    Creative Technologies for Behaviour Change

MI scripts aimed to develop discrepancy, evoke solutions and promote self-efficacy. FIT scripts included multi-sensory mental imagery exercises at key points in the MI scripts. Four methods of delivery were developed: a human video-counsellor, a NAO robot programmed with Choregraphe software, a video robot counsellor for comparison with the human video-counsellor, and a life-sized two dimensional 'holographic' projection. Four empirical studies tested these developments in participants wanting to become more physically active. Study 1 (N=18) and Study 2 (N=20) used qualitative methods to explore the usability and acceptability of MI delivered by a pre-recorded human video counsellor and NAO robot respectively. Analysis of participants' verbal dialogue with the video counsellor in Study 1 showed high levels of change talk, an important ingredient of effective MI. In both studies, participants reported that voicing their goals aloud was helpful but they were somewhat frustrated by the lack of personalised response. Participants positively appraised the non-judgemental aspect of the interview with the robot. Study 3 tested if virtual FIT would be more acceptable and effective than virtual MI. Ninety-eight participants received FIT or MI delivered by a video robot, and compared to a wait-list control group. In Study 4, 104 participants were randomized to a monologue version of FIT delivered by a human counsellor projected as a two-dimensional life-size hologram, or on a computer screen, or a wait-list control condition. Neither Study 3 or 4 found any quantitative effect of virtual counselling on physical activity, self-efficacy, or motivation. As in studies 1 and 2, although participants found the technological interaction somewhat impersonal, qualitative responses were largely positive: participants liked the opportunity to voice their goals, reported a motivational boost, and perceived the virtual coaches as non-judgmental. This research has shown that people perceive benefits from speaking aloud about their goals and problems, and even engaging silently in imagery-based counselling. There is potential to deliver a brief motivational intervention that is fully-automated and acceptable to participants

    Assessment of the Treatment Performance of an Open-Air Green Wall Fed with Graywater under Winter Conditions

    Graywater (GW), i.e., the portion of household wastewater that excludes toilet flushes, is an interesting wastewater type because it requires only mild treatment. Green walls have been proposed as example of a nature-based solution for GW treatment due to low energy requirement and high ecological/societal benefits; however, indications about their treatment performances remain limited. This work presents experimental results of a laboratory modular green wall for GW treatment. Experiments have been performed outdoors during the winter season for three months. Each panel included four vertical columns of planted pots, and it was fed with 100 L of synthetic GW per day. Removal efficiencies were as follows (average values): 40% chemical oxygen demand, 97% biochemical oxygen demand, 61% total Kjeldhal nitrogen, 56% NO3–-N, 57% total phosphorus, 99% Escherichia coli, and 63% anionic surfactants. This work proved the potential of an open-air green wall for treating GW, even under challenging conditions for biological treatment processes and with high hydraulic loading rates

    KLF9 and JNK3 Interact to Suppress Axon Regeneration in the Adult CNS

    Neurons in the adult mammalian CNS decrease in intrinsic axon growth capacity during development in concert with changes in Krüppel-like transcription factors (KLFs). KLFs regulate axon growth in CNS neurons including retinal ganglion cells (RGCs). Here, we found that knock-down of KLF9, an axon growth suppressor that is normally upregulated 250-fold in RGC development, promotes long-distance optic nerve regeneration in adult rats of both sexes. We identified a novel binding partner, MAPK10/JNK3 kinase, and found that JNK3 (c-Jun N-terminal kinase 3) is critical for KLF9\u27s axon-growth-suppressive activity. Interfering with a JNK3-binding domain or mutating two newly discovered serine phosphorylation acceptor sites, Ser106 and Ser110, effectively abolished KLF9\u27s neurite growth suppression in vitro and promoted axon regeneration in vivo. These findings demonstrate a novel, physiologic role for the interaction of KLF9 and JNK3 in regenerative failure in the optic nerve and suggest new therapeutic strategies to promote axon regeneration in the adult CNS

    The impact of surgical delay on resectability of colorectal cancer: An international prospective cohort study

    AIM: The SARS-CoV-2 pandemic has provided a unique opportunity to explore the impact of surgical delays on cancer resectability. This study aimed to compare resectability for colorectal cancer patients undergoing delayed versus non-delayed surgery. METHODS: This was an international prospective cohort study of consecutive colorectal cancer patients with a decision for curative surgery (January-April 2020). Surgical delay was defined as an operation taking place more than 4 weeks after treatment decision, in a patient who did not receive neoadjuvant therapy. A subgroup analysis explored the effects of delay in elective patients only. The impact of longer delays was explored in a sensitivity analysis. The primary outcome was complete resection, defined as curative resection with an R0 margin. RESULTS: Overall, 5453 patients from 304 hospitals in 47 countries were included, of whom 6.6% (358/5453) did not receive their planned operation. Of the 4304 operated patients without neoadjuvant therapy, 40.5% (1744/4304) were delayed beyond 4 weeks. Delayed patients were more likely to be older, men, more comorbid, have higher body mass index and have rectal cancer and early stage disease. Delayed patients had higher unadjusted rates of complete resection (93.7% vs. 91.9%, P = 0.032) and lower rates of emergency surgery (4.5% vs. 22.5%, P < 0.001). After adjustment, delay was not associated with a lower rate of complete resection (OR 1.18, 95% CI 0.90-1.55, P = 0.224), which was consistent in elective patients only (OR 0.94, 95% CI 0.69-1.27, P = 0.672). Longer delays were not associated with poorer outcomes. CONCLUSION: One in 15 colorectal cancer patients did not receive their planned operation during the first wave of COVID-19. Surgical delay did not appear to compromise resectability, raising the hypothesis that any reduction in long-term survival attributable to delays is likely to be due to micro-metastatic disease

    The evolving SARS-CoV-2 epidemic in Africa: Insights from rapidly expanding genomic surveillance

    INTRODUCTION Investment in Africa over the past year with regard to severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) sequencing has led to a massive increase in the number of sequences, which, to date, exceeds 100,000 sequences generated to track the pandemic on the continent. These sequences have profoundly affected how public health officials in Africa have navigated the COVID-19 pandemic. RATIONALE We demonstrate how the first 100,000 SARS-CoV-2 sequences from Africa have helped monitor the epidemic on the continent, how genomic surveillance expanded over the course of the pandemic, and how we adapted our sequencing methods to deal with an evolving virus. Finally, we also examine how viral lineages have spread across the continent in a phylogeographic framework to gain insights into the underlying temporal and spatial transmission dynamics for several variants of concern (VOCs). RESULTS Our results indicate that the number of countries in Africa that can sequence the virus within their own borders is growing and that this is coupled with a shorter turnaround time from the time of sampling to sequence submission. Ongoing evolution necessitated the continual updating of primer sets, and, as a result, eight primer sets were designed in tandem with viral evolution and used to ensure effective sequencing of the virus. The pandemic unfolded through multiple waves of infection that were each driven by distinct genetic lineages, with B.1-like ancestral strains associated with the first pandemic wave of infections in 2020. Successive waves on the continent were fueled by different VOCs, with Alpha and Beta cocirculating in distinct spatial patterns during the second wave and Delta and Omicron affecting the whole continent during the third and fourth waves, respectively. Phylogeographic reconstruction points toward distinct differences in viral importation and exportation patterns associated with the Alpha, Beta, Delta, and Omicron variants and subvariants, when considering both Africa versus the rest of the world and viral dissemination within the continent. Our epidemiological and phylogenetic inferences therefore underscore the heterogeneous nature of the pandemic on the continent and highlight key insights and challenges, for instance, recognizing the limitations of low testing proportions. We also highlight the early warning capacity that genomic surveillance in Africa has had for the rest of the world with the detection of new lineages and variants, the most recent being the characterization of various Omicron subvariants. CONCLUSION Sustained investment for diagnostics and genomic surveillance in Africa is needed as the virus continues to evolve. This is important not only to help combat SARS-CoV-2 on the continent but also because it can be used as a platform to help address the many emerging and reemerging infectious disease threats in Africa. In particular, capacity building for local sequencing within countries or within the continent should be prioritized because this is generally associated with shorter turnaround times, providing the most benefit to local public health authorities tasked with pandemic response and mitigation and allowing for the fastest reaction to localized outbreaks. These investments are crucial for pandemic preparedness and response and will serve the health of the continent well into the 21st century

    Produção científica: reflexos da avaliação nos programas de pós-graduação em Física

    Based on the analysis of the scientific literature in graduate programs in the field of Physics at the Universities Federal de Minas Gerais (UFMG), Pernambuco (UFPE) and Rio de Janeiro (UFRJ), in the period 2007-2009 it used methodological triangulation to contemplate easing of quantitative methods, bibliometric and scientometric for scientific production, with qualitative and interpretive content analysis, for interviews. The main results found the existence of incentive instruments and control of scientific production in these institutions, the predominance of teachers’ work published in journals that are well regarded by the scientific community and are well ranked, according to the classification system of agency promotion (Qualis B); concern predominance of evaluation of scientific literature, in a purely quantitative, and policy assessment CAPES the time that encourages, pushes researchers through documents to a context of scientific productivity.Partindo da análise da produção científica nos programas de pós-graduação na área de Física das Universidades Federais de Minas Gerais, Pernambuco e Rio de Janeiro, no período de 2007 a 2009, utilizou-se a triangulação metodológica por contemplar a flexibilização dos métodos de natureza quantitativa (bibliometria e cientometria) para a produção científica; e qualitativa e interpretativa (análise de conteúdo) para documentos normatizadores e entrevistas. Os principais resultados constataram a existência de instrumentos de incentivo e controle da produção científica nas referidas instituições; a predominância de trabalhos dos docentes publicados em periódicos que são bem considerados perante a comunidade científica e bem classificados segundo o sistema de classificação da agência de fomento (Qualis B); preocupação quanto à predominância de avaliação da produção científica de forma puramente quantitativa; e política de avaliação da CAPES que ao mesmo tempo estimula e pressiona os pesquisadores por meio de documentos a um contexto de produtividade científica.

    Adipose Tissue Fibrosis In Human Cancer Cachexia: The Role Of Tgf Beta Pathway

Loss of white adipose tissue (WAT) is one of the key characteristics of cachexia. WAT wasting is paralleled by microarchitectural remodeling in cachectic cancer patients. Fibrosis results from uncontrolled ECM synthesis, a process in which, transforming growth factor-beta (TGF beta) plays a pivotal role. So far, the mechanisms involved in adipose tissue (AT) re-arrangement, and the role of TGF beta in inducing AT remodeling in weight-losing cancer patients are poorly understood. This study examined the modulation of ECM components mediated by TGF beta pathway in fibrotic AT obtained from cachectic gastrointestinal cancer patients. Methods: After signing the informed consent form, patients were enrolled into the following groups: cancer cachexia (CC, n = 21), weight-stable cancer (WSC, n = 17), and control (n = 21). The total amount of collagen and elastic fibers in the subcutaneous AT was assessed by histological analysis and by immunohistochemistry. TGF beta isoforms expression was analyzed by Multiplex assay and by immunohistochemistry. Alpha-smooth muscle actin (aSMA), fibroblast-specific protein (FSP1), Smad3 and 4 were quantified by qPCR and/or by immunohistochemistry. Interleukin (IL) 2, IL5, IL8, IL13 and IL17 content, cytokines known to be associated with fibrosis, was measured by Multiplex assay. Results: There was an accumulation of collagen and elastic fibers in the AT of CC, as compared with WSC and controls. Collagens type I, III, VI, and fibronectin expression was enhanced in the tissue of CC, compared with both WSC and control. The pronounced expression of aSMA in the surrounding of adipocytes, and the increased mRNA content for FSP1 (20-fold) indicate the presence of activated myofibroblasts; particularly in CC. TGF beta 1 and TGF beta 3 levels were up-regulated by cachexia in AT, as well in the isolated adipocytes. Smad3 and Smad4 labeling was found to be more evident in the fibrotic areas of CC adipose tissue.
