Xanthine oxidoreductase regulates macrophage IL1ÎČ secretion upon NLRP3 inflammasome activation.
Activation of the NLRP3 inflammasome by microbial ligands or tissue damage requires intracellular generation of reactive oxygen species (ROS). We present evidence that macrophage secretion of IL1ÎČ upon stimulation with ATP, crystals or LPS is mediated by a rapid increase in the activity of xanthine oxidase (XO), the oxidized form of xanthine dehydrogenase, resulting in the formation of uric acid as well as ROS. We show that XO-derived ROS, but not uric acid, is the trigger for IL1ÎČ release and that XO blockade results in impaired IL1ÎČ and caspase1 secretion. XO is localized to both cytoplasmic and mitochondrial compartments and acts upstream to the PI3K-AKT signalling pathway that results in mitochondrial ROS generation. This pathway represents a mechanism for regulating NLRP3 inflammasome activation that may have therapeutic implications in inflammatory diseases
Conceptualizing the adventure-sports coach
As a comparatively recent development, the adventure-sports coach struggles for a clear and distinct identity. The generic term âinstructorâ no longer characterizes the role and function of this subgroup of outdoor professionals. Indeed, although the fields of adventure/outdoor education and leadership are comparatively well researched, the arrival of this ânew kid on the blockâ appears to challenge both the adventure-sports old guard and traditional views of sports coaching. In an attempt to offer clarity and stimulate debate, this paper attempts to conceptualize the adventure-sports coach in the context of the existing roles in the field and current motivations for activity in the outdoors. We identify issues that are specific to the adventure-sports coach while also recognizing those skills and competencies shared with other professionals, both in the adventure sports profession and traditional sports coaching fields. Based on this review, we offer a conceptual model which may be used to focus debate, stimulate research and, at a possible later stage, to underpin accreditation, training and professional development
Wavelength-scale stationary-wave integrated Fourier-transform spectrometry
Spectrometry is a general physical-analysis approach for investigating
light-matter interactions. However, the complex designs of existing
spectrometers render them resistant to simplification and miniaturization, both
of which are vital for applications in micro- and nanotechnology and which are
now undergoing intensive research. Stationary-wave integrated Fourier-transform
spectrometry (SWIFTS)-an approach based on direct intensity detection of a
standing wave resulting from either reflection (as in the principle of colour
photography by Gabriel Lippmann) or counterpropagative interference
phenomenon-is expected to be able to overcome this drawback. Here, we present a
SWIFTS-based spectrometer relying on an original optical near-field detection
method in which optical nanoprobes are used to sample directly the evanescent
standing wave in the waveguide. Combined with integrated optics, we report a
way of reducing the volume of the spectrometer to a few hundreds of cubic
wavelengths. This is the first attempt, using SWIFTS, to produce a very small
integrated one-dimensional spectrometer suitable for applications where
microspectrometers are essential

Size and emotion or depth and emotion? Evidence, using Matryoshka (Russian) dolls, of children using physical depth as a proxy for emotional charge
Background: The size and emotion effect is the tendency for children to draw people and other objects with a positive emotional charge larger than those with a negative or neutral charge. Here we explored the novel idea that drawing size might be acting as a proxy for depth (proximity).Methods: Forty-two children (aged 3-11 years) chose, from 2 sets of Matryoshka (Russian) dolls, a doll to represent a person with positive, negative or neutral charge, which they placed in front of themselves on a sheet of A3 paper. Results: We found that the children used proximity and doll size, to indicate emotional charge. Conclusions: These findings are consistent with the notion that in drawings, children are using size as a proxy for physical closeness (proximity), as they attempt with varying success to put positive charged items closer to, or negative and neutral charge items further away from, themselves

Associations of Blood Pressure and Cholesterol Levels During Young Adulthood With Later Cardiovascular Events.
BackgroundBlood pressure (BP) and cholesterol are major modifiable risk factors for cardiovascular disease (CVD), but effects of exposures during young adulthood on later life CVD risk have not been well quantified.ObjectiveThe authors sought to evaluate the independent associations between young adult exposures to risk factors and later life CVD risk, accounting for later life exposures.MethodsThe authors pooled data from 6 U.S. cohorts with observations spanning the life course from young adulthood to later life, and imputed risk factor trajectories for low-density lipoprotein (LDL) and high-density lipoprotein cholesterols, systolic and diastolic BP starting from age 18 years for every participant. Time-weighted average exposures to each risk factor during young (age 18 to 39 years) and later adulthood (age â„40 years) were calculated and linked to subsequent risks of coronary heart disease (CHD), heart failure (HF), or stroke.ResultsA total of 36,030 participants were included. During a median follow-up of 17 years, there were 4,570 CHD, 5,119 HF, and 2,862 stroke events. When young and later adult risk factors were considered jointly in the model, young adult LDL â„100 mg/dl (compared with <100 mg/dl) was associated with a 64% increased risk for CHD, independent of later adult exposures. Similarly, young adult SBP â„130 mm Hg (compared with <120 mm Hg) was associated with a 37% increased risk for HF, and young adult DBP â„80 mm Hg (compared with <80 mm Hg) was associated with a 21% increased risk.ConclusionsCumulative young adult exposures to elevated systolic BP, diastolic BP and LDL were associated with increased CVD risks in later life, independent of later adult exposures
Construction and Calibration of Optically Efficient LCD-based Multi-Layer Light Field Displays
Near-term commercial multi-view displays currently employ ray-based 3D or 4D light field techniques. Conventional approaches to ray-based display typically include lens arrays or heuristic barrier patterns combined with integral interlaced views on a display screen such as an LCD panel. Recent work has placed an emphasis on the co-design of optics and image formation algorithms to achieve increased frame rates, brighter images, and wider fields-of-view using optimization-in-the-loop and novel arrangements of commodity LCD panels. In this paper we examine the construction and calibration methods of computational, multi-layer LCD light field displays. We present several experimental configurations that are simple to build and can be tuned to sufficient precision to achieve a research quality light field display. We also present an analysis of moiré interference in these displays, and guidelines for diffuser placement and display alignment to reduce the effects of moiré. We describe a technique using the moiré magnifier to fine-tune the alignment of the LCD layers
