    Vitamin C (ascorbic acid) requirements of Heterobranchus longifilis fingerlings

    A feeding experiment was conducted to quantify the minimum dietary vitamin C requirement of Heterobranchus longifilis fingerlings. Fish were fed a basal diet with 42.5% crude protein for a conditioning period of 2 weeks. Following conditioning, fingerlings with initial mean weight, 2.3 ± 0.3 g were stocked as groups of 20 fingerlings into 30 litre tanks in a mini-flow through experimental system. Graded levels (0, 50, 100, 150, 200 and 250) mg of L-ascorbic acid kg-1 diet was included into the basal diet by replacing part of the silica component and fed to triplicate groups for 20 weeks. Fish fed the control (0 mg vitamin C kg-1) diet exhibited deficiency signs including lordosis, caudal fin deformity,skin erosion and significantly (P < 0.05) suppressed weight gain and higher condition factor. Protein efficiency ratio and specific growth rate were significantly (P < 0.05) improved with increasing levels ofvitamin C up to 200 mg kg-1 diets. Tissues (liver, kidney, gills and muscle) ascorbate concentration generally reflected dietary inclusion levels with significant (P < 0.05) lowest level occurring in thecontrol groups. The dietary requirement based on least mean squares error regression analysis of weight gain and specific growth rate data on inclusion level of vitamin C was 82.2 ± 0.2 mg vitamin C kg-1 diet which corresponds to 100 mg of vitamin C kg-1 diet

    Dimerization-Induced Fermi-Surface Reconstruction in IrTe2

    We report a de Haas-van Alphen (dHvA) oscillation study on IrTe2 single crystals showing complex dimer formations. By comparing the angle dependence of dHvA oscillations with band structure calculations, we show distinct Fermi surface reconstruction induced by a 1/5-type and a 1/8-type dimerizations. This verifies that an intriguing quasi-two-dimensional conducting plane across the layers is induced by dimerization in both cases. A phase transition to the 1/8 phase with higher dimer density reveals that local instabilities associated with intra-and interdimer couplings are the main driving force for complex dimer formations in IrTe2.X11149sciescopu

    Successive spin-flop transitions of a Neel-type antiferromagnet Li2MnO3 single crystal with a honeycomb lattice

    We have carried out high magnetic field studies of single-crystalline Li2MnO3, a honeycomb lattice antiferromagnet. Its magnetic phase diagram was mapped out using magnetization measurements at applied fields up to 35 T. Our results show that it undergoes two successive meta-magnetic transitions around 9 T fields applied perpendicular to the ab plane (along the c* axis). These phase transitions are completely absent in the magnetization measured with the field applied along the ab plane. In order to understand this magnetic phase diagram, we developed a mean-field model starting from the correct Neel-type magnetic structure, consistent with our single crystal neutron diffraction data at zero field. Our model calculations succeeded in explaining the two meta-magnetic transitions that arise when Li2MnO3 enters two different spin-flop phases from the zero field Neel phase.open1187Nsciescopu

    Quantum Oscillation Signatures of Pressure-induced Topological Phase Transition in BiTeI

    We report the pressure-induced topological quantum phase transition of BiTeI single crystals using Shubnikov-de Haas oscillations of bulk Fermi surfaces. The sizes of the inner and the outer FSs of the Rashba-split bands exhibit opposite pressure dependence up to P=3.35 GPa, indicating pressure-tunable Rashba effect. Above a critical pressure P similar to 2 GPa, the Shubnikov-de Haas frequency for the inner Fermi surface increases unusually with pressure, and the Shubnikov-de Haas oscillations for the outer Fermi surface shows an abrupt phase shift. In comparison with band structure calculations, we find that these unusual behaviors originate from the Fermi surface shape change due to pressure-induced band inversion. These results clearly demonstrate that the topological quantum phase transition is intimately tied to the shape of bulk Fermi surfaces enclosing the time-reversal invariant momenta with band inversion.11117Ysciescopu

    The Hanle Effect in 1D, 2D and 3D

    This paper addresses the problem of scattering line polarization and the Hanle effect in one-dimensional (1D), two-dimensional (2D) and three-dimensional (3D) media for the case of a two-level model atom without lower-level polarization and assuming complete frequency redistribution. The theoretical framework chosen for its formulation is the QED theory of Landi Degl'Innocenti (1983), which specifies the excitation state of the atoms in terms of the irreducible tensor components of the atomic density matrix. The self-consistent values of these density-matrix elements is to be determined by solving jointly the kinetic and radiative transfer equations for the Stokes parameters. We show how to achieve this by generalizing to Non-LTE polarization transfer the Jacobi-based ALI method of Olson et al. (1986) and the iterative schemes based on Gauss-Seidel iteration of Trujillo Bueno and Fabiani Bendicho (1995). These methods essentially maintain the simplicity of the Lambda-iteration method, but their convergence rate is extremely high. Finally, some 1D and 2D model calculations are presented that illustrate the effect of horizontal atmospheric inhomogeneities on magnetic and non-magnetic resonance line polarization signals.Comment: 14 pages and 5 figure

    An Australian longitudinal pilot study examining health determinants of cardiac outcomes 12 months post percutaneous coronary intervention

    Background Percutaneous coronary intervention (PCI) is a very common revascularisation procedure for coronary artery disease (CAD). The purpose of this study was to evaluate cardiac outcomes, health related quality of life (HRQoL), resilience and adherence behaviours in patients who have undergone a PCI at two time points (6 and 12 months) following their procedure. Methods A longitudinal pilot study was conducted to observe the cardiac outcomes across a cohort of patients who had undergone a percutaneous coronary intervention (PCI). Participants who had undergone PCI 6 months prior were invited. Those participants who met the inclusion criteria and provided consent then completed a telephone survey (time point 1). These participants were then contacted 6 months later (i.e. 12 months post-intervention, time point 2) and the measures were repeated. Results All patients (n = 51) were recorded as being alive at time point 1. The multiple model indicated that controlling for other factors, gender was significantly associated with a linear combination of outcome measures (p = 0.004). The effect was moderate in magnitude (partial-η2 = 0.303), where males performed significantly better than females 6 months after the PCI procedure physically and with mood. Follow-up univariate ANOVAs indicated that gender differences were grounded in the scale measuring depression (PHQ9) (p = 0.005) and the physical component score of the short form measuring HRQoL (SF12-PCS) (p = 0.003). Thirteen patients were lost to follow-up between time points 1 and 2. One patient was confirmed to have passed away. The pattern of correlations between outcome measures at time point 2 revealed statistically significant negative correlation between the PHQ instrument and the resilience scale (CD-RISC) (r = -0.611; p < 0.001); and the physical component score of the SF-12 instrument (r = -0.437; p = 0.054). Conclusions Men were performing better than women in the 6 months post-PCI, particularly in the areas of mood (depression) and physical health. This pilot results indicate gender-sensitive practices are recommended particularly up to 6 months post-PCI. Any gender differences observed at 6 month appear to disappear at 12 months post-PCI. Further research into the management of mood particularly for women post-PCI is warranted. A more detailed inquiry related to access/attendance to secondary prevention is also warranted

    Developing an intervention to facilitate family communication about inherited genetic conditions, and training genetic counsellors in its delivery.

    Many families experience difficulty in talking about an inherited genetic condition that affects one or more of them. There have now been a number of studies identifying the issues in detail, however few have developed interventions to assist families. The SPRinG collaborative have used the UK Medical Research Council's guidance on Developing and Evaluating Complex Interventions, to work with families and genetic counsellors (GCs) to co-design a psycho-educational intervention to facilitate family communication and promote better coping and adaptation to living with an inherited genetic condition for parents and their children (<18 years). The intervention is modelled on multi-family discussion groups (MFDGs) used in psychiatric settings. The MFDG was developed and tested over three phases. First focus groups with parents, young people, children and health professionals discussed whether MFDG was acceptable and proposed a suitable design. Using evidence and focus group data, the intervention and a training manual were developed and three GCs were trained in its delivery. Finally, a prototype MFDG was led by a family therapist and co-facilitated by the three GCs. Data analysis showed that families attending the focus groups and intervention thought MFDG highly beneficial, and the pilot sessions had a significant impact on their family' functioning. We also demonstrated that it is possible to train GCs to deliver the MFDG intervention. Further studies are now required to test the feasibility of undertaking a definitive randomised controlled trial to evaluate its effectiveness in improving family outcomes before implementing into genetic counselling practice.The National Institute of Health Research funded the study but any views expressed do not necessarily reflect those of the Authority.     Upregulation of the cell-cycle regulator RGC-32 in Epstein-Barr virus-immortalized cells

    Epstein-Barr virus (EBV) is implicated in the pathogenesis of multiple human tumours of lymphoid and epithelial origin. The virus infects and immortalizes B cells establishing a persistent latent infection characterized by varying patterns of EBV latent gene expression (latency 0, I, II and III). The CDK1 activator, Response Gene to Complement-32 (RGC-32, C13ORF15), is overexpressed in colon, breast and ovarian cancer tissues and we have detected selective high-level RGC-32 protein expression in EBV-immortalized latency III cells. Significantly, we show that overexpression of RGC-32 in B cells is sufficient to disrupt G2 cell-cycle arrest consistent with activation of CDK1, implicating RGC-32 in the EBV transformation process. Surprisingly, RGC-32 mRNA is expressed at high levels in latency I Burkitt's lymphoma (BL) cells and in some EBV-negative BL cell-lines, although RGC-32 protein expression is not detectable. We show that RGC-32 mRNA expression is elevated in latency I cells due to transcriptional activation by high levels of the differentially expressed RUNX1c transcription factor. We found that proteosomal degradation or blocked cytoplasmic export of the RGC-32 message were not responsible for the lack of RGC-32 protein expression in latency I cells. Significantly, analysis of the ribosomal association of the RGC-32 mRNA in latency I and latency III cells revealed that RGC-32 transcripts were associated with multiple ribosomes in both cell-types implicating post-initiation translational repression mechanisms in the block to RGC-32 protein production in latency I cells. In summary, our results are the first to demonstrate RGC-32 protein upregulation in cells transformed by a human tumour virus and to identify post-initiation translational mechanisms as an expression control point for this key cell-cycle regulator

    Improving statistical inference on pathogen densities estimated by quantitative molecular methods: malaria gametocytaemia as a case study

    BACKGROUND: Quantitative molecular methods (QMMs) such as quantitative real-time polymerase chain reaction (q-PCR), reverse-transcriptase PCR (qRT-PCR) and quantitative nucleic acid sequence-based amplification (QT-NASBA) are increasingly used to estimate pathogen density in a variety of clinical and epidemiological contexts. These methods are often classified as semi-quantitative, yet estimates of reliability or sensitivity are seldom reported. Here, a statistical framework is developed for assessing the reliability (uncertainty) of pathogen densities estimated using QMMs and the associated diagnostic sensitivity. The method is illustrated with quantification of Plasmodium falciparum gametocytaemia by QT-NASBA. RESULTS: The reliability of pathogen (e.g. gametocyte) densities, and the accompanying diagnostic sensitivity, estimated by two contrasting statistical calibration techniques, are compared; a traditional method and a mixed model Bayesian approach. The latter accounts for statistical dependence of QMM assays run under identical laboratory protocols and permits structural modelling of experimental measurements, allowing precision to vary with pathogen density. Traditional calibration cannot account for inter-assay variability arising from imperfect QMMs and generates estimates of pathogen density that have poor reliability, are variable among assays and inaccurately reflect diagnostic sensitivity. The Bayesian mixed model approach assimilates information from replica QMM assays, improving reliability and inter-assay homogeneity, providing an accurate appraisal of quantitative and diagnostic performance. CONCLUSIONS: Bayesian mixed model statistical calibration supersedes traditional techniques in the context of QMM-derived estimates of pathogen density, offering the potential to improve substantially the depth and quality of clinical and epidemiological inference for a wide variety of pathogens
