    This paper addresses the fraught relationships among commodity trade, urban economic development and the environment in the world’s largest rainforest reserve, in a historical narrative fashion. The conceptual framework in which we position this narrative is provided by Hesse (2010), in the “site” and “situation” dimensions of the interaction between places or locales on the one hand, and material flows or global value chains on the other. It is argued that the assemblage of both site and situation is what shapes the wealth of cities. The case study of Manaus and Belém shows how the rapid urbanization of the Amazon rainforest is accompanied by the growth of shipping as “new” commodities are being extracted from the jungle interior

    Diversity, seasonality and sites of attachment of adult ixodid ticks on dogs in the central region of the Free State Province, South Africa

    Several surveys of ticks infesting dogs belonging to owners in resource-limited and more affluent communities have been conducted in South Africa, but no such investigation has been carried out in the Free State Province of this country. The present study was initiated to meet this shortcoming. Ticks were collected from dogs at six localities in, and to the east of the city of Bloemfontein in the central region of the province. Three of these localities could be classed as resource-limited and two as affluent, while the sixth locality was an animal shelter serving all members of the public. Adult ticks belonging to nine ixodid tick species were collected, of which Rhipicephalus sanguineus was the most numerous. Significantly more R. sanguineus was collected from dogs at resource-limited than at more affluent localities. The greatest proportions of these ticks attached to the backs and necks of the dogs, with the proportions being larger in long-haired than in short-haired dogs. Most R. sanguineus were collected during the warmer months particularly from January to April. The greatest proportions of Haemaphysalis leachi, the next most numerous species, were also collected from the backs and the necks of the dogs.     Compositional specification of functionality and timing of manufacturing systems

    In this paper, a formal modeling approach is introduced for compositional specification of both functionality and timing of manufacturing systems. Functionality aspects can be considered orthogonally to the timing. The functional aspects are specified using two abstraction levels; high-level activities and lower level actions. Design of a functionally correct controller is possible by looking only at the activity level, abstracting from the different execution orders of actions. Furthermore, the specific timing of actions is not needed. As a result, controller designcan be performed on a much smaller state space compared to an explicit model where timing and actions are present. The performance of the controller can be analyzed and optimizedby taking into account the timing characteristics. Since formal semantics are given in terms of a (max, +) state space, various existing performance analysis techniques can be used.     Physical activity and diet associations with the gut microbiota in the Coronary Artery Risk Development in Young Adults (CARDIA) study

    Background: Gut microbiota may influence metabolic pathways related to chronic health conditions. Evidence for physical activity and diet influences on gut microbial composition exists, but data from diverse population-based cohort studies are limited. Objectives: We hypothesized that gut microbial diversity and genera are associated with physical activity and diet quality. Methods: Data were from 537 participants in the Coronary Artery Risk Development in Young Adults (CARDIA) Study, a prospective cohort, who attended the year 30 follow-up examination (2015–2016; aged 47–61 y; 45% Black race/55% White race; 45% men/55% women). The 16S ribosomal RNA marker gene was sequenced from stool DNA, and genus-level taxonomy was assigned. Within-person microbial diversity (α-diversity) was assessed with Shannon diversity index and richness scores; between-person diversity (β-diversity) measures were generated with principal coordinates analysis (PCoA). Current and long-term physical activity and diet quality measures were derived from data collected over 30 y of follow-up. Multivariable-adjusted regression analysis controlled for: sociodemographic variables (age, race, sex, education, and field center), other health behaviors (smoking, alcohol consumption, and medication use), and adjusted for multiple comparisons with the false discovery rate (<0.20). Results: Based on PCoA β-diversity, participants’ microbial community compositions differed significantly (P < 0.001), with respect to both current and long-term physical activity and diet quality. α-Diversity was associated only with current physical activity (positively) in multivariable-adjusted analysis. Multiple genera (n = 45) were associated with physical activity and fewer with diet (n = 5), including positive associations with Lachnospiraceae UCG-001 and Ruminococcaceae Incertae Sedis with both behaviors. Conclusions: Physical activity and diet quality were associated with gut microbial composition among 537 participants in the CARDIA study. Multiple genera were associated with physical activity. Physical activity and diet quality were associated with genera consistent with pathways related to inflammation and short-chain fatty acid production

    Nacubactam enhances meropenem activity against carbapenem-resistant klebsiella pneumoniae producing KPC

    Carbapenem-resistant Enterobacteriaceae (CRE) are resistant to most antibiotics, making CRE infections extremely difficult to treat with available agents. Klebsiella pneumoniae carbapenemases (KPC-2 and KPC-3) are predominant carbapenemases in CRE in the United States. Nacubactam is a bridged diazabicyclooctane (DBO) -lactamase inhibitor that inactivates class A and C -lactamases and exhibits intrinsic antibiotic and -lactam “enhancer” activity against Enterobacteriaceae. In this study, we examined a collection of meropenem-resistant K. pneumoniae isolates carrying blaKPC-2 or blaKPC-3; meropenem-nacubactam restored susceptibility. Upon testing isogenic Escherichia coli strains producing KPC-2 variants with single-residue substitutions at important Ambler class A positions (K73, S130, R164, E166, N170, D179, K234, E276, etc.), the K234R variant increased the meropenem-nacubactam MIC compared to that for the strain producing KPC-2, without increasing the meropenem MIC. Correspondingly, nacubactam inhibited KPC-2 (apparent Ki [Kiapp] 31 3 M) more efficiently than the K234R variant (Kiapp 270 27 M) and displayed a faster acylation rate (k2/K), which was 5,815 582 M1 s1 for KPC-2 versus 247 25 M1 s1 for the K234R variant. Unlike avibactam, timed mass spectrometry revealed an intact sulfate on nacubactam and a novel peak (337 Da) with the K234R variant. Molecular modeling of the K234R variant showed significant catalytic residue (i.e., S70, K73, and S130) rearrangements that likely interfere with nacubactam binding and acylation. Nacubactam’s aminoethoxy tail formed unproductive interactions with the K234R variant’s active site. Molecular modeling and docking observations were consistent with the results of biochemical analyses. Overall, the meropenem-nacubactam combination is effective against carbapenem-resistant K. pneumoniae. Moreover, our data suggest that -lactamase inhibition by nacubactam proceeds through an alternative mechanism compared to that for avibactam
