    Advanced Gas Turbine (AGT) powertrain system

    A 74.5 kW(100 hp) advanced automotive gas turbine engine is described. A design iteration to improve the weight and production cost associated with the original concept is discussed. Major rig tests included 15 hours of compressor testing to 80% design speed and the results are presented. Approximately 150 hours of cold flow testing showed duct loss to be less than the design goal. Combustor test results are presented for initial checkout tests. Turbine design and rig fabrication is discussed. From a materials study of six methods to fabricate rotors, two have been selected for further effort. A discussion of all six methods is given

    Racial Dynamics in Counselor Training: The Racial Identity Social Interaction Model

    Counselors frequently receive their initial training about the dynamics of race and culture in the counseling process in didactic group settings, such as multicultural courses and experiential skills-building labs. Whereas multicultural and diversity courses reportedly have been growth promoting for students, counselor educators describe several difficulties that arise as they attempt to teach these courses. Yet virtually no research has focused on examination of instructors’ difficulties from a theoretical perspective. To examine the complex, intersecting dynamics that occur when teaching groups of counselor trainees about race and culture, we used Directed Content Analysis with theoretical guidance from the Racial Identity Social Interaction Model. We analyzed interviews obtained from instructors (n = 8) who had led small-group counseling skills labs with a multicultural and social justice perspective. Problematic dynamics occurred in three major domains, group, leader, and institutional dynamics. Implications for teaching about race and culture in group settings are discussed

    A Fascinating Polynomial Sequence arising from an Electrostatics Problem on the Sphere

    A positive unit point charge approaching from infinity a perfectly spherical isolated conductor carrying a total charge of +1 will eventually cause a negatively charged spherical cap to appear. The determination of the smallest distance ρ(d)\rho(d) (dd is the dimension of the unit sphere) from the point charge to the sphere where still all of the sphere is positively charged is known as Gonchar's problem. Using classical potential theory for the harmonic case, we show that 1+ρ(d)1+\rho(d) is equal to the largest positive zero of a certain sequence of monic polynomials of degree 2d12d-1 with integer coefficients which we call Gonchar polynomials. Rather surprisingly, ρ(2)\rho(2) is the Golden ratio and ρ(4)\rho(4) the lesser known Plastic number. But Gonchar polynomials have other interesting properties.     Response of selected microorganisms to experimental planetary environments

    The anaerobic utilization of phosphite or phosphine and the significance of this conversion to potential contamination of Jupiter were investigated. A sporeforming organism was isolated from Cape Canaveral soil which anaerobically converts hypophosphite to phosphate. This conversion coincides with an increase in turbidity of the culture and with phosphate accumulation in the medium. Investigations of omnitherms (organisms which grow over a broad temperature range, i.e. 3 -55 C were also conducted. The cellular morphology of 28 of these isolates was investigated, and all were demonstrated to be sporeformers. Biochemical characterizations are also presented. Procedures for replicate plating were evaluated, and those results are also presented. The procedures for different replicate-plating techniques are presented, and these are evaluated on the basis of reproducibility, percentage of viable transfer, and ease of use. Standardized procedures for the enumeration of microbial populations from ocean-dredge samples from Cape Canaveral are also presented

    The transcriptome of utricle hair cell regeneration in the avian inner ear

    Sensory hair cell loss is the major cause of hearing and balance disorders. Mammals are incapable of sustained hair cell regeneration, but lower vertebrates can regenerate these mechano-electrical transducers. We present the first comprehensive transcriptome (by mRNA-Seq) of hair cell regeneration in the chick utricle. We provide pathway and pattern annotations and correlate these with the phenotypic events that occur during regeneration. These patterns are surprisingly synchronous and highly punctuated. We show how these patterns are a new resource for identifying components of the hair cell transcriptome and identify 494 new putative hair-cell-specific genes and validate three of these (of three tested) by immunohistochemical staining. We describe many surprising new components and dynamic expression patterns, particularly within NOTCH signaling. For example, we show that HES7 is specifically expressed during utricle hair cell regeneration and closely parallels the expression of HES5. Likewise, the expression of ATOH1 is closely correlated with HEYL and the HLH inhibitory transcription factors ID1, ID2, and ID4. We investigate the correlation between fibroblast growth factor signaling and supporting cell proliferation and show that FGF20 inhibits supporting cell proliferation. We also present an analysis of 212 differentially expressed transcription factor genes in the regenerative time course that fall into nine distinct gene expression patterns, many of which correlate with phenotypic events during regeneration and represent attractive candidates for future analysis and manipulation of the regenerative program in sensory epithelia and other vertebrate neuroepithelia

    Structure and oxidation kinetics of the Si(100)-SiO2 interface

    We present first-principles calculations of the structural and electronic properties of Si(001)-SiO2 interfaces. We first arrive at reasonable structures for the c-Si/a-SiO2 interface via a Monte-Carlo simulated annealing applied to an empirical interatomic potential, and then relax these structures using first-principles calculations within the framework of density-functional theory. We find a transition region at the interface, having a thickness on the order of 20\AA, in which there is some oxygen deficiency and a corresponding presence of sub-oxide Si species (mostly Si^+2 and Si^+3). Distributions of bond lengths and bond angles, and the nature of the electronic states at the interface, are investigated and discussed. The behavior of atomic oxygen in a-SiO2 is also investigated. The peroxyl linkage configuration is found to be lower in energy than interstitial or threefold configurations. Based on these results, we suggest a possible mechanism for oxygen diffusion in a-SiO2 that may be relevant to the oxidation process.Comment: 7 pages, two-column style with 6 postscript figures embedded.     Habitat Use by Middle Mississippi River Pallid Sturgeon

    Little is known about the habitat preferences and needs of pallid sturgeon Scaphirhynchus albus, which was federally listed as endangered in 1990. To learn more about habitat use and selection by pallid sturgeon, sonic transmitters were surgically implanted in 27 individuals from the middle Mississippi River. Study fish were located 184 times (1–23 times/individual) from November 1995 to December 1999. Of the seven macrohabitats identified, pallid sturgeon were found most often in main-channel habitats (39% of all relocations) and main-channel border habitats (26%); the between-wing-dam habitats were used less often (14%). Strauss’s linear selectivity index (Li) values indicated that study fish exhibited positive selection for the main-channel border, downstream island tips, between-wing-dam, and wing-dam-tip habitats; they showed negative selection for main-channel, downstream of wing dams, and upstream of wing dam habitats. Comparison of Li values for four temperature ranges and three daily mean discharge ranges revealed little change in habitat selection due to temperature or discharge. Habitat use patterns also were similar across seasons and discharge regimes, except during spring months when between-wingdam habitats saw greater use and main-channel and main-channel border habitat use declined. These changes may have been a response to high river stages associated with spring flooding, which may create favorable feeding areas in the between-wing-dam habitats. Enhancement and restoration of habitat diversity, particularly downstream island tip and between-wing-dam habitats, may be necessary for the recovery of pallid sturgeon in the middle Mississippi River

    The fate of porcine sperm CRISP2 from the perinuclear theca before and after in vitro fertilization

    In a previous study, we reported that porcine sperm cysteine-rich secretory protein 2 (CRISP2) is localized in the post-acrosomal sheath-perinuclear theca (PT) as reduction-sensitive oligomers. In the current study, the decondensation and removal of CRISP2 was investigated during in vitro sperm capacitation, after both the induction of the acrosome reaction and in vitro fertilization. Confocal immunofluorescent imaging revealed that additional CRISP2 fluorescence appeared on the apical ridge and on the equatorial segment (EqS) of the sperm head following capacitation, likely due to cholesterol removal. After an ionophore A23187-induced acrosome reaction, CRISP2 immunofluorescence disappeared from the apical ridge and the EqS area partly not only owing to the removal of the acrosomal shroud vesicles, but to its presence in a subdomain of EqS. The fate of sperm head CRISP2 was further examined post-fertilization. In vitro matured porcine oocytes were co-incubated with boar sperm cells for 6-8 h and the zygotes were processed for CRISP2 immunofluorescent staining. Notably, decondensation of CRISP2, and thus of the sperm PT, occurred while the sperm nucleus was still fully condensed. CRISP2 was no longer detectable in fertilized oocytes in which sperm nuclear decondensation and paternal pronucleus formation were apparent. This rapid dispersal of CRISP2 in the PT is likely regulated by redox reactions for which its cysteine-rich domain is sensitive.
