    Проектування віртуальної навчальної лабораторії для майбутніх педагогів професійного навчання з харчових технологій

    Аrticle analyzes the definition of the concept of “virtual laboratory”, considers the features of using virtual laboratories at various stages of laboratory work, gives examples and indicates the advantages of creating them for the training system for teachers of vocational education in food technology. A design model for the functioning of a virtual training laboratory using algorithms for the operation of technological equipment is proposed. The ways of implementing modern approaches in the preparation of future teachers of vocational training in food technology are presented. The algorithm of virtual laboratory work is considered on the example of the discipline "Equipment of restaurant facilities" in the simulation format. The implementation of laboratory work in virtual laboratories contributes to the formation of the intellectual development of students, allows pedagogically balanced use of modern software in the educational process and the assimilation of knowledge at a more conscious and deep level. The positive dynamics of the qualitative indicators of the formation of professional competence of future teachers of vocational training was followed after classes using virtual laboratory work. In the process of developing the virtual laboratory work we presented, psychological requirements were taken into account, the essence of which is a phased solution of problem situations programmed by the teacher, which strengthen the students' semantic and logical activity. A thorough mastery of the knowledge gained during the execution of the virtual laboratory complex provides an appropriate level of development of scientific developments and technological management of the educational process. The structure of virtual laboratory work presented by us makes it possible to provide such components of the learning process as enhancing knowledge to perform research and control knowledge.Keywords: virtual laboratory, vocational training teachers, food technology, laboratory work, technological equipment.У статті аналізується визначення поняття «віртуальна лабораторія», розглянуті особливості використання віртуальних лабораторій на різних етапах проведення лабораторної роботи, наведені приклади та вказуються переваги їх створення для системи підготовки педагогів професійного навчання з харчових технологій. Запропоновано проектування моделі функціонування віртуальної навчальної лабораторії з використанням алгоритмів роботи технологічного обладнання. Представлено шляхи реалізації сучасних підходів у підготовці майбутніх педагогів професійного навчання з харчових технологій. Розглядається алгоритм виконання віртуальної лабораторної роботи на прикладі дисципліни «Устаткування закладів ресторанного господарства» у форматі симуляції. Виконання лабораторних робіт в віртуальних лабораторіях сприяє формуванню інтелектуального розвитку студентів, що дає змогу педагогічно-виважено застосовувати сучасне програмне забезпечення в навчальному процесі та засвоєнню знань на більш свідомому та глибокому рівні. Була простежена позитивна динаміка якісних показників сформованості професійної компетенції майбутніх педагогів професійного навчання після проведення занять за допомогою віртуальних лабораторних робіт. У процесі розробки представленої нами віртуальної лабораторної роботи враховані психологічні вимоги , суть яких полягає в поетапному вирішенню запрограмованих викладачем проблемних ситуацій, які посилюють смислову та логічну діяльність студентів. Ґрунтовне оволодіння знаннями, отриманими в процесі виконання віртуального лабораторного комплексу забезпечує відповідний рівень розвитку наукових розробок і технологічного керування навчальним процесом. Представлена нами структура віртуальної лабораторної роботи дає можливість забезпечити такі складові процесу навчання як активізація знань до виконання дослідження і контроль знань. Ключові слова: віртуальна лабораторія, педагоги професійного навчання, харчові технології, лабораторна робота, технологічне обладнання

    Investigation of the marine compound spongistatin 1 links the inhibition of PKCα translocation to nonmitotic effects of tubulin antagonism in angiogenesis

    The aims of the study were to meet the demand of new tubulin antagonists with fewer side effects by characterizing the antiangiogenic properties of the experimental compound spongistatin 1, and to elucidate nonmitotic mechanisms by which tubulin antagonists inhibit angiogenesis. Although tubulin-inhibiting drugs and their antiangiogenic properties have been investigated for a long time, surprisingly little is known about their underlying mechanisms of action. Antiangiogenic effects of spongistatin 1 were investigated in endothelial cells in vitro, including functional cell-based assays, live-cell imaging, and a kinome array, and in the mouse cornea pocket assay in vivo. Spongistatin 1 inhibited angiogenesis at nanomolar concentrations (IC50: cytotoxicity>50 nM, proliferation 100 pM, migration 1.0 nM, tube formation 1.0 nM, chemotaxis 1.0 nM, aortic ring sprouting 500 pM, neovascularization in vivo 10 μg/kg). Further, a kinome array and validating data showed that spongistatin 1 inhibits the phosphorylation activity of protein kinase Cα (PKCα), an essential kinase in angiogenesis, and its translocation to the membrane. Thus, we conclude that PKCα might be an important target for the antiangiogenic effects of tubulin antagonism. In addition, the data from the kinase array suggest that different tubulin antagonists might have individual intracellular actions.—Rothmeier, A. Siendo declarada patrimonio nacional de la humanidad y al ser el lugar de trabajo de una comunidad de personas dedicadas a la cantería, se convierte en un PATRIMONIO PRODUCTIVO cuya preservación es necesaria, asimismo la necesidad de darle un HABITAT DIGNO a la población adyacente que ha hecho de esta zona su lugar de trabajo y hogar. Para este objetivo se utilizó como herramienta el PROCESO PARTICIPATIVO, logrando juntar en un solo equipo de trabajo a profesionales, pobladores, autoridades y agentes externos; que permitieron conocer el territorio a los usuarios a fondo, manejar una hoja de ruta a través de talleres de participación que nos permitiera conoces los conflictos y potencialidades de la zona. A través de este proceso se pudo obtener unas PREMISAS DE DISEÑO, que nos permitieron realizar un REORDENAMIENTO TERRITORIAL adecuado para la zona y cumpliendo los objetivos planteados. Asimismo, nos adentramos en el ámbito constructivo de la localidad para darles un nuevo sistema de construcción mediante un PROTOTIPO de sillar auto portante para el desarrollo de sus viviendas de INTERES SOCIAL y EVOLUTIVAS. Finalmente, pudimos satisfacer la necesidad de la zona y de la población y además dándoles un aporte innovador que podrán utilizar tanto para la construcción de su sector como para impulsar el desarrollo de su negocio.     Influence of VEGF deprivation upon vascular formation by endothelium in the presence of macrophages

    Development of angiogenesis depends on the functional state of endothelial cells, as well as on the balanced secretion of cytokines, growth factors and chemokines by endothelial cells and cells of microenvironment. Macrophages represent an essential component of the microenvironment and take part in the formation of blood vessels both due to the production of cytokines and due to contact interactions with endothelial cells. VEGF is among the most important cytokines that control angiogenesis at all its stages. Currently, the role of VEGF in the intercellular interactions of endothelial cells and macrophages is not well described. The aim of our study was to investigate the effect of VEGF deprivation using monoclonal antibodies on angiogenesis under conditions of co-cultivation of endothelium and macrophages. Materials and methods: monoclonal antibodies to VEGF-A were used for VEGF deprivation in monoculture of endothelial cells and in co-culture of endothelial cells with macrophages. The IL-1β, IL-6 and TNFα cytokines were used as inducers. When VEGF-A was removed from the medium, endothelial cells show plasticity and form longer vessels, they modify the expression of VEGF receptors. Macrophages regulate endothelial cell activity through the secretion of cytokines, including VEGF, and through contact interactions with endothelial cells. THP-1 cells increase the sensitivity of endothelial cells to VEGF by stimulating the VEGFR1 and VEGFR3 expression, this effect is VEGF-A-independent. The IL-1β, IL-6, TNFa cytokines independently stimulate non-branching angiogenesis, increasing the length of the vessels. At the same time, IL-ip increases the VEGFR1 expression on the surface of endothelial cells. In contrast, IL-6 and TNFα decrease it, thereby regulating the sensitivity of endothelial cells to VEGF. The effects of these cytokines are not dependent on VEGF-A. The IL-1β, IL-6, TNFα cytokines promote acquisition of anti-angiogenic properties by THP-1 cells that is independent on VEGF-A, as well as on expression of its receptors by endothelial cells. Thus, VEGF is an important, but not the sole factor controlling angiogenesis. Under conditions of VEGF-A deficiency, either endothelial cells or microenvironment cells are able to compensate for its functional load due to the production of other growth factors

    A homebuilt ESE spectrometer on the basis of a high-power Q-band microwave bridge

    We present a Q-band spectrometer which was built recently at the Institute of Physical Chemistry of the University of Stuttgart. It allows us to perform the field-sweep electron spin echo (ESE), pulsed electron-nuclear double resonance (ENDOR), relaxation and electron spin echo envelope modulation experiments both at room and low (down to 1.5 K) temperatures. The spectrometer consists of an electromagnet, digital field controller, pulsed microwave bridge, probehead, cryostat, radio frequency unit, pulse programmer and data acquisition electronics. The Q-band microwave bridge with 10.8 W output power is based on a two-stage IMPATT-diode pulse amplifier. The commercial Varian electromagnet system is controlled by a 24-bit home-built digital controller. The external devices are interfaced to the two PCs via GPIB and LAN. The spectrometer control software was developed in Visual C++. It consists of two programs running synchronously on the control PCs. The spectrometer is equipped with a cylindrical TE 011 cavity constructed both for ESE and for pulsed ENDOR. The cavity fits into a liquid He cryostat thus allowing low-temperature experiments. An 8-bit data acquisition digitizer is used to collect the echo signals, and the PBESR-PRO-400 digital word generator orchestrates the pulse experiments and sets pulse sequences of the microwave bridge. The spectrometer performance is demonstrated on nitrogen impurities in a polycrystalline synthetic diamond, on silver clusters supported on NaA zeolite and electron-irradiated tooth enamel. © 2008 Springer-Verlag

    Microbiome Responses to an Uncontrolled Short-Term Diet Intervention in the Frame of the Citizen Science Project

    Personalized nutrition is of increasing interest to individuals actively monitoring their health. The relations between the duration of diet intervention and the effects on gut microbiota have yet to be elucidated. Here we examined the associations of short-term dietary changes, long-term dietary habits and lifestyle with gut microbiota. Stool samples from 248 citizen-science volunteers were collected before and after a self-reported 2-week personalized diet intervention, then analyzed using 16S rRNA sequencing. Considerable correlations between long-term dietary habits and gut community structure were detected. A higher intake of vegetables and fruits was associated with increased levels of butyrate-producing Clostridiales and higher community richness. A paired comparison of the metagenomes before and after the 2-week intervention showed that even a brief, uncontrolled intervention produced profound changes in community structure: resulting in decreased levels of Bacteroidaceae, Porphyromonadaceae and Rikenellaceae families and decreased alpha-diversity coupled with an increase of Methanobrevibacter, Bifidobacterium, Clostridium and butyrate-producing Lachnospiraceae- as well as the prevalence of a permatype (a bootstrapping-based variation of enterotype) associated with a higher diversity of diet. The response of microbiota to the intervention was dependent on the initial microbiota state. These findings pave the way for the development of an individualized diet.</p
