent PMN interaction with HUVEC. Although both anti-Lex mAbs lysed PMN in suspension and adhered to HUVEC, PMN aggregation was only induced by mAb FC-2.15. Blotting studies revealed that the endothelial scavenger receptor C-type lectin (SRCL), which binds Lex-trisaccharide, interacts with specific glycoproteins of Mr␣∼␣28 kD and 10 kD from MCF-7 cells.     Radio continuum emission in the northern Galactic plane: Sources and spectral indices from the THOR survey

    Radio continuum surveys of the Galactic plane can find and characterize HII regions, supernova remnants (SNRs), planetary nebulae (PNe), and extragalactic sources. A number of surveys at high angular resolution (<25") at different wavelengths exist to study the interstellar medium (ISM), but no comparable high-resolution and high-sensitivity survey exists at long radio wavelengths around 21cm. We observed a large fraction of the Galactic plane in the first quadrant of the Milky Way (l=14.0-67.4deg and |b| < 1.25deg) with the Karl G. Jansky Very Large Array (VLA) in the C-configuration covering six continuum spectral windows. These data provide a detailed view on the compact as well as extended radio emission of our Galaxy and thousands of extragalactic background sources. We used the BLOBCAT software and extracted 10916 sources. After removing spurious source detections caused by the sidelobes of the synthesised beam, we classified 10387 sources as reliable detections. We smoothed the images to a common resolution of 25" and extracted the peak flux density of each source in each spectral window (SPW) to determine the spectral indices α\alpha (assuming I(ν)ναI(\nu)\propto\nu^\alpha). By cross-matching with catalogs of HII regions, SNRs, PNe, and pulsars, we found radio counterparts for 840 HII regions, 52 SNRs, 164 PNe, and 38 pulsars. We found 79 continuum sources that are associated with X-ray sources. We identified 699 ultra-steep spectral sources (α<1.3\alpha < -1.3) that could be high-redshift galaxies. Around 9000 of the sources we extracted are not classified specifically, but based on their spatial and spectral distribution, a large fraction of them is likely to be extragalactic background sources. More than 7750 sources do not have counterparts in the SIMBAD database, and more than 3760 sources do not have counterparts in the NED database

    Construction of non-polar mutants in Haemophilus influenzae using FLP recombinase technology

    Background Nontypeable Haemophilus influenzae (NTHi) is a gram-negative bacterium that causes otitis media in children as well as other infections of the upper and lower respiratory tract in children and adults. We are employing genetic strategies to identify and characterize virulence determinants in NTHi. NTHi is naturally competent for transformation and thus construction of most mutants by common methodologies is relatively straightforward. However, new methodology was required in order to construct unmarked non-polar mutations in poorly expressed genes whose products are required for transformation. We have adapted the lambda red/FLP-recombinase-mediated strategy used in E. coli for use in NTHi. Results A cassette containing a spectinomycin resistance gene and an rpsL gene flanked by FRT sites was constructed. A PCR amplicon containing 50 base pairs of DNA homologous to the 5' and 3' ends of the gene to be disrupted and the cassette was generated, then recombineered into the target NTHi gene, cloned on a plasmid, using the lambda recombination proteins expressed in E. coli DY380. Thus, the gene of interest was replaced by the cassette. The construct was then transformed into a streptomycin resistant NTHi strain and mutants were selected on spectinomycin-containing growth media. A plasmid derived from pLS88 with a temperature sensitive replicon expressing the FLP recombinase gene under the control of the tet operator/repressor was constructed. This plasmid was electroporated into the NTHi mutant at the permissive temperature and FLP expression was induced using anhydrotetracycline. The recombinase recognizes the FRT sites and eliminates the antibiotic cassette by site-specific recombination, creating the unmarked non-polar mutation. The plasmid is cured by growth of cells at the restrictive temperature. Conclusion The products of the genes in the NTHi pilABCD operon are required for type IV pilus biogenesis and have a role in transformation. We demonstrated the utility of our methodology by the construction of a non-polar pilA mutation in NTHi strain 2019 and complementation of the mutation with a plasmid containing the pilA gene.
