    Primeros resultados sobre la calidad de la canal de los corderos de la raza autóctona montesina

    En el presente trabajo se ha realizado la caracterización de la canal de los corderos machos de raza Montesina sacrificados con aproximadamente 20 y 25 kg de peso vivo. Para ello se han realizado las siguientes determinaciones: valor de pH, peso de la canal fría, estado de engrasamiento y conformación. Además, se han obtenido algunas medidas zoométricas, como el espesor de la grasa dorsal. El color de la canal se ha valorado instrumentalmente mediante espectrocolorímetro. Por último, se ha calculado el contenido de piezas de 1ª, 2ª y 3ª categoría mediante despiece comercial normalizado. Las características de las canales de corderos de raza Montesina se aproximan a las de los ovinos ligeros producidos en la cuenca mediterránea con pesos de canal por debajo de los 13 kg, de conformación ligera (O y P) y reducido contenido de grasa. El porcentaje de piezas de primera categoría ha sido de aproximadamente del 46% lo que hace pensar en el buen rendimiento carnicero de la raza. El incremento de peso de las canales de raza Montesina desde 20 a 25 kg de peso vivo supone, fundamentalmente, una mejora en la conformación de las mismas, sin embargo, otros aspectos de la canal no se modifican significativamenteMontesina Sheep breed, as endangered breed, must be characterized for developing an useful expansion plan.     Vortex flow reactor assessment for the purification of monoclonal antibodies from unclarified broths

    The vortex flow reactor (VFR) can be used in many chemical engineering applications. This paper assesses its novel use in the purification of monoclonal antibodies from cell broth. To this end, the IgG2a antibody was purified from the unclarified fermentation broth of transgenic mouse 55/6 hybridoma cells. Visual experiments showed that the VFR worked in the laminar vortices flow regime and the vortices displaced slightly faster than the axial flow. The VFR has the advantage of creating two sorts of flows: axial flow to produce the expanded bed and an extra vortex flow to avoid channeling and stabilize the expanded bed, the hydrodynamic behavior of which is plug flow with an experimental Pèclet number higher than 20. The pH was adjusted in the untreated fermentation broth, which was directly introduced into the reactor thus reducing the number of stages. The IgG2a purification was carried out in a single device via two steps: antibody adsorption in the expanded bed and antibody elution in the settled bed using Streamline rProtein A. A thirty-fold increase in the high-purity antibody concentration was achieved at the top of the pH5 elution peak with a total recovery of 93.1% (w/w) between elution peaks pH 5 and 3

    The addition of albumin improves Schwann cells viability in nerve cryopreservation

    [Abstract] The purpose of the current study was to establish a valid protocol for nerve cryopreservation, and to evaluate if the addition of albumin supposed any advantage in the procedure. We compared a traditional cryopreservation method that uses dimethyl sulfoxide (DMSO) as cryoprotectant, to an alternative method that uses DMSO and albumin. Six Wistar Lewis rats were used to obtain twelve 20 mm fragments of sciatic nerve. In the first group, six fragments were cryopreserved in 199 media with 10% DMSO, with a temperature decreasing rate of 1 °C per minute. In the second group, six fragments were cryopreserved adding 4% human albumin. The unfreezing process consisted of sequential washings with saline in the first group, and saline and 20% albumin in the second group at 37 °C until the crioprotectant was removed. Structural evaluation was performed through histological analysis and electronic microscopy. The viability was assessed with the calcein-AM (CAM) and 4′,6-diamino-2-fenilindol (DAPI) staining. Histological results showed a correct preservation of peripheral nerve architecture and no significant differences were found between the two groups. However, Schwann cells viability showed in the CAM-DAPI staining was significantly superior in the albumin group. The viability of Schwann cells was significantly increased when albumin was added to the nerve cryopreservation protocol. However, no significant structural differences were found between groups. Further studies need to be performed to assess the cryopreserved nerve functionality using this new method

    Calidad de la canal de corderos (ternasco y lechal) de la raza autóctona andaluza churra lebrijana

    En la raza Churra Lebrijana (catalogada como “de protección oficial”) se ha estudiado por primera vez el potencial productivo de corderos lechales frente a ternascos (el peso vivo de estos animales fue de 11,38 y 18,48 Kg respectivamente). Para ello se estudiaron en 8 corderos machos de cada tipo las características de sus canales en relación a parámetros de pH, color, rendimiento comercial, medidas zoometricas, engrasamiento y despiece. Del trabajo se deduce que los ternascos ofrecen unas mejores características cárnicas que los lechales (tipo que es el que más se comercializa actualmente) por lo que es interesante su comercialización en este tipo comercial.In Churra Lebrijana breed (catalogued as "of official protection") the productive potential of suckling lambs has studied for the first time front to light lamb (the alive weight of these animals was of 11.38 and 18.48 kg respectively). For it the characteristics of their carcasses in relation to parameters of pH studied in 8 male lambs of each type, color, commercial yield, zoometric measures, fattening and quartering. Of the work it is deduced that the light lambs offer better meat characteristics than the suckling lambs (type that are the one that is commercialized more at the moment) reason why its commercialization in this commercial type is interesting.INIA RZ03-01

    Intraneural IFG-1 in cryopreserved nerve isografts increase neural regeneration and functional recovery in the rat sciatic nerve

    [Abstract] Background: Insulin-like growth factor 1 (IGF-1) was found to stimulate Schwann cell mitosis. Exogenous IGF-1 may improve nerve regeneration after cryopreservation. Objective: To evaulate the effect of intraneural administration of IGF-1 in cryopreserved nerve isografts. Methods: Eighteen millimeter grafts were used for bridging an 18-mm defect in the rat sciatic nerve. A total of 57 rats were randomly divided into three groups: (1) autograft (Group 1); (2) cryopreserved isograft (Group 2); (3) cryopreserved isograft with intraneural IGF-1 administration (Group 3). 12 weeks after surgery, functional recovery (Sciatic functional index [SFI], Swing speed [SS], nerve conduction velocity [NCV], amplitude of compound motor action potentials [CMAP], and gastrocnemius muscle index [GMI]) and nerve regeneration (myelin sheath area, total fiber counts, fiber density, and fiber width) were all evaluated. Results: The intraneural injection of IGF-1 significantly improved SFI and SS at weeks 10 and 12. There were no statistical differences between Groups 1 and 3 in any of the SFI or SS evaluations. CMAP and NCV in Group 1 were significantly higher than in Groups 2 and 3, and Group 3 had significantly higher CMAP and NCV compared to Group 2. No significant differences were found in fiber width. The number of nerve fibers, percentage of myelinated fibers, fiber density, and GMI was significantly higher in Group 1 compared to Group 2, but no significant differences were found between Groups 1 and 3. Conclusion: The results show that intraneural injection of IGF-1 in an 18 mm cryopreserved isograft improve axonal regeneration and functional recovery

    Clinical Validation of a 3-Dimensional Ultrafast Cardiac Magnetic Resonance Protocol Including Single Breath-Hold 3-Dimensional Sequences

    Objectives: This study sought to clinically validate a novel 3-dimensional (3D) ultrafast cardiac magnetic resonance (CMR) protocol including cine (anatomy and function) and late gadolinium enhancement (LGE), each in a single breath-hold. Background: CMR is the reference tool for cardiac imaging but is time-consuming. Methods: A protocol comprising isotropic 3D cine (Enhanced sensitivity encoding [SENSE] by Static Outer volume Subtraction [ESSOS]) and isotropic 3D LGE sequences was compared with a standard cine+LGE protocol in a prospective study of 107 patients (age 58 ± 11 years; 24% female). Left ventricular (LV) mass, volumes, and LV and right ventricular (RV) ejection fraction (LVEF, RVEF) were assessed by 3D ESSOS and 2D cine CMR. LGE (% LV) was assessed using 3D and 2D sequences. Results: Three-dimensional and LGE acquisitions lasted 24 and 22 s, respectively. Three-dimensional and LGE images were of good quality and allowed quantification in all cases. Mean LVEF by 3D and 2D CMR were 51 ± 12% and 52 ± 12%, respectively, with excellent intermethod agreement (intraclass correlation coefficient [ICC]: 0.96; 95% confidence interval [CI]: 0.94 to 0.97) and insignificant bias. Mean RVEF 3D and 2D CMR were 60.4 ± 5.4% and 59.7 ± 5.2%, respectively, with acceptable intermethod agreement (ICC: 0.73; 95% CI: 0.63 to 0.81) and insignificant bias. Both 2D and 3D LGE showed excellent agreement, and intraobserver and interobserver agreement were excellent for 3D LGE. Conclusions: ESSOS single breath-hold 3D CMR allows accurate assessment of heart anatomy and function. Combining ESSOS with 3D LGE allows complete cardiac examination in less than 1 min of acquisition time. This protocol expands the indication for CMR, reduces costs, and increases patient comfort.     Effects of functional remediation on neurocognitively impaired bipolar patients: enhancement of verbal memory

    Background: functional remediation is a novel intervention with demonstrated efficacy at improving functional outcome in euthymic bipolar patients. However, in a previous trial no significant changes in neurocognitive measures were detected. The objective of the present analysis was to test the efficacy of this therapy in the enhancement of neuropsychological functions in a subgroup of neurocognitively impaired bipolar patients. Method: a total of 188 out of 239 DSM-IV euthymic bipolar patients performing below two standard deviations from the mean of normative data in any neurocognitive test were included in this subanalysis. Repeated-measures analyses of variance were conducted to assess the impact of the treatment arms [functional remediation, psychoeducation, or treatment as usual (TAU)] on participants' neurocognitive and functional outcomes in the subgroup of neurocognitively impaired patients. Results: patients receiving functional remediation (n = 56) showed an improvement on delayed free recall when compared with the TAU (n = 63) and psychoeducation (n = 69) groups as shown by the group × time interaction at 6-month follow-up [F 2,158 = 3.37, degrees of freedom (df) = 2, p = 0.037]. However, Tukey post-hoc analyses revealed that functional remediation was only superior when compared with TAU (p = 0.04), but not with psychoeducation (p = 0.10). Finally, the patients in the functional remediation group also benefited from the treatment in terms of functional outcome (F 2,158 = 4.26, df = 2, p = 0.016). Conclusions: functional remediation is effective at improving verbal memory and psychosocial functioning in a sample of neurocognitively impaired bipolar patients at 6-month follow-up. Neurocognitive enhancement may be one of the active ingredients of this novel intervention, and, specifically, verbal memory appears to be the most sensitive function that improves with functional remediation
