    Understanding communicative actions:A repetitive TMS study

    Previous work has shown that the right posterior superior temporal sulcus (RpSTS) is involved in inferring both instrumental goals and communicative intentions of observed actions (1, 2), using previous knowledge to generate perceptual and/or conceptual inferences (3, 4). Here, we disturb neural activity in RpSTS to test whether this region is necessary for understanding the meaning of novel communicative actions. Thirteen subjects received two 20 min sessions of low-frequency (1 Hz) repetitive transcranial magnetic stimulation (rTMS), either over RpSTS (50,-42,14) or over a control region (area MT+, -43,-70,10), before performance of a communicative game and a control task. The game involved controlled non-verbal communicative interactions between pairs of subjects (40 trials). Each pair was asked to jointly create a goal configuration of two geometrical tokens, using the movements of the tokens on a gameboard as the only available communicative channel (5). One participant (a confederate) knew the goal configuration, and she moved her token on the gameboard to inform an addressee (a participant) where and how to position his token. The control task was a visual search paradigm that involved the same stimuli, responses, joint attention, and inter-subjects dependencies, but no communicative necessities. Performance was indexed by Task Efficiency, defined as the number of correct responses per unit of planning time, and by Efficiency Rate, defined as the rate of change (across trials) in Task Efficiency. After rTMS over RpSTS, but not left MT+, the Efficiency Rate of the addresses was reduced in the communicative game, but not in the visual search. In contrast, after rTMS over left MT+, subjects were not able to benefit from experience gained during the previous trials of the visual search task (Task X Site interaction, p<0.05). There were no corresponding interactions between tasks and site of rTMS intervention on the Task Efficiency parameter. These findings qualify how RpSTS contributes to understanding the meaning of non-verbal communicative actions. Repetitive TMS over RpSTS did not disrupt the ability of addressees to interpret novel communicative actions. Rather, this region appears to be necessary for incorporating previous knowledge, accumulated during interactions with a communicative partner, to constrain the inferential process that leads to action understanding

    Kansverdelingen van extreme neerslag zijn doorgaans afgeleid van puntmetingen van regenmeternetwerken en gelden voor een locatie. In het waterbeheer is echter vaak de extreme gebiedsneerslag van belang. Die is voor een bepaalde herhalingstijd lager dan de extreme neerslag op een locatie in dat gebied voor dezelfde herhalingstijd. Door deze gebiedsreductie leiden de gangbare neerslagstatistieken tot een onderschatting van de herhalingstijd voor extreme gebiedsneerslag. Daarom is het noodzakelijk om de kansverdeling van extreme gebiedsneerslag te bestuderen. Dit is maar beperkt mogelijk met de netwerken van regenmeters in Nederland. Recent hebben het Koninklijk Nederlands Meteorologisch Instituut (KNMI) en de leerstoelgroep Hydrologie en Kwantitatief Waterbeheer van Wageningen Universiteit een elfjarige radardataset van neerslaghoeveelheden geanalyseerd. Hierdoor zijn nu neerslagwaarnemingen met een hoge resolutie in tijd en ruimte beschikbaar voor heel Nederland. Met behulp van deze dataset zijn kansverdelingen van extreme gebiedsneerslag afgeleid

    Migraine and sleep disorders: a systematic review

    Migraine and sleep disorders are common and often burdensome chronic conditions with a high prevalence in the general population, and with considerable socio-economic impact and costs. The existence of a relationship between migraine and sleep disorders has been recognized from centuries by clinicians and epidemiological studies. Nevertheless, the exact nature of this association, the underlying mechanisms and interactions are complex and not completely understood. Recent biochemical and functional imaging studies identified central nervous system structures and neurotransmitters involved in the pathophysiology of migraine and also important for the regulation of normal sleep architecture, suggesting a possible causative role, in the pathogenesis of both disorders, of a dysregulation in these common nervous system pathways. This systematic review summarizes the existing data on migraine and sleep disorders with the aim to evaluate the existence of a causal relationship and to assess the presence of influencing factors. The identification of specific sleep disorders associated with migraine should induce clinicians to systematically assess their presence in migraine patients and to adopt combined treatment strategies

    Engineering Azobenzene Derivatives to Control the Photoisomerization Process

    In this work, we show how the structural features of photoactive azobenzene derivatives can influence the photoexcited state behavior and the yield of the trans/cis photoisomerization process. By combining high-resolution transient absorption experiments in the vis-NIR region and quantum chemistry calculations (TDDFT and RASPT2), we address the origin of the transient signals of three poly-substituted push-pull azobenzenes with an increasing strength of the intramolecular interactions stabilizing the planar trans isomer (absence of intramolecular H-bonds, methyl, and traditional H-bond, respectively, for 4-diethyl-4′-nitroazobenzene, Disperse Blue 366, and Disperse Blue 165) and a commercial red dye showing keto-enol tautomerism involving the azo group (Sudan Red G). Our results indicate that the intramolecular H-bonds can act as a “molecular lock” stabilizing the trans isomer and increasing the energy barrier along the photoreactive CNNC torsion coordinate, thus preventing photoisomerization in the Disperse Blue dyes. In contrast, the involvement of the azo group in keto-enol tautomerism can be employed as a strategy to change the nature of the lower excited state and remove the nonproductive symmetric CNN/NNC bending pathway typical of the azo group, thus favoring the productive torsional motion. Taken together, our results can provide guidelines for the structural design of azobenzene-based photoswitches with a tunable excited state behavior

    Efficient and scalable generation of primordial germ cells in 2D culture using basement membrane extract overlay

    Current methods to generate human primordial germ cell-like cells (hPGCLCs) from human pluripotent stem cells (hPSCs) can be inefficient, and it is challenging to generate sufficient hPGCLCs to optimize in vitro gametogenesis. We present a differentiation method that uses diluted basement membrane extract (BMEx) and low BMP4 concentration to efficiently induce hPGCLC differentiation in scalable 2D cell culture. We show that BMEx overlay potentiated BMP/SMAD signaling, induced lumenogenesis, and increased expression of key hPGCLC-progenitor markers such as TFAP2A and EOMES. hPGCLCs that were generated using the BMEx overlay method were able to upregulate more mature germ cell markers, such as DAZL and DDX4, in human fetal ovary reconstitution culture. These findings highlight the importance of BMEx during hPGCLC differentiation and demonstrate the potential of the BMEx overlay method to interrogate the formation of PGCs and amnion in humans, as well as to investigate the next steps to achieve in vitro gametogenesis.Stem cells & developmental biolog

    Recognition and diagnosis of sleep disorders in Parkinson's disease

    Contains fulltext : 109296.pdf (publisher's version ) (Open Access)Sleep disturbances are among the most frequent and incapacitating non-motor symptoms of Parkinson's disease (PD), and are increasingly recognized as an important determinant of impaired quality of life. Here we review several recent developments regarding the recognition and diagnosis of sleep disorders in PD. In addition, we provide a practical and easily applicable approach to the diagnostic process as a basis for tailored therapeutic interventions. This includes a stepwise scheme that guides the clinical interview and subsequent ancillary investigations. In this scheme, the various possible sleep disorders are arranged not in order of prevalence, but in a 'differential diagnostic' order. We also provide recommendations for the use of sleep registrations such as polysomnography. Furthermore, we point out when a sleep specialist could be consulted to provide additional diagnostic and therapeutic input. This structured approach facilitates early detection of sleep disturbances in PD, so treatment can be initiated promptly
