Arctic smoke - aerosol characteristics during a record smoke event in the European Arctic and its radiative impact
In early May 2006 a record high air pollution event was observed at Ny-Ă
lesund, Spitsbergen. An atypical weather pattern established a pathway for the rapid transport of biomass burning aerosols from agricultural fires in Eastern Europe to the Arctic. Atmospheric stability was such that the smoke was constrained to low levels, within 2 km of the surface during the transport. A description of this smoke event in terms of transport and main aerosol characteristics can be found in Stohl et al. (2007). This study puts emphasis on the radiative effect of the smoke. The aerosol number size distribution was characterised by lognormal parameters as having an accumulation mode centered around 165â185 nm and almost 1.6 for geometric standard deviation of the mode. Nucleation and small Aitken mode particles were almost completely suppressed within the smoke plume measured at Ny-Ă
lesund. Chemical and microphysical aerosol information obtained at Mt. Zeppelin (474 m a.s.l) was used to derive input parameters for a one-dimensional radiation transfer model to explore the radiative effects of the smoke. The daily mean heating rate calculated on 2 May 2006 for the average size distribution and measured chemical composition reached 0.55 K dayâ1 at 0.5 km altitude for the assumed external mixture of the aerosols but showing much higher heating rates for an internal mixture (1.7 K dayâ1). In comparison a case study for March 2000 showed that the local climatic effects due to Arctic haze, using a regional climate model, HIRHAM, amounts to a maximum of 0.3 K dayâ1 of heating at 2 km altitude (Treffeisen et al., 2005)
In vivo alkaline comet assay: Statistical considerations on historical negative and positive control data
The alkaline comet assay is frequently used as in vivo follow-up test within different regulatory environments to characterize the DNA-damaging potential of different test items. The corresponding OECD Test guideline 489 highlights the importance of statistical analyses and historical control data (HCD) but does not provide detailed procedures. Therefore, the working group âStatisticsâ of the German-speaking Society for Environmental Mutation Research (GUM) collected HCD from five laboratories and >200 comet assay studies and performed several statistical analyses. Key results included that (I) observed large inter-laboratory effects argue against the use of absolute quality thresholds, (II) > 50% zero values on a slide are considered problematic, due to their influence on slide or animal summary statistics, (III) the type of summarizing measure for single-cell data (e.g., median, arithmetic and geometric mean) may lead to extreme differences in resulting animal tail intensities and study outcome in the HCD. These summarizing values increase the reliability of analysis results by better meeting statistical model assumptions, but at the cost of information loss. Furthermore, the relation between negative and positive control groups in the data set was always satisfactorily (or sufficiently) based on ratio, difference and quantile analyses
Galanin pathogenic mutations in temporal lobe epilepsy
Temporal lobe epilepsy (TLE) is a common epilepsy syndrome with a complex etiology. Despite evidence for the participation of genetic factors, the genetic basis of TLE remains largely unknown. A role for the galanin neuropeptide in the regulation of epileptic seizures has been established in animal models more than two decades ago. However, until now there was no report of pathogenic mutations in GAL, the galanin-encoding gene, and therefore its role in human epilepsy was not established. Here, we studied a family with a pair of monozygotic twins affected by TLE and two unaffected siblings born to healthy parents. Exome sequencing revealed that both twins carried a novel de novo mutation (p.A39E) in the GAL gene. Functional analysis revealed that the p.A39E mutant showed antagonistic activity against galanin receptor 1 (GalR1)-mediated response, and decreased binding affinity and reduced agonist properties for GalR2. These findings suggest that the p.A39E mutant could impair galanin signaling in the hippocampus, leading to increased glutamatergic excitation and ultimately to TLE. In a cohort of 582 cases, we did not observe any pathogenic mutations indicating that mutations in GAL are a rare cause of TLE. The identification of a novel de novo mutation in a biologically-relevant candidate gene, coupled with functional evidence that the mutant protein disrupts galanin signaling, strongly supports GAL as the causal gene for the TLE in this family. Development of a non-contact drive for conveyor belts

Durch diese Art des Antriebs wird die LĂ€nge, die Lebensdauer oder die Förderleistung des Bandförderers begrenzt. Uric Acid Is a Genuine Metabolite of Penicillium cyclopium and Stimulates the Expression of Alkaloid Biosynthesis in This Fungus
On searching for endogenous, low-molecular-weight effectors of benzodiazepine alkaloid biosynthesis in Penicillium cyclopium uric acid was isolated from ethanolic or autoclaved mycelial extracts of this fungus. The isolation was based on a three-step high-pressure liquid chromatography procedure guided by a microplate bioassay, and uric acid was identified by mass spectrometry and the uricase reaction. Conidiospore suspensions that were treated with this compound during the early phase of outgrowth developed emerged cultures with an enhanced rate of alkaloid production. Uric acid treatment did not increase the in vitro measurable activity of the rate-limiting biosynthetic enzyme, cyclopeptine synthetase. However, these cultures displayed a reduced rate of uptake of the alkaloid precursor l-phenylalanine into the vacuoles of the hyphal cells as assayed in situ. It is suggested that the depressed capacity of vacuolar uptake caused by the contact of outgrowing spores with uric acid liberated from hyphal cells results in an enhanced availability of the precursor l-phenylalanine in the cytoplasm and thus accounts at least in part for the increase in alkaloid production
