    Sequence Analysis of the Angiotensin II Type 1 Receptor (AGTR1) Gene for Mutations Contributing to Pulmonary Hypertension in the Chicken (Gallus gallus)

    Our multidisciplinary group at the University of Arkansas has been investigating the suitability of the chicken as a medical model for pulmonary arterial hypertension (PAH) in humans. There are several forms of PAH in humans arising from elevated pulmonary arterial pressure and pulmonary vascular resistance. Spontaneous cases of PAH are known as idiopathic PAH (IPAH), where the exact physiological causes are not known. IPAH patients that do not respond to standard treatments have a prognosis of only a few years. Currently, there is no acceptable animal model for IPAH. As part of our effort to pursue the chicken as model for IPAH, we have mapped chromosomal regions associated with susceptibility to ascites, an industry term for PAH in chickens. One region identified contains the angiotensin II type 1 receptor gene (AGTR1), a gene known to be associated with particular forms of PAH in humans. My project was to sequence the exonic regions of AGTR1 from chickens selected for susceptibility and resistance to ascites. I identified a total of 15 single nucleotide polymorphisms (SNPs) in intronic regions and 9 affecting exonic sequences that distinguish resistant and susceptible birds. One of the SNPs alters the encoded protein, but more sequencing data is needed to confirm the presence of this SNP. The 24 SNPs have become the basis for further genotypic efforts using quantitative polymerase chain reaction high-resolution melt (qPCR-HRM) assays to determine the association of alternative alleles of AGTR1 with PAH in the chicken. This knowledge will help the poultry industry in genetic selection to reduce incidence of ascites. Characterization of the genetic determinants of ascites in the chicken will also identify specific gene networks and further our understanding of PAH in humans

    Taxonomic description and phylogenetic placement of two new species of Spalangiopelta (Hymenoptera: Pteromalidae: Ceinae) from Eocene Baltic amber

    Spalangiopelta is a small genus of chalcid wasps that has received little attention despite the widespread distribution of its extant species. The fossil record of the genus is restricted to a single species from Miocene Dominican amber. We describe two new fossil species, Spalangiopelta darlingi sp. n. and Spalangiopelta semialba sp. n. from Baltic amber. The species can be placed within the extant genus Spalangiopelta based on the distinctly raised hind margin of the mesopleuron. 3D models reconstructed from ”CT data were utilized to assist in the descriptions. Furthermore, we provide a key for the females of all currently known Spalangiopelta species. The phylogenetic placement of the fossils within the genus is analyzed using parsimony analysis based on morphological characters. Phylogenetic and functional relevance of two wing characters, admarginal setae and the hyaline break, are discussed. The newly described Baltic amber fossils significantly extend the minimum age of Spalangiopelta to the Upper Eocene

    Simulation of Fixed– and Variable–Rate Application of Granular Materials

    Research has shown that application errors exist with variable–rate technology (VRT) systems. Consequently, using prescription maps for economic and agronomic analyses can generate misleading results. The intent of this article was to develop and validate a spatial data model for generating “as–applied” maps to support the advancement of precision agriculture practices. Previous research modified ASAE Standard S341.2 to include a 2–D matrix of collection pans to assess fixed–rate and variable–rate (VR) deposition of granular fertilizers and agricultural lime from a spinner disc spreader. The “as–applied” spatial data model uses GIS functionality to generate “as–applied” surfaces by merging distribution patterns and a spatial field application file (FAF) into an “as–applied” surface representing the actual distribution of granular fertilizer or agricultural lime across a field. To validate the “as–applied” spatial data model, field studies were conducted by randomly placing collection pans across two fields. Murate of potash was then applied using a VR spinner spreader. The “as–applied” spatial data model was used to predict the amount of material each pan should have received. Comparisons were made between the actual and predicted application rates for two fields, with R2 values of 0.45 (field A) and 0.58 (field B) computed. However, R2 values of 0.16 (field A) and 0.21 (field B) were observed when comparing the actual application rates and prescription maps. These low R2 values indicated poor application by the spinner spreader but demonstrated that the “as–applied” model did a better job of representing the distribution of murate of potash when contrasted with the prescription maps. “As–applied” surfaces provide a means for evaluating fixed–rate and VR application of granular products while enhancing researchers’ ability to compare VR management approaches

    Ion irradiation of Fe-Fe oxide core-shell nanocluster films: Effect of interface on stability of magnetic properties

    A cluster deposition method was used to produce films of loosely aggregated nanoclusters (NC) of Fe core-Fe3O4 shell or fully oxidized Fe3O4. Films of these NC on Si(100) or MgO(100)/Fe3O4(100) were irradiated to 10^16 Si2+/cm2 near room temperature using an ion accelerator. Ion irradiation creates structural change in the NC film with corresponding chemical and magnetic changes which depend on the initial oxidation state of the cluster. Films were characterized using magnetometry (hysteresis, first order reversal curves), microscopy (transmission electron, helium ion), and x-ray diffraction. In all cases, the particle sizes increased due to ion irradiation, and when a core of Fe is present, irradiation reduces the oxide shells to lower valent Fe species. These results show that ion irradiated behavior of the nanocluster films depends strongly on the initial nanostructure and chemistry, but in general saturation magnetization decreases slightly.Comment: 25 pages, 4 tables, 6 figure

    Suprarenal fixation of endovascular aortic stent grafts: assessment of medium-term to long-term renal function by analysis of juxtarenal stent morphology.

    Objective: The effect of supra-renal stent fixation during endovascular aortic aneurysm repair (EVAR) on renal function remains unclear. Using a unique validated 3D intraluminal imaging technique, we analysed the effect of suprarenal stent position relative to renal artery orifices and its medium to long term effects on renal function.Methods: Thirty-four consecutive patients from a single institution were studied following EVAR using the Zenith endograft system from September 1999 to March 2002. The precise locations of the uncovered supra-renal stent struts were assessed by a virtual endoscopic imaging technique. Anatomical and quantitative categorisation of patients was made according to the configuration of uncovered stent struts across the renal artery ostia (RAO). The anatomical subgroups were defined as struts located centrally or peripherally across both RAO. The quantitative subgroups were defined as RAO crossed by multiple struts, a solitary strut or no struts. The subgroups were compared for their renal function as assessed by temporal measurements of serum creatinine concentration and creatinine clearance, and renal parenchymal perfusion defects as assessed using contrast-enhanced computerised tomography (CT).Results: Mean follow-up was 52.7 months. Separate subgroup analysis for both anatomical and quantitative configurations did not demonstrate any significant difference in renal function between the different strut permutations (p>0.05). Follow-up imaging confirmed one case of renal infarction secondary to an occluded accessory renal artery, although, this patient had normal creatinine levels.Conclusion: RAO coverage by supra-renal uncovered stents does not appear to have a significant effect on renal function as evaluated by creatinine measurements in patients with normal pre-operative renal function

    Financial literacy, present bias and alternative mortgage products

    Choosing a mortgage is one of the most important financial decisions made by a household. Financial innovation has given rise to more complex mortgage products with back-loaded payments, known as ‘Alternative Mortgage Products’ (AMPs), or ‘Interest-Only Mortgages’. Using a specially designed question module in a representative survey of UK mortgage holders, we investigate the effect of consumer financial sophistication on the decision to choose an AMP instead of a standard repayment mortgage. We show poor financial literacy and present bias raise the likelihood of choosing an AMP. Financially literate individuals are also more likely to choose an Adjustable Rate Mortgage (ARM), suggesting they avoid paying the term premium of a fixed rate mortgage

    World Allergy Organization-McMaster University Guidelines for Allergic Disease Prevention (GLAD-P): Probiotics

    Background: Prevalence of allergic diseases in infants, whose parents and siblings do not have allergy, is approximately 10% and reaches 20–30% in those with an allergic first-degree relative. Intestinal microbiota may modulate immunologic and inflammatory systemic responses and, thus, influence development of sensitization and allergy. Probiotics have been reported to modulate immune responses and their supplementation has been proposed as a preventive intervention. Objective: The World Allergy Organization (WAO) convened a guideline panel to develop evidence-based recommendations about the use of probiotics in the prevention of allergy. Methods: We identified the most relevant clinical questions and performed a systematic review of randomized controlled trials of probiotics for the prevention of allergy. We followed the Grading of Recommendations Assessment, Development and Evaluation (GRADE) approach to develop recommendations. We searched for and reviewed the evidence about health effects, patient values and preferences, and resource use (up to November 2014). We followed the GRADE evidence-to-decision framework to develop recommendations. Results: Currently available evidence does not indicate that probiotic supplementation reduces the risk of developing allergy in children. However, considering all critical outcomes in this context, the WAO guideline panel determined that there is a likely net benefit from using probiotics resulting primarily from prevention of eczema. The WAO guideline panel suggests: a) using probiotics in pregnant women at high risk for having an allergic child; b) using probiotics in women who breastfeed infants at high risk of developing allergy; and c) using probiotics in infants at high risk of developing allergy. All recommendations are conditional and supported by very low quality evidence. Conclusions: WAO recommendations about probiotic supplementation for prevention of allergy are intended to support parents, clinicians and other health care professionals in their decisions whether to use probiotics in pregnancy and during breastfeeding, and whether to give them to infants
