    Landscape of overlapping gene expression in the equine placenta

    Increasing evidence suggests that overlapping genes are much more common in eukaryotic genomes than previously thought. These different-strand overlapping genes are potential sense-antisense (SAS) pairs, which might have regulatory effects on each other. In the present study, we identified the SAS loci in the equine genome using previously generated stranded, paired-end RNA sequencing data from the equine chorioallantois. We identified a total of 1261 overlapping loci. The ratio of the number of overlapping regions to chromosomal length was numerically higher on chromosome 11 followed by chromosomes 13 and 12. These results show that overlapping transcription is distributed throughout the equine genome, but that distributions differ for each chromosome. Next, we evaluated the expression patterns of SAS pairs during the course of gestation. The sense and antisense genes showed an overall positive correlation between the sense and antisense pairs. We further provide a list of SAS pairs with both positive and negative correlation in their expression patterns throughout gestation. This study characterizes the landscape of sense and antisense gene expression in the placenta for the first time and provides a resource that will enable researchers to elucidate the mechanisms of sense/antisense regulation during pregnancy

    The Diboson Excess: Experimental Situation and Classification of Explanations; A Les Houches Pre-Proceeding

    We examine the `diboson' excess at ∼2\sim 2 TeV seen by the LHC experiments in various channels. We provide a comparison of the excess significances as a function of the mass of the tentative resonance and give the signal cross sections needed to explain the excesses. We also present a survey of available theoretical explanations of the resonance, classified in three main approaches. Beyond that, we discuss methods to verify the anomaly, determining the major properties of the various surpluses and exploring how different models can be discriminated. ISBN 978-0-9557497-0-4An international round-robin study of fire modelling was conducted prior to the Dalmarnock Fire Tests in order to assess the state-of-the-art of fire modelling in real scenarios. The philosophy behind the Dalmarnock Fire Tests was to provide instrumentation density suitable for comparison to field models and designed the scenario for maximum test reproducibility. Each participating team independently simulated a priori the test using a common detailed description of the compartment geometry, fuel packages, ignition source and ventilation conditions. The aim of the exercise was to forecast the test results as accurately as possible, and not to provide an engineering analysis with adequate conservative assumptions or safety factors. The modelling results and experimental measurements are compared among themselves, allowing for conclusions on the robustness, reliability and accuracy of current modelling practices. The results indicate large scatter and considerable disparity among predicted fires and also differing from the experimental data. The Dalmarnock Fire Test One was benchmarked against a second test to establish the potential experimental variability. The scatter of the simulations is much larger than the experimental error and the experimental variability. The study emphasises on the inherent difficulty of predicting fire dynamics and demonstrates that the main source of scatter is originated in the many degrees of freedom and the uncertainty in the input parameters. The conclusions from the study are made public to encourage debate and exchange of views on the topic of fire modelling

    Peer-reviewed journal paper published in 2009 about the international modelling exercise conducted in 2006.An international study of fire modelling was conducted prior to the Dalmarnock Fire Test One in order to assess the state-of-the-art of fire simulations using a round-robin approach. This test forms part of the Dalmarnock Fire Tests, a series of experiments conducted in 2006 in a high-rise building. The philosophy behind the tests was to provide measurements in a realistic fire scenario involving multiple fuel packages and non-trivial fire growth, and with an instrumentation density suitable for comparison with computational fluid dynamics models. Each of the seven round-robin teams independently simulated the test scenario a priori using a common detailed description of the compartment geometry, fuel packages, ignition source and ventilation conditions. The aim of the exercise was to forecast the fire development as accurately as possible and compare the results. The aim was not to provide an engineering analysis with conservative assumptions or safety factors. Comparison of the modelling results shows a large scatter and considerable disparity among the predictions, and between predictions and experimental measurements. The scatter of the simulations is much larger than the error and variability expected in the experiments. The study emphasises on the inherent difficulty of modelling fire dynamics in complex fire scenarios like Dalmarnock, and shows that the accuracy to predict fire growth (i.e. evolution of the heat released rate) is, in general, poor

    Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead

    Termite sensitivity to temperature affects global wood decay rates.

    Deadwood is a large global carbon store with its store size partially determined by biotic decay. Microbial wood decay rates are known to respond to changing temperature and precipitation. Termites are also important decomposers in the tropics but are less well studied. An understanding of their climate sensitivities is needed to estimate climate change effects on wood carbon pools. Using data from 133 sites spanning six continents, we found that termite wood discovery and consumption were highly sensitive to temperature (with decay increasing >6.8 times per 10°C increase in temperature)-even more so than microbes. Termite decay effects were greatest in tropical seasonal forests, tropical savannas, and subtropical deserts. With tropicalization (i.e., warming shifts to tropical climates), termite wood decay will likely increase as termites access more of Earth's surface

    IntroductionRegional anesthesia offers an alternative to general anesthesia and may be advantageous in low resource environments. There is a paucity of data regarding the practice of regional anesthesia in low- and middle-income countries. Using access data from a free Android app with curated regional anesthesia learning modules, we aimed to estimate global interest in regional anesthesia and potential applications to clinical practice stratified by World Bank income level.MethodsWe retrospectively analyzed data collected from the free Android app "Anesthesiologist" from December 2015 to April 2020. The app performs basic anesthetic calculations and provides links to videos on performing 12 different nerve blocks. Users of the app were classified on the basis of whether or not they had accessed the links. Nerve blocks were also classified according to major use (surgical block, postoperative pain adjunct, rescue block).ResultsPractitioners in low- and middle-income countries accessed the app more frequently than in high-income countries as measured by clicks. Users from low- and middle-income countries focused mainly on surgical blocks: ankle, axillary, infraclavicular, interscalene, and supraclavicular blocks. In high-income countries, more users viewed postoperative pain blocks: adductor canal, popliteal, femoral, and transverse abdominis plane blocks. Utilization of the app was constant over time with a general decline with the start of the COVID-19 pandemic.ConclusionThe use of an in app survey and analytics can help identify gaps and opportunities for regional anesthesia techniques and practices. This is especially impactful in limited-resource areas, such as lower-income environments and can lead to targeted educational initiatives
