    Arousal and Executive Alterations in Attention Deficit Hyperactivity Disorder (ADHD)

    Attention deficit hyperactivity disorder (ADHD) is the most common neurobehavioral disorder in childhood and can significantly affect a child’s personal and social development and academic achievement. Taking into account the model of attentional networks proposed by Posner et al., the aim of the present study was to review the literature regarding two main explicative models of ADHD, i.e., the inhibition model and the cognitive-energetic model, by discussing behavioral and neurological evidence of both models and the limitations of each model. The review highlights evidence that favors the energetic model and points to an unstable arousal as a potential pathogenetic factor in ADHD

    Contribution of microscopy for understanding the mechanism of action against trypanosomatids

    Transmission electron microscopy (TEM) has proved to be a useful tool to study the ultrastructural alterations and the target organelles of new antitrypanosomatid drugs. Thus, it has been observed that sesquiterpene lactones induce diverse ultrastructural alterations in both T. cruzi and Leishmania spp., such as cytoplasmic vacuolization, appearance of multilamellar structures, condensation of nuclear DNA, and, in some cases, an important accumulation of lipid vacuoles. This accumulation could be related to apoptotic events. Some of the sesquiterpene lactones (e.g., psilostachyin) have also been demonstrated to cause an intense mitochondrial swelling accompanied by a visible kinetoplast deformation as well as the appearance of multivesicular bodies. This mitochondrial swelling could be related to the generation of oxidative stress and associated to alterations in the ergosterol metabolism. The appearance of multilamellar structures and multiple kinetoplasts and flagella induced by the sesquiterpene lactone psilostachyin C indicates that this compound would act at the parasite cell cycle level, in an intermediate stage between kinetoplast segregation and nuclear division. In turn, the diterpene lactone icetexane has proved to induce the external membrane budding on T. cruzi together with an apparent disorganization of the pericellar cytoskeleton. Thus, ultrastructural TEM studies allow elucidating the possible mechanisms and the subsequent identification of molecular targets for the action of natural compounds on trypanosomatids.Fil: Lozano, Esteban Sebastián.     Host adaptive immunity deficiency in severe pandemic influenza

    INTRODUCTION: Pandemic A/H1N1/2009 influenza causes severe lower respiratory complications in rare cases. The association between host immune responses and clinical outcome in severe cases is unknown. METHODS: We utilized gene expression, cytokine profiles and generation of antibody responses following hospitalization in 19 critically ill patients with primary pandemic A/H1N1/2009 influenza pneumonia for identifying host immune responses associated with clinical outcome. Ingenuity pathway analysis 8.5 (IPA) (Ingenuity Systems, Redwood City, CA) was used to select, annotate and visualize genes by function and pathway (gene ontology). IPA analysis identified those canonical pathways differentially expressed (P < 0.05) between comparison groups. Hierarchical clustering of those genes differentially expressed between groups by IPA analysis was performed using BRB-Array Tools v.3.8.1. RESULTS: The majority of patients were characterized by the presence of comorbidities and the absence of immunosuppressive conditions. pH1N1 specific antibody production was observed around day 9 from disease onset and defined an early period of innate immune response and a late period of adaptive immune response to the virus. The most severe patients (n = 12) showed persistence of viral secretion. Seven of the most severe patients died. During the late phase, the most severe patient group had impaired expression of a number of genes participating in adaptive immune responses when compared to less severe patients. These genes were involved in antigen presentation, B-cell development, T-helper cell differentiation, CD28, granzyme B signaling, apoptosis and protein ubiquitination. Patients with the poorest outcomes were characterized by proinflammatory hypercytokinemia, along with elevated levels of immunosuppressory cytokines (interleukin (IL)-10 and IL-1ra) in serum. CONCLUSIONS: Our findings suggest an impaired development of adaptive immunity in the most severe cases of pandemic influenza, leading to an unremitting cycle of viral replication and innate cytokine-chemokine release.     Identifying metabolite markers for preterm birth in cervicovaginal fluid by magnetic resonance spectroscopy

    Introduction Preterm birth (PTB) may be preceded by changes in the vaginal microflora and metabolite profiles. Objectives We sought to characterise the metabolite profile of cervicovaginal fluid (CVF) of pregnant women by 1H NMR spectroscopy, and assess their predictive value for PTB. Methods A pair of high-vaginal swabs was obtained from pregnant women with no evidence of clinical infection and grouped as follows: asymptomatic low risk (ALR) women with no previous history of PTB, assessed at 20–22 gestational weeks, g.w., n = 83; asymptomatic high risk (AHR) women with a previous history of PTB, assessed at both 20–22 g.w., n = 71, and 26–28 g.w., n = 58; and women presenting with symptoms of preterm labor (PTL) (SYM), assessed at 24–36 g.w., n = 65. Vaginal secretions were dissolved in phosphate buffered saline and scanned with a 9.4 T NMR spectrometer. Results Six metabolites (lactate, alanine, acetate, glutamine/glutamate, succinate and glucose) were analysed. In all study cohorts vaginal pH correlated with lactate integral (r = -0.62, p\0.0001). Lactate integrals were higher in the term ALR compared to the AHR (20–22 g.w.) women (p = 0.003). Acetate integrals were higher in the preterm versus term women for the AHR (20–22 g.w.) (p = 0.048) and SYM (p = 0.003) groups; and was predictive of PTB\37 g.w. (AUC 0.78; 95 % CI 0.61–0.95), and delivery within 2 weeks of the index assessment (AUC 0.84; 95 % CI 0.64–1) in the SYM women, whilst other metabolites were not. Conclusion High CVF acetate integral of women with symptoms of PTL appears predictive of preterm delivery, as well as delivery within 2 weeks of presentation

    Establecimiento y manejo del frijol terciopelo (Mucuna sp. L.) como cobertura viva para el combate de malezas en maíz (Zea mays L.)

Sc) -- CATIE, Turrialba (Costa Rica), 1984El combate de malezas en maíz en zonas tropicales húmedas, requiere de un enfoque que combine los principios de combate existentes en los sistemas de Producción del pequeño agricultor, con las nuevas técnicas disponibles para el desarrollo de sistemas eficientes en el manejo de las malezas. En este sentido, el presente estudio realizado en las instalaciones del CATIE, Turrialba, Costa Rica, entre octubre de 1983 y abril de 1984, tuvo como objetivo evaluar el comportamiento y manejo de la leguminosa Mucuna sp. (125.000 pl/ha) como cobertura viva para el combate de malezas en maíz cv. Tuxpeño C-7 (50.000 pl/ha). La cobertura viva se sembró 40 y 55 días antes de la siembra del maíz. Para facilitar la siembra del maíz se aplicó paraquat (0,5 kg i.a/ha) en franjas sobre la cobertura de 55 días de edad. El mismo día se sembró el maíz entre las hileras de la cobertura de 40 días de edad. Algunos tratamientos con coberturas vivas recibieron aplicaciones de dicamba 3 días después de la emergencia del maíz, en dosis de 150 g i.a/ha. Cuarenta días después, parte de estos tratamientos tuvieron una segunda aplicación de dicamba en dosis de 75 g i.a/ha, para "regular" el crecimiento de la cobertura. Se consideraron además tratamientos con "mulch" de Mucuna sp., suprimidas por medios físicos y químicos, y combate de malezas manual y químico (sin coberturas). Las principales variables evaluadas en los 4 períodos de muestreo (25, 40, 55 y 120 DDSM) fueron: Incidencia y Producción de biomasa de malezas por especie y categoría (hoja ancha, hoja angosta y ciperáceas) índice de área foliar, biomasa y peso seco de nódulos para la cobertura viva índice de área foliar, altura de planta, biomasa y rendimiento en grano para el maíz. Los resultados mostraron una mayor incidencia (N{eth} malezas/m{ostrok}) y Producción de biomasa de malezas en las parcelas con mulch obtenido mediante corte manual, seguido de los tratamientos con coberturas vivas establecidas 40 días antes de la siembra del maíz. La menor incidencia y biomasa de malezas se halló en el tratamiento con mulch (con aplicaciones de paraquat) sin diferir del combate químico. Dos aplicaciones de dicamba en las coberturas vivas, provocaron muerte de las plantas a los 120 DDSM. En cambio una sola aplicación, presentó disminución en la biomasa y el IAF de la cobertura viva. La cobertura sin aplicación de dicamba, presentó los mayores valores de índice de área foliar y biomasa, provocando una drástica disminución en la altura de planta, biomasa y rendimientos del maíz. Los otros tratamientos no mostraron diferencias significativas en el rendimiento del maíz. Sin embargo, se puedo establecer tendencias en favor de los tratamientos con mulch. El tratamiento de mulch (paraquat), evidenció un adecuado combate de malezas, principalmente gramíneas

    The Validity of a Mixed Reality-Based Automated Functional Mobility Assessment

    Functional mobility assessments (i.e., Timed Up and Go) are commonly used clinical tools for mobility and fall risk screening in older adults. In this work, we proposed a new Mixed Reality (MR)-based assessment that utilized a Microsoft HoloLensTM headset to automatically lead and track the performance of functional mobility tests, and subsequently evaluated its validity in comparison with reference inertial sensors. Twenty-two healthy adults (10 older and 12 young adults) participated in this study. An automated functional mobility assessment app was developed, based on the HoloLens platform. The mobility performance was recorded with the headset built-in sensor and reference inertial sensor (Opal, APDM) taped on the headset and lower back. The results indicate that the vertical kinematic measurements by HoloLens were in good agreement with the reference sensor (Normalized RMSE ~ 10%, except for cases where the inertial sensor drift correction was not viable). Additionally, the HoloLens-based test completion time was in perfect agreement with the clinical standard stopwatch measure. Overall, our preliminary investigation indicates that it is possible to use an MR headset to automatically guide users (without severe mobility deficit) to complete common mobility tests, and this approach has the potential to provide an objective and efficient sensor-based mobility assessment that does not require any direct research/clinical oversight
