    Highlighting when animals expend excessive energy for travel using dynamic body acceleration

    Travel represents a major cost for many animals so there should be selection pressure for it to be efficient ā€“ at minimum cost. However, animals sometimes exceed minimum travel costs for reasons that must be correspondingly important. We use Dynamic Body Acceleration (DBA), an acceleration-based metric, as a proxy for movement-based power, in tandem with vertical velocity (rate of change in depth) in a shark (Rhincodon typus) to derive the minimum estimated power required to swim at defined vertical velocities. We show how subtraction of measured DBA from the estimated minimum power for any given vertical velocity provides a ā€œproxy for power above minimumā€ metric (PPAmin), highlighting when these animals travel above minimum power. We suggest that the adoption of this metric across species has value in identifying where and when animals are subject to compelling conditions that lead them to deviate from ostensibly judicious energy expenditure

    Two-dimensional turbulence appears to be a more formidable problem than three-dimensional turbulence despite the numerical advantage of working with one less dimension. In the present paper we review recent numerical investigations of the phenomenology of two-dimensional turbulence as well as recent theoretical breakthroughs by various leading researchers. We also review efforts to reconcile the observed energy spectrum of the atmosphere (the spectrum) with the predictions of two-dimensional turbulence and quasi-geostrophic turbulence.Comment: Invited review; accepted by J.     Multi-scale waves in sound-proof global simulations with EULAG

    EULAG is a computational model for simulating flows across a wide range of scales and physical scenarios. A standard option employs an anelastic approximation to capture nonhydrostatic effects and simultaneously filter sound waves from the solution. In this study, we examine a localized gravity wave packet generated by instabilities in Held-Suarez climates. Although still simplified versus the Earthā€™s atmosphere, a rich set of planetary wave instabilities and ensuing radiated gravity waves can arise. Wave packets are observed that have lifetimes ā‰¤ 2 days, are negligibly impacted by Coriolis force, and do not show the rotational effects of differential jet advection typical of inertia-gravity waves. Linear modal analysis shows that wavelength, period, and phase speed fit the dispersion equation to within a mean difference of āˆ¼ 4%, suggesting an excellent fit. However, the group velocities match poorly even though a propagation of uncertainty analysis indicates that they should be predicted as well as the phase velocities. Theoretical arguments suggest the discrepancy is due to nonlinearity ā€” a strong southerly flow leads to a critical surface forming to the southwest of the wave packet that prevents the expected propagation

    Structure and Function of the Hair Cell Ribbon Synapse

    Faithful information transfer at the hair cell afferent synapse requires synaptic transmission to be both reliable and temporally precise. The release of neurotransmitter must exhibit both rapid on and off kinetics to accurately follow acoustic stimuli with a periodicity of 1 ms or less. To ensure such remarkable temporal fidelity, the cochlear hair cell afferent synapse undoubtedly relies on unique cellular and molecular specializations. While the electron microscopy hallmark of the hair cell afferent synapse ā€” the electron-dense synaptic ribbon or synaptic body ā€” has been recognized for decades, dissection of the synapseā€™s molecular make-up has only just begun. Recent cell physiology studies have added important insights into the synaptic mechanisms underlying fidelity and reliability of sound coding. The presence of the synaptic ribbon links afferent synapses of cochlear and vestibular hair cells to photoreceptors and bipolar neurons of the retina. This review focuses on major advances in understanding the hair cell afferent synapse molecular anatomy and function that have been achieved during the past years

    Discovery and functional prioritization of Parkinson's disease candidate genes from large-scale whole exome sequencing.

    BACKGROUND: Whole-exome sequencing (WES) has been successful in identifying genes that cause familial Parkinson's disease (PD). However, until now this approach has not been deployed to study large cohorts of unrelated participants. To discover rare PD susceptibility variants, we performed WES in 1148 unrelated cases and 503 control participants. Candidate genes were subsequently validated for functions relevant to PD based on parallel RNA-interference (RNAi) screens in human cell culture and Drosophila and C. elegans models. RESULTS: Assuming autosomal recessive inheritance, we identify 27 genes that have homozygous or compound heterozygous loss-of-function variants in PD cases. Definitive replication and confirmation of these findings were hindered by potential heterogeneity and by the rarity of the implicated alleles. We therefore looked for potential genetic interactions with established PD mechanisms. Following RNAi-mediated knockdown, 15 of the genes modulated mitochondrial dynamics in human neuronal cultures and four candidates enhanced Ī±-synuclein-induced neurodegeneration in Drosophila. Based on complementary analyses in independent human datasets, five functionally validated genes-GPATCH2L, UHRF1BP1L, PTPRH, ARSB, and VPS13C-also showed evidence consistent with genetic replication. CONCLUSIONS: By integrating human genetic and functional evidence, we identify several PD susceptibility gene candidates for further investigation. Our approach highlights a powerful experimental strategy with broad applicability for future studies of disorders with complex genetic etiologies
