    Velocity spectra and coherence estimates in the marine atmospheric boundary layer

    Two years of continuous sonic anemometer measurements conducted in 2007 and 2008 at the FINO1 platform are used to investigate the characteristics of the single- and two-point velocity spectra in relation to the atmospheric stability in the marine atmospheric boundary layer. The goals are to reveal the limits of current turbulence models for the estimation of wind loads on offshore structures, and to propose a refined description of turbulence at altitudes where Monin–Obukhov similarity theory may be limited. Using local similarity theory, a composite spectrum model, combining a pointed and a blunt model, is proposed to describe the turbulence spectrum for unstable, neutral and stable conditions. Such a model captures the −1 power law followed by the velocity spectra at an intermediate frequency range in the marine atmospheric boundary layer. For the Monin–Obukhov similarity parameter ζ<0.3, the Davenport coherence model captures the vertical coherence of the horizontal velocity components well. A two-parameter exponential decay function is found more appropriate for modelling the coherence of the vertical velocity component. Under increasingly stable conditions, the size of the eddies in the vertical coordinate reduces, such that smaller separation distances than that covered in the present dataset may be required to study the coherence with sufficient accuracy.acceptedVersio

    Offshore wind turbine wake characteristics using scanning doppler lidar

    Within an offshore wind park, wind flow characteristics are quite complex and govern both the energy production and the structural wind turbine response. An experimental study focussed on assessing the spatial variability of winds near the German offshore wind energy platform FINO1 was conducted using multiple remote sensing devices. This study focuses on measuring the wind turbine wake characteristics, such as velocity deficit, the extent (length and width) of the wake and wake meandering under various atmospheric conditions using the data collected from a single scanning Doppler Lidar for several months in 2016. A new algorithm based on using a Gaussian model to measure the downwind wake characteristics is developed. The wind turbine downwind wake deficits compared well to previous models at far-wake regions, while at near-wake regions the models deviated due to different instruments & methodologies used in measuring the wake characteristics. It was also observed that the length of the Alpha Ventus wind turbine wake varied from 3 to 15 times the Rotor Diameter (RD), and the maximum velocity deficit varied from 55% to 75% of the free-stream wind speed, depending on mean wind speed and atmospheric stability.     First record of the opal chimaera, Chimaera opalescens (Holocephali: Chimaeridae) and revision of the occurrence of the rabbitfish Chimaera monstrosa in the Azores waters

    The presence of the opal chimaera, Chimaera opalescens, is reported for the first time in the deep waters of the Azores, with the capture of four specimens by fishermen and the video recording of an additional five individuals. Species identification was supported by the 646 bp sequenced fragment of the mitochondrial gene cytochrome oxidase subunit I. Because C. opalescens is a recently recognised species that had been recurrently misidentified as rabbitfish, Chimaera monstrosa, the historical data of C. monstrosa in the Azores were reviewed to assess the possible presence of both Chimaera species in the region. Although several authors have reported the occurrence of C. monstrosa in the Azorean waters since the 1800s, the majority of these are based on only three specimens caught during the late 1800s. The investigation performed using literature and examination of the museum specimens still available concluded that the most likely scenario is that C. monstrosa is absent from the Azores and past records of that species in the region are most likely misidentifications of C. opalescens.publishedVersio

    Benthic community zonation from mesophotic to deep sea: Description of first deep-water kelp forest and coral gardens in the Madeira archipelago (central NE Atlantic)

    The Madeira archipelago has a unique underwater landscape that is characterised by narrow shelves, steep slopes and a large submarine tributary system that boosts primary productivity in oligotrophic waters and thus offers a potential for hotspots of biodiversity. Despite this, there have been limited deep-water exploration activities with less than five expeditions since the 1960s. Here, we investigated the seabed on the southern side of the Madeira-Desertas Ridge using a manned submersible along a 3.8 km long transect starting at 366 m depth up the ridge shelf until its top at 73 m. Benthic habitats and community composition were documented with video along a depth gradient from mesophotic to deep sea. Six distinct biotopes were recognised (three deeper, and three shallower than 115 m depth). Our results showed a rich biodiversity with deep biotopes characterised by sponges and non-reef-building corals (e.g., Pachastrella monilifera, Viminella flagellum, Eunicella verrucosa) and shallow biotopes comprising macroalgae and the gorgonian Paramuricea cf. grayi. The pronounced benthic zonation reflects the steep environmental gradient that includes high topographic variation, heterogeneous substrates, and bidirectional regular wave-motion at the shallow mesophotic part. Together with biotic factors, such as low density of sea urchins and presence of predatory fish, this environment with unusual deep light penetration, a mesoscale cyclonic eddy, and deep wave-motion, has allowed the establishment of a mature deep-water kelp population of Laminaria ochroleuca in the plateau (max. >100 individuals p/100 m2). At the same time, a conspicuous coral fauna was observed on a wide range of soft to hard bottoms with several species taking advantage of the favourable hydrodynamic regime and seawater properties together with substratum availability to create coral gardens. These habitats were previously not known from Madeira, and their newfound discovery in the archipelago merit further investigation and protection.publishedVersio

    A randomized trial of alendronate as prophylaxis against loss in bone mineral density following lymphoma treatment

    Lymphoma patients often receive high glucocorticoid doses as part of standard therapy. Observational studies have shown a substantial risk of glucocorticoid-induced osteoporosis (GIO) with associated fractures. The aim of the SIESTA trial was to determine if oral alendronate (ALN) is a safe and effective prophylaxis against GIO in lymphoma. SIESTA was a single-center, randomized, double-blinded, phase 2 study of lymphoma patients planned for glucocorticoid-containing chemotherapy. After randomization, patients received weekly ALN 70 mg or placebo for a total of 52 weeks. Bone mineral density (BMD) was assessed at baseline, after completion of chemotherapy (end of treatment [EOT]) (4 to 6 months), and at the end of the study (EOS) (12 months). Vertebral fracture and biomarkers were assessed at baseline and EOS. Patients with baseline BMD assessment and at least 1 follow-up BMD assessment were analyzed for efficacy. The primary endpoint was a change in lumbar spine T-score from baseline to EOS. Of the 59 patients enrolled, 23 of 30 in the ALN arm and 24 of 29 in the placebo arm were analyzed for efficacy. The mean change in T-score from baseline to 12 months at the lumbar spine was +0.15 for ALN and -0.12 for placebo (P = .023). The difference in ΔT(EOS) between the ALN and placebo groups was larger among females (ALN 0.28; placebo -0.28; P = .01). Biomarker analyses confirmed reduced bone resorption in ALN-treated patients. In conclusion, ALN is a safe and effective primary prophylaxis against loss in BMD following glucocorticoid-containing chemotherapy. Despite reduced BMD loss in the ALN arm, the treatment did not influence fracture risk in this small cohort of patients

    Complexity and variability analyses of motor activity distinguish mood states in Bipolar Disorder

    Changes in motor activity are core symptoms of mood episodes in bipolar disorder. The manic state is characterized by increased variance, augmented complexity and irregular circadian rhythmicity when compared to healthy controls. No previous studies have compared mania to euthymia intra-individually in motor activity. The aim of this study was to characterize differences in motor activity when comparing manic patients to their euthymic selves. Motor activity was collected from 16 bipolar inpatients in mania and remission. 24-h recordings and 2-h time series in the morning and evening were analyzed for mean activity, variability and complexity. Lastly, the recordings were analyzed with the similarity graph algorithm and graph theory concepts such as edges, bridges, connected components and cliques. The similarity graph measures fluctuations in activity reasonably comparable to both variability and complexity measures. However, direct comparisons are difficult as most graph measures reveal variability in constricted time windows. Compared to sample entropy, the similarity graph is less sensitive to outliers. The little-understood estimate Bridges is possibly revealing underlying dynamics in the time series. When compared to euthymia, over the duration of approximately one circadian cycle, the manic state presented reduced variability, displayed by decreased standard deviation (p = 0.013) and augmented complexity shown by increased sample entropy (p = 0.025). During mania there were also fewer edges (p = 0.039) and more bridges (p = 0.026). Similar significant changes in variability and complexity were observed in the 2-h morning and evening sequences, mainly in the estimates of the similarity graph algorithm. Finally, augmented complexity was present in morning samples during mania, displayed by increased sample entropy (p = 0.015).     Objective assessment of motor activity in a clinical sample of adults with attention-deficit/hyperactivity disorder and/or cyclothymic temperament

    Background Most research on patterns of motor activity has been conducted on adults with mood disorders, but few studies have investigated comorbid attention-deficit/hyperactivity disorder (ADHD) or temperamental factors that may influence the clinical course and symptoms. Cyclothymic temperament (CT) is particularly associated with functional impairment. Clinical features define both disorders, but objective, biological markers for these disorders could give important insights with regard to pathophysiology and classification. Methods Seventy-six patients, requiring diagnostic evaluation of ADHD, mood or anxiety disorders were recruited. A comprehensive diagnostic evaluation, including the CT scale of the Temperament Evaluation of Memphis, Pisa, Paris and San Diego – Auto-questionnaire (TEMPS-A), neuropsychological tests and actigraphy, was performed. ADHD was diagnosed according to the DSM-IV criteria. There was a range of different conditions in this clinical sample, but here we report on the presence of CT and ADHD in relation to motor activity. Twenty-nine healthy controls were recruited. We analyzed motor activity time series using linear and nonlinear mathematical methods, with a special focus on active and inactive periods in the actigraphic recordings. Results Forty patients fulfilled the criteria for ADHD, with the remainder receiving other psychiatric diagnoses (clinical controls). Forty-two patients fulfilled the criteria for CT. Twenty-two patients fulfilled the criteria for ADHD and CT, 18 patients met the criteria for ADHD without CT, and 15 patients had neither. The ratio duration of active/inactive periods was significantly lower in patients with CT than in patients without CT, in both the total sample, and in the ADHD subsample. Conclusions CT is associated with objectively assessed changes in motor activity, implying that the systems regulating motor behavior in these patients are different from both healthy controls and clinical controls without CT. Findings suggest that actigraphy may supplement clinical assessments of CT and ADHD, and may provide an objective marker for CT.publishedVersio

    Mammalian tissues defective in nonsense-mediated mRNA decay display highly aberrant splicing patterns

    BACKGROUND: Nonsense-mediated mRNA decay (NMD) affects the outcome of alternative splicing by degrading mRNA isoforms with premature termination codons. Splicing regulators constitute important NMD targets; however, the extent to which loss of NMD causes extensive deregulation of alternative splicing has not previously been assayed in a global, unbiased manner. Here, we combine mouse genetics and RNA-seq to provide the first in vivo analysis of the global impact of NMD on splicing patterns in two primary mouse tissues ablated for the NMD factor UPF2. RESULTS: We developed a bioinformatic pipeline that maps RNA-seq data to a combinatorial exon database, predicts NMD-susceptibility for mRNA isoforms and calculates the distribution of major splice isoform classes. We present a catalog of NMD-regulated alternative splicing events, showing that isoforms of 30% of all expressed genes are upregulated in NMD-deficient cells and that NMD targets all major splicing classes. Importantly, NMD-dependent effects are not restricted to premature termination codon+ isoforms but also involve an abundance of splicing events that do not generate premature termination codons. Supporting their functional importance, the latter events are associated with high intronic conservation. CONCLUSIONS: Our data demonstrate that NMD regulates alternative splicing outcomes through an intricate web of splicing regulators and that its loss leads to the deregulation of a panoply of splicing events, providing novel insights into its role in core- and tissue-specific regulation of gene expression.     Diurnal variation of motor activity in adult ADHD patients analyzed with methods from graph theory

    Attention-deficit /hyperactivity disorder (ADHD) is a common neurodevelopmental syndrome characterized by age-inappropriate levels of motor activity, impulsivity and attention. The aim of the present study was to study diurnal variation of motor activity in adult ADHD patients, compared to healthy controls and clinical controls with mood and anxiety disorders. Wrist-worn actigraphs were used to record motor activity in a sample of 81 patients and 30 healthy controls. Time series from registrations in the morning and evening were analyzed using measures of variability, complexity and a newly developed method, the similarity algorithm, based on transforming time series into graphs. In healthy controls the evening registrations showed higher variability and lower complexity compared to morning registrations, however this was evident only in the female controls. In the two patient groups the same measures were not significantly different, with one exception, the graph measure bridges. This was the measure that most clearly separated morning and evening registrations and was significantly different both in healthy controls and in patients with a diagnosis of ADHD. These findings suggest that actigraph registrations, combined with mathematical methods based on graph theory, may be used to elucidate the mechanisms responsible for the diurnal regulation of motor activity.publishedVersio
