    Genetic variability of kernel provitamin-A in sub-tropically adapted maize hybrids possessing rare allele of β-carotene hydroxylase

    Vitamin-A deficiency is a major health concern. Traditional yellow maize possesses low provitamin-A (proA). Mutant crtRB1 gene significantly enhances proA. 24 experimental hybrids possessing crtRB1 allele were evaluated for β-carotene (BC), β-cryptoxanthin (BCX), lutein (LUT), zeaxanthin (ZEA), total carotenoids (TC) and grain yield at multi-locations. BC (0.64–17.24 µg/g), BCX (0.45–6.84 µg/g), proA (0.86–20.46 µg/g), LUT (9.60–31.03 µg/g), ZEA (1.24–12.73 µg/g) and TC (20.60–64.02 µg/g) showed wide variation. No significant genotype × location interaction was observed for carotenoids. The mean BC (8.61 µg/g), BCX (4.04 µg/g) and proA (10.63 µg/g) in crtRB1-based hybrids was significantly higher than normal hybrids lacking crtRB1-favourable allele (BC: 1.73 µg/g, BCX: 1.29 µg/g and proA: 2.37 µg/g). Selected crtRB1-based hybrids possessed 33% BC and 40% BCX compared to 6% BC and 5% BCX in normal hybrids. BC showed positive correlation with BCX (r = 0.90), proA (r = 0.99) and TC (r = 0.64) among crtRB1-based hybrids. Carotenoids didn't show association with grain yield. Average yield potential of proA rich hybrids (6794 kg/ha) was at par with normal hybrids (6961 kg/ha). PROAH-13, PROAH-21, PROAH-17, PROAH-11, PROAH-23, PROAH-24 and PROAH-3 were the most promising with >12 µg/g proA and >6000 kg/ha grain yield.     Urinary active transforming growth factor ß in feline chronic kidney disease

    The cytokine transforming growth factor beta 1 (TGF-β1) has been widely implicated in the development and progression of renal fibrosis in chronic kidney disease (CKD) in humans and in experimental models. The aims of this study were to assess the association between urinary active TGF-β1 and (a) development of CKD in a cross-sectional study, (b) deterioration of renal function over 1 year in a longitudinal study, and (c) renal histopathological parameters in cats. A human active TGF-β1 ELISA was validated for use in feline urine. Cross-sectional analysis revealed no significant difference in urinary active TGF-β1:creatinine ratio (aTGF-β1:UCr) between groups with differing renal function. Longitudinally, non-azotaemic cats that developed CKD demonstrated a significant (P = 0.028) increase in aTGF-β1:UCr approximately 6 months before the development of azotaemia, which remained elevated (P = 0.046) at diagnosis (approximately 12 months prior, 8.4 pg/mg; approximately 6 months prior, 22.2 pg/mg; at CKD diagnosis, 24.6 pg/mg). In the histopathology study, aTGF-β1:UCr was significantly higher in cats with moderate (P = 0.02) and diffuse (P = 0.005) renal fibrosis than in cats without fibrosis. Cats with moderate renal inflammation had significantly higher urinary active aTGF-β1 concentrations than cats with mild (P = 0.035) or no inflammatory change (P = 0.004). The parameter aTGF-β1:UCr was independently associated with Log urine protein:creatinine ratio in a multivariable analysis of clinicopathological parameters and interstitial fibrosis score in a multivariable analysis of histopathological features. These results suggest that urinary aTGF-β1 reflects the severity of renal pathology. Increases in urinary aTGF-β1 followed longitudinally in individual cats may indicate the development of CKD

    Percutaneous Mitral Valve Annuloplasty in Patients With Secondary Mitral Regurgitation and Severe Left Ventricular Enlargement.

    This study sought to determine the effect of percutaneous mitral valve annuloplasty with the Carillon device versus guideline-directed medical therapy (GDMT) alone in patients with secondary mitral regurgitation (MR) and severe left ventricular (LV) enlargement.The clinical impact of the Carillon device in patients with severe LV dilation is not well established.This is a pooled analysis involving 3 prospective trials (TITAN [Transcatheter Implantation of Carillon Mitral Annuloplasty Device], TITAN II, and REDUCE FMR [CARILLON Mitral Contour System for Reducing Functional Mitral Regurgitation] trials) in which patients with functional MR and severe LV enlargement (LV end-diastolic diameter65 mm) were treated with GDMT and the Carillon device versus GDMT alone. Key outcomes of this analysis were changes over 1 year of follow-up in mitral valve and LV echocardiographic parameters, functional outcome, quality of life, mortality, and heart failure hospitalization (HFH).A total of 95 patients (67 in the Carillon group, 28 in the GDMT group) with severe LV enlargement were included. In the Carillon group, all mitral valve and LV morphology parameters were significantly improved at 1 year. Regurgitant volume decreased by 12 ml (p 0.001), MR grade decreased by 0.6 U (p 0.001), LV end-diastolic volume decreased by 25 cmIn patients with functional MR and severe LV enlargement, the Carillon device improved mitral valve function, LV morphology, and functional outcome compared with patients receiving GDMT only. Preoperative LV dimension should not be a limiting factor when evaluating patient eligibility or anticipated response to therapy with the Carillon device

    Endothelial miR-30c suppresses tumor growth via inhibition of TGF-β–induced Serpine1

    In tumors, extravascular fibrin forms provisional scaffolds for endothelial cell (EC) growth and motility during angiogenesis. We report that fibrin-mediated angiogenesis was inhibited and tumor growth delayed following postnatal deletion of Tgfbr2 in the endothelium of Cdh5-CreERT2 Tgfbr2fl/fl mice (Tgfbr2iECKOmice). ECs from Tgfbr2iECKO mice failed to upregulate the fibrinolysis inhibitor plasminogen activator inhibitor 1 (Serpine1, also known as PAI-1), due in part to uncoupled TGF-β–mediated suppression of miR-30c. Bypassing TGF-β signaling with vascular tropic nanoparticles that deliver miR-30c antagomiRs promoted PAI-1–dependent tumor growth and increased fibrin abundance, whereas miR-30c mimics inhibited tumor growth and promoted vascular-directed fibrinolysis in vivo. Using single-cell RNA-Seq and a NanoString miRNA array, we also found that subtypes of ECs in tumors showed spectrums of Serpine1 and miR-30c expression levels, suggesting functional diversity in ECs at the level of individual cells; indeed, fresh EC isolates from lung and mammary tumor models had differential abilities to degrade fibrin and launch new vessel sprouts, a finding that was linked to their inverse expression patterns of miR-30c and Serpine1 (i.e., miR-30chi Serpine1lo ECs were poorly angiogenic and miR-30clo Serpine1hi ECs were highly angiogenic). Thus, by balancing Serpine1 expression in ECs downstream of TGF-β, miR-30c functions as a tumor suppressor in the tumor microenvironment through its ability to promote fibrin degradation and inhibit blood vessel formation

    Effects of drying methods on the characteristics of rambutan (Nephelium lappaceum L.) seed fat: an optimisation approach

    The pre-treatment of oilseeds prior to extraction process may affect oil yield and quality. The aim of this study was to investigate the effects of two drying methods on rambutan seed fat (RSF) yield and their oxidative stability, physicochemical properties, and crystal morphology. Response surface methodology (RSM) was used in the optimisation and investigation of the effects of three process conditions: seed weight (g), extraction time (min), and solvent volume (ml) on RSF yield. Under optimal conditions, a maximum RSF yield of 44.14% was obtained. The differences between RSF pre-treated with oven-drying and RSF with freeze-drying methods in slip melting point (38.3◦C to 39.7◦C), free fatty acid (3.13 to 3.50 mg KOH/g fat), peroxide value (1.04 to 1.67 meq of O2/kg of fat), p-anisidine value (1.10 to 1.56), and total oxidation value (4.21 to 5.67) were significant (p < 0.05). Both fats showed needle-like shaped crystals. Our results provide usefu linformation in the pre-treatment of RSF, which has potential to be used as blending component with palm oil for cocoa butter equivalent formulation in chocolate and confectionery industries

    Lattice Study of the Massive Schwinger Model with a θ\theta term under L\"uscher's "Admissibility" condition

    We present a numerical study of the massive two-flavor QED in two dimensions with the gauge action proposed by L\"uscher, which allows only ``admissible'' gauge fields. We find that the admissibility condition does not allow any topology changes by the local updation in Hybrid Monte Carlo algorithm so that the configurations in each topological sector can be generated separately. By developing a new method to sum over different topological sectors, we investigate θ\theta vacuum effects. Combining with domain-wall fermion action, we obtain the fermion mass dependence and θ\theta dependence of the meson masses, which are consistent with the analytic results by mass perturbation in the continuum theory.Comment: 3 pages, Lattice2003(chiral

    Nucleon axial and pseudoscalar form factors from the covariant Faddeev equation

    We compute the axial and pseudoscalar form factors of the nucleon in the Dyson-Schwinger approach. To this end, we solve a covariant three-body Faddeev equation for the nucleon wave function and determine the matrix elements of the axialvector and pseudoscalar isotriplet currents. Our only input is a well-established and phenomenologically successful ansatz for the nonperturbative quark-gluon interaction. As a consequence of the axial Ward-Takahashi identity that is respected at the quark level, the Goldberger-Treiman relation is reproduced for all current-quark masses. We discuss the timelike pole structure of the quark-antiquark vertices that enters the nucleon matrix elements and determines the momentum dependence of the form factors. Our result for the axial charge underestimates the experimental value by 20-25% which might be a signal of missing pion-cloud contributions. The axial and pseudoscalar form factors agree with phenomenological and lattice data in the momentum range above Q^2 ~ 1...2 GeV^2.Comment: 17 pages, 7 figures, 1 tabl

They however remain observationally elusive, thus raising a question about characteristic observational signatures associated with these systems.
