Presuming his profession to be a herald of an integrated U.S. nation-state, Bache guided organizations such as the United States Coast Survey, then the country's largest scientific enterprise. In this analytical biography, Axel Jansen explains Bache's efforts to build and shape public institutions as a national foundation for a universalistic culture - efforts that culminated during the Civil War when Bache helped found the National Academy of Sciences as a symbol for the continued viability of an American nation

    Archazolid and apicularen: Novel specific V-ATPase inhibitors

    BACKGROUND: V-ATPases constitute a ubiquitous family of heteromultimeric, proton translocating proteins. According to their localization in a multitude of eukaryotic membranes, they energize many different transport processes. Since their malfunction is correlated with various diseases in humans, the elucidation of the properties of this enzyme for the development of selective inhibitors and drugs is one of the challenges in V-ATPase research. RESULTS: Archazolid A and B, two recently discovered cytotoxic macrolactones produced by the myxobacterium Archangium gephyra, and apicularen A and B, two novel benzolactone enamides produced by different species of the myxobacterium Chondromyces, exerted a similar inhibitory efficacy on a wide range of mammalian cell lines as the well established plecomacrolidic type V-ATPase inhibitors concanamycin and bafilomycin. Like the plecomacrolides both new macrolides also prevented the lysosomal acidification in cells and inhibited the V-ATPase purified from the midgut of the tobacco hornworm, Manduca sexta, with IC(50 )values of 20–60 nM. However, they did not influence the activity of mitochondrial F-ATPase or that of the Na(+)/K(+)-ATPase. To define the binding sites of these new inhibitors we used a semi-synthetic radioactively labelled derivative of concanamycin which exclusively binds to the membrane V(o )subunit c. Whereas archazolid A prevented, like the plecomacrolides concanamycin A, bafilomycin A(1 )and B(1), labelling of subunit c by the radioactive I-concanolide A, the benzolactone enamide apicularen A did not compete with the plecomacrolide derivative. CONCLUSION: The myxobacterial antibiotics archazolid and apicularen are highly efficient and specific novel inhibitors of V-ATPases. While archazolid at least partly shares a common binding site with the plecomacrolides bafilomycin and concanamycin, apicularen adheres to an independent binding site

    Detection of Emission from the CN Radical in the Cloverleaf Quasar at z=2.56

    We report the detection of CN(N=3-2) emission towards the Cloverleaf quasar (z=2.56) based on observations with the IRAM Plateau de Bure Interferometer. This is the first clear detection of emission from this radical at high redshift. CN emission is a tracer of dense molecular hydrogen gas (n(H2) > 10^4 cm^{-3}) within star-forming molecular clouds, in particular in regions where the clouds are affected by UV radiation. The HCN/CN intensity ratio can be used as a diagnostic for the relative importance of photodissociation regions (PDRs) in a source, and as a sensitive probe of optical depth, the radiation field, and photochemical processes. We derive a lensing-corrected CN(N=3-2) line luminosity of L'(CN(3-2) = (4.5 +/- 0.5) x 10^9 K km/s pc^2. The ratio between CN luminosity and far-infrared luminosity falls within the scatter of the same relationship found for low-z (ultra-) luminous infrared galaxies. Combining our new results with CO(J=3-2) and HCN(J=1-0) measurements from the literature and assuming thermal excitation for all transitions, we find a CO/CN luminosity ratio of 9.3 +/- 1.9 and a HCN/CN luminosity ratio of 0.95 +/- 0.15. However, we find that the CN(N=3-2) line is likely only subthermally excited, implying that those ratios may only provide upper limits for the intrinsic 1-0 line luminosity ratios. We conclude that, in combination with other molecular gas tracers like CO, HCN, and HCO+, CN is an important probe of the physical conditions and chemical composition of dense molecular environments at high redshift.Comment: 6 pages, 5 figures, 1 table, to appear in ApJ (accepted May 23, 2007

    Increased incidence of syphilis in men who have sex with men and risk management strategies, Germany, 2015.

    In Germany, the number of reported syphilis cases increased between 11% and 22% per year between 2010 and 2014. We analysed syphilis surveillance data and data of four behavioural surveys on men who have sex with men (MSM) in Germany (2003, 2007, 2010, 2013) to assess if this rise is ongoing and to find possible explanations for it. Syphilis notifications increased in 2015 by 19% to a total of 6,834. This was mainly due to increasing notifications in MSM of all age groups in larger German cities. Data from the behavioural surveys on MSM in Germany showed a simultaneous increase of selective condom use as HIV-status-bases risk management strategy and the number of syphilis cases. MSM diagnosed with HIV reported condomless anal intercourse with non-steady partners more frequent than MSM not diagnosed with HIV or untested for HIV, but the latter also reported higher frequencies of this behaviour in the more recent surveys. Transmission in HIV-positive MSM probably plays an important, but not exclusive role, for the syphilis dynamics in Germany. A risk adapted routine screening for sexually active MSM and potentially innovative approaches to increase early screening and treatment of syphilis such as internet counselling, home sampling, home testing and broadening venue-based (rapid) testing, should be critically evaluated to effectively reduce syphilis infections

    Development of a high-throughput colorimetric Zika virus infection assay

    Zika virus (ZIKV) is an emerging pathogen that causes congenital infections which may result in birth defects, such as microcephaly. Currently, no approved treatment or vaccination is available. ZIKV can be readily detected in cell culture where virally infected cells are normally stained by specific antibodies. As ZIKV regularly causes a cytopathic effect, we were wondering whether this viral property can be used to quantitatively determine viral infectivity. We here describe the use of an 3-[4,5-dimethyl-2-thiazolyl]-2,5-diphenyl-2H-tetrazolium bromide-(MTT)-based cell viability assay that allows to determine ZIKV-induced cell death. We show that this colorimetric assay quantifies ZIKV infection over a broad range of viral dilutions in both monkey and human cells. It allows to determine inhibitory activities of antivirals that block ZIKV or to define the neutralizing antibody titers of ZIKV antisera. This MTT-based ZIKV detection assay can be evaluated by naked eye or computational tools, has a broad linear range, does not require large equipment or costly reagents, and thus represents a promising alternative to antibody-based assays, in particular in resource-poor settings. We propose to use this simple, fast, and cheap method for quantification of ZIKV neutralizing antibodies and testing of antiviral compounds.Peer reviewe

    Recent genetic studies found the A allele of the variant rs1006737 in the alpha 1C subunit of the L-type voltage-gated calcium channel (CACNA1C) gene to be overrepresented in patients suffering from bipolar disorder, schizophrenia or major depression. While the functions underlying the pathophysiology of these psychiatric disorders are yet unknown, impaired performance in verbal fluency tasks is an often replicated finding. We investigated the influence of the rs1006737 single nucleotide polymorphism (SNP) on verbal fluency and its neural correlates.Brain activation was measured with functional magnetic resonance imaging (fMRI) during a semantic verbal fluency task in 63 healthy male individuals. They additionally performed more demanding verbal fluency tasks outside the scanner. All subjects were genotyped for CACNA1C rs1006737.For the behavioral measures outside the scanner, rs1006737genotype had an effect on semantic but not on lexical verbal fluency with decreased performance in risk-allele carriers. In the fMRI experiment, while there were no differences in behavioural performance, increased activation in the left inferior frontal gyrus as well as the left precuneus was found in risk-allele carriers in the semantic verbal fluency task.The rs1006737 variant does influence language production on a semantic level in conjunction with the underlying neural systems. These findings are in line with results of studies in bipolar disorder, schizophrenia and major depression and may explain some of the cognitive and brain activation variation found in these disorders
