    First Detection of Mycobacterium ulcerans DNA in Environmental Samples from South America

    The occurrences of many environmentally-persistent and zoonotic infections are driven by ecosystem changes, which in turn are underpinned by land-use modifications that alter the governance of pathogen, biodiversity and human interactions. Our current understanding of these ecological changes on disease emergence however remains limited. Buruli ulcer is an emerging human skin disease caused by the mycobacterium, Mycobacterium ulcerans, for which the exact route of infection remains unclear. It can have a devastating impact on its human host, causing extensive necrosis of the skin and underlying tissue, often leading to permanent disability. The mycobacterium is associated with tropical aquatic environments and incidences of the disease are significantly higher on floodplains and where there is an increase of human aquatic activities. Although the disease has been previously diagnosed in South America, until now the presence of M. ulcerans DNA in the wild has only been identified in Australia where there have been significant outbreaks and in western and central regions of Africa where the disease is persistent. Here for the first time, we have identified the presence of the aetiological agent's DNA in environmental samples from South America. The DNA was positively identified using Real-time Polymerase Chain Reaction (PCR) on 163 environmental samples, taken from 23 freshwater bodies in French Guiana (Southern America), using primers for both IS2404 and for the ketoreductase-B domain of the M. ulcerans mycolactone polyketide synthase genes (KR). Five samples out of 163 were positive for both primers from three different water bodies. A further nine sites had low levels of IS2404 close to a standard CT of 35 and could potentially harbour M. ulcerans. The majority of our positive samples (8/14) came from filtered water. These results also reveal the Sinnamary River as a potential source of infection to humans. © 2014 Morris et al

    Beyond insulin: Unraveling the complex interplay of ER stress, oxidative damage, and CFTR modulation in CFRD

    \ua9 2024 The Author(s)CF-related diabetes (CFRD) is a prevalent comorbidity in people with Cystic Fibrosis (CF), significantly impacting morbidity and mortality rates. This review article critically evaluates the current understanding of CFRD molecular mechanisms, including the role of CFTR protein, oxidative stress, unfolded protein response (UPR) and intracellular communication. CFRD manifests from a complex interplay between exocrine pancreatic damage and intrinsic endocrine dysfunction, further complicated by the deleterious effects of misfolded CFTR protein on insulin secretion and action. Studies indicate that ER stress and subsequent UPR activation play critical roles in both exocrine and endocrine pancreatic cell dysfunction, contributing to β-cell loss and insulin insufficiency. Additionally, oxidative stress and altered calcium flux, exacerbated by CFTR dysfunction, impair β-cell survival and function, highlighting the significance of antioxidant pathways in CFRD pathogenesis. Emerging evidence underscores the importance of exosomal microRNAs (miRNAs) in mediating inflammatory and stress responses, offering novel insights into CFRD\u27s molecular landscape. Despite insulin therapy remaining the cornerstone of CFRD management, the variability in response to CFTR modulators underscores the need for personalized treatment approaches. The review advocates for further research into non-CFTR therapeutic targets, emphasizing the need to address the multifaceted pathophysiology of CFRD. Understanding the intricate mechanisms underlying CFRD will pave the way for innovative treatments, moving beyond insulin therapy to target the disease\u27s root causes and improve the quality of life for individuals with CF

    Glucose enhancement of memory is modulated by trait anxiety in healthy adolescent males

    Glucose administration is associated with memory enhancement in healthy young individuals under conditions of divided attention at encoding. While the specific neurocognitive mechanisms underlying this ‘glucose memory facilitation effect’ are currently uncertain, it is thought that individual differences in glucoregulatory efficiency may alter an individual’s sensitivity to the glucose memory facilitation effect. In the present study, we sought to investigate whether basal hypothalamic–pituitary–adrenal axis function (itself a modulator of glucoregulatory efficiency), baseline self-reported stress and trait anxiety influence the glucose memory facilitation effect. Adolescent males (age range = 14–17 years) were administered glucose and placebo prior to completing a verbal episodic memory task on two separate testing days in a counter-balanced, within-subjects design. Glucose ingestion improved verbal episodic memory performance when memory recall was tested (i) within an hour of glucose ingestion and encoding, and (ii) one week subsequent to glucose ingestion and encoding. Basal hypothalamic–pituitary–adrenal axis function did not appear to influence the glucose memory facilitation effect; however, glucose ingestion only improved memory in participants reporting relatively higher trait anxiety. These findings suggest that the glucose memory facilitation effect may be mediated by biological mechanisms associated with trait anxiety

    "Michael Douglas" effect

    Background: In June 2013, celebrity actor Michael Douglas announced that he had received a diagnosis of throat cancer, which he attributed to the human papillomavirus (HPV). HPV is the most common sexually transmitted disease, however the HPV vaccination rates are lower than other vaccines. Given the health disclosure of Michael Douglas's cancer, we queried whether this event had an effect on public awareness for HPV-related head and neck cancers (HNCs) or its prevention.Methods: Using Google Trends and Twitter, we retrospectively analyzed search trends and Tweets for the keywords throat cancer, oral cancer, hpv vaccine, and human papillomavirus infection. We extracted data and compared it to an expected forecast found using an autoregressive integrated moving algorithm (ARIMA).Results: During the week of disclosure, Google Trends data demonstrated an increase of 62.5% in hpv infection inquiries, 47.0% in hpv vaccine inquiries, 47.0% in oral cancer inquiries, and 81.5% in throat cancer inquiries above the expected. Tweets referencing hpv increased 127.2%; tweets referencing hpv (vaccine OR vaccination) increased 98.8%; tweets referencing hpv awareness increased 778.4%; tweets referencing oral cancer increased 1038.7%; and tweets referencing throat cancer increased 811.7% compared to the rest of 2013.Conclusions: Data from Google Trends and Twitter indicated that Michael Douglas's disclosure of HPV-related HNC resulted in a substantial increase in public awareness and prevention methods for HPV-related HNC. Celebrity health disclosures may thus prove to be a cost-effective strategy to advocate for public health literacy, and positively influence public interest of specific health issues

    PHYLOGENETIC ANALYSIS OF RED COTTON BUG SPECIES (HEMIPTERA: PYRRHOCORIDAE) IN PUNJAB, PAKISTAN

    Traditional techniques for identifying the red cotton bugs Dysdercus koenigii and Dysdercus cingulatus (Pyrrhocoridae: Hemiptera) depend on phenotypic and morphological data. These methods are suitable when species have well-defined structures and when huge taxonomic knowledge is at hand. In this study, the mitochondrial cytochrome oxidase I (COI) gene was selected for the molecular identification of D. koenigii and D. cingulatus. The sequence data showed that one species of the red cotton bug sample was identified as Dysdercus koenigii with 35-100% similarity to other Dysdercus species present in the NCBI database, while the other species was identified as Dysdercus cingulatus with 88-94% similarity to other red cotton bugs species present in the NCBI database. The phylogenetic analysis revealed that our     D. koenigii Seq (>180319003-A02-102-DK-.ab1) shares the same cluster with four D. koenigii isolates (ZSI/SRC_I.28B, ZSI/SRC_I.28, GQ306227.1, KJ459924.1). While the D. cingulatus Seq (>180319003-A02-103-DC-.abl) shares a cluster with D. cingulatus voucher (RO_DC2015). This is the first molecular identification of red cotton bugs (D. koenigii and     D. cingulatus) reported in Pakistan. Thus, in the current study, molecular identification has been accepted as a reliable method for the identification of these agriculturally important insect pests

    ABSTRACT When the European Monetary System was negotiated in 1978, governments in France, Britain, and Italy took very different approaches to this new international institution for coordinating exchange rate policies. The French government actively supported the creation of the European Monetary System, the Italian government entered the system but on weaker terms than the French, and the British government refused to enter the system, preferring to allow the pound to float. To explain these different policy choices, I analyze the impact of domestic politics and institutions on exchange rate policy, paying particular attention to how the organization of bank-industry relations and government instability shape policymakers' policy preferences and their abilities to implement these preferences. There are many approaches for explaining international economic policy coordination.
