    Analysis of material reliability of AlSi17Cu5 alloy using statistical weibull distribution

    Analysis of reliability of AlSi17Cu5 alloy was carried out using the so-called „permanence measure”. Two-parameter Weibull distribution, defining stability of material features, is its numerical equivalent. Mechanical properties (HB hardness HB, tensile strength Rm, and yield strength R0,2) were selected as parameters for statistical evaluation, whereas inoculation with phosphorus (CuP10) and overheating to 920 oC (separately and jointly) together with intense cooling of the alloy were the factors shaping the influence of technology type. Statistical calculations, preceded by verification of hypotheses on conformity of distribution of the investigated resultant characteristics with normal distribution, were supported with variance estimators, and correlation and regression analyses

    Un hallazgo de monedas califales en una de las múltiples viviendas excavadas en la última década al oeste de la medina, parte del extenso arrabal que se genera desde época emiral y que eclosiona en época de ‘Abd al-Raḥmān III en el año 929. Se emplaza a medio camino del lienzo occidental de la muralla de la capital y la ciudad palatina de madinat al-Zahra, en un paisaje suburbano densamente urbanizado.     The theory of regular variation, in its Karamata and Bojani´c-Karamata/de Haan forms, is long established and makes essential use of the Cauchy functional equation. Both forms are subsumed within the recent theory of Beurling regular variation, developed elsewhere. Various generalizations of the Cauchy equation, including the Gołab–Schinzel functional equation (GS) and Goldie's equation (GBE) below, are prominent there. Here we unify their treatment by algebraicization: extensive use of group structures introduced by Popa and Javor in the 1960s turn all the various (known) solutions into homomorphisms, in fact identifying them 'en passant', and show that (GS) is present everywhere, even if in a thick disguise

    Classification, substrate specificity and structural features of D-2-hydroxyacid dehydrogenases: 2HADH knowledgebase

    BACKGROUND: The family of D-isomer specific 2-hydroxyacid dehydrogenases (2HADHs) contains a wide range of oxidoreductases with various metabolic roles as well as biotechnological applications. Despite a vast amount of biochemical and structural data for various representatives of the family, the long and complex evolution and broad sequence diversity hinder functional annotations for uncharacterized members. RESULTS: We report an in-depth phylogenetic analysis, followed by mapping of available biochemical and structural data on the reconstructed phylogenetic tree. The analysis suggests that some subfamilies comprising enzymes with similar yet broad substrate specificity profiles diverged early in the evolution of 2HADHs. Based on the phylogenetic tree, we present a revised classification of the family that comprises 22 subfamilies, including 13 new subfamilies not studied biochemically. We summarize characteristics of the nine biochemically studied subfamilies by aggregating all available sequence, biochemical, and structural data, providing comprehensive descriptions of the active site, cofactor-binding residues, and potential roles of specific structural regions in substrate recognition. In addition, we concisely present our analysis as an online 2HADH enzymes knowledgebase. CONCLUSIONS: The knowledgebase enables navigation over the 2HADHs classification, search through collected data, and functional predictions of uncharacterized 2HADHs. Future characterization of the new subfamilies may result in discoveries of enzymes with novel metabolic roles and with properties beneficial for biotechnological applications

    The activity of hydrolytic enzymes in the digestive system of Acanthobdellida, Branchiobdellida and Hirudinida (Annelida,Clitellata) – considerations on similarity and phylogeny

    Activities of nineteen hydrolases were measured in the digestive systems of predatory and blood-feeding true leeches (Hirudinida) and their closest relatives, Branchiobdellida and Acanthobdellida. Hydrolase activities were analyzed in different parts of the digestive systems: the species-specific anterior part, i.e. jaws, pharynx or proboscis, crop and intestine. The results obtained suggest that food digestion and possible absorption predominate in the intestine of most of the studied Hirudinida and A. peledina, whereas in B. astaci these processes take place in the anterior part of the digestive system and crop. In Erpobdellidae and Piscicola respirans, the activity of acid and alkaline phosphatases, N-acetyl-β-glucosaminidase, leucine and valine arylamidases, and α-fucosidase was also detected in the anterior part of the digestive system. We also detected differences in enzyme occurrence between the studied species, which are probably connected with their different food preferences. Moreover, the presence of the whole spectrum of enzymes in predatory leeches and the absence of trypsin and α-chymotrypsin activity in the crop of all the leeches support the hypothesis that the leech ancestor was a blood-feeder. Our study showed that “Rhynchobdellida” constitute a paraphyletic group which confirms the previous results based on molecular phylogenetics, while Arhynchobdellida appears to be a non-monophyletic group which is not consistent with previous molecular results

    Guess who? Taxonomic problems in the genus Eiseniella revisited by integrated approach

    Eiseniella neapolitana is a semi-aquatic, diploid earthworm that for many years was related to the cosmopolitan species Eiseniella tetraedra and even considered a subspecies of it. Norealidys andaluciana was described in Spain and is usually synonymized with E. neapolitana. We collected 69 specimens from Italy, Spain, and Cyprus and studied fve molecular markers (COI, 16S, 28S, 12S, and ND1) and their morphology to solve this taxonomic problem. Phylogenetic analyses reveal the possible existence of two separate genera confounded under the name Eiseniella, but the study of more molecular markers and species of the genus would be necessary to confrm this. Therefore, the synonymy between Eiseniella and Norealidys is maintained. Various genetic analyses, including species delimitation, confrm the separation between E. neapolitana and E. andaluciana (=N. andaluciana) and excluded that E. neapolitana is a subspecies of E. tetraedra. The resemblance in external appearance despite clear genetic diferences of the three species could be explained by convergent adaptation to the aquatic habitat. Despite the expected low haplotype diversity based on the 28S gene, we found a surprisingly high variability in the E. andaluciana (=N. andaluciana) population in Spain. However, its stable predicted secondary structure and its high content of G+C reject the presence of a pseudogene

    The Gaia alerted fading of the FUor-type star Gaia21elv

    FU Orionis objects (FUors) are eruptive young stars, which exhibit outbursts that last from decades to a century. Due to the duration of their outbursts, and to the fact that only about two dozens of such sources are known, information on the end of their outbursts is limited. Here we analyse follow-up photometry and spectroscopy of Gaia21elv, a young stellar object, which had a several decades long outburst. It was reported as a Gaia science alert due to its recent fading by more than a magnitude. To study the fading of the source and look for signatures characteristic of FUors, we have obtained follow-up near infrared (NIR) spectra using Gemini South/IGRINS, and both optical and NIR spectra using VLT/X-SHOOTER. The spectra at both epochs show typical FUor signatures, such as a triangular shaped HH-band continuum, absorption-line dominated spectrum, and P Cygni profiles. In addition to the typical FUor signatures, [OI], [FeII], and [SII] were detected, suggesting the presence of a jet or disk wind. Fitting the spectral energy distributions with an accretion disc model suggests a decrease of the accretion rate between the brightest and faintest states. The rapid fading of the source in 2021 was most likely dominated by an increase of circumstellar extinction. The spectroscopy presented here confirms that Gaia21elv is a classical FUor, the third such object discovered among the Gaia science alerts.Comment: Accepted to MNRA

    Phylogenetic groups, virulence genes and quinolone resistance of integron-bearing Escherichia coli strains isolated from a wastewater treatment plant

    We investigated phylogenetic affiliation, occurrence of virulence genes and quinolone resistance in 109 integron-containing strains of Escherichia coli isolated from a wastewater treatment plant. Selection for integron-bearing strains caused a shift toward phylogroup D, which was most numerous, followed by A, B1 and B2. Phylogroups D and B2, both of which are reported to include virulent extraintestinal pathotypes, made up 50.5% of all isolates and were present in every stage of wastewater treatment, including final effluent. Diarrheagenic pathotypes made up 21% of the strains. The average virulence factor genes score was low (1.40) and the range was from 0 to 5. Quinolone and fluoroquinolone resistance was observed in 56.0% and 50.4% of the strains, respectively; however, it was not associated with virulence factor score. Although the average virulence factor score was low, 17.4% of strains had three and more virulence genes. They were isolated mostly from raw sewage, but 30% of them were cultured from final effluent. Release of multiresistant integron-bearing E. coli strains with virulence traits into the environment may create potential threat and be of public health concern

    Clear cell variant of diffuse large B-cell lymphoma: a case report

    <p>Abstract</p> <p>Introduction</p> <p>Diffuse large B-cell lymphoma is a diffuse proliferation of large neoplastic B lymphoid cells with a nuclear size equal to or exceeding the normal macrophage nuclei. We report a case of a clear cell variant of diffuse large B-cell lymphoma involving a lymph node in the neck, which was clinically suspected of being metastatic carcinoma.</p> <p>Case presentation</p> <p>A 39-year-old Caucasian ethnic Albanian man from Kosovo presented with a rapidly enlarging lymph node in his neck, but he also disclosed B symptoms and fatigue. A cytological aspirate of the lymph node revealed pleomorphic features. Our patient underwent a cervical lymph node biopsy (large excision). The mass was homogeneously fish-flesh, pale white tissue replacing almost the whole structure of the lymph node. The lymph node biopsy showed a partial alveolar growth pattern, which raised clinical suspicion that it was an epithelial neoplasm. With regard to morphological and phenotypic features, we discovered large nodules in diffuse areas, comprising large cells with slightly irregular nuclei and clear cytoplasm admixed with a few mononuclear cells. In these areas, there was high mitotic activity, and in some areas there were macrophages with tangible bodies. Staining for cytokeratins was negative. These areas had the following phenotypes: cluster designation marker 20 (CD20) positive, B-cell lymphoma (Bcl)-2-positive, Bcl-6<sup>-</sup>, CD5<sup>-</sup>, CD3<sup>-</sup>, CD21<sup>+ </sup>(in alveolar patterns), prostate-specific antigen-negative, human melanoma black marker 45-negative, melanoma marker-negative, cytokeratin-7-negative and multiple myeloma marker 1-positive in about 30% of cells, and exhibited a high proliferation index marker (Ki-67, 80%).</p> <p>Conclusion</p> <p>According to the immunohistochemical findings, we concluded that this patient has a clear cell variant of diffuse large B-cell lymphoma of activated cell type, post-germinal center cell origin. Our patient is undergoing R-CHOP chemotherapy treatment.</p
