    In ovo validation model to assess the efficacy of commercial prebiotics on broiler performance and oxidative stability of meat

    Abstract The purpose of this study was to examine the effect of in ovo injection of 2 different prebiotics, DiNovo (DN; Laminaria spp., extract containing laminarin and fucoidan) and Bi2 tos (BI; non-digestive trans-galactooligosaccharides from milk lactose digested with Bifidobacterium bifidum NCIMB 41171), on growth, slaughter traits, intramuscular fat percentage (IF) and muscle fiber diameter, and lipid oxidation of meat in chickens reared under commercial conditions, following an in ovo trial protocol. On d 12 of embryonic incubation, 350,560 Ross 308 eggs were randomly divided into 3 experimental groups and automatically injected in ovo with: physiological saline (control group), BI at dose of 3.5 mg/embryo and DN at dose of 0.88 mg/embryo. Hatched chicks (males and females) were allocated dependent on treatment group into 3 poultry houses on each farm (3 farms in total) with a stocking density of 21.2 to 21.5 chicks/m2. At 42 d of age, 14 randomly chosen birds (7 males and 7 females), per each treatment from each farm, were individually weighed and slaughtered. The results showed no significant differences of final number of chickens/chicken house, mortality, BW per treatment, stocking density (kg/m2), feed intake, feed conversion rate (FCR), and European Broiler Index among 3 experimental groups. Treatments with BI and DN were associated with slight increases (P > 0.05) in average BW and a minor improvement (P > 0.05) of FCR in BI group. Slaughtered chickens from DN and BI treated groups had significantly increase of BW, carcass weight, carcass yield, and breast muscle weight compared with the control group. IF and muscle fiber diameter were similar among groups. Males had significantly higher slaughter traits compared to females, except for breast muscle yield. The prebiotic treatments led to a higher lipid oxidation in meat, even if the detected TBA reactive substances were below the critical value recognized for meat acceptability. In conclusion, in ovo administration of prebiotics was associated with improvements in a number of parameters of relevance to commercial poultry production

    Wybrane cechy mikrostruktury mięśnia piersiowego powierzchownego oraz podstawowe wartości użytkowości rzeźnej gęsi mieszańców

    The aim of study was to determine of basic values slaughter meat production and histological parameters of pectoral muscle in quadruple 24 weeks old crossbred geese. The hybrids produced using Graylag, White Kołuda and Slovakian geese. The geese were divided to 4 groups differing crosses scheme. The body weight before slaughter was in the range from 4,752 g to 4,921 g, weight of carcass with neck from 3,101 g to 3,175 g and weight of breast muscles from 649 g to 698 g. Histological analysis of pectoral muscle showed that diameters of white fibers (αW) was in the range from 43 μm to 46 μm, red fibers (βR) from 19.4 μm to 22.1 μm. The percentage share of αW muscle fibers was in the range from 25.3% to 28.9%, βR fibers from 71.1% to 74.7%. The number of muscle fibers per unit area was in the range from 256 to 316 and intramuscular fat content from 3.9% to 6.7%. The results of evaluation of microstructural traits of musculus pectoralis superficialis and meat production parameters suggest that the breast muscles of the crossbred geese are raw material of high quality. The quadruple crossbred geese with graylag geese it is an alternative for production of high-quality meat.Celem pracy było określenie podstawowych wskaźników użytkowości mięsnej i cech histologicznych mięśnia piersiowego 24-tygodniowych poczwórnych mieszańców gęsi. Mieszańce pochodziły z krzyżowania dzikiej gęsi gęgawej, Białej Kołudzkiej i Słowackiej. Gęsi przydzielono do 4 grup różniących się systemem krzyżowania. Masa ciała ptaków przed ubojem mieściła się w graniacach od 4 752 g do 4 921 g, masa tuszki patroszonej z szyją od 3 101 g do 3 175 g i masa mięśni piersiowych od 649 g do 698 g. Ocena histologiczna mięśnia piersiowego wykazała, że średnica włókien białych (αW) wynosiła od 43 μm do 46 μm, włókien czerwonych (βR) od 19,4 μm do 22,1 μm. Procentowy udział włókien αW mieścił się w granicach od 25,3% do 28,9%, βR od 71,1% do 74,7%. Ilość włókien na jednostce powierzchni mieściła się w granicach od 256 do 316, a udział tłuszczu śródmięśniowego wynosił od 3,9% do 6,7%. Wyniki oceny cech mikrostruktury musculus pectoralis superficialis i cech użytkowości mięsnej sugerują, że mięśnie piersiowe gęsi mieszańców są wysokiej jakości surowcem rzeźnym. Poczwórne mieszańce wytworzone z udziałem dzikiej gęsi gęgawy stanowią alternatywę w produkcji mięsa o wysokiej jakości

    Influence of rearing system and sex on carcass traits and meat quality of broiler chickens

    The trial was conducted in order to evaluate the effect of the rearing system and sex on live body weight, daily weight gain, carcass traits and meat quality of broiler chickens. Sixty 1-d-old Ross 308 chickens (males and females) were randomly divided, according to the sex, into 2 experimental groups based on rearing system: intensive system (IS; n = 30), birds reared till 42 d of age and semi-intensive system (SIS; n = 30), birds reared till 56 d of age. Compared with SIS group, IS group had higher (P < 0.01) daily weight gain, weight and yield of pectoral muscle (PM). Differently, SIS group had higher (P < 0.05) carcass weight and carcass yield. Males have usually a better performance than females. In comparison with IS, SIS exhibited a lower (P < 0.01) breast meat pH and higher (P < 0.01) lightness and yellowness values. Pectoral muscle from IS broiler chickens showed a greater (P < 0.01) fibre diameter and significantly higher shear force values than PM from SIS chicken. PM from IS chicken group had higher (P < 0.01) dry matter and protein content and slight fat content than PM from SIS chickens. The sex influence was not observed on physicochemical characteristics of PM in chickens reared in both system

    Biomarkers of brain ischemia-reperfusion injury after carotid endarterectomy

    Abstract Background: Endarterectomy of the internal carotid artery (CEA) is a surgical procedure used to prevent cerebral ischemic stroke. Available data from previous literature indicates that CEA may lead to complications in the form of cerebral ischemia-reperfusion syndrome and oxidative stress. The aim of this study was to evaluate the serum levels of oxidative stress-related biomarkers such as 8-hydroxydeoxyguanosine (8-OHdG) and malondialdehyde (MDA) in patients who underwent CEA. Material and methods. Twenty-four patients with severe internal carotid artery stenosis participated in the study. Serum samples were taken from patients at three different times: within 24 hours preoperatively to CEA, 12 hours postoperatively, and 48 hours postoperatively. Serum 8-OHdG and MDA levels were measured using a commercially available enzyme-linked immunosorbent assay. Results: Serum 8-OHdG levels indicated statistically significant elevation 12 hours after surgery when compared to preoperative levels (p&lt;0.05). A further increase in the concentration of this parameter was observed 48 hours after surgery when compared to the previous measurement, however, this increase was no longer statistically significant (p=0.05). Furthermore, serum MDA levels also indicated a statistically significantly elevation 48 hours after surgery when compared to preoperative levels (p&lt;0.05). Conclusion: Our study showed that CEA causes an increase in blood levels of 8-OHdG and MDA, which may be related to the occurrence of oxidative stress during cerebral ischemia-reperfusion injury. Therefore, 8-OHdG and MDA may represent corresponding markers of cerebral ischemia-reperfusion complications in patients undergoing CEA

    Medieval multilingualism in Poland:Creating a corpus of Greater Poland court oaths (ROThA)

    In this paper we introduce the research plan for the preparation of a searchable electronic repository of the earliest extant legal oaths from medieval Poland drawing on the expertise in historical corpus-building developed for the history of English. The oaths survive in the overwhelmingly Latin land books from the period between 1386 and 1446 for six localities Greater Poland, in which the land courts operated: Poznań, Kościan, Pyzdry, Gniezno, Konin and Kalisz. A diplomatic edition of the oaths was published in five volumes by Polish historical linguists (Kowalewicz & Kuraszkiewicz 1959–1966). The edition is the only comprehensive resource of considerable scope (over 6300 oaths from the years 1386–1446) for the study of the earliest attestations of the Polish language beyond glosses. Recognising some limitations, but most of all its unparalleled coverage of the coexistence of Latin and the vernacular, the ROThA project embarks on transforming the edition into an open up-to-date digital resource. We thus aim to facilitate research into the history of Polish and Latin as well as of the legal system and the related social and linguistic issues of the period

    Mitochondrial theory of skeletal muscle ageing –new facts, new doubts

    For many years, scientists have been pursuing research on skeletal muscle ageing both in humans and animals. Studies on animal models have extended our knowledge of this mechanism in humans. Most researchers agree that the major processes of muscle ageing occur in the mitochondria as the major energy production centres in muscle cells. It is believed that decisive changes occur at the enzymatic activity level as well as in protein synthesis and turnover ability. Deregulation of ion channels and oxidative stress also play significant roles. In particular, in recent years the free radical theory of ageing has undergone considerable modification; researchers are increasingly highlighting the partly positive effects of free radicals on processes occurring in cells. In addition, the influence of diet and physical activity on the rate of muscle cell ageing is widely debated as well as the possibility of delaying it through appropriate physical exercise and diet programmes. Numerous studies, especially those related to genetic processes, are still being conducted, and in the near future the findings could provide valuable information on muscle ageing. The results of ongoing research could answer the perennial question of whether and how we can influence the rate of ageing both in animals and humans

    Growth Performance, Carcass Characteristics and Meat Quality of Organically Reared Broiler Chickens Depending on Sex

    Given the growing interest of consumers in naturally produced meat, we decided to examine the muscles of Ross 308 broiler chickens kept in an ecological system, with the division into two research groups depending on sex. All the analyses were carried out using the appropriate methods recommended by the AOAC and in accordance with the Polish standards (PN), which are described in detail in the relevant section of the publication. The aim of the experiment was to investigate the effect of sex on the growth performance, carcass traits and meat quality, as well as the fatty acid profile and histological parameters, of the pectoral muscles from organic broiler chickens. A total of 60 one-day-old Ross 308 chickens (half males and half females) were divided into two groups, according to the sex, and reared under organic conditions (Org.) until 82 days of age (ten birds, i.e., five males and five females in each of three pens; replications for experimental groups). Compared with the female group, the male group had a higher final BW and carcass weight (p &lt; 0.05). The males had a better growth and slaughter performance than the females. The meat quality traits and fatty acids content were also affected by sex. The meat from females showed a significantly higher (p &lt; 0.05) protein, dry matter, fiber diameter and shear force and a significantly lower (p &lt; 0.05) fat level than the male group. In this research, the suitability of these fast-growing broilers for natural and organic production systems has been researched with regard to the performance, meat quality and histological characteristics of the muscles

    Effects of Probiotics, Prebiotics and Synbiotics Injected in Ovo on the Microstructure of the Breast Muscle in Different Chicken Genotypes

    The aim of the study was to analyse the effect of probiotics, prebiotics and synbiotics injected in ovo on day 12 of embryonic development on the microstructure of the superficial pectoral muscle (musculus pectoralis superficialis) from 42-day-old chickens of different genotypes: broilers (Ross 308) and general-purpose type (green-legged partridge (GP) chickens Zk-11, native chickens). Incubated eggs were divided into four groups (each genotype separately) depending on the substance injected in ovo: normal saline (C, control); Lactococcus lactis subsp. cremoris (PRO); galactooligosaccharides, GOS (PRE) or GOS + L. lactis (SYN). After hatching, chicks were placed in eight replicated pens (four pens/genotype group). There were eight birds per pen. In total, 64 birds were used in the experiment. Birds were slaughtered at the age of 42 days, and samples of superficial pectoral muscles were taken for analysis. The microstructure of the pectoral muscles was evaluated using the cryosectioning (frozen tissue sectioning) technique and staining with haematoxylin and eosin. Statistical analysis revealed that the in ovo injection of probiotics, prebiotics and synbiotics had no significant effect on the diameter of muscle fibres from chickens of the two genotypes. The number of fibres in the muscles from green-legged partridge chickens was about three-fold higher than the fibre density in the muscles from broiler chickens, with the fibre diameter being two-fold smaller. This fact may indicate a greater tenderness of meat from GP chickens compared to the meat from Ross 308 broilers. In the case of broilers, a prebiotic (GOS) was the most effective bioactive substance in reducing the number of histopathological changes. Considering muscles from GP chickens, the number of normal fibres was highest in birds treated with the probiotic. These findings indicate that the microstructural features of pectoral muscles depend not only on the type of the injected bioactive substance but also on the genotype of chickens

    The ide (Leuciscus idus) is a native European species of rheophilic cyprinid fish whose wild population status is dependent on conservation efforts, particularly regular restocking. This study aimed to evaluate the effects of including insect meals as a component in the diet of ide juveniles on their growth performance, feed utilization, and nutriphysiological status. Four diets were formulated: three with insect meals, HI–with 20% Hermetia illucens meal, TM–with 20% Tenebrio molitor meal, and ZM–with 20% Zophobas morio meal, and the control group diet, CON–fish meal with no insect component. The effects of the various diets on the efficiency of rearing ide juveniles were assessed based on fish growth parameters, feed utilization parameters, somatic indices, and intestinal and hepatopancreatic histomorphology. The highest increase in fish weight gain and the protein efficiency ratio was observed in the HI and TM groups, while the lowest values were observed in the CON and ZM groups. Comparable results were noted for the feed conversion ratio, which was most favorable in the HI and TM groups and increased in the ZM group. The use of black soldier fly and mealworm larval meal in the diets of ide juveniles had a positive effect on rearing results and overall fish health

    Effects of Effective Microorganisms on Meat Quality, Microstructure of the Longissimus Lumborum Muscle, and Electrophoretic Protein Separation in Pigs Fed on Different Diets

    The aim of the study was to determine how effective microorganisms influence meat quality, the microstructure of the longissimus lumborum muscle, and electrophoretic protein separation. The study group consisted of 150 piglets divided into three feeding groups: C, E1, and E2. The feeding groups included C&mdash;a standard fodder blend with a full share of post-extracted soya meal; E1&mdash;a 50%/50% mix of pea and lupine/soya bean in phase I of fattening and a 75%/25% mix of pea and lupine/soya bean in phase II of fattening; and E2&mdash;a 50%/50% mix of pea and lupine/soya bean in phase I of fattening and in 100% pea and lupine in phase II of fattening. The experimental factor was the addition of the EM Carbon Bokashi probiotic to the diet (C + EM, E1 + EM, E2 + EM). Influence of the feeding system on the following parameters was also estimated. After slaughter, the meat quality, LL muscle microstructure, and electrophoretic protein separation were assessed. In the C + EM group, a lower water-holding capacity was demonstrated. Meat from pigs fed the effective microorganism additive was much harder in the E1+EM group compared to meat from pigs from the E1 group. A beneficial effect of effective microorganism was found in the E2 + EM group, where less thermal leakage from the meat was demonstrated. A beneficial effect of the feeding system on thermal leakage and loin eye area in the E2 + EM group was demonstrated. In the C + EM group, a lower total number of muscle fibers was demonstrated. The addition of effective microorganism caused an increase in the diameter of fast twitch fibers in the E1 + EM group. In the same group of pigs, effective microorganisms caused a lower proportion of fiber fission. This nutritional variant appears to be the most appropriate for proteins as well, because it led to the most favorable percentage of individual proteins after effective microorganisms supplementation in the longissimus lumborum muscle
