    Cryogenic Thermal Emittance Measurements on Small-Diameter Stainless Steel Tubing

    The Mid Infrared Instrument aboard the James Webb Space Telescoep includes a mechanical cryocooler which cools its detectors to their 6 K operating temperature. The refrigerant flows through several meters of approximately 2 mm diameter 304L stainless steel tubing, with some sections gold plated, and some not, which are exposed to their environment. An issue of water freezing onto the tube surfaces is mitigated by a running a warm gas through the lines to sublimate the water. To model the effect of this process on nearby instruments, an accurate measure of the tube emittance is needed. Previously we reported the abosprtance of the gold plated stainless steel tubing as a function of source temperature (i.e. its environment). In this work the thermal emittance of the uncoated tubing is measured as a function of its temperature between 100 and 300 K. This value leads to an accurate prediction of the minimum length of time required to thermally recycle the system. We report the technique and present the results

    Fano resonance resulting from a tunable interaction between molecular vibrational modes and a double-continuum of a plasmonic metamolecule

    Coupling between tuneable broadband modes of an array of plasmonic metamolecules and a vibrational mode of carbonyl bond of poly(methyl methacrylate) is shown experimentally to produce a Fano resonance, which can be tuned in situ by varying the polarization of incident light. The interaction between the plasmon modes and the molecular resonance is investigated using both rigorous electromagnetic calculations and a quantum mechanical model describing the quantum interference between a discrete state and two continua. The predictions of the quantum mechanical model are in good agreement with the experimental data and provide an intuitive interpretation, at the quantum level, of the plasmon-molecule coupling

    Growth performance, duodenal morphology and the caecal microbial population in female broiler chickens fed glycine-fortified low protein diets under heat stress conditions

    This study was undertaken to examine the effect of feeding glycine (Gly)-fortified low protein (LP) diets on the growth performance, duodenal morphology and caecal microbial populations of broiler chickens raised under unheated, cyclic or constant heat stress environmental conditions. 2. From d 1 to 21 (starter phase), an equivalent number of birds were fed either a normal protein (NP) diet or a LP diet fortified with Gly. From d 22 to 42 (grower phase), an equivalent number of birds from each starter diet were distributed to one of the following dietary groups: (i) an NP diet during the starter and grower phases (NPNP), (ii) an NP diet during the starter phase and a LP diet during the grower phase (NPLP), (iii) an LP diet during the starter phase and an NP diet during the grower phase (LPNP) or (iv) LP diets during both phases (LPLP). 3. Commencing from d 22, an equivalent number of birds from each dietary group were exposed to (i) 23 ± 1°C throughout (unheated), (ii) 34 ± 1°C for 7 h each day from 10:00 to 17:00 (cyclic heat) or (iii) 34 ± 1°C throughout (constant heat). 4. Feeding the LP diet during the starter phase resulted in feed intake (FI), weight gain (WG), feed conversion ratios (FCR) and energy efficiency ratios (EER) similar to those for the NP diet. The birds fed the LP diet had a significantly higher protein efficiency ratio (PER) compared with the birds fed the NP diet. 5. During the grower phase, there were significant diet × temperature interactions for F, WG, FCR, PER, EER, villus height, crypt depth and caecal Clostridia. The birds fed the NPLP and LPLP diets had lower FI, WG and EER, higher FCR, shorter villus height and crypt depth and higher caecal Clostridia compared with the birds fed LPNP and NPNP diets under constant heat stress. However, feeding birds the NPLP and LPLP diets resulted in FI, WG, EER, FCR, morphology parameters and caecal Clostridia equivalent to the birds fed LPNP and NPNP diets, as well as improved PER, under unheated and cyclic heat stress conditions. 6. In conclusion, our results indicate that Gly-fortified LP diets can be fed to broilers under normal and acute heat stress environmental conditions without any adverse effects on performance. However, the use of such LP diets can be detrimental to broilers under chronic heat stress conditions

    Interferometric speckle visibility spectroscopy (ISVS) for human cerebral blood flow monitoring

    Infrared light scattering methods have been developed and employed to non-invasively monitor human cerebral blood flow (CBF). However, the number of reflected photons that interact with the brain is low when detecting blood flow in deep tissue. To tackle this photon-starved problem, we present and demonstrate the idea of interferometric speckle visibility spectroscopy (ISVS). In ISVS, an interferometric detection scheme is used to boost the weak signal light. The blood flow dynamics are inferred from the speckle statistics of a single frame speckle pattern. We experimentally demonstrated the improvement of measurement fidelity by introducing interferometric detection when the signal photon number is insufficient. We apply the ISVS system to monitor the human CBF in situations where the light intensity is \sim100-fold less than that in common diffuse correlation spectroscopy (DCS) implementations. Due to the large number of pixels (2×105\sim 2\times 10^5) used to capture light in the ISVS system, we are able to collect a similar number of photons within one exposure time as in normal DCS implementations. Our system operates at a sampling rate of 100 Hz. At the exposure time of 2 ms, the average signal photon electron number is \sim0.95 count/pixel, yielding a single pixel interferometric measurement signal-to-noise ratio (SNR) of \sim0.97. The total 2×105\sim 2\times 10^5 pixels provide an expected overall SNR of 436. We successfully demonstrate that the ISVS system is able to monitor the human brain pulsatile blood flow, as well as the blood flow change when a human subject is doing a breath holding task

    Perceptual snoring as a basis for a psychoacoustical modeling and clinical patient profiling

    Purpose The perceptual burden and social nuisance for mainly the co-sleeper can affect the relationship between snorer and bedpartner. Mandibular advancement devices (MAD) are commonly recommended to treat sleep-related breathing such as snoring or sleep apnea. There is no consensus about the definition of snoring particularly with MAD, which is essential for assessing the effectiveness of treatment. We aimed to stablish a notion of perceptual snoring with MAD in place. Methods Sound samples, each 30 min long, were recorded during in-home, overnight, automatic mandibular repositioning titration studies in a population of 29 patients with obstructive sleep apnea syndrome (OSAS) from a clinical trial carried out to validate the MATRx plus. Three unspecialized and calibrated raters identified sound events and classified them as noise, snore, or breathing as well as providing scores for classification certainty and annoyance. Data were analyzed with respect to expiration-inspiration, duration, annoyance, and classification certainty. Results A Fleiss' kappa (>0.80) and correlation duration of events (>0.90) between raters were observed. Prevalence of all breath sounds: snore 55.6% (N = 6398), breathing sounds 31.7% (N = 3652), and noise 9.3% (N = 1072). Inspiration occurs in 88.3% of events, 96.8% contained at least on expiration phase. Snore and breath events had similar duration, respectively 2.58s (sd 1.43) and 2.41s (sd 1.22). Annoyance is lowest for breathing events (8.00 sd 0.98) and highest for snore events (4.90 sd 1.92) on a VAS from zero to ten. Conclusion Perceptual sound events can be a basis for analysis in a psychosocial context. Perceived snoring occurs during both expiration as well as inspiration. Substantial amount of snoring remains despite repositioning of the mandible aimed at the reduction of AHI-ODI

    Purpose: The effect of snoring on the bed partner can be studied through the evaluation of in situ sound records by the bed partner or unspecialized raters as a proxy of real-life snoring perception. The aim was to characterize perceptual snore events through acoustical features in patients with obstructive sleep apnea (OSA) with an advanced mandibular position. Methods: Thirty-minute sound samples of 29 patients with OSA were retrieved from overnight, in-home recordings of a study to validate the MATRx plus® dynamic mandibular advancement system. Three unspecialized raters identified sound events and classified them as noise, snore, or breathing. The raters provided ratings for classification certainty and annoyance. Data were analyzed with respect to respiratory phases, and annoyance. Results: When subdividing perceptual events based on respiratory phase, the logarithm-transformed Mean Power, Spectral Centroid, and Snore Factor differed significantly between event types, although not substantially for the spectral centroid. The variability within event type was high and distributions suggested the presence of subpopulations. The general linear model (GLM) showed a significant patient effect. Inspiration segments occurred in 65% of snore events, expiration segments in 54%. The annoyance correlated with the logarithm of mean power (r = 0.48) and the Snore Factor (0.46). Conclusion: Perceptual sound events identified by non-experts contain a non-negligible mixture of expiration and inspiration phases making the characterization through acoustical features complex. The present study reveals that subpopulations may exist, and patient-specific features need to be introduced

    Planarian Cholinesterase: Molecular And Functional Characterization Of An Evolutionarily Ancient Enzyme To Study Organophosphorus Pesticide Toxicity

    The asexual freshwater planarian Dugesia japonica has emerged as a medium-throughput alternative animal model for neurotoxicology. We have previously shown that D. japonica are sensitive to organophosphorus pesticides (OPs) and characterized the in vitro inhibition profile of planarian cholinesterase (DjChE) activity using irreversible and reversible inhibitors. We found that DjChE has intermediate features of acetylcholinesterase (AChE) and butyrylcholinesterase (BChE). Here, we identify two candidate genes (Djche1 and Djche2) responsible for DjChE activity. Sequence alignment and structural homology modeling with representative vertebrate AChE and BChE sequences confirmed our structural predictions, and show that both DjChE enzymes have intermediate sized catalytic gorges and disrupted peripheral binding sites. Djche1 and Djche2 were both expressed in the planarian nervous system, as anticipated from previous activity staining, but with distinct expression profiles. To dissect how DjChE inhibition affects planarian behavior, we acutely inhibited DjChE activity by exposing animals to either an OP (diazinon) or carbamate (physostigmine) at 1 µM for 4 days. Both inhibitors delayed the reaction of planarians to heat stress. Simultaneous knockdown of both Djche genes by RNAi similarly resulted in a delayed heat stress response. Furthermore, chemical inhibition of DjChE activity increased the worms’ ability to adhere to a substrate. However, increased substrate adhesion was not observed in Djche1/Djche2 (RNAi) animals or in inhibitor-treated day 11 regenerates, suggesting this phenotype may be modulated by other mechanisms besides ChE inhibition. Together, our study characterizes DjChE expression and function, providing the basis for future studies in this system to dissect alternative mechanisms of OP toxicity

    Gene bank preparation from the endemic shrimps and crustaceans of Persian Gulf and Oman Sea

    Genetic knowledge and Gene bank preparation can help to protect biodiversity and detect , species identify , fishing offenses , genetic classification and also identification the faliure cross hybridizations of marine animals. In this study, sampling was performed from Jask, guatr and Hormuz areas, which is the most important habitats for the species studied using bottom trawl. Total DNA extraction was performed using phenol- chloroform method. After relevant studies on this gene primers were designed and in use. After editing the sequences, nucleotide BLAST (Basic Local Alignment Search Tool) was performed using NCBI blast main page. The sequences obtained from each sample were aligned and corrected from any ambiguities and assembled using Bio edit program. Trees were generated using maximum parsimony (MP), a character-based algorithm and neighbor joining (NJ) a distance-based algorithm for phenetic analysis. The distance matrix option of MEGA4 was used to calculate genetic distance according to the Kimura 2-parameter model of sequence evolution. Based on the results obtained, the optical density of 260 to 280 nm in the samples was recorded between 1/8 - 2, indicating good quality DNA samples. Optimized PCR reaction to 16SrRNA and COI gene amplification using the gradient between 48 - 60° C showed that the most suitable criteria for binding primers, 48 and 54 Celsius degrees respectively. The project objectives including the identification of the genetic structure of the species, and draw the phylogenetic trees using two genes 16SrRNA and COI, making identification and registration of specified computer storage and regulate the structure and management of mentioned species by focus on genetic resources 5 species of shrimp (P. semisulcatus، P. indicus، P. merguiensis ، P. monodon، M. affinis) in the Persia Gulf and Oman Sea through the creation of an integrated network of aquatic genetic resources in the region to try to identify genetic resources and aquatic gene bank. Molecular investigation of mitochondria DNA (mtDNA) using partial sequences of 16S rRNA gene showed relatively low genetic differences between the P. semisulcatus morphotypes. These sequences were able to distinguish between the two morphotypes, and separated them into two distinct clades. Also genetic divergence detected by COI gene analysis was consistently higher. High genetic divergence for COI was observed between the two morphotypes of P.semeisulcatus which emphesise that the gene bank preparation should be perform for this morphotype of this species. This type of analysis could be considered as an important tool to be used in broodstock selection in breeding programs. In this case, different management in broodstocking programs should be performed for two morphotypes of P. semisulcatus which were detected in Persian Gulf. The results of this study show that two Morphotype of P.semisulcatus can be considered as two separate species from genetic aspects. In this regard, it can be assumed that the genetic composition of the studied species is very close together and we've no seen a huge difference in the species except in the green tiger species
