Reactive electrospinning is capable of efficiently producing in situ crosslinked scaffolds resembling the natural extracellular matrix with tunable characteristics. In this study, we aimed to synthesize, characterize, and investigate the in vitro cytocompatibility of electrospun fibers of acrylated poly(1,10-decanediol-co-tricarballylate) copolymer prepared utilizing the photoreactive electrospinning process with ultraviolet radiation for crosslinking, to be used for cardiac tissue engineering applications. Chemical, thermal, and morphological characterization confirmed the successful synthesis of the polymer used for production of the electrospun fibrous scaffolds with more than 70% porosity. Mechanical testing confirmed the elastomeric nature of the fibers required to withstand cardiac contraction and relaxation. The cell viability assay showed no significant cytotoxicity of the fibers on cultured cardiomyoblasts and the cell-scaffolds interaction study showed a significant increase in cell attachment and growth on the electrospun fibers compared to the reference. This data suggests that the newly synthesized fibrous scaffold constitutes a promising candidate for cardiac tissue engineering applications. - 2018 by the authors. We would like to thank Fatima Mraiche for supplying the H9C2 cardiomyoblasts and Nabeel Abdulrahman for his support in the fluorescent imaging. Response to Dr Greenwood’s Comments on “Extending the Double-Hertz Model to Allow Modeling of an Adhesive Elliptical Contact”

© 2018, The Author(s). An adhesive elliptical contact is normally found in microscale applications that involve cylindrical solids, crossing at an angle between 0° and 90°. Currently, only one model is available to describe the elliptical contact’s surface interaction: the approximate Johnson–Kendall–Roberts (JKR) model which is limited to soft materials. In this paper, a new adhesive elliptical model is developed for a wide range of adhesive contacts by extending the double-Hertz theory, where adhesion is modeled by the difference between two Hertzian pressure distributions. Both Hertzian pressures are assumed to have an equivalent shape of contact areas, the only difference being in size. Assuming that the annular adhesive region is obtained by the area difference between the two Hertzian contact areas, the pull-off force curves can be calculated. In the limiting case of an adhesive circular contact, the results are very close to results from the existing models. However, for an adhesive elliptical contact in the JKR domain, lower pull-off forces are predicted when compared to the JKR values. Unlike the developed model, the shape of the JKR contact area varies throughout contact. Results show, particularly for conditions close to the JKR domain, that it is important to take into account that the adhesive region is the result of the two Hertzian contact areas having a non-equivalent shape

The mechano-ubiquitinome of articular cartilage: differential ubiquitination and activation of a group of ER-associated DUBs and ER stress regulators

Understanding how connective tissue cells respond to mechanical stimulation is important to human health and disease processes in musculoskeletal diseases. Injury to articular cartilage is a key risk factor in predisposition to tissue damage and degenerative osteoarthritis. Recently, we have discovered that mechanical injury to connective tissues including murine and porcine articular cartilage causes a significant increase in Lysine 63- polyubiquitination. Here we identified the ubiquitin signature that is unique to injured articular cartilage tissue post mechanical injury (the “mechano-ubiquitinome”). A total of 463 ubiquitinated peptides were identified, with an enrichment of ubiquitinated peptides of proteins involved in protein processing in the endoplasmic reticulum (ER), also known as the ER-associated degradation (ERAD) response, including YOD1, BRCC3, ATXN3 and USP5 as well as the ER stress regulators, RAD23B, VCP/p97 and Ubiquilin 1. Enrichment of these proteins suggested an injury-induced ER stress response and, for instance, ER stress markers DDIT3/CHOP and BIP/GRP78 were upregulated following cartilage injury on the protein and gene expression levels. Similar ER stress induction was also observed in response to tail fin injury in zebrafish larvae, suggesting a generic response to tissue injury. Furthermore, a rapid increase in global DUB activity following injury and significant activity in human osteoarthritic cartilage was observed using DUB specific activity probes. Combined, these results implicate the involvement of ubiquitination events and activation of a set of DUBs and ER stress regulators in cellular responses to cartilage tissue injury and in osteoarthritic cartilage tissues. This link through the ERAD pathway makes this protein set attractive for further investigation in in vivo models of tissue injury and for targeting in osteoarthritis and related musculoskeletal diseases

Development and characterization of epoxy nanocomposites based on nano-structured oil palm ash.

The aim of this study is to utilize the bio-agricultural waste as filler material for composite production which are abundantly available and low cost compared to the silica, alumina etc. The lacks of sufficient scientific information about the utilization of the oil palm ash (OPA) on composites production were the driving force for the choice of this work. Furthermore, the effect of filler loading percentage on physical, mechanical, thermal and morphology properties of the epoxy nanocomposites were studied. It was concluded that the size of the OPA had been successfully reduced from macromolecular to the nano-size range by high energy ball milling and was confirmed by TEM analysis. The density of the nano-structured OPA filled epoxy composites revealed that increasing filler loading will eventually increase the density. The tensile and flexural strength attained maximum value when the filler loading was 3%. Also, increase in the thermal stability was observed in case of 3% filler loading and was attributed to the increase in cross-linking of the epoxy resin in the presence of nano-stuctured OPA and having minimum particle to particle interaction and well dispersed nanoparticles

Investigation of incorporating dielectric pocket (DP) on Vertical Strained-SiGe Impact Ionization MOSFET (VESIMOS-DP)

    Get PDF
The Vertical Strained Silicon Germanium (SiGe) Impact Ionization MOSFET with Dielectric Pocket (VESIMOS-DP) has been successfully developed and analyzed in this paper. Due to the DP layer, improve stability of threshold voltage, VT was found for VESIMOS-DP device of various DP size ranging from 20nm to 80nm. The stability is due to the reducing charge sharing effects between source and drain region. However, the presence of DP layer has introduced another potential barrier in addition to δp+ triangular potential barrier. Thus, increased amount of gate source voltage for lowering both barriers and allows the electron to move from source to drain. Accordingly, slight different and consistency of VESIMOS-DP sub-threshold value as compared to VESIMOS has revealed to give advantages for incorporating DP layer near the drain end. Moreover, the DP layer has suppressed the parasitic bipolar transistor effect with higher breakdown voltage as compared to without DP layer

Asymmetric dimethylation of ribosomal S6 kinase 2 regulates its cellular localisation and pro-survival function

Ribosomal S6 kinases (S6Ks) are critical regulators of cell growth, homeostasis, and survival, with dysregulation of these kinases found to be associated with various malignancies. While S6K1 has been extensively studied, S6K2 has been neglected despite its clear involvement in cancer progression. Protein arginine methylation is a widespread post-translational modification regulating many biological processes in mammalian cells. Here, we report that p54-S6K2 is asymmetrically dimethylated at Arg-475 and Arg-477, two residues conserved amongst mammalian S6K2s and several AT-hook-containing proteins. We demonstrate that this methylation event results from the association of S6K2 with the methyltransferases PRMT1, PRMT3, and PRMT6 in vitro and in vivo and leads to nuclear the localisation of S6K2 that is essential to the pro-survival effects of this kinase to starvation-induced cell death. Taken together, our findings highlight a novel post-translational modification regulating the function of p54-S6K2 that may be particularly relevant to cancer progression where general Arg-methylation is often elevated
