Legionella in Puerto Rico cooling towers
Water samples from air conditioning cooling towers receiving different treatment protocols on five large municipal buildings in San Juan, Puerto Rico were assayed for various species and serogroups of Legionella spp. using direct immunofluorescence. Several water quality parameters were also measured with each sample. Guinea pigs were inoculated with water samples to confirm pathogenicity and recover viable organisms. Legionella pneumophila (1-6), L. bozemanii, L. micdadei, L. dumoffii, and L. gormanii were observed in at least one of the cooling towers. L. pneumophila was the most abundant species, reaching 10{sup 5} cells/ml, within the range that is considered potentially pathogenic to humans. A significantly higher density of L. pneumophila was observed in the cooling tower water that was not being treated with biocides. Percent respiration (INT) and total cell activity (AODC), were inversely correlated with bacterial density. This study demonstrates that Legionella spp. are present in tropical air-conditioning cooling systems, and without continuous biocide treatment may reach densities that present a health risk
A global assessment of the potential distribution of naturalized and planted populations of the ornamental alien tree Schinus molle
Estudio sobre la potencial invasión del pirul, una planta naturalizada cuyo origen es el sur de América.The Peruvian Peppertree (Schinus molle L.) is an evergreen tree native to semiarid environments of Peru and Bolivia in South America. This tree has been introduced and widely planted for ornamental and forestry purposes in several semiarid regions of the world because its seedlings are easily established and have a high survival rate; it also grows quickly, and it is tolerant of dry climates. We compared the global and regional niches of naturalized and planted populations of S. molle in order to examine the invasive stages and potential distribution of this species in four regions of the world. This work provides a novel approach for understanding the invasion dynamics of S. molle in these areas and elucidates the ecological processes that bring about such invasions. Most naturalized and planted populations were found to be in equilibrium with the environment. In its native range as well as in Australia and South Africa the models of the coverage area of habitat suitability for natural populations were the highest, whereas the coverage area of planted populations was lower. For planted populations in Australia and South Africa, a large percentage of predicted presences fell within sink populations. The invasion stages of S. molle vary across regions in its adventive range; this result may be attributable to residence time as well as climatic and anthropic factors that have contributed to the spread of populations.SEMARNAT-CONACYT [Grant FSSEMARNAT01-C-2018-1-A3-S-80837]
Cervical cancer cell lines expressing NKG2D-ligands are able to down-modulate the NKG2D receptor on NKL cells with functional implications
<p>Abstract</p> <p>Background</p> <p>Cervical cancer represents the third most commonly diagnosed cancer and the fourth leading cause of cancer-related deaths in women worldwide. Natural killer (NK) cells play an important role in the defense against viruses, intracellular bacteria and tumors. NKG2D, an activating receptor on NK cells, recognizes MHC class I chain-related molecules, such as MICA/B and members of the ULBP/RAET1 family. Tumor-derived soluble NKG2D-ligands have been shown to down-modulate the expression of NKG2D on NK cells. In addition to the down-modulation induced by soluble NKG2D-ligands, it has recently been described that persistent cell-cell contact can also down-modulate NKG2D expression. The goal of this study was to determine whether the NKG2D receptor is down-modulated by cell-cell contact with cervical cancer cells and whether this down-modulation might be associated with changes in NK cell activity.</p> <p>Results</p> <p>We demonstrate that NKG2D expressed on NKL cells is down-modulated by direct cell contact with cervical cancer cell lines HeLa, SiHa, and C33A, but not with non-tumorigenic keratinocytes (HaCaT). Moreover, this down-modulation had functional implications. We found expression of NKG2D-ligands in all cervical cancer cell lines, but the patterns of ligand distribution were different in each cell line. Cervical cancer cell lines co-cultured with NKL cells or fresh NK cells induced a marked diminution of NKG2D expression on NKL cells. Additionally, the cytotoxic activity of NKL cells against K562 targets was compromised after co-culture with HeLa and SiHa cells, while co-culture with C33A increased the cytotoxic activity of the NKL cells.</p> <p>Conclusions</p> <p>Our results suggest that differential expression of NKG2D-ligands in cervical cancer cell lines might be associated with the down-modulation of NKG2D, as well as with changes in the cytotoxic activity of NKL cells after cell-cell contact with the tumor cells.</p

Effect of lipid-lowering treatment in cardiovascular disease prevalence in familial hypercholesterolemia
Background and aims: The impact on heterozygous familial hypercholesterolemia (HeFH) health led by high-intensity lipid-lowering therapy (HILLT) is unknown, and the question remains if there is still an unacceptably high residual risk to justify treatment with new lipid-lowering drugs.
Methods: This observational, retrospective, multicenter, national study in Spain, whose information was obtained from a national dyslipemia registry, was designed to establish the current prevalence of cardiovascular disease (CVD) in HeFH and to define the impact of HILLT on CVD in this population. Odds were estimated using several logistic regression models with progressive adjustment.
Results: 1958 HeFH, mean age 49.3 ± 14.3 years, were included in the analysis. At inclusion in the registry, 295 patients (15.1%) had suffered CVD and 164 (55.6%) had suffered the first event before the onset lipid-lowering treatment. Exposition to treatment associated more than ten times lower odds for CVD than in subjects naïve to treatment (OR 0.085, 95% CI 0.063–0.114, p < 0.001). A first CVD event after a mean treatment period of 9.1 ± 7.2 years occurred in 131 out of 1615 (8.1%) HeFH subjects, and 115 (87.8%) of them were on HILLT.
Conclusions: Current prevalence of CVD among HeFH is one third of that reported before the statins era. Early initiation and prolonged lipid-lowering treatment was associated with a reduction in CVD. New cases of CVD, in spite of HILLT, appeared mostly among patients accumulating risk factors and probably they may be considered for further lipid-lowering drugs
Evaluation of different bowel preparations for small bowel capsule endoscopy: a prospective, randomized, controlled study
To obtain an adequate view of the whole small
intestine during capsule endoscopy (CE) a clear liquid diet and overnight fasting
is recommended. However, intestinal content can hamper vision in spite of these
measures. Our aim was to evaluate tolerance and degree of intestinal cleanliness
during CE following three types of bowel preparation. PATIENTS AND METHODS: This
was a prospective, multicenter, randomized, controlled study. Two-hundred
ninety-one patients underwent one of the following preparations: 4 L of clear
liquids (CL) (group A; 92 patients); 90 mL of aqueous sodium phosphate (group B;
89 patients); or 4 L of a polyethylene glycol electrolyte solution (group C; 92
patients). The degree of cleanliness of the small bowel was classified by blinded
examiners according to four categories (excellent, good, fair or poor). The
degree of patient satisfaction, gastric and small bowel transit times, and
diagnostic yield were measured. RESULTS: The degree of cleanliness did not differ
significantly between the groups (P = 0.496). Interobserver concordance was fair
(k = 0.38). No significant differences were detected between the diagnostic
yields of the CE (P = 0.601). Gastric transit time was 35.7 +/- 3.7 min (group
A), 46.1 +/- 8.6 min (group B) and 34.6 +/- 5.0 min (group C) (P = 0.417).
Small-intestinal transit time was 276.9 +/- 10.7 min (group A), 249.7 +/- 13.1
min (group B) and 245.6 +/- 11.6 min (group C) (P = 0.120). CL was the best
tolerated preparation. Compliance with the bowel preparation regimen was lowest
in group C (P = 0.008). CONCLUSIONS: A clear liquid diet and overnight fasting is
sufficient to achieve an adequate level of cleanliness and is better tolerated by
patients than other forms of preparation
Carcinoma-derived interleukin-8 disorients dendritic cell migration without impairing T-cell stimulation
BACKGROUND:
Interleukin-8 (IL-8, CXCL8) is readily produced by human malignant cells. Dendritic cells (DC) both produce IL-8 and express the IL-8 functional receptors CXCR1 and CXCR2. Most human colon carcinomas produce IL-8. IL-8 importance in malignancies has been ascribed to angiogenesis promotion.
PRINCIPAL FINDINGS:
IL-8 effects on human monocyte-derived DC biology were explored upon DC exposure to recombinant IL-8 and with the help of an IL-8 neutralizing mAb. In vivo experiments were performed in immunodeficient mice xenografted with IL-8-producing human colon carcinomas and comparatively with cell lines that do not produce IL-8. Allogenic T lymphocyte stimulation by DC was explored under the influence of IL-8. DC and neutrophil chemotaxis were measured by transwell-migration assays. Sera from tumor-xenografted mice contained increasing concentrations of IL-8 as the tumors progress. IL-8 production by carcinoma cells can be modulated by low doses of cyclophosphamide at the transcription level. If human DC are injected into HT29 or CaCo2 xenografted tumors, DC are retained intratumorally in an IL-8-dependent fashion. However, IL-8 did not modify the ability of DC to stimulate T cells. Interestingly, pre-exposure of DC to IL-8 desensitizes such cells for IL-8-mediated in vitro or in vivo chemoattraction. Thereby DC become disoriented to subsequently follow IL-8 chemotactic gradients towards malignant or inflamed tissue.
CONCLUSIONS:
IL-8 as produced by carcinoma cells changes DC migration cues, without directly interfering with DC-mediated T-cell stimulation
Characterization of oral yeasts isolated from healthy individuals attended in different Colombian dental clinics
The aim of this study was to identify the most frequent yeasts in the oral cavity of adult individuals without immune disorders and to associate the presence of these oral yeasts with different characteristics of each individual. Oral rinse samples were obtained from 96 healthy adults and cultured in Sabouraud dextrose agar media and CHROMagar. Yeasts were identified by sequencing the D1/D2 region of the 28S rRNA gene. Probable association among the socio-demographic characteristics, body mass index, family and personal medical history, oral hygiene, tobacco and/or alcohol consumption habits and presence of oral fungi was analyzed. Contingency tables and logistic regression were employed to evaluate possible relationships between the presence of oral fungi and mixed colonization with these variables. 57.3% of the healthy individuals had oral yeasts and 21.8% had mixed colonization. The most prevalent yeasts were Candida albicans (52%), C. parapsilosis (17.9%), and C. dubliniensis (7.57%). Yeasts with most frequently mixed colonization were C. albicans and C. parapsilosis. No relationships were found among the variables analyzed. However, the presence of mixed colonization was related to the presence of dental prostheses (P less than 0.006), dental apparatuses (P=0.016) and O'Leary index (P=0.012). This is the first study that characterized oral yeasts in Colombian healthy individuals, determined the most prevalent oral yeasts C. albicans, C. parapsilosis and C. dublinensis and an association of mixed colonization with the use of dental prostheses and aparatology and poor hygiene. © 2019 by the Journal of Biomedical Research
Functional characterization of a melon alcohol acyl-transferase gene family involved in the biosynthesis of ester volatiles. Identification of the crucial role of a threonine residue for enzyme activity
Volatile esters, a major class of compounds contributing to the aroma of many fruit, are synthesized by
alcohol acyl-transferases (AAT). We demonstrate here that, in Charentais melon (Cucumis melo var.
cantalupensis), AAT are encoded by a gene family of at least four members with amino acid identity ranging
from 84% (Cm-AAT1/Cm-AAT2) and 58% (Cm-AAT1/Cm-AAT3) to only 22% (Cm-AAT1/Cm-AAT4).
All encoded proteins, except Cm-AAT2, were enzymatically active upon expression in yeast and show
differential substrate preferences. Cm-AAT1 protein produces a wide range of short and long-chain acyl
esters but has strong preference for the formation of E-2-hexenyl acetate and hexyl hexanoate. Cm-AAT3
also accepts a wide range of substrates but with very strong preference for producing benzyl acetate.
Cm-AAT4 is almost exclusively devoted to the formation of acetates, with strong preference for cinnamoyl
acetate. Site directed mutagenesis demonstrated that the failure of Cm-AAT2 to produce volatile esters is
related to the presence of a 268-alanine residue instead of threonine as in all active AAT proteins. Mutating
268-A into 268-T of Cm-AAT2 restored enzyme activity, while mutating 268-T into 268-A abolished
activity of Cm-AAT1. Activities of all three proteins measured with the prefered substrates sharply increase
during fruit ripening. The expression of all Cm-AAT genes is up-regulated during ripening and inhibited in
antisense ACC oxidase melons and in fruit treated with the ethylene antagonist 1-methylcyclopropene
(1-MCP), indicating a positive regulation by ethylene. The data presented in this work suggest that the
multiplicity of AAT genes accounts for the great diversity of esters formed in melon
Effectiveness of rotavirus vaccination in Spain
With the aim of determining rotavirus vaccine effectiveness (RVVE) in Spain, from Oct-2008/Jun-2009, 467 consecutive children below 2 years old with acute gastroenteritis (AGE) were recruited using a pediatric research network (ReGALIP-www.regalip.org) that includes primary, emergency and hospital care settings. Of 467 enrolled children, 32.3% were rotavirus positive and 35.0% had received at least one dose of any rotavirus vaccine. RRVE to prevent any episode of rotavirus AGE was 91.5% (95% CI: 83.7%-95.6%). RVVE to prevent hospitalization by rotavirus AGE was 95.6% (85.6-98.6%).
