    Body condition assessment using digital images.

    This project assessed the ability to assign a body condition score (BCS) to a dairy cow from digital photographs or videos. Images were taken from the rear of the cow at a 0 to 20 degrees angle relative to the tail head. Four observers assigned a BCS to each of 57 cows at a farm visit (live, farm 1) and later from a photograph (photo). Means +/- standard deviations of BCS by method and observer were as follows: live = 3.25 +/- 0.51, 3.42 +/- 0.49, 3.32 +/- 0.58, 3.13 +/- 0.62; photo = 3.36 +/- 0.52, 3.32 +/- 0.43, 3.44 +/- 0.62, 3.14 +/- 0.6 for observers 1 to 4, respectively. Body condition score means differed across observers for live (observer 2 higher and observer 4 lower, compared with observers 1 and 3) and photo methods (observer 3 lower, compared with observers 1, 2, and 3); however, within observer, the mean live BCS did not differ from the mean photo BCS. Correlation coefficients between BCS assigned live and from photos were 0.84, 0.82, 0.82, and 0.90 for observers 1 to 4, respectively. Subsequently, observer 1 visited 2 farms, assigned a live BCS, and digitally photographed 187 cows (56 and 131 cows from farms 2 and 3, respectively). Observers 2, 3, and 4 assigned a BCS from the photographs. Means +/- standard deviations of BCS by observer (method) were 1 (live) 3.35 +/- 0.55; 2 (photo) 3.33 +/- 0.49; 3 (photo) 3.60 +/- 0.54; and 4 (photo) 3.26 +/- 0.62. The mean BCS for observer 3 was higher and that for observer 4 was lower than for observers 1 and 2. Correlation coefficients between observer 1 and observers 2 through 4 were 0.78, 0.76, and 0.79, respectively. Observer 1 assigned a BCS to 41 cows at a farm visit and 3 wk later assessed the BCS of cows from a video taken at a farm visit by a different individual. Cows were restrained in headlocks at a feed bunk when assessing BCS and for video production. No difference was detected for the mean BCS, for the standard deviation of the mean BCS, or in the distribution of BCS between the live and video assessments. Mean and SD for 17 groups of Holstein cows from 20 farms were used to generate 10,000 random samples of BCS. Groups of 25, 50, 100, and 150 cows were created from the random samples, and estimates of mean BCS were determined by sampling 3 to 80% of the group. Estimates of mean BCS with a sample size of 30% or more from a group of cows fell within the 95% confidence limit of the true mean more than 98% of the time. Digital photographs provide adequate imaging for assessment of BCS. Sampling 30% of a group should be adequate to assess the mean BCS. Video imaging allowed a rapid assessment of BCS but did not permit identification of individual cows

    Characterization of DNA binding, transcriptional activation, and regulated nuclear association of recombinant human NFATp

    BACKGROUND: NFATp is one member of a family of transcriptional activators whose nuclear accumulation and hence transcriptional activity is regulated in mammalian cells. Human NFATp exists as a phosphoprotein in the cytoplasm of naive T cells. Upon antigen stimulation, NFATp is dephosphorylated, accumulates in nuclei, and functions to regulate transcription of genes including those encoding cytokines. While the properties of the DNA binding domain of NFATp have been investigated in detail, biochemical studies of the transcriptional activation and regulated association with nuclei have remained unexplored because of a lack of full length, purified recombinant NFATp. RESULTS: We developed methods for expressing and purifying full length recombinant human NFATp that has all of the properties known to be associated with native NFATp. The recombinant NFATp binds DNA on its own and cooperatively with AP-1 proteins, activates transcription in vitro, is phosphorylated, can be dephosphorylated by calcineurin, and exhibits regulated association with nuclei in vitro. Importantly, activation by recombinant NFATp in a reconstituted transcription system required regions of the protein outside of the central DNA binding domain. CONCLUSIONS: We conclude that NFATp is a bona fide transcriptional activator. Moreover, the reagents and methods that we developed will facilitate future studies on the mechanisms of transcriptional activation and nuclear accumulation by NFATp, a member of an important family of transcriptional regulatory proteins

    Exploring the Impact of Efficacy Messages on Cessation-Related Outcomes Using Ecological Momentary Assessment

    Introduction:Observational studies indicate that cigarette package inserts with efficacy messages about the benefits of quitting (i.e. response efficacy) and recommendations for successful cessation increase smokers’ self-efficacy to quit and promote sustained cessation. However, the effects of inserts with such efficacy messages have not been studied using experimental designs. This study used ecological momentary assessment (EMA) to assess smokers’ responses to efficacy inserts.Material and Methods:In a randomized case-crossover study among smokers from the United States (n=23), participants received a one-week supply of cigarettes with efficacy inserts and a one-week supply without any inserts, and were randomized to use the packs with inserts on either the first or second week of the study. For 14 consecutive days, participants used a smartphone to answer brief surveys on cessation-related variables during smoking sessions and at the beginning of each day. Multilevel mixed-effects linear and logistic regression models compared responses during the insert period to those of the non-insert period.Results:The insert period was associated with greater desire to quit (b=0.21, p=0.012), motivation to quit (b=0.18, p=0.001), self-efficacy to cut down (b=0.26, p\u3c0.001) and to quit (b=0.28, p\u3c0.000), and response efficacy/ perceived benefits of quitting (b=0.13, p=0.012). Insert exposure was not significantly associated with forgoing cigarettes (OR=1.9, p=0.2).Conclusions:Results from this EMA study suggest that inserts with efficacy messages may promote determinants of smoking cessation. This is consistent with observational research in Canada, which is the only country to use inserts with efficacy messages as well as pictorial warnings about smoking risks on the outside of packs. Future studies should assess the extent to which efficacy inserts can not only be used to communicate health information to smokers but also work in synergy with pictorial warnings

    A Selective Irreversible Inhibitor of Furin Does Not Prevent Pseudomonas Aeruginosa Exotoxin A-Induced Airway Epithelial Cytotoxicity

    Many bacterial and viral pathogens (or their toxins), including Pseudomonas aeruginosa exotoxin A, require processing by host pro-protein convertases such as furin to cause disease. We report the development of a novel irreversible inhibitor of furin (QUB-F1) consisting of a diphenyl phosphonate electrophilic warhead coupled with a substrate-like peptide (RVKR), that also includes a biotin tag, to facilitate activity-based profiling/visualisation. QUB-F1 displays greater selectivity for furin, in comparison to a widely used exemplar compound (furin I) which has a chloromethylketone warhead coupled to RVKR, when tested against the serine trypsin-like proteases (trypsin, prostasin and matriptase), factor Xa and the cysteine protease cathepsin B. We demonstrate QUB-F1 does not prevent P. aeruginosa exotoxin A-induced airway epithelial cell toxicity; in contrast to furin I, despite inhibiting cell surface furin-like activity to a similar degree. This finding indicates additional proteases, which are sensitive to the more broad-spectrum furin I compound, may be involved in this process

    Breaking the Curve with CANDELS: A Bayesian Approach to Reveal the Non-Universality of the Dust-Attenuation Law at High Redshift

    Dust attenuation affects nearly all observational aspects of galaxy evolution, yet very little is known about the form of the dust-attenuation law in the distant Universe. Here, we model the spectral energy distributions (SEDs) of galaxies at z = 1.5--3 from CANDELS with rest-frame UV to near-IR imaging under different assumptions about the dust law, and compare the amount of inferred attenuated light with the observed infrared (IR) luminosities. Some individual galaxies show strong Bayesian evidence in preference of one dust law over another, and this preference agrees with their observed location on the plane of infrared excess (IRX, LTIR/LUVL_{\text{TIR}}/L_{\text{UV}}) and UV slope (β\beta). We generalize the shape of the dust law with an empirical model, Aλ,δ=E(BV) kλ (λ/λV)δA_{\lambda,\delta}=E(B-V)\ k_\lambda\ (\lambda/\lambda_V)^\delta where kλk_\lambda is the dust law of Calzetti et al. (2000), and show that there exists a correlation between the color excess E(BV){E(B-V)} and tilt δ\delta with δ=(0.62±0.05)log(E(BV)){\delta=(0.62\pm0.05)\log(E(B-V))}+ (0.26 ± 0.02){(0.26~\pm~0.02)}. Galaxies with high color excess have a shallower, starburst-like law, and those with low color excess have a steeper, SMC-like law. Surprisingly, the galaxies in our sample show no correlation between the shape of the dust law and stellar mass, star-formation rate, or β\beta. The change in the dust law with color excess is consistent with a model where attenuation is caused by by scattering, a mixed star-dust geometry, and/or trends with stellar population age, metallicity, and dust grain size.     A capsid is the protein coat surrounding a virus' genome that ensures its protection and transport. The capsid of murine polyomavirus (muPy) consists of one major (VP1) and two minor (VP2/3) proteins, from which just VP1 is sufficient to form the capsid when expressed recombinantly (1). From a material engineering point of view, viral capsids are of interest because they present a paradigm for complex self-assembly on the nanometer scale. Understanding and controlling these assembly dynamics will allow the construction of nanoscale structures using a self-assembly process. The first step in this direction was the discovery that capsids of several viruses can be reversibly disassembled into their building blocks and reassembled using the same building blocks by simply changing the buffer conditions (2, 3). Such capsids already find applications as targeted in vivo delivery vectors for genes, proteins or small molecular drugs (4, 5), as optical probes for biomedical imaging and sensing purposes with unprecedented resolution and sensitivity and can potentially be used as templates for nanoelectronics (6, 7)

    Faint AGN and the Ionizing Background

    We determine the evolution of the faint, high-redshift, optical luminosity function (LF) of AGN implied by several observationally-motivated models of the ionizing background. Our results depend crucially on whether we use the total ionizing rate measured by the proximity effect technique or the lower determination from the flux decrement distribution of Ly alpha forest lines. Assuming a faint-end LF slope of 1.58 and the SDSS estimates of the bright-end slope and normalization, we find that the LF must break at M_B*=-24.2,-22.3, -20.8 at z=3,4, 5 if we adopt the lower ionization rate and assume no stellar contribution to the background. The break must occur at M_B*=-20.6,-18.7, -18.7 for the proximity effect background estimate. These values brighten by as much as ~2 mag if high-z galaxies contribute to the background with an escape fraction of ionizing photons consistent with recent estimates: f_e=0.16. By comparing to faint AGN searches, we find that the typically-quoted proximity effect estimates of the background imply an over-abundance of faint AGN (even with f_e=1). Even adopting the lower bound on proximity effect measurements, the stellar escape fraction must be high: f_e>0.2. Conversely, the lower flux- decrement-derived background requires a limited stellar contribution: f_e<0.05. Our derived LFs together with the locally-estimated black hole density suggest that the efficiency of converting mass to light in optically-unobscured AGN is somewhat lower than expected, <0.05. Comparison with similar estimates based on X-ray counts suggests that more than half of all AGN are obscured in the UV/optical. We also derive lower limits on typical AGN lifetimes and obtain >10^7 yrs for favored cases.Comment: 19 pages, 16 figures. Accepted by Astrophysical Journa

    Combustion Joining of Regolith Tiles for In-Situ Fabrication of Launch/Landing Pads on the Moon and Mars

    To mitigate dust problems during launch-landing operations in lunar and Mars missions, it is desired to build solid pads on the surface. Recently, strong tiles have been fabricated from lunar regolith simulants using high-temperature sintering. The present work investigates combustion joining of these tiles through the use of exothermic intermetallic reactions. Specifically, nickel aluminum (1:1 mole ratio) mixture was placed in a gap between the tiles sintered from JSC-1A lunar regolith simulant. Upon ignition by a laser, a self-sustained propagation of the combustion front over the mixture occurred. Joining was improved with increasing the tile thickness from 6.3 mm to 12.7 mm. The temperatures sufficient for melting the glass phase of JSC-1A were recorded for 12.7-mm tiles, which explains the observed better joining

    <p>Abstract</p> <p>Background</p> <p>It has been suggested that studying non-science subjects at A-level should be compulsory for medical students. Our admissions criteria specify only Biology, Chemistry and one or more additional subjects. This study aimed to determine whether studying a non-science subject for A-level is an independent predictor of achievement on the undergraduate medical course.</p> <p>Methods</p> <p>The subjects of this retrospective cohort study were 164 students from one entry-year group (October 2000), who progressed normally on the 5-year undergraduate medical course at Nottingham. Pre-admission academic and socio-demographic data and undergraduate course marks were obtained. T-test and hierarchical multiple linear regression analyses were undertaken to identify independent predictors of five course outcomes at different stages throughout the course.</p> <p>Results</p> <p>There was no evidence that the choice of science or non-science as the third or fourth A-level subject had any influence on course performance. Demographic variables (age group, sex, and fee status) had some predictive value but ethnicity did not. Pre-clinical course performance was the strongest predictor in the clinical phases (pre-clinical Themes A&B (knowledge) predicted Clinical Knowledge, p < 0.001, and pre-clinical Themes C&D (skills) predicted Clinical Skills, p = < 0.01).</p> <p>Conclusion</p> <p>This study of one year group at Nottingham Medical School provided no evidence that the admissions policy on A-level requirements should specify the choice of third or fourth subject.</p
