    On the Definition of Effective Permittivity and Permeability For Thin Composite Layers

    The problem of definition of effective material parameters (permittivity and permeability) for composite layers containing only one-two parallel arrays of complex-shaped inclusions is discussed. Such structures are of high importance for the design of novel metamaterials, where the realizable layers quite often have only one or two layers of particles across the sample thickness. Effective parameters which describe the averaged induced polarizations are introduced. As an explicit example, we develop an analytical model suitable for calculation of the effective material parameters ϵeff\epsilon_{\rm{eff}} and μeff\mu_{\rm{eff}} for double arrays of electrically small electrically polarizable scatterers. Electric and magnetic dipole moments induced in the structure and the corresponding reflection and transmission coefficients are calculated using the local field approach for the normal plane-wave incidence, and effective parameters are introduced through the averaged fields and polarizations. In the absence of losses both material parameters are purely real and satisfy the Kramers-Kronig relations and the second law of thermodynamics. We compare the analytical results to the simulated and experimental results available in the literature. The physical meaning of the introduced parameters is discussed in detail.Comment: 6 pages, 5 figure

    Europium-based high-temperature superconductors studied by x-ray diffraction and 151Eu Mössbauer spectroscopy

    Isotropic powders and magnetically aligned crystallites of EuBa2Cu3O7−δ (1:2:3) and europium-doped Bi2Sr2CaCu2O8 (2:2:1:2) were studied by means of x-ray diffraction and Eu151 Mössbauer spectroscopy. The degree of crystallite orientation of the samples and the values of the lattice constants were determined by x-ray diffraction. The Mössbauer spectra were analyzed considering the full hyperfine Hamiltonian of the nuclear states of the 21.5-keV γ transition. The Mössbauer hyperfine parameters obtained from the superconducting and semiconducting phases are presented. A small change is seen in the Eu151 isomer shift when the oxygen deficiency δ of the 1:2:3 compound is varied. The shift can be explained by a decrease in the s-electron density due to lattice expansion. The changes in the oxidation state of the copper atoms with varying δ were determined from the Mössbauer data: The Cu(2) atoms retain their oxidation state, whereas the Cu(1) atoms adjust their valence according to the value of δ. In the 2:2:1:2 samples, the Eu concentration clearly affected the value of the electric-field gradient at the Eu nucleus. Using a standard procedure, magnetically aligned 2:2:1:2 samples were prepared. The preferred direction of the crystal c axis changed from parallel to perpendicular alignment with the external magnetic field, when the Eu concentration exceeded 20% of the Ca atoms.Peer reviewe

    COVID-19 outbreak at a reception centre for asylum seekers in Espoo, Finland

In April 2020, an increasing number of asylum seekers at a reception centre in Espoo, Finland presented with COVID-19 despite earlier implementation of preventive measures. We decided to screen the entire population of the centre for SARS-CoV-2. Methods we offered nasopharyngeal swab collection and SARS-CoV-2 real-time polymerase chain reaction (RT-PCR) analysis to the centre's clients. Symptoms were recorded at the time of diagnostic sample collection using electronic forms and followed up for two weeks through phone interviews and a review of medical records. Findings 260 clients were screened. Of them, 96 (37%) were found positive for SARS-CoV-2 and isolated. The high attack rate prompted the local public health authority to set the other clients in quarantine for 14 days to prevent further spread. Of the positive cases, 61 (64%) reported having had symptoms at the time of the screening or one week prior. Of the 35 initially asymptomatic individuals, 12 developed symptoms during follow-up, while 23 (or 18% of all screened SARS-CoV-2 positive clients) remained asymptomatic. No widespread transmission of COVID-19 was detected after the quarantine was lifted. Interpretation in this large COVID-19 outbreak, voluntary mass screening provided valuable information about its extent and helped guide the public health response. Comprehensive quarantine and isolation measures were likely instrumental in containing the outbreak.     Cholesterol-Dependent Anaplasma phagocytophilum Exploits the Low-Density Lipoprotein Uptake Pathway

    In eukaryotes, intracellular cholesterol homeostasis and trafficking are tightly regulated. Certain bacteria, such as Anaplasma phagocytophilum, also require cholesterol; it is unknown, however, how this cholesterol-dependent obligatory intracellular bacterium of granulocytes interacts with the host cell cholesterol regulatory pathway to acquire cholesterol. Here, we report that total host cell cholesterol increased >2-fold during A. phagocytophilum infection in a human promyelocytic leukemia cell line. Cellular free cholesterol was enriched in A. phagocytophilum inclusions as detected by filipin staining. We determined that A. phagocytophilum requires cholesterol derived from low-density lipoprotein (LDL), because its replication was significantly inhibited by depleting the growth medium of cholesterol-containing lipoproteins, by blocking LDL uptake with a monoclonal antibody against LDL receptor (LDLR), or by treating the host cells with inhibitors that block LDL-derived cholesterol egress from late endosomes or lysosomes. However, de novo cholesterol biosynthesis is not required, since inhibition of the biosynthesis pathway did not inhibit A. phagocytophilum infection. The uptake of fluorescence-labeled LDL was enhanced in infected cells, and LDLR expression was up-regulated at both the mRNA and protein levels. A. phagocytophilum infection stabilized LDLR mRNA through the 3′ UTR region, but not through activation of the sterol regulatory element binding proteins. Extracellular signal–regulated kinase (ERK) was up-regulated by A. phagocytophilum infection, and inhibition of its upstream kinase, MEK, by a specific inhibitor or siRNA knockdown, reduced A. phagocytophilum infection. Up-regulation of LDLR mRNA by A. phagocytophilum was also inhibited by the MEK inhibitor; however, it was unclear whether ERK activation is required for LDLR mRNA up-regulation by A. phagocytophilum. These data reveal that A. phagocytophilum exploits the host LDL uptake pathway and LDLR mRNA regulatory system to accumulate cholesterol in inclusions to facilitate its replication

    Ingraft chimerism in lung transplantation - a study in a porcine model of obliterative bronchiolitis

    <p>Abstract</p> <p>Background</p> <p>Bronchial epithelium is a target of the alloimmune response in lung transplantation, and intact epithelium may protect allografts from rejection and obliterative bronchiolitis (OB). Herein we study the influence of chimerism on bronchial epithelium and OB development in pigs.</p> <p>Methods</p> <p>A total of 54 immunosuppressed and unimmunosuppressed bronchial allografts were serially obtained 2-90 days after transplantation. Histology (H&E) was assessed and the fluorescence in situ hybridization (FISH) method for Y chromosomes using pig-specific DNA-label was used to detect recipient derived cells in graft epithelium and bronchial wall, and donor cell migration to recipient organs. Ingraft chimerism was studied by using male recipients with female donors, whereas donor cell migration to recipient organs was studied using female recipients with male donors.</p> <p>Results</p> <p>Early appearance of recipient-derived cells in the airway epithelium appeared predictive of epithelial destruction (<it>R </it>= 0.610 - 0.671 and <it>p </it>< 0.05) and of obliteration of the bronchial lumen (<it>R </it>= 0.698 and <it>p </it>< 0.01). All allografts with preserved epithelium showed epithelial chimerism throughout the follow-up. Antirejection medication did not prevent, but delayed the appearance of Y chromosome positive cells in the epithelium (<it>p </it>< 0.05), or bronchial wall (<it>p </it>< 0.05).</p> <p>Conclusions</p> <p>In this study we demonstrate that early appearance of Y chromosomes in the airway epithelium predicts features characteristic of OB. Chimerism occurred in all allografts, including those without features of OB. Therefore we suggest that ingraft chimerism may be a mechanism involved in the repair of alloimmune-mediated tissue injury after transplantation.</p
