The Ins and Outs of TAPBPR.
Peptide presentation on MHC class I molecules (MHC-I) is central to mounting effective antiviral and antitumoral immune responses. The tapasin-related protein TAPBPR is an MHC-I peptide editor which shapes the final peptide repertoire displayed on the cell surface. Here, we review recent findings which further elucidate the mechanisms by which TAPBPR performs peptide editing on a molecular level, and how glycosylation on MHC-I influences the interaction with TAPBPR and the peptide loading complex. We also explore how the function of TAPBPR can be utilized to promote exogenous peptide loading directly onto plasma-membrane expressed MHC-I. This has led to the development of new assays to investigate TAPBPR-mediated peptide editing and uncovered translational opportunities of utilizing TAPBPR to treat human disease.Wellcom

Este trabalho de investigação científica, coordenado pela Faculdade Mauá, busca estratégias coletivas que viabilizem a proposta de uma Escola de Básica cidadã com responsabilidade social. Para desenvolver a pesquisa tornou-se necessário montar uma equipe interdisciplinar de pesquisadores e escolher um tema e problemas de pesquisa para cujas soluções os docentes e discentes dos cursos envolvidos - Administração, Serviço Social e Pedagogia - pudessem trazer contribuições significativas
Castro Alves e Cecília Meireles: a África como espaço do exílio e da liberdade
Neste texto apresentamos uma reflexão sobre as imagens da África em poemas de Castro Alves e Cecília Meireles, demonstrando como esses poetas exploraram o tema do exílio e da liberdade em momentos distintos da literatura brasileira. Nos poemas “Vozes d´África”, “Navio Negreiro” e “Cachoeira de Paulo Afonso”, Castro Alves constrói várias imagens da África, país da liberdade, em contraponto com as Américas, espaço da escravidão e do desterro. No Romanceiro da Inconfidência, Cecília Meireles, ao recriar o espaço das Minas Gerais do século XVIII, traz à cena dos romances os povos africanos, seres marginalizados e excluídos pela sociedade, mas que ajudaram a construir a nossa história e também retrata o exílio e o desterro dos inconfidentes em Moçambique e Angola

Molecular basis of TAPBPR-mediated peptide editing on MHC class I molecules
Major histocompatibility complex (MHC) class I molecules present fragments of the cellular proteome, in the form of short peptides, to the cell surface for the inspection by cytotoxic T cells. This process is a crucial immunosurveillance mechanism used to induce appropriate immune responses against intracellular pathogens and cancer. In order to generate optimal T cell-mediated immune responses, prior to their export to the cell surface, MHC class I molecules undergo a process known as peptide selection. Optimal peptide selection is facilitated by two intracellular peptide editors, tapasin and TAPBPR. TAPBPR was shown to shape the peptide repertoire presented on MHC class I at the cell surface, either by directly catalysing peptide exchange on MHC class I molecules or by associating with the quality control enzyme UDP-glycoprotein glucosyltransferase 1 (UGT1), which selects optimally-loaded MHC class I molecules for export to the cell surface.
Given that unlike tapasin, TAPBPR could catalyse peptide editing on MHC class I on its own in solution, I sought to test whether TAPBPR could also function as a peptide exchange catalyst on MHC class I molecules present on the surface of cells. By examining the artefactual expression of TAPBPR at the cell surface upon over-expression, I developed two novel cellular assays which allowed me to explore the function of TAPBPR as a peptide exchange catalyst on plasma membrane-expressed MHC class I molecules. I showed that, when given access the cell surface, TAPBPR can promote efficient peptide exchange on surface expressed MHC class I molecules. These assays allowed me to demonstrate that the 22-35 loop of TAPBPR was essential for its peptide exchange function. Moreover, I revealed that residue L30 within the loop was both necessary and sufficient for the efficient ability of TAPBPR to dissociate peptides from MHC class I molecules that typically accommodate hydrophobic anchor residues in their F pocket. This enabled me to propose a new mechanistic model for TAPBPR-mediated peptide editing. I further addressed the molecular basis governing the compatibility between TAPBPR and MHC class I molecules, by screening a wide panel of human leukocyte antigen (HLA) class I allotypes for their relative propensities to undergo peptide editing by TAPBPR. TAPBPR displayed a clear functional preference for HLA-A molecules, particularly for members of the A2 and A24 supertypes, over HLA-B and -C molecules. This preference appears to be driven by specific molecular features of the MHC class I F pocket, in particular residues H114 and Y116.
Finally, I explored the potential translational applications of using TAPBPR as a peptide exchange catalyst on surface-expressed MHC class I molecules. I demonstrated that recombinant TAPBPR can be utilised to load immunogenic peptides of choice directly onto plasma-membrane expression MHC class I, thus overriding the internal antigen presentation pathway. Subsequently, I revealed that, TAPBPR can be used to induce T cell-mediated killing of tumour cells. These findings highlight a potential therapeutic application of TAPBPR in increasing the immune recognition of tumours.My PhD studentship was integrally funded by Wellcome
Identity affirmation in the text “Black dolls from the quilombo Conceição das Crioulas”
Neste artigo, sob a perspectiva da semiótica greimasiana, refletimos sobre a seguinte questão: O o que é e como se produz o efeito de sentido de afirmação da identidade? A partir de discussão teórica e de sua incidência na análise do texto bonecas pretas do Quilombo de Conceição das Crioulas, observamos que a identidade semiótica é um simulacro e para o efeito de sua afirmação são necessários elementos sintáxicos que produzam a repetição que, no discurso das bonecas, realiza-se, principalmente, com o recurso à isotopia, à aspectualização e à tensividade.In this article, from the perspective of greimasian semiotics, we reflect about the question: what is it and how is produced the effect of sense of the identity affirmation? From theoretical discussion and its impact on the analysis of the text black dolls from the Quilombo Conceição das Crioulas, we observe that the semiotic identity is a simulacrum and to the effect of its affirmation are necessary syntactic elements that can produce repeating that in the discourse of the dolls performs mainly with the use of isotope, aspectualization and tensivity
Programa Nacional de Assistência Estudantil - PNAES: disputa entre assistencialização e permanência
Apresenta uma análise sobre a política de Assistência Estudantil no Brasil por meio de análise histórica e política do modo de produção capitalista e o desenvolvimento de suas estratégias, que explicam as relações com o mundo do trabalho em tempo de crise neoliberal. O recorte priorizado foram as “reformas” universitárias e a demanda colocada para a Assistência Estudantil nos anos 2000 através da disputa entre assistencialização, demanda do Estado capitalista; e permanência, demanda dos estudantes. Para tanto, traremos na introdução um resumo sobre o modo de produção capitalista, as respostas dadas pelo capitalismo às suas crises e discutiremos a perspectiva do Estado no capitalismo neoliberal, sua contrarreforma; a contrarreforma na seguridade social brasileira de 1990 e 2000; bem como a contrarreforma universitária. Por fim, analisaremos os avanços e limites do Programa Nacional de Assistência Estudantil decretado em Julho de 2010
Deslocamento para o trabalho de industriários: avaliação de uma intervenção educativa para promoção do uso de bicicleta
Tese (doutorado) - Universidade Federal de Santa Catarina, Centro de Desportos, Programa de Pós-Graduação em Educação Física, Florianópolis, 2014.Os benefícios para a saúde devido à utilização regular da bicicleta no deslocamento ao trabalho e lazer, são evidentes. O principal benefício individual é que enquanto se desloca de bicicleta pela cidade o cidadão exerce uma atividade física que faz bem a sua saúde, proporcionado bem estar físico e psicológico. Nesse sentido, este estudo teve como objetivo avaliar a efetividade de uma intervenção educativa para promoção do uso da bicicleta no deslocamento para o trabalho. As análises foram realizadas utilizando-se o banco de dados do projeto  Deslocamento ativo dos trabalhadores, com uso da bicicleta, na indústria , o estudo foi realizado pelo SESI/SC e, financiado pelo SESI/SC/DN e CNPq, por meio do Edital SENAI/SESI de Inovação 2010.A amostra foi constituída de 932 trabalhadores, de uma empresa metal mecânica, situada no município de Joinville em Santa Catarina, Brasil. Foram sorteados aleatoriamente 464 indivíduos para compor o grupo intervenção (GI) e 468 para compor o grupo controle (GC).Os grupos intervenção e controle foram constituídos de trabalhadores que usavam e não usavam a bicicleta no deslocamento ao trabalho. Estes trabalhadores foram alocados aleatoriamente nos grupos intervenção e controle. O grupo de intervenção participou de 23 encontros durante 6 meses. A intervenção consistiu em estratégias baseadas no Modelo Transteórico de Mudança de Comportamento (MTMC). A intervenção constou de atividades educativas experienciais e lúdicas (palestras, vídeos, jogos). Os instrumentos de pesquisa foram compostos de questionário e protocolos validados. Foram coletados dados de variáveis sociodemográficas, comportamentais e antropométricas. As informações coletadas foram digitalizadas a partir da leitura ótica dos questionários (Software SPHYNX). As análises estatísticas foram realizadas no programa estatístico Stata 11.0 (Stata Corporation, College Station, Texas, USA). Foi utilizada a estatística descritiva, regressão logística, Qui-quadrado e McNemar com nível de confiança de 95%. Do total de 932 trabalhadores do início do estudo a perda amostral foi de 6,2%, totalizando 876 sujeitos ao final do estudo. A prevalência de uso da bicicleta no deslocamento ao trabalho foi de 44,1%. O uso da bicicleta foi associado com baixa renda e nível de escolaridade, assim como, com a distância da residência até o trabalho menor ou igual a 5 km e posse de bicicleta. Em relação à efetividade da intervenção educativa observou-se que não foram encontradas diferenças significativas entre os trabalhadores que utilizavam a bicicleta antes (p = 0,49) e depois da intervenção (p = 0,10) entre os grupos controle e intervenção. Foi observada diferença significativa pré e pós-intervenção (45,3% vs 47,5% p = 0,04) para o grupo intervenção com aumento de (2,2%) no uso da bicicleta. Conclusão: Nossos resultados evidenciaram que a intervenção para promoção do uso da bicicleta baseada no modelo transteórico foi considerada uma boa estratégia metodológica para aumentar o uso da bicicleta no deslocamento ao trabalho. Sugerimos levar em consideração estes achados em outros estudos para melhor compreender o papel da intervenção educativa sobre o deslocamento ativo de bicicleta.Abstract : The health benefits brought by the regular use of the bicycle while commuting to work or leisure are evident. The main individual benefit lies on the fact that while one commutes by bicycle, s/he engages in a healthy physical activity, which promotes physical and psychological well being. In this sense, this study had as its objective to evaluate the effectiveness of an educational intervention to promote the use of the bicycle while commuting to work. The analysis was conducted through the use of the database of a project entitled  Active commuting by bicycle of industry workers . The study was carried out by SESI/SC and it was financed by SESI/SC/DN and CNPq, through the official public notice SENAI/SESI of Innovation from 2010. The sample encompassed 932 workers of a metal mechanics enterprise located in the city of Joinville in Santa Catarina, Brazil. Through a raffle, 464 individuals were chosen to be part of the intervention group (GI) and 468 formed the control group (GC). Both groups were composed of workers that used and that did not make use of the bicycle to commute to work. These workers had their groups assigned randomly. The intervention group participated of 23 meetings during 6 months. The intervention consisted of strategies based on the Transtheoretical Model of Change and Behavior (TMCB). The intervention consisted of educational activities such as lectures, videos, and games. The research instruments were a questionnaire and validated protocols. Data of socio demographic, behavioral and anthropometric variables were collected. All the gathered information was scanned through the optical reading of the questionnaires (Software SPHYNX). The statistical analysis was conducted in the statistics software Stata 11.0 (Stata Corporation College Station, Texas, USA). The descriptive statistics was used, logistical regression, Chi-squared distribution and McNemar with trust level of 95%. From the total of 932 workers from the beginning of the study, 6,2% gave up, making up a total of 876 participants at the end of the study. The prevalence of the use of the bicycle while commuting to work was of 44,1%. The use of the bicycle was associated with low income and educational level as well as to a distance from home to work equal to or less than 5 km and to the possession of a bicycle. Regarding the effectiveness of the educational intervention, no significant differences could be observed between the workers who made use of the bicycle before (p = 0,49) and after the intervention (p = 0,10) in the control and intervention groups. A significant difference was observed while considering the pre and post-intervention stages (45,3% vs 47,5% p = 0,04) for the intervention group with an increase of 2,2% in the use of the bicycle. Our results showed that the intervention to promote bicycle commuting based on the transtheoretical model was considered an effective methodological strategy to increase the use of bicycle commuting to work. Therefore, it is possible to suggest that these findings should be taken into account in other studies so as to better understand the role of the educational intervention regarding the active commuting by bicycle
