    Ingested bovine amniotic fluid enhances morphine antinociception in rats

    Ingestion by rats of rat placenta or amniotic fluid enhances opioid-mediated, or partly opioid-mediated, antinociception produced by morphine injection, vaginal or cervical stimulation, late pregnancy, and foot shock. This phenomenon is believed to be produced by a placental\ud opioid-enhancing factor (POEF). Ingestion by rats of human or dolphin placenta has also been shown to enhance opioid antinociception, suggesting that POEF may be common to many mammalian species. We tested bovine amniotic fluid (BAF) for its capacity to enhance morphine antinociception in female Long-Evans rats, as determined by percentage change from baseline tail-flick latency in response to radiant heat, and we report that 0.50 mL BAF effectively enhanced morphine antinociception but did not by itself produce antinociception. The efficacy of POEF across species suggests that POEF may have been functionally (and structurally) conserved during evolution. Furthermore, the availability of POEF at parturition, as well as its ability to enhance pregnancy-mediated antinociception without\ud disrupting maternal behavior, offers a tenable explanation for the long-debated ultimate causality of placentophagia

    Ingested placenta blocks the effect of morphine on gut transit in Long–Evans rats

    Opioids produce antinociception, and ingested placenta or amniotic fluid modifies that antinociception. More specifically, ingested placenta enhances the antinociception produced by selective activation of central n-opioid or y-opioid receptors but attenuates that produced by activation of central A-opioid receptors. Opioids also slow gut transit by acting on central or peripheral A-opioid receptors. Therefore, we hypothesized that ingested placenta would reverse the slowing of gut transit that is produced by morphine, a preferential A-opioid-receptor agonist. Rats were injected with morphine either centrally or systemically and fed placenta, after which gastrointestinal transit was evaluated. We report here that ingested placenta reversed the slowing of gut transit produced by centrally administered morphine but did not affect the slowing of gut transit produced by systemically administered morphine. These results suggest another likely consequence of placentophagia at parturition in mammals—reversal of opioid-mediated, pregnancy-based disruption of gastrointestinal function—as well as an important consideration in opioid-based treatments for pain in humans—enhancement of desirable effects with attenuation of adverse effects

    The Impact of Supervisor Servant Leadership on Counselor Supervisee Burnout and Secondary Traumatic Stress

    This study investigates the prediction of supervisee burnout and secondary traumatic stress by perceived supervisor servant leadership. Authors hypothesized that the servant leadership of supervisors would predict diminished burnout and secondary traumatic stress of supervisees. A sample of 241 counseling supervisees participated in the cross-sectional study and completed instruments measuring burnout, secondary traumatic stress, and perceived servant leadership of their direct supervisors. Data were analyzed with two simple linear regressions, and a one-way MANOVA was performed to determine if supervisee burnout, supervisee secondary traumatic stress, and perceived servant leadership of supervisors differed significantly according to supervisor type (i.e., clinical, administrative, or dual role). Results confirmed the main hypothesis, and administrative supervisors were perceived to demonstrate significantly less servant leadership qualities than the other two supervisor types. Limitations, avenues for future research, and implications for counselor education and supervision are discussed

    Spatio-temporally Resolved Methane Fluxes From the Los Angeles Megacity

    We combine sustained observations from a network of atmospheric monitoring stations with inverse modeling to uniquely obtain spatiotemporal (3‐km, 4‐day) estimates of methane emissions from the Los Angeles megacity and the broader South Coast Air Basin for 2015–2016. Our inversions use customized and validated high‐fidelity meteorological output from Weather Research Forecasting and Stochastic Time‐Inverted Lagrangian model for South Coast Air Basin and innovatively employ a model resolution matrix‐based metric to disentangle the spatiotemporal information content of observations as manifested through estimated fluxes. We partially track and constrain fluxes from the Aliso Canyon natural gas leak and detect closure of the Puente Hills landfill, with no prior information. Our annually aggregated fluxes and their uncertainty excluding the Aliso Canyon leak period lie within the uncertainty bounds of the fluxes reported by the previous studies. Spatially, major sources of CH_4 emissions in the basin were correlated with CH_4‐emitting infrastructure. Temporally, our findings show large seasonal variations in CH_4 fluxes with significantly higher fluxes in winter in comparison to summer months, which is consistent with natural gas demand and anticorrelated with air temperature. Overall, this is the first study that utilizes inversions to detect both enhancement (Aliso Canyon leak) and reduction (Puente Hills) in CH_4 fluxes due to the unintended events and policy decisions and thereby demonstrates the utility of inverse modeling for identifying variations in fluxes at fine spatiotemporal resolution

    Creating a multilayered representational ‘package’ for subcontracted workers: the case of cleaners at Ben-Gurion University

    Research into the organising of subcontracted workers tends to focus on how such campaigns contribute to union revitalisation, the shortcomings of non-union organisations in comparison to classic unionism, and opposition rather than complementarity between strategies. Analysing the organising of subcontracted cleaners at a university, this article shifts this focus, evaluating the campaign in terms of how it assisted the workers, regardless of whether it contributed to union renewal, and in terms of complementarity between new and traditional IR actors. Drawing on the power resources approach, it asserts that collaboration across different ‘paths to representation’ can create a multi-layered representational ‘package’ in which different organisations with different power resources take on different aspects of what was once a (single) union’s role, covering each other’s shortcomings in a kind of de facto representational ‘division of labour.

    Life History Response of Mediterranean Fruit Flies to Dietary Restriction

    The purpose of this study was to investigate medfly longevity and reproduction across a broad spectrum of diet restriction using a protocol similar to those applied in most rodent studies. Age-specific reproduction and age of death were monitored for 1,200 adult males and 1,200 females, each individually maintained on one of 12 diets from ad libitum to 30% of ad libitum. Diet was provided in a fixed volume of solution that was fully consumed each day, ensuring control of total nutrient consumption for every fly. Contrary to expectation and precedence, increased longevity was not observed at any level of diet restriction. Among females, reproduction continued across all diet levels despite the cost in terms of increased mortality. Among males, life expectancy exceeded that of females at most diet levels. However, in both sexes, mortality increased more sharply and the pattern of survival changed abruptly once the diet level fell to 50% of ad libitum or below, even though the energetic demands of egg production has no obvious counterpart in males. We believe that a more complete picture of the life table response to dietary restriction will emerge when studies are conducted on a wider range of species and include both sexes, more levels of diet, and the opportunity for mating and reproduction

    Metabolomics in Early Alzheimer's Disease: Identification of Altered Plasma Sphingolipidome Using Shotgun Lipidomics

    The development of plasma biomarkers could facilitate early detection, risk assessment and therapeutic monitoring in Alzheimer's disease (AD). Alterations in ceramides and sphingomyelins have been postulated to play a role in amyloidogensis and inflammatory stress related neuronal apoptosis; however few studies have conducted a comprehensive analysis of the sphingolipidome in AD plasma using analytical platforms with accuracy, sensitivity and reproducibility.We prospectively analyzed plasma from 26 AD patients (mean MMSE 21) and 26 cognitively normal controls in a non-targeted approach using multi-dimensional mass spectrometry-based shotgun lipidomics to determine the levels of over 800 molecular species of lipids. These data were then correlated with diagnosis, apolipoprotein E4 genotype and cognitive performance. Plasma levels of species of sphingolipids were significantly altered in AD. Of the 33 sphingomyelin species tested, 8 molecular species, particularly those containing long aliphatic chains such as 22 and 24 carbon atoms, were significantly lower (p<0.05) in AD compared to controls. Levels of 2 ceramide species (N16:0 and N21:0) were significantly higher in AD (p<0.05) with a similar, but weaker, trend for 5 other species. Ratios of ceramide to sphingomyelin species containing identical fatty acyl chains differed significantly between AD patients and controls. MMSE scores were correlated with altered mass levels of both N20:2 SM and OH-N25:0 ceramides (p<0.004) though lipid abnormalities were observed in mild and moderate AD. Within AD subjects, there were also genotype specific differences.In this prospective study, we used a sensitive multimodality platform to identify and characterize an essentially uniform but opposite pattern of disruption in sphingomyelin and ceramide mass levels in AD plasma. Given the role of brain sphingolipids in neuronal function, our findings provide new insights into the AD sphingolipidome and the potential use of metabolomic signatures as peripheral biomarkers

    Probing the Interstellar Medium in Early type galaxies with ISO observations

    Four IRAS-detected early type galaxies were observed with ISO. With the exception of the 15 micron image of NGC1052, the mid-IR emission from NGC1052, NGC1155, NGC5866 and NGC6958 at 4.5, 7 and 15 microns show extended emission. Mid-IR emission from NGC1052, NGC1155, and NGC6958 follows a de Vaucouleurs profile. The ratio of 15/7 micron flux decreases with radius in these galaxies, approaching the values empirically observed for purely stellar systems. In NGC5866, the 7 and 15 micron emission is concentrated in the edge-on dust lane. All the galaxies are detected in the [CII] line, and the S0s NGC1155 and NGC5866 are detected in the [OI] line as well. The ISO-LWS observations of the [CII] line are more sensitive measures of cool, neutral ISM than HI and CO by about a factor of 10-100. Three of four early type galaxies, namely NGC1052, NGC6958 and NGC5866, have low ratio FIR/Blue and show a lower [CII]/FIR, which is due to a softer radiation field from old stellar populations. The low [CII]/CO ratio in NGC5866 ([CII]/CO(1-0) < 570) confirms this scenario. We estimate the UV radiation expected from the old stellar populations in these galaxies and compare it to that needed to heat the gas to account for the cooling observed [CII] and [OI] lines. In three out of four galaxies, NGC1052, NGC5866 and NGC6958, the predicted UV radiation falls short by a factor of 2-3. In view of the observed intrinsic scatter in the "UV-upturn" in elliptical galaxies and its great sensitivity to age and metallicity effects, this is not significant. However, the much larger difference (about a factor of 20) between the UV radiation from old stars and that needed to produce the FIR lines for NGC 1155 is strong evidence for the presence of young stars, in NGC1155.Comment: To appear in the Astrophysical Journal.
