C-peptide, Na+,K+-ATPase, and Diabetes
Na+,K+-ATPase is an ubiquitous membrane enzyme
that allows the extrusion of three sodium ions from the cell
and two potassium ions from the extracellular fluid. Its activity
is decreased in many tissues of streptozotocin-induced
diabetic animals. This impairment could be at least partly
responsible for the development of diabetic complications.
Na+,K+-ATPase activity is decreased in the red blood cell
membranes of type 1 diabetic individuals, irrespective of the
degree of diabetic control. It is less impaired or even normal
in those of type 2 diabetic patients. The authors have
shown that in the red blood cells of type 2 diabetic patients,
Na+,K+-ATPase activity was strongly related to blood C-peptide
levels in non–insulin-treated patients (in whom C-peptide
concentration reflects that of insulin) as well as in
insulin-treated patients. Furthermore, a gene-environment
relationship has been observed. The alpha-1 isoform of the
enzyme predominant in red blood cells and nerve tissue is
encoded by the ATP1A1 gene.Apolymorphism in the intron
1 of this gene is associated with lower enzyme activity in patients
with C-peptide deficiency either with type 1 or type
2 diabetes, but not in normal individuals. There are several
lines of evidence for a low C-peptide level being responsible
for low Na+,K+-ATPase activity in the red blood cells.
Short-term C-peptide infusion to type 1 diabetic patients
restores normal Na+,K+-ATPase activity. Islet transplantation,
which restores endogenous C-peptide secretion, enhances
Na+,K+-ATPase activity proportionally to the rise
in C-peptide. This C-peptide effect is not indirect. In fact,
incubation of diabetic red blood cells with C-peptide at
physiological concentration leads to an increase of Na+,K+-ATPase activity. In isolated proximal tubules of rats or
in the medullary thick ascending limb of the kidney, C-peptide stimulates in a dose-dependent manner Na+,K+-ATPase activity. This impairment in Na+,K+-ATPase activity,
mainly secondary to the lack of C-peptide, plays probably
a role in the development of diabetic complications.
Arguments have been developed showing that the diabetesinduced
decrease in Na+,K+-ATPase activity compromises
microvascular blood flow by two mechanisms: by affecting
microvascular regulation and by decreasing red blood cell
deformability, which leads to an increase in blood viscosity.
C-peptide infusion restores red blood cell deformability
and microvascular blood flow concomitantly with Na+,K+-ATPase activity. The defect in ATPase is strongly related to
diabetic neuropathy. Patients with neuropathy have lower
ATPase activity than those without. The diabetes-induced
impairment in Na+,K+-ATPase activity is identical in red
blood cells and neural tissue. Red blood cell ATPase activity
is related to nerve conduction velocity in the peroneal
and the tibial nerve of diabetic patients. C-peptide infusion
to diabetic rats increases endoneural ATPase activity in rat.
Because the defect in Na+,K+-ATPase activity is also probably
involved in the development of diabetic nephropathy and
cardiomyopathy, physiological C-peptide infusion could be
beneficial for the prevention of diabetic complications
Ab initio phonon dispersion curves and interatomic force constants of barium titanate
The phonon dispersion curves of cubic BaTiO_3 have been computed within a
first-principles approach and the results compared to the experimental data.
The curves obtained are very similar to those reported for KNbO_3 by Yu and
Krakauer [Phys. Rev. Lett. 74, 4067 (1995)]. They reveal that correlated atomic
displacements along chains are at the origin of the ferroelectric
instability. A simplified model illustrates that spontaneous collective
displacements will occur when a dozen of aligned atoms are coupled. The
longitudinal interatomic force constant between nearest neighbour Ti and O
atoms is relatively weak in comparison to that between Ti atoms in adjacent
cells. The small coupling between Ti and O displacements seems however
necessary to reproduce a ferroelectric instability.Comment: 12 pages, 4 figure

Fishery-specific solutions to seabird bycatch in the U.S. West Coast sablefish fishery
Bird scaring lines (BSLs) protect longline fishing gear from seabird attacks, save bait, reduce incidental seabird mortality and are the most commonly prescribed seabird bycatch mitigation measure worldwide. We collaborated with fishermen to assess the efficacy of applying BSL regulations from the demersal longline sablefish fishery in Alaska to a similar fishery along the U.S West Coast. In contrast to Alaska, some U.S. West Coast vessels use floats along the line to keep hooks off the seafloor, where scavengers degrade the bait and the target catch. Our results confirmed that BSL regulations from Alaska were sufficient to protect baits from bird attacks on longlines without floats, but not baits on longlines with floats. Longlines with floats sank below the reach of albatrosses (2 m depth) at a distance astern (157.7 m ± 44.8 95% CI) that was 2.3 times farther than longlines without floats (68.8 m ± 37.8 95% CI). The floated longline distance was well beyond the protection afforded by BSLs, which is approximately 40 m of aerial extent. Black-footed albatross attacked floated longlines at rates ten times more (2.7 attacks/1000 hooks, 0.48–4.45 95%CI) than longlines without floats (0.20 attacks/1000 hooks, 0.01–0.36 95% CI). Retrospective analysis of NOAA Fisheries Groundfish Observer Program data suggested that seabird bycatch occurs in a few sablefish longline fishing sectors and a minority of vessels, but is not confined to larger vessels. Analysis also confirmed fishermen testimonials that night setting reduced albatross bycatch by an order of magnitude compared to daytime setting, without reducing target catch. Night setting could be an effective albatross bycatch prevention practice if applied to the U.S. West Coast sablefish longline fishery and provide a practical alternative for vessels that elect to use floated longlines. These results highlight the importance of understanding region-specific longline gear modifications to identify effective bycatch reduction tools and the value of working collaboratively with fishermen to craft solutions
In C. elegans, High Levels of dsRNA Allow RNAi in the Absence of RDE-4
C. elegans Dicer requires an accessory double-stranded RNA binding protein, RDE-4, to enact the first step of RNA interference, the cleavage of dsRNA to produce siRNA. While RDE-4 is typically essential for RNAi, we report that in the presence of high concentrations of trigger dsRNA, rde-4 deficient animals are capable of silencing a transgene. By multiple criteria the silencing occurs by the canonical RNAi pathway. For example, silencing is RDE-1 dependent and exhibits a decrease in the targeted mRNA in response to an increase in siRNA. We also find that high concentrations of dsRNA trigger lead to increased accumulation of primary siRNAs, consistent with the existence of a rate-limiting step during the conversion of primary to secondary siRNAs. Our studies also revealed that transgene silencing occurs at low levels in the soma, even in the presence of ADARs, and that at least some siRNAs accumulate in a temperature-dependent manner. We conclude that an RNAi response varies with different conditions, and this may allow an organism to tailor a response to specific environmental signals
Thermal conductivity of refractory glass fibres
In the present study, the current international
standards and corresponding apparatus for measuring the
thermal conductivity of refractory glass fibre products have
been reviewed. Refractory glass fibres are normally produced
in the form of low-density needled mats. A major
issue with thermal conductivity measurements of these
materials is lack of reproducibility in the test results due to
transformation of the test material during the test. Also
needled mats are inherently inhomogeneous, and this poses
additional problems. To be able to compare the various
methods of thermal conductivity measurement, a refractory
reference material was designed which is capable of
withstanding maximum test temperatures (1673 K) with
minimum transformation. The thermal conductivity of this
reference material was then measured using various
methods according to the different standards surveyed. In
order to compare different materials, samples have been
acquired from major refractory glass fibre manufacturers
and the results have been compared against the newly
introduced reference material. Materials manufactured by
melt spinning, melt blowing and sol–gel have been studied,
and results compared with literature values
RACK1 Is a Ribosome Scaffold Protein for β-actin mRNA/ZBP1 Complex
In neurons, specific mRNAs are transported in a translationally repressed manner along dendrites or axons by transport ribonucleic-protein complexes called RNA granules. ZBP1 is one RNA binding protein present in transport RNPs, where it transports and represses the translation of cotransported mRNAs, including β-actin mRNA. The release of β-actin mRNA from ZBP1 and its subsequent translation depends on the phosphorylation of ZBP1 by Src kinase, but little is known about how this process is regulated. Here we demonstrate that the ribosomal-associated protein RACK1, another substrate of Src, binds the β-actin mRNA/ZBP1 complex on ribosomes and contributes to the release of β-actin mRNA from ZBP1 and to its translation. We identify the Src binding and phosphorylation site Y246 on RACK1 as the critical site for the binding to the β-actin mRNA/ZBP1 complex. Based on these results we propose RACK1 as a ribosomal scaffold protein for specific mRNA-RBP complexes to tightly regulate the translation of specific mRNAs
ADAM33, a New Candidate for Psoriasis Susceptibility
Psoriasis is a chronic skin disorder with multifactorial etiology. In a recent study, we reported results of a genome-wide scan on 46 French extended families presenting with plaque psoriasis. In addition to unambiguous linkage to the major susceptibility locus PSORS1 on Chromosome 6p21, we provided evidence for a susceptibility locus on Chromosome 20p13. To follow up this novel psoriasis susceptibility locus we used a family-based association test (FBAT) for an association scan over the 17 Mb candidate region. A total of 85 uncorrelated SNP markers located in 65 genes of the region were initially investigated in the same set of large families used for the genome wide search, which consisted of 295 nuclear families. When positive association was obtained for a SNP, candidate genes nearby were explored more in detail using a denser set of SNPs. Thus, the gene ADAM33 was found to be significantly associated with psoriasis in this family set (The best association was on a 3-SNP haplotype P = 0.00004, based on 1,000,000 permutations). This association was independent of PSORS1. ADAM33 has been previously associated with asthma, which demonstrates that immune system diseases may be controlled by common susceptibility genes with general effects on dermal inflammation and immunity. The identification of ADAM33 as a psoriasis susceptibility gene identified by positional cloning in an outbred population should provide insights into the pathogenesis and natural history of this common disease
Receptor for Activated Protein Kinase C: Requirement for Efficient MicroRNA Function and Reduced Expression in Hepatocellular Carcinoma
MicroRNAs (miRNAs) are important regulators of gene expression that control physiological and pathological processes. A global reduction in miRNA abundance and function is a general trait of human cancers, playing a causal role in the transformed phenotype. Here, we sought to newly identify genes involved in the regulation of miRNA function by performing a genetic screen using reporter constructs that measure miRNA function and retrovirus-based random gene disruption. Of the six genes identified, RACK1, which encodes “receptor for activated protein kinase C” (RACK1), was confirmed to be necessary for full miRNA function. RACK1 binds to KH-type splicing regulatory protein (KSRP), a member of the Dicer complex, and is required for the recruitment of mature miRNAs to the RNA-induced silencing complex (RISC). In addition, RACK1 expression was frequently found to be reduced in hepatocellular carcinoma. These findings suggest the involvement of RACK1 in miRNA function and indicate that reduced miRNA function, due to decreased expression of RACK1, may have pathologically relevant roles in liver cancers
Genetic polymorphisms of MMP1, MMP3 and MMP7 gene promoter and risk of colorectal adenoma
BACKGROUND: Matrix metalloproteinases (MMP) have been shown to play a role in colorectal cancer (CRC). More recently, MMP1, MMP3 and MMP7 functional gene promoter polymorphisms have been found to be associated with CRC occurrence and prognosis. To document the role of MMP polymorphisms in the early step of colorectal carcinogenesis, we investigated their association with colorectal adenoma risk in a case-control study comprising 295 patients with large adenomas (LA), 302 patients with small adenomas (SA) and 568 polyp-free (PF) controls. METHODS: Patients were genotyped using automated fragment analysis for MMP1 -1607 ins/del G and MMP3 -1612 ins/delA (MMP3.1) polymorphisms and allelic discrimination assay for MMP3 -709 A/G (MMP3.2) and MMP7 -181 A/G polymorphisms. Association between MMP genotypes and colorectal adenomas was first tested for each polymorphism separately and then for combined genotypes using the combination test. Adjustment on relevant variables and estimation of odds ratios were performed using unconditional logistic regression. RESULTS: No association was observed between the polymorphisms and LA when compared to PF or SA. When comparing SA to PF controls, analysis revealed a significant association between MMP3 -1612 ins/delA polymorphism and SA with an increased risk associated with the 6A/6A genotype (OR = 1.67, 95%CI: 1.20–2.34). Using the combination test, the best association was found for MMP3.1-MMP1 (p = 0.001) with an OR of 1.88 (95%CI: 1.08–3.28) for the combined genotype 2G/2G-6A/6A estimated by logistic regression. CONCLUSION: These data show a relation between MMP1 -1607 ins/del G and MMP3 -1612 ins/delA combined polymorphisms and risk of SA, suggesting their potential role in the early steps of colorectal carcinogenesis
Melanocortin-1 Receptor, Skin Cancer and Phenotypic Characteristics (M-SKIP) Project: Study Design and Methods for Pooling Results of Genetic Epidemiological Studies
Background: For complex diseases like cancer, pooled-analysis of individual data represents a powerful tool to investigate the joint contribution of genetic, phenotypic and environmental factors to the development of a disease. Pooled-analysis of epidemiological studies has many advantages over meta-analysis, and preliminary results may be obtained faster and with lower costs than with prospective consortia. Design and methods: Based on our experience with the study design of the Melanocortin-1 receptor (MC1R) gene, SKin cancer and Phenotypic characteristics (M-SKIP) project, we describe the most important steps in planning and conducting a pooled-analysis of genetic epidemiological studies. We then present the statistical analysis plan that we are going to apply, giving particular attention to methods of analysis recently proposed to account for between-study heterogeneity and to explore the joint contribution of genetic, phenotypic and environmental factors in the development of a disease. Within the M-SKIP project, data on 10,959 skin cancer cases and 14,785 controls from 31 international investigators were checked for quality and recoded for standardization. We first proposed to fit the aggregated data with random-effects logistic regression models. However, for the M-SKIP project, a two-stage analysis will be preferred to overcome the problem regarding the availability of different study covariates. The joint contribution of MC1R variants and phenotypic characteristics to skin cancer development will be studied via logic regression modeling. Discussion: Methodological guidelines to correctly design and conduct pooled-analyses are needed to facilitate application of such methods, thus providing a better summary of the actual findings on specific fields
