    Is Screening Cargo Containers for Smuggled Nuclear Threats Worthwhile?

    In recent years, Customs and Border Protection (CBP) has installed radiation sensors to screen cargo containers entering theUnited States. They are concerned that terrorists could use containers to smuggle radiological material into the country and carry out attacks with dirty bombs or a nuclear device. Recent studies have questioned the value of improving this screening system with new sensor technology. The cost of delays caused by frequent false alarms outweighs any reduction in the probability of an attack in an expected cost analysis. We extend existing methodology in three ways to demonstrate how additional factors affect the value of screening investments. We examine the effect that screening has in discouraging terrorists. We model multiple levels of screening. Finally, we consider additional objectives beyond cost. We find that the conclusion about screening depends on key inputs to the probability model (reflecting uncertainties) and to the value function (reflecting the stakeholders’ fundamental objectives)

    Evidence of Varroa-mediated Deformed Wing virus spillover in Hawaii

    Varroa destructor, a parasitic mite of honey bees, is also a vector for viral diseases. The mite displays high host specificity and requires access to colonies of Apis spp. to complete its lifecycle. In contrast, the Deformed Wing Virus (DWV), one of the many viruses transmitted by V. destructor, appears to have a much broader host range. Previous studies have detected DWV in a variety of insect groups that are not directly parasitized by the mite. In this study, we take advantage of the discrete distribution of the Varroa mite in the Hawaiian archipelago to compare DWV prevalence on non-Apis flower visitors, and test whether Varroa presence is linked to a “viral spillover”. We selected two islands with different viral landscapes: Oahu, where V. destructor has been present since 2007, and Maui, where the mite is absent. We sampled individuals of Apis mellifera, Ceratina smaragdula, Polistes aurifer, and Polistes exclamens, to assess and compare the DWV prevalence in the Hymenoptera community of the two islands. The results indicated that, as expected, honey bee colonies on Oahu have much higher incidence of DWV compared to Maui. Correspondingly, DWV was detected on the Non-Apis Hymenoptera collected from Oahu, but was absent in the species examined on Maui. The study sites selected shared a similar geography, climate, and insect fauna, but differed in the presence of the Varroa mite, suggesting an indirect, but significant, increase on DWV prevalence in the Hymenoptera community on mite-infected islands

    Pilot Study of Endurance Runners and Brain Responses Associated with Delay Discounting

High levels of endurance training have been associated with potentially negative health outcomes and addictive-like symptoms such as exercise in the presence of injury and higher levels of impulsivity. This pilot study examined the relationships among self-report measures of addictive symptoms related to exercise and behavioral and neural measures of impulsivity in endurance runners. We hypothesized endurance runners would have increased preference for immediate rewards and greater activation of cognitive control regions when making decisions involving delayed rewards. Twenty endurance runners (at least 20 miles/week) were recruited to undergo measures of self-report exercise addiction symptoms, impulsive decision-making (delay discounting) and functional magnetic resonance imaging (fMRI). During behavioral and fMRI examinations, participants chose between a small hypothetical amount of money given immediately (0–100)comparedtoalargerhypotheticalamountofmoney(0 – 100) compared to a larger hypothetical amount of money (100) given after a delay (2-12 weeks). On half of the trials participants were instructed that if they chose the delayed reward they would not be able to exercise during the delay period. Eighteen participants were included in the analysis. Results indicated that 94% of endurance runners reported high levels of exercise addiction symptoms, and 44% were “at-risk” for exercise addiction. In addition, endurance runners demonstrated increased preference for immediately available compared to delayed rewards (p \u3c 0.001) and greater recruitment of cognitive control regions (dorsomedial prefrontal cortex and anterior cingulate) when making decisions involving rewards when exercise was delayed (p \u3c 0.05). Together, these results indicate that endurance runners not only report addictive symptoms related to exercise, but also demonstrate addictive-like behaviors

    Gambogic Acid, a Natural Product Inhibitor of Hsp90

    A high-throughput screening of natural product libraries identified (−)-gambogic acid (1), a component of the exudate of Garcinia harburyi, as a potential Hsp90 inhibitor, in addition to the known Hsp90 inhibitor celastrol (2). Subsequent testing established that 1 inhibited cell proliferation, brought about the degradation of Hsp90 client proteins in cultured cells, and induced the expression of Hsp70 and Hsp90, which are hallmarks of Hsp90 inhibition. Gambogic acid also disrupted the interaction of Hsp90, Hsp70, and Cdc37 with the heme-regulated eIF2α kinase (HRI, an Hsp90-dependent client) and blocked the maturation of HRI in vitro. Surface plasmon resonance spectroscopy indicated that 1 bound to the N-terminal domain of Hsp90 with a low micromolar Kd, in a manner that was not competitive with the Hsp90 inhibitor geldanamycin (3). Molecular docking experiments supported the posit that 1 binds Hsp90 at a site distinct from Hsp90s ATP binding pocket. The data obtained have firmly established 1 as a novel Hsp90 inhibitor and have provided evidence of a new site that can be targeted for the development of improved Hsp90 inhibitors

    Unilateral Disruptions in the Default Network with Aging in Native Space

    BACKGROUND: Disruption of the default-mode network (DMN) in healthy elders has been reported in many studies. METHODS: In a group of 51 participants (25 young, 26 elder) we examined DMN connectivity in subjects' native space. In the native space method, subject-specific regional masks (obtained independently for each subject) are used to extract regional fMRI times series. This approach substitutes the spatial normalization and subsequent smoothing used in prevailing methods, affords more accurate spatial localization, and provides the power to examine connectivity separately in the two hemispheres instead of averaging regions across hemispheres. RESULTS: The native space method yielded new findings which were not detectable by the prevailing methods. The most reliable and robust disruption in elders' DMN connectivity were found between supramarginal gyrus and superior-frontal cortex in the right hemisphere only. The mean correlation between these two regions in young participants was about 0.5, and dropped significantly to 0.04 in elders (P = 2.1 x 10(-5)). In addition, the magnitude of functional connectivity between these regions in the right hemisphere correlated with memory (P = 0.05) and general fluid ability (P = 0.01) in elder participants and with speed of processing in young participants (P = 0.008). These relationships were not observed in the left hemisphere. CONCLUSION: These findings suggest that analysis of DMN connectivity in subjects' native space can improve localization and power and that it is important to examine connectivity separately in each hemisphere

    Visual attentional load influences plasticity in the human motor cortex

    Neural plasticity plays a critical role in learning, memory, and recovery from injury to the nervous system. Although much is known about the physical and physiological determinants of plasticity, little is known about the influence of cognitive factors. In this study, we investigated whether selective attention plays a role in modifying changes in neural excitability reflecting long-term potentiation (LTP)like plasticity. We induced LTP-like effects in the hand area of the human motor cortex using transcranial magnetic stimulation (TMS). During the induction of plasticity, participants engaged in a visual detection task with either low or high attentional demands. Changes in neural excitability were assessed by measuring motor-evoked potentials in a small hand muscle before and after the TMS procedures. In separate experiments plasticity was induced either by paired associative stimulation (PAS) or intermittent theta-burst stimulation (iTBS). Because these procedures induce different forms of LTP-like effects, they allowed us to investigate the generality of any attentional influence on plasticity. In both experiments reliable changes in motor cortex excitability were evident under low-load conditions, but this effect was eliminated under high-attentional load. In a third experiment we investigated whether the attentional task was associated with ongoing changes in the excitability of motor cortex, but found no difference in evoked potentials across the levels of attentional load. Our findings indicate that in addition to their role in modifying sensory processing, mechanisms of attention can also be a potent modulator of cortical plasticity

    Evaluating a Modular Decision Support Application For Colorectal Cancer Screening

    BACKGROUND: There is a need for health information technology evaluation that goes beyond randomized controlled trials to include consideration of usability, cognition, feedback from representative users, and impact on efficiency, data quality, and clinical workflow. This article presents an evaluation illustrating one approach to this need using the Decision-Centered Design framework. OBJECTIVE: To evaluate, through a Decision-Centered Design framework, the ability of the Screening and Surveillance App to support primary care clinicians in tracking and managing colorectal cancer testing. METHODS: We leveraged two evaluation formats, online and in-person, to obtain feedback from a range primary care clinicians and obtain comparative data. Both the online and in-person evaluations used mock patient data to simulate challenging patient scenarios. Primary care clinicians responded to a series of colorectal cancer-related questions about each patient and made recommendations for screening. We collected data on performance, perceived workload, and usability. Key elements of Decision-Centered Design include evaluation in the context of realistic, challenging scenarios and measures designed to explore impact on cognitive performance. RESULTS: Comparison of means revealed increases in accuracy, efficiency, and usability and decreases in perceived mental effort and workload when using the Screening and Surveillance App. CONCLUSION: The results speak to the benefits of using the Decision-Centered Design approach in the analysis, design, and evaluation of Health Information Technology. Furthermore, the Screening and Surveillance App shows promise for filling decision support gaps in current electronic health records

    Prey of reintroduced fishers and their habitat relationships in the Cascades T Range, Washington

    Conservation and recovery of forest carnivores requires an understanding of their habitat requirements, as well as requirements of their prey. In much of the western United States, trapping and habitat loss led to extirpations of fishers (Pekania pennanti) by the mid-20th century, and reintroductions are ongoing to restore fishers to portions of their former range. Fisher recovery in Washington State has been limited by isolation from other populations, but other potentially important factors, such as diet of fishers in this region and prey availability, have not been thoroughly investigated. We collected hair samples from potential prey and fishers for stable isotope analysis to identify important prey items for fishers within a reintroduction area in southern Washington. We then estimated the abundance of prey species at 21 sites across a gradient of forest structural classes within the fisher reintroduction area, and assessed the effects of forest age and vegetation on the prey community using permutational multivariate analysis of variance and non-metric multidimensional scaling. Stable isotopes revealed that larger prey items, including snowshoe hares (Lepus americanus) and/or mountain beavers (Aplodontia rufa), were the most important prey item(s) for fishers in the southern Cascades. We found distinct but equally diverse prey communities in old-growth (unmanaged) and young (heavily managed) forest stands, with snowshoe hares and mountain beavers most common in young forests, while chipmunks (Neotamius spp.) and small mammals were more common in older forests. Our results suggest a discrepancy between the habitats where important fisher prey are most abundant and habitat requirements of fishers. Snowshoe hares and mountain beavers were most abundant in young forests, whereas fishers are associated with landscapes dominated by older forest stands or those that provide large woody structures, which fishers use for denning and resting. Our results add to growing evidence that forest landscape mosaics provide valuable habitat for fishers in the Pacific Northwest, suggesting that both mature and younger forest stands are important for fishers and fisher recovery
