    Qualitative and quantitative analysis of milk for the detection of adulteration by Laser Induced Breakdown Spectroscopy (LIBS)

    The present work focuses on the development of a fast and cost effective method based on Laser Induced Breakdown Spectroscopy (LIBS) to the quality control, traceability and detection of adulteration in milk. Two adulteration cases have been studied; a qualitative analysis for the discrimination between different milk blends and quantification of melamine in adulterated toddler milk powder. Principal Component Analysis (PCA) and neural networks (NN) have been used to analyze LIBS spectra obtaining a correct classification rate of 98% with a 100% of robustness. For the quantification of melamine, two methodologies have been developed; univariate analysis using CN emission band and multivariate calibration NN model obtaining correlation coefficient (R2) values of 0.982 and 0.999 respectively. The results of the use of LIBS technique coupled with chemometric analysis are discussed in terms of its potential use in the food industry to perform the quality control of this dairy product

    Local and remote sources of airborne suspended particulate matter in the antarctic region

    Quantification of suspended particulate matter (SPM) measurements-together with statistical tools, polar contour maps and backward air mass trajectory analyses-were implemented to better understand the main local and remote sources of contamination in this pristine region. Field campaigns were carried out during the austral summer of 2016-2017 at the "Gabriel de Castilla" Spanish Antarctic Research Station, located on Deception Island (South Shetland Islands, Antarctic). Aerosols were deposited in an air filter through a low-volume sampler and chemically analysed using Inductively Coupled Plasma-Mass Spectrometry (ICP-MS) and Inductively Coupled Plasma-Atomic Emission Spectroscopy (ICP-AES). Elements such as Al, Ca, Fe, K, Mg, Na, P, S, Cu, Pb, Sr, Ti, Zn, Hf, Zr, V, As, Ti, Mn, Sn and Cr were identified. The statistical tools together with their correlations (Sr/Na, Al/Ti, Al/Mn, Al/Sr, Al/Pb, K/P) suggest a potentially significant role of terrestrial inputs for Al, Ti, Mn, Sr and Pb; marine environments for Sr and Na; and biological inputs for K and P. Polar contour graphical maps allowed reproducing wind maps, revealing the biological local distribution of K and P (penguin colony). Additionally, backward trajectory analysis confirmed previous affirmations and atmospheric air masses following the Antarctic circumpolar pattern

    Determination of lanthanides in fossil samples using laser induced breakdown spectroscopy

    As being a fast, simple and relatively non-destructive analytical technique Laser-induced breakdown spectroscopy (LIBS) has a large variety of applications including the analysis of paleontological samples. In this work LIBS is employed for the quantitative determination of lanthanides (Ce, Dy, Er, Eu, Gd, Ho, La, Lu, Nd, Sm, Tb, Tm and Yb) in vertebrate fossil samples comprising teeth, disarticulated complete or fragmented bones, eggshell fragments, and coprolites of dinosaurs, mammals and crocodiles. For emission line data, standard AnalaR grade salts of lanthanides were used. The major components: Iron, calcium, magnesium, silicon and aluminum in the samples were also determined. The analytical information may be helpful in studying the samples for their age, formation environment and other paleontological properties

    Assessment of the variability of atmospheric pollution in national parks of mainland Spain

    Air quality in nine National Parks in mainland Spain was assessed analysing SO2, NOx, O3, PM10 and PM2.5 data from background stations. As emissions in and around parks are limited, the levels of primary pollutants are low. Concentrations of secondary pollutants are high especially in summer due to photochemical production. The geographical variability of pollutants responds to regional emission patterns and the dominant circulation regimes in different regions resulting in west-east gradients for O3 and PM. Seasonal variability of pollutants was also interpreted in virtue of transport scenarios, changes in photochemical activity and emissions variability. NOx and SO2, maximize in winter due to higher emissions while O3 and PM do it in summer due to photochemical production, lower precipitation and, in the case of PM, the occurrence of African dust outbreaks. The diurnal evolution was interpreted in virtue of variability in emissions and changes in the Planetary Boundar Layer height

    Multielemental analysis of Antarctic soils using calibration free laser-induced breakdown spectroscopy

    Laser-induced breakdown spectroscopy (LIBS) is a quick technique that allows the analysis of all types of samples without destroying them and with much reduced sample treatment. One of its many applications is the study of geological samples such as soils. Because of the complexity of the matrix, it is very difficult to find or manufacture standards for these types of samples. Therefore, a good alternative is to make use of a methodology, called Calibration Free (CF), where instead of using standards, the physical parameters of the plasma created by the interaction of the laser with the sample are studied and related to the elements and species that compose it. This methodology is followed to perform a multielemental quantitative analysis of soil samples from Antarctica. Two studies were made, differing in the optimization of the instrumental parameters in order to obtain the best possible spectra in the chosen spectral lines. In both cases, the signal to noise ratio (SNR) was used to evaluate the quality of the spectra, but in the second study a full factorial design 23 with center and axial points was developed to get better results. The choice of spectral lines was based on a series of criteria, being stricter in the second study. The samples were mainly composed of the following oxides: SiO2, Al2O3, Fe2O3, CaO, MgO, Na2O, TiO and K2O. In the second study, it was also possible to determine the species present in lower concentrations: Mn, Cr, V, Sr, Zr, BA and Li. The results were compared with those provided by ICP-OES analysis, obtaining close values for most oxides, especially in the second study. For minority elements, the CF-LIBS and the ICP-OES results were within the same order of magnitude in all cases except the Cr case. These results show that CF-LIBS can be very useful in the characterization of complex samples from remote regions, such as Antarctic soils

    Thyroid hormone activation of retinoic acid synthesis in hypothalamic tanycytes

    yesThyroid hormone (TH) is essential for adult brain function and its actions include several key roles in the hypothalamus. Although TH controls gene expression via specific TH receptors of the nuclear receptor class, surprisingly few genes have been demonstrated to be directly regulated by TH in the hypothalamus, or the adult brain as a whole. This study explored the rapid induction by TH of retinaldehyde dehydrogenase 1 (Raldh1), encoding a retinoic acid (RA)-synthesizing enzyme, as a gene specifically expressed in hypothalamic tanycytes, cells that mediate a number of actions of TH in the hypothalamus. The resulting increase in RA may then regulate gene expression via the RA receptors, also of the nuclear receptor class. In vivo exposure of the rat to TH led to a significant and rapid increase in hypothalamic Raldh1 within 4 hours. That this may lead to an in vivo increase in RA is suggested by the later induction by TH of the RA-responsive gene Cyp26b1. To explore the actions of RA in the hypothalamus as a potential mediator of TH control of gene regulation, an ex vivo hypothalamic rat slice culture method was developed in which the Raldh1-expressing tanycytes were maintained. These slice cultures confirmed that TH did not act on genes regulating energy balance but could induce Raldh1. RA has the potential to upregulate expression of genes involved in growth and appetite, Ghrh and Agrp. This regulation is acutely sensitive to epigenetic changes, as has been shown for TH action in vivo. These results indicate that sequential triggering of two nuclear receptor signalling systems has the capability to mediate some of the functions of TH in the hypothalamus

    Raloxifene inhibits tumor growth and lymph node metastasis in a xenograft model of metastatic mammary cancer

    <p>Abstract</p> <p>Background</p> <p>The effects of raloxifene, a novel selective estrogen receptor modulator, were studied in a mouse metastatic mammary cancer model expressing cytoplasmic ERα.</p> <p>Methods</p> <p>Mammary tumors, induced by inoculation of syngeneic BALB/c mice with BJMC3879luc2 cells, were subsequently treated with raloxifene at 0, 18 and 27 mg/kg/day using mini-osmotic pumps.</p> <p>Results</p> <p><it>In vitro </it>study demonstrated that the ERα in BJMC3879luc2 cells was smaller (between 50 and 64 kDa) than the normal-sized ERα (66 kDa) and showed cytoplasmic localization. A statistically significant but weak estradiol response was observed in this cell line. When BJMC3879luc2 tumors were implanted into mice, the ERα mRNA levels were significantly higher in females than in males. <it>In vitro </it>studies showed that raloxifene induced mitochondria-mediated apoptosis and cell-cycle arrest in the G1-phase and a decrease in the cell population in the S-phase. In animal experiments, tumor volumes were significantly suppressed in the raloxifene-treated groups. The multiplicity of lymph node metastasis was significantly decreased in the 27 mg/kg group. Levels of apoptosis were significantly increased in the raloxifene-treated groups, whereas the levels of DNA synthesis were significantly decreased in these groups. No differences in microvessel density in tumors were observed between the control and raloxifene-treated groups. The numbers of dilated lymphatic vessels containing intraluminal tumor cells were significantly reduced in mammary tumors in the raloxifene-treated groups. The levels of ERα mRNA in mammary tumors tended to be decreased in the raloxifene-treated groups.</p> <p>Conclusion</p> <p>These results suggest that the antimetastatic activity of raloxifene in mammary cancer expressing cytoplasmic ERα may be a crucial finding with clinical applications and that raloxifene may be useful as an adjuvant therapy and for the chemoprevention of breast cancer development.</p
