    Senegalese sole (Solea senegalensis) ISG15: molecular characterization and in vivo interplay with viral infections

    The interferon-stimulated gene 15 (Isg15) is strongly induced by type I interferon (IFN I), viral infection, and double-stranded RNA (poly I:C) in several fish species, suggesting that Isg15 protein could play a key role in fish innate immunity against viral diseases. Thus, the aim of the present study was to characterize the molecular structure and transcription pattern of the Senegalese sole (Solea senegalensis) Isg15 gene in response to viral infections. The molecular characterization shows that the Senegalese sole Isg15 gene codes for a typical Isg15 protein of 165 aa, containing two ubiquitin-like domains and one conserved LRLRGG conjugating motif at the C-terminal end. The untranslated 5´-end region exhibited the structure of an IFN-stimulated gene promoter, with two interferon stimulated response elements (ISRE). Pairwise alignments based on deduced amino acid sequences showed homologous relationships (72.5-74.2%) between the Isg15 of Senegalese sole and other pleuronectiforms. The Isg15 transcription has been studied in head kidneys of Senegalese sole inoculated with poly I:C and with different fish viruses: two Viral Haemorrhagic Septicaemia Virus (VHSV) isolates (highly pathogenic and non-pathogenic to sole), and one reassortant Viral Nervous Necrosis Virus (VNNV) isolate, composed of a RGNNV-type RNA1 and a SJNNV-type RNA2 (pathogenic to sole). These challenges showed that poly I:C induces Isg15 transcription from 3 to 72 h post-injection (p.i.), whereas the induction in response to viral infections started at 24-48 h p.i. The fast induction of Isg15 indicates the potential implication of this ISG in the antiviral state stablished by the IFN I system. On the other hand, the interaction between each virus and the IFN I system was evaluated in fish inoculated with poly I:C and subsequently (24 h later) challenged with the different viruses. This challenge showed a viral multiplication decrease in poly I:C treated animals compared with untreated fish. Besides, results showed that only both pathogenic isolates interfered negatively with the Isg15 stimulation triggered by poly I:C. These results suggest that the Isg15 might play an important role in host defense against RNA virus infection, and the pathogenic isolates used in this study may have mechanisms to evade or limit the Senegalese sole innate host defenses.Universidad de Málaga.     El lenguado senegalés es susceptible a la infección por el Virus de la Necrosis Nerviosa Viral (VNNV). Los betanodavirus se clasifican en cuatro genotipos, siendo los genotiposSJNNV, RGNNV y virus recombinantes RGNNV-SJNNV los que causan mortalidad en lenguado. En condiciones experimentales es el recombinante el que provoca mayor mortalidad. Las diferencias en la tasa de mortalidad pueden indicar variaciones en la interacción entre los distintos genotipos y el sistema inmune de lenguado. El sistema del interferón tipo I es un componente esencial de la respuesta inmune frente a infecciones virales, induciendo la expresión de genes que codifican proteínas antivíricas, tales como la Mx, la ISG15 y la PKR.     Sequential determination of heavy metals in environmental water samples by flow injection-chemical vapour generation-inductively coupled plasma mass spectrometry

    The toxicity of “heavy metals” has been well recognized for a long time. Often the non-specific term “heavy metals” is used for three of the metals, cadmium, mercury and lead. These have large bioconcentration factors in marine organism, are highly toxic and, unlike many of the transition elements have no known natural biological functions. For these reasons these metals generate the greatest concern for the general public and therefore also for environmental agencies in the majority of states. The monitoring and control of these trace elements in the environment requires powerful analytical methods to accurately characterize their abundance and to reach reliable conclusions. In this work, an inductively coupled plasma mass spectrometry (ICP-MS) method has been developed for the sequential determination of Pb, Cd and Hg in natural waters, including sea-water, using an on-line preconcentration flow injection chemical vapour generation system (FI-CVG). It is difficult to simultaneously determine these elements by CVG, because their conditions of CVG are different. Thus, the system was based on the use of two minicolumns packed with 1-(di-2-pyridyl)methylene thiocarbonohydrazide chelating resin which were placed in two injection valves of a simple flow manifold to be loaded simultaneously. A third valve was arranged to select the reagent for the selective vapour generation of the analytes and, thus make possible the sequential determination of the three metals. By using this device, diverse advantages are attained: increase of the sensitivity and reduction of the interferences by the preconcentration and the vapour generation. The detection limits achieved (3 min sample loading time) were: 9, 17 and 12 ngL-1 for Pb, Cd and Hg, respectively, with a sample throughput about 10.4 h-1. The accuracy of the proposed method was checked with three certified reference materials (CRMs): TMDA-54.4 fortified lake water, LGC6016 estuarine water and CASS-5 oceanic water and the results obtained were in good agreement with the certified values.     La narrativa como herramientas para el abordaje psicosocial en escenarios de violencia.

    El presente documento consta de la fase final del Diplomado de Profundización en Acompañamiento Psicosocial en Escenarios de Violencia, estudiando las diferentes unidades propuestas por el mismo, con el objetivo de profundizar conocimientos teóricos y pertinentes propios de la psicología. Se presenta un análisis del relato de José Ignacio Medina, tomado del libro Voces: historias de violencia y esperanza en Colombia; el cual muestra como la violencia influyo en su vida, la de su familia y una comunidad en general. Este caso es uno más de los tantos en Colombia que pretende mostrar como la violencia, los grupos al margen de la ley y las desigualdades políticas dejan traumas, desplazamientos, abandonos, enfermedades físicas y psicológicas en la población vulnerable, pero que además como estas comunidades y protagonistas de las historias tienen la capacidad de superación a pesar de la adversidad. Es en estos escenarios donde desde la psicología implementa estrategias psicosociales entre se fomenta la participación comunitaria para el bienestar de los mismos. Seguidamente se realiza una propuesta de abordaje psicosocial en el caso de la comunidad de Cacarica, la cual ha sido afectada por la violencia de grupos armados generando un impacto en la salud mental de la comunidad, como también desplazamientos y afectación económica. Por último se plasma un informe analítico y reflexivo desde la experiencia de la foto voz en diferentes Departamentos de ColombiaAbstrac This document consists of the final phase corresponding to the Diploma of Deepening in Psychosocial Accompaniment in Violence Scenarios, studying the different units proposed by it, with the aim of deepening theoretical and relevant knowledge of psychology. An analysis of José Ignacio Medina's story, taken from the book Voces: histories of violence and hope in Colombia, is presented; which shows how violence influenced his life, his family and a community in general. This case is one of many in Colombia that aims to show how violence, groups outside the law and political inequalities leave traumas, displacements, abandonments, physical and psychological diseases in the vulnerable population, but also how these communities and protagonists of the stories have the ability to excel despite adversity. It is in these scenarios where psychology implements psychosocial strategies among community participation is encouraged for the welfare of them. Then a proposal of psychosocial approach is made in the case of the community of Cacarica, which has been affected by the violence of armed groups, generating an impact on the mental health of the community, as well as displacement and economic affectation.     Dendrometric analysis of Tamarix africana L.,species of river and wetlands of the Mediterranean area. Characterisation of biomass

    A dendrometric and energetic characterisation of Tamarix africana L. was carried out in this study. This is an indigenous plant species in the Mediterranean area, and it can survive in semi-arid areas with saline soils and occasional floods. Owing to these characteristics, this species is a good candidate for its use in the restoration of several environments, such as the marshes. This opens up the possibility of improving the management of biomass, which can be used for obtaining economic benefits from its energetic use. For the stems, biomass volume functions were determined from their diameter and length with r(2) of 0.96. For the whole plant, biomass volume functions were determined from the diameter of the projected area in the soil and the height of the plant with r(2) of 0.95. For energetic characterisation, the density, higher heat value, and the composition of this wood were measured in terms of following elements: carbon, nitrogen, hydrogen, sulphur and chlorine, volatiles and ash. The determination of its composition in C allows us to evaluate the CO2 fixed by the plant during its growth. The average catchment was estimated to be 2.58 kg/plant

    Ontogeny of Sex-Related Differences in Foetal Developmental Features, Lipid Availability and Fatty Acid Composition

    Sex-related differences in lipid availability and fatty acid composition during swine foetal development were investigated. Plasma cholesterol and triglyceride concentrations in the mother were strongly related to the adequacy or inadequacy of foetal development and concomitant activation of protective growth in some organs (brain, heart, liver and spleen). Cholesterol and triglyceride availability was similar in male and female offspring, but female foetuses showed evidence of higher placental transfer of essential fatty acids and synthesis of non-essential fatty acids in muscle and liver. These sex-related differences affected primarily the neutral lipid fraction (triglycerides), which may lead to sex-related postnatal differences in energy partitioning. These results illustrate the strong influence of the maternal lipid profile on foetal development and homeorhesis, and they confirm and extend previous reports that female offspring show better adaptive responses to maternal malnutrition than male offspring. These findings may help guide dietary interventions to ensure adequate fatty acid availability for postnatal development

    Spanish cross-cultural adaptation and validation of the Oslo Sports Trauma Research Centre (OSTRC) Overuse Injury Questionnaire in Handball Players.

    Objectives: The aim of this study was the cultural adaptation, Spanish translation and validation of the Oslo Sports Trauma Research Centre (OSTRC) Overuse Injury Questionnaire in an adult population. Design: In this study, a cross-sectional design was used. Methods: This study was divided into two phases: (1) cross-cultural adaptation of the original version of the OSTRC to a Spanish version (OSTRC-Sp) and (2) analysis of the psychometric properties of the OSTRC-Sp. A total of 427 handball players of both sexes and over 18 years of age participated in the study. Results: The translated version of the questionnaire showed a very high internal consistency (Cronbach’s α = 0.954), while the subscales showed an internal consistency between 0.832 and 0.961, with the endmost values being for shoulder and low back pain, respectively. On the other hand, when analysing item responses, the OSTRC-Sp showed ICC values ranging from 0.844 to 0.956, the former being for the first back question (back_1), and the latter for the fourth shoulder question (shoulder_4), in line with most published versions. Conclusion: The Spanish version of the OSTRC is a reliable and valid tool that can be used by researchers and clinicians in a Spanish-speaking population with musculoskeletal disorders

    Sequential oral antibiotic in uncomplicated Staphylococcus aureus bacteraemia: a propensity-matched cohort analysis

    Objectives: We aimed to analyse the efficacy and safety of oral sequential therapy (OST) in uncomplicated Staphylococcus aureus bacteraemia (SAB). Methods: Single-centre observational cohort at a tertiary hospital in Spain, including all patients with the first SAB episode from January 2015 to December 2020. We excluded patients with complicated SAB and those who died during the first week. Patients were classified into the OST group (patients who received oral therapy after initial intravenous antibiotic therapy [IVT]), and IVT group (patients who received exclusively IVT). We performed a propensity-score matching to balance baseline differences. The primary composite endpoint was 90-day mortality or microbiological failure. Secondary endpoints included 90-day SAB relapse. Results: Out of 407 SAB first episodes, 230 (56.5%) were included. Of these, 112 (n = 48.7%) received OST and 118 (51.3%) IVT exclusively. Transition to oral therapy was performed after 7 days (interquartile range, 4–11). The primary endpoint occurred in 10.7% (11/112) in OST vs. 30.5% (36/118) in IVT (p < 0.001). SAB relapses occurred in 3.6% (4/112) vs. 1.7% (2/118) (p 0.436). None of the deaths in OST were related to SAB or its complications. After propensity-score matching, the primary endpoint was not more frequent in the OST group (relative risk, 0.42; 95% CI, 0.22–0.79). Ninety-day relapses occurred similarly in both groups (relative risk, 1.35; 95% CI, 0.75–2.39). Discussion: After an initial intravenous antibiotic, patients with uncomplicated SAB can safely be switched to oral antibiotics without apparent adverse outcomes. This strategy could save costs and complications of prolonged hospital stays. Prospective randomized studies are needed

    Synthetic antigenic determinants of clavulanic acid induce dendritic cell maturation and specific T cell proliferation in patients with immediate hypersensitivity reactions

    Background Immediate drug hypersensitivity reactions (IDHRs) to clavulanic acid (CLV) have increased in the last decades due to a higher consumption alongside amoxicillin (AX). Due to its chemical instability, diagnostic procedures to evaluate IDHRs to CLV are difficult, and current in vitro assays do not have an optimal sensitivity. The inclusion of the specific metabolites after CLV degradation, which are efficiently recognised by the immune system, could help to improve sensitivity of in vitro tests. Methods Recognition by dendritic cells (DCs) of CLV and the synthetic analogues of two of its hypothesised antigenic determinants (ADs) was evaluated by flow cytometry in 27 allergic patients (AP) and healthy controls (HC). Their ability to trigger the proliferation of T cells was also analysed by flow cytometry. Results The inclusion of synthetic analogues of CLV ADs, significantly increased the expression of maturation markers on DCs from AP compared to HC. A different recognition pattern could be observed with each AD, and, therefore, the inclusion of both ADs achieves an improved sensitivity. The addition of synthetic ADs analogues increased the proliferative response of CD4+Th2 compared to the addition of native CLV. The combination of results from both ADs increased the sensitivity of proliferative assays from 19% to 65% with a specificity higher than 90%. Conclusions Synthetic ADs from CLV are efficiently recognised by DCs with ability to activate CD4+Th2 cells from AP.
