    EVALUATION OF A RAPID TEST TO DETECT CARBAPENEMASE PRODUCTION AMONG GRAM-NEGATIVE BACILLI.

    Objective: There is a need for an early detection of carbapenemase-producing Gram-negative bacilli to reduce the morbidity and mortality associated with it. The objective of this study is to evaluate the usefulness of the Carba NP test in detecting carbapenemase-producing Gram-negative bacilli.Methods: This prospective experimental study was conducted in 2013 at PSG Institute of Medical Sciences and Research, Coimbatore. In this study, 120 consecutive Gram-negative bacilli clinical isolates identified as carbapenem resistant as per the Clinical and Laboratory Standard Institute guidelines were subjected to Carba NP.Results: About 62.5% of the carbapenem-resistant isolates were found to be carbapenemase producers by modified Hodge test. The sensitivity of the Carba NP test was found to be 92% and the specificity was 100%. The most common bacterial isolate was Acinetobacter baumannii, and the most common sample was urine.Conclusion: The Carba NP test is a simple inexpensive, rapid, reproducible, sensitive test that can be performed directly from the colonies. It improves early detection of patients infected or colonized with carbapenemase in the hospital

    Straminipile genus Phytophthora cause significant disease losses to global cocoa production. Phytopthtora palmivora is one of the major constraints in cocoa production causes significant pod losses. P. palmivora has a complex disease cycle involving several sources of primary inoculum and several modes of dissemination of secondary inoculum. This results in explosive epidemics during favorable environmental conditions. Highest severity of Seedling blight was observed in raised bed nurseries as compared to poly bag nurseries in Shivamogga district (59.26%) followed by Chikkamagluru (53.85%) of the state Karnataka , respectively. Further, the highest incidence of 72.00, 70.83 and 70.00% of black pod rot disease was recorded in Udupi, Dakshina Kannada and Shivamogga district followed by Chikkamagaluru (65.22%), Kodagu (64.00%) and Davanagere (55.56%) district respectively. Due to continuous rain fall or high moisture conditions and the crop was grown as intercrop with arecanut is vulnerable for the attack of pathogen due to the presence of pathogenic variability

    We are sure about inspiring white-collar employees who do technical, managerial, or administrative work and are defined as qualified manual workers in the middle class. Motivation usually plays an important role in the workforce and influences workers efficiency, happiness, absenteeism, relocation, and citizenship actions. Job is carried out in a workplace by white-collar employees, and they are often searching for advancement and professional growth. Blue-collar employees have little management position and are not even linked to increasing professional advancement. They also have no precise occupation to associate their personality with their jobs. They will not be satisfied with their employment. I thought that it was important to establish a plan to improve employees morale from my point of view. It is a complicated and often messy query since it cannot be solved immediately. Many workers have diverse outlooks and characters, and what motivates X does not inspire Y. The dynamic phenomenon here is recognizing and anticipating the driving factors that will satisfy and inspire any employee in the job.     Exploiting metabolic acidosis in solid cancers using a tumor-agnostic pH-activatable nanoprobe for fluorescence-guided surgery

    Cancer cell metabolism leads to a uniquely acidic microenvironment in solid tumors, but exploiting the labile extracellular pH differences between cancer and normal tissues for clinical use has been challenging. Here we describe the clinical translation of ONM-100, a nanoparticle-based fluorescent imaging agent. This is comprised of an ultra-pH sensitive amphiphilic polymer, conjugated with indocyanine green, which rapidly and irreversibly dissociates to fluoresce in the acidic extracellular tumor microenvironment due to the mechanism of nanoscale macromolecular cooperativity. Primary outcomes were safety, pharmacokinetics and imaging feasilibity of ONM-100. Secondary outcomes were to determine a range of safe doses of ONM-100 for intra-operative imaging using commonly used fluorescence camera systems. In this study (Netherlands National Trial Register #7085), we report that ONM-100 was well tolerated, and four solid tumor types could be visualized both in- and ex vivo in thirty subjects. ONM-100 enables detection of tumor-positive resection margins in 9/9 subjects and four additional otherwise missed occult lesions. Consequently, this pH-activatable optical imaging agent may be clinically beneficial in differentiating previously unexploitable narrow physiologic differences

    Critical adsorption on curved objects

    A systematic fieldtheoretic description of critical adsorption on curved objects such as spherical or rodlike colloidal particles immersed in a fluid near criticality is presented. The temperature dependence of the corresponding order parameter profiles and of the excess adsorption are calculated explicitly. Critical adsorption on elongated rods is substantially more pronounced than on spherical particles. It turns out that, within the context of critical phenomena in confined geometries, critical adsorption on a microscopically thin `needle' represents a distinct universality class of its own. Under favorable conditions the results are relevant for the flocculation of colloidal particles.Comment: 52 pages, 10 figure

    Topoisomerase 1 inhibition reversibly impairs synaptic function

    Topoisomerases are enzymes that resolve DNA supercoiling during cell division and gene transcription. Inhibitors of these enzymes are used to treat multiple forms of cancer. Recently we found that topoisomerase inhibitors have profound effects on synaptic genes expressed in the brain. Here we examine the contribution of a clinically used topoisomerase inhibitor on the expression of synaptic proteins and synaptic transmission. We find that inhibition of topoisomerase 1 (TOP1) dampens excitatory and inhibitory synaptic transmission in cortical neurons. Additionally, these effects are fully reversible, because synaptic protein levels and synaptic transmission recover upon washout of the TOP1 inhibitor. These findings provide insights into how inhibition of TOP1 impacts synaptic function in neurons

    Haplotype-based stratification of Huntington's disease

    Huntington’s disease (HD) is an autosomal dominant neurodegenerative disease caused by expansion of a CAG trinucleotide repeat in HTT, resulting in an extended polyglutamine tract in huntingtin. We and others have previously determined that the HD-causing expansion occurs on multiple different haplotype backbones, reflecting more than one ancestral origin of the same type of mutation. In view of the therapeutic potential of mutant allele-specific gene silencing, we have compared and integrated two major systems of HTT haplotype definition, combining data from 74 sequence variants to identify the most frequent disease-associated and control chromosome backbones and revealing that there is potential for additional resolution of HD haplotypes. We have used the large collection of 4078 heterozygous HD subjects analyzed in our recent genome-wide association study of HD age at onset to estimate the frequency of these haplotypes in European subjects, finding that common genetic variation at HTT can distinguish the normal and CAG-expanded chromosomes for more than 95% of European HD individuals. As a resource for the HD research community, we have also determined the haplotypes present in a series of publicly available HD subject-derived fibroblasts, induced pluripotent cells, and embryonic stem cells in order to facilitate efforts to develop inclusive methods of allele-specific HTT silencing applicable to most HD patients. Our data providing genetic guidance for therapeutic gene-based targeting will significantly contribute to the developments of rational treatments and implementation of precision medicine in HD
