    TB117: Techniques for Using the Growth and Behavior of Imprinted Ducklings to Evaluate Habitat Quality

    We developed a technique for evaluating duckling habitat quality that is based on two assumptions. In good habitat young birds (1) grow rapidly and thus are better able t o survive stresses such as inclement weather, and (2) spend relatively less time moving about in search of food and more time resting and thus are less conspicuous to predators. We imprinted artificially incubated and hatched ducklings by being present at the time of hatching; i.e., the ducklings thought we were their mother. Ducklings were split into broods and placed on ponds where their growth was measured and their behavior monitored for several days. Comparisons of growth rates and behavioral time budgets allowed us to determine which ponds were better habitat.     University of Maine NSF-ADVANCE Project Outcomes Report

    The University of Maine ADVANCE IT project proposes a three-pronged approach to institutional gender equity for STEM women faculty. Specific goals and objectives include increasing the percentage of women in the STEM disciplines, supporting professional development activities, addressing recruitment, retention and advancement of women faculty and engaging other campuses in the University of Maine System, as well as the faculty union, through the dissemination of information regarding ADVANCE outcomes. Intellectual Merit. The proposed activities are rooted in a conceptual framework that focuses on faculty job satisfaction, which plays a major role in retention. This project also presents a unique perspective to institutional transformation in that it takes into consideration the faculty union and its impact on faculty advancement and job satisfaction. Broader Impact. The University of Maine ADVANCE IT project , because of its emphasis on seven other institutions in the state of Maine, has the capacity to impact women faculty beyond the proposing institution. It is expected that project results will be broadly disseminated across Maine and to other institutions through traditional means including national presentations and scholarly journal articles. As such, the University of Maine will contribute new knowledge to the field of institutional transformation

    Impact of Face Masks on Audiovisual Word Recognition in Young Children with Hearing Loss During the Covid-19 Pandemic

    Objective: To investigate effects of surgical and transparent face masks during the Covid-19 pandemic on audiovisual speech recognition of words for deaf and hard-of-hearing children. Design: Recorded Word Intelligibility by Picture Identification test (WIPI) was presented in quiet via a computer monitor to children in a quiet test room. The acoustic power spectra of each mask type were compared to the baseline no mask condition. Percent correct word recognition was recorded for four mask conditions (no mask, surgical mask, transparent apron mask and ClearMask) in counterbalanced order. Repeated measures ANOVA was used to test for significant differences in word recognition scores across mask types. Study Sample: Thirteen children (3 to 7 years) in a private auditory oral school wearing hearing aids, bone-anchored hearing aids or cochlear implants. Children were excluded if English was not their primary language or if they had a severe speech-language delay, uncorrected vision loss, or developmental disorder that would affect the results. No children had been exposed to or had contracted the Covid-19 virus. Results: Acoustic spectra showed a decrease in the 2000-8000 Hz region for the transparent apron mask. The surgical mask and ClearMask showed fewer acoustic effects. Children with hearing aids performed similarly to children with cochlear implants. Word recognition was significantly poorer for surgical masks and transparent apron masks. The ClearMask condition was not significantly worse than the no mask condition for words in quiet. Conclusions: Standard surgical and custom apron shield masks significantly hampered word recognition, even in quiet conditions. The commercially available ClearMask did not significantly affect scores in quiet for young deaf and hard-of-hearing children, but scores were highly variable

    dsRNA expression in the mouse elicits RNAi in oocytes and low adenosine deamination in somatic cells

    Double-stranded RNA (dsRNA) can enter different pathways in mammalian cells, including sequence-specific RNA interference (RNAi), sequence-independent interferon (IFN) response and editing by adenosine deaminases. To study the routing of dsRNA to these pathways in vivo, we used transgenic mice ubiquitously expressing from a strong promoter, an mRNA with a long hairpin in its 3′-UTR. The expressed dsRNA neither caused any developmental defects nor activated the IFN response, which was inducible only at high expression levels in cultured cells. The dsRNA was poorly processed into siRNAs in somatic cells, whereas, robust RNAi effects were found in oocytes, suggesting that somatic cells lack some factor(s) facilitating siRNA biogenesis. Expressed dsRNA did not cause transcriptional silencing in trans. Analysis of RNA editing revealed that a small fraction of long dsRNA is edited. RNA editing neither prevented the cytoplasmic localization nor processing into siRNAs. Thus, a long dsRNA structure is well tolerated in mammalian cells and is mainly causing a robust RNAi response in oocytes

    Effect of Deutetrabenazine on Chorea Among Patients With Huntington Disease A Randomized Clinical Trial

    Importance Deutetrabenazine is a novel molecule containing deuterium, which attenuates CYP2D6 metabolism and increases active metabolite half-lives and may therefore lead to stable systemic exposure while preserving key pharmacological activity. Objective To evaluate efficacy and safety of deutetrabenazine treatment to control chorea associated with Huntington disease. Design, Setting, and Participants Ninety ambulatory adults diagnosed with manifest Huntington disease and a baseline total maximal chorea score of 8 or higher (range, 0-28; lower score indicates less chorea) were enrolled from August 2013 to August 2014 and randomized to receive deutetrabenazine (n = 45) or placebo (n = 45) in a double-blind fashion at 34 Huntington Study Group sites. Interventions Deutetrabenazine or placebo was titrated to optimal dose level over 8 weeks and maintained for 4 weeks, followed by a 1-week washout. Main Outcomes and Measures Primary end point was the total maximal chorea score change from baseline (the average of values from the screening and day-0 visits) to maintenance therapy (the average of values from the week 9 and 12 visits) obtained by in-person visits. This study was designed to detect a 2.7-unit treatment difference in scores. The secondary end points, assessed hierarchically, were the proportion of patients who achieved treatment success on the Patient Global Impression of Change (PGIC) and on the Clinical Global Impression of Change (CGIC), the change in 36-Item Short Form– physical functioning subscale score (SF-36), and the change in the Berg Balance Test. Results Ninety patients with Huntington disease (mean age, 53.7 years; 40 women [44.4%]) were enrolled. In the deutetrabenazine group, the mean total maximal chorea scores improved from 12.1 (95% CI, 11.2-12.9) to 7.7 (95% CI, 6.5-8.9), whereas in the placebo group, scores improved from 13.2 (95% CI, 12.2-14.3) to 11.3 (95% CI, 10.0-12.5); the mean between-group difference was –2.5 units (95% CI, –3.7 to –1.3) (P < .001). Treatment success, as measured by the PGIC, occurred in 23 patients (51%) in the deutetrabenazine group vs 9 (20%) in the placebo group (P = .002). As measured by the CGIC, treatment success occurred in 19 patients (42%) in the deutetrabenazine group vs 6 (13%) in the placebo group (P = .002). In the deutetrabenazine group, the mean SF-36 physical functioning subscale scores decreased from 47.5 (95% CI, 44.3-50.8) to 47.4 (44.3-50.5), whereas in the placebo group, scores decreased from 43.2 (95% CI, 40.2-46.3) to 39.9 (95% CI, 36.2-43.6), for a treatment benefit of 4.3 (95% CI, 0.4 to 8.3) (P = .03). There was no difference between groups (mean difference of 1.0 unit; 95% CI, –0.3 to 2.3; P = .14), for improvement in the Berg Balance Test, which improved by 2.2 units (95% CI, 1.3-3.1) in the deutetrabenazine group and by 1.3 units (95% CI, 0.4-2.2) in the placebo group. Adverse event rates were similar for deutetrabenazine and placebo, including depression, anxiety, and akathisia. Conclusions and Relevance Among patients with chorea associated with Huntington disease, the use of deutetrabenazine compared with placebo resulted in improved motor signs at 12 weeks. Further research is needed to assess the clinical importance of the effect size and to determine longer-term efficacy and safety

    Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead

    The real and imagined legacy of the ancient Celts has shaped modern identities across the British Isles and retains a powerful hold over the popular imagination. Furthermore, Celtic art is one of Europe’s great artistic traditions, with the skills of Celtic craftspeople standing alongside the best of the ancient and medieval worlds. But who were the Celts? Recent research and new archaeological discoveries are continuing to transform our understanding of the idea of the Celts – a subject involving much controversy and academic debate since the late 1990s. Drawing on the latest scholarship, the authors explore how the Celts have been defined differently from ancient times to the modern day, by people with different perspectives and agendas. They look, too, at what is meant by Celtic art, from its origins c.500 BC in western Europe, through its transformations and revivals in the Roman, Anglo-Saxon and medieval periods, to its rediscovery in Britain in the eighteenth and nineteenth centuries. Over 250 remarkable objects have been selected from the collections of the British Museum, the National Museums of Scotland and other key European museums to richly illustrate the narrative and highlight the artistic accomplishments of craftspeople through the centuries.
